One thousand one hundred eighty-seven compounds were cherry-picked for confirmation; actives identified in the primary HCS and compounds selected by a structural similarity search of the remaining MLSCN library using hits identified in phases I and II of the screen. Actives were confirmed in pSTAT3 IC50 assays, and an IFNγ-induced pSTAT1 activation assay was used to prioritize selective inhibitors of STAT3 activation that would not inhibit STAT1 tumor suppressor functions. Two hundred three concentration-dependent inhibitors of IL-6-induced pSTAT3 activation were identified and 89 of these also produced IC50s against IFN-γ-induced pSTAT1 activation. Forty-nine compounds met our hit criteria: they reproducibly inhibited IL-6-induced pSTAT3 activation by ≥70% at 20 μM; their pSTAT3 activation IC50s were ≤25 μM; they were ≥2-fold selective for pSTAT3 inhibition over pSTAT1 inhibition; a cross target query of PubChem indicated that they were not biologically promiscuous; and they were ≥90% pure. Twenty-six chemically tractable hits that passed filters for nuisance compounds and had acceptable drug-like and ADME-Tox properties by computational evaluation were purchased for characterization. The hit structures were distributed among 5 clusters and 8 singletons. Twenty-four compounds inhibited IL-6-induced pSTAT3 activation with IC50s ≤20 μM and 13 were ≥3-fold selective versus inhibition of pSTAT1 activation. Eighteen hits inhibited the growth of HNSCC cell lines with average IC50s ≤ 20 μM. Four chemical series were progressed into lead optimization: the guanidinoquinazolines, the triazolothiadiazines, the amino alcohols, and an oxazole-piperazine singleton. (shrink)

Current COVID-19 pandemic and previous SARS/MERS outbreaks have caused a series of major crises to global public health. We must integrate the large and exponentially growing amount of heterogeneous coronavirus data to better understand coronaviruses and associated disease mechanisms, in the interest of developing effective and safe vaccines and drugs. Ontologies have emerged to play an important role in standard knowledge and data representation, integration, sharing, and analysis. We have initiated the development of the community-based Coronavirus Infectious Disease Ontology (CIDO). (...) As an Open Biomedical Ontology (OBO) library ontology, CIDO is an open source and interoperable with other existing OBO ontologies. In this article, the general architecture and the design patterns of the CIDO are introduced, CIDO representation of coronaviruses, phenotypes, anti-coronavirus drugs and medical devices (e.g. ventilators) are illustrated, and an application of CIDO implemented to identify repurposable drug candidates for effective and safe COVID-19 treatment is presented. As fundamental researchers in the neuroethology of efference copy, we were stimulated by Grush's bold and original synthesis. In the following critique, we draw attention to ways in which it might be tested in the future, we point out an avoidable conceptual error concerning emulation that Grush seems to share with other workers in the field, and we raise questions about the neural correlates of Grush's schemata that might be probed by neurophysiologists.

Tversky and Koehler's support theory attempts to explain why probability judgments are affected by the manner in which formally similar events are described. Support theory suggests that as the explicitness of a description increases, an event will be judged to be more likely. In the present experiment, experienced decision-makers from large, international accounting firms were given case-specific information about an audit client and asked to provide a series of judgments regarding the perceived likelihood of events. Unpacking a hypothesis into four (...) formally equivalent hypotheses more than doubled its perceived likelihood. These results are largely consistent with support theory, extending its generalizability to applied contexts involving business professionals. Further, the paper finds that task-specific factors, a previously untested area, can also affect support theory interpretations. (shrink)

The most searching elaboration of the detective image of the historian has come from the pen of R. G. Collingwood. His short detective story "Who Killed John Doe?" implied that, in spite of the often tentative nature of the question-answer process in a successful historical investigation, the pieces of the puzzle fit together and their coherence becomes self-evident. The predominance of physical evidence in Collingwood's detective story had its counterpart in his research on Hadrian's Wall. In examining the questions (...) raised by his investigations, and distinguishing between direct and circumstantial evidence, Collingwood was able to formulate a comprehensive theory concerning the date of the Wall's construction, the purpose of the Wall, the date of the related turf wall at Birdoswald, the chronological position of the fourteen forts along the Wall, and the role and date of the Vallum following the Walls' course. The pattern of research into the mysteries of Hadrian's Wall has hardly conformed to the linear, step-by-step schema of Collingwood's logic of question and answer. As with much detective work, it has embodied its share of informed guesswork, mistaken inference, false leads, and fortuitous revelations. (shrink)

The device I use is to emphasize the major error Ockham wants to avoid: "platonism" rather than scepticism. In the second part, I try to show how difficulties raised by some recent (...) commentary result from the wrong orientation. This very limited project involves neither exposition nor assessment of Ockham's proposed solutions for the philosophical problems. Although the great majority of genes are not subject to cell‐cycle controls, those that are could play a very important role in regulation of the cell cycle itself. The tubulin and histone genes of the naturally synchronous myxomycete, Physarum polycephalum, provide an excellent paradigm for such regulation. The transcription of both is highly periodic within the Physarum cycle, and curiously, both sets of genes appear to be activated at the same time. This activation appears to function as part of a (...) developmental program, since it serves to meet future needs for each gene's product. The significance of this activation event for cell‐cycle regulation is discussed with particular regard to the nature of the mitotic oscillator. (shrink)

It may seem odd to review a New York social club's yearbook, with its list of members’ addresses and series of committee reports. But such books sometimes contain material of more general interest. The latest one from the Century Association, for example, devotes 250 of its 685 pages to “Century Memorials”—that is, biographical sketches of recently deceased members, written by other members. Among the well-known figures taken up in these eighty-three sketches are the artists Richard Anuszkiewicz and Robert Motherwell; the (...) architects Henry N. Cobb and Charles A. Platt; the newspaper editors Harold Evans and Whitelaw Reid; the historians Henry F. Graff and William McFeely; the translator and literary scholar Donald Keene; the collector of literary and musical manuscripts Frederick Koch; the popular writer on Russian history Robert K. Massie; the clergyman James Parks Morton; the financier Felix G. Rohatyn; the lawyers Whitney North Seymour Jr. and Isaac N. Phelps Stokes; the literary editor Elisabeth Sifton; the university president Michael Sovern; the museum director Evan H. Turner; and the broadcast journalist Sander Vanocur. The sketches are generally well written and often richly evocative: the writers include Louis Begley, Kenneth T. Jackson, D. T. Max, and Honor Moore.The long shelf of Century volumes like this one clearly constitutes a valuable contribution to prosopographical literature. And the Century is not the only institution with such a tradition (other examples are the British Academy and the American Philosophical Society); but their publications are a frequently overlooked source, one not always available in research libraries. Unfortunately, it is also a source that seems to be diminishing: the American Antiquarian Society and the John Simon Guggenheim Memorial Foundation are examples of organizations, each with a history of providing biographical information, that are no longer doing so. Of course, the great previous runs of the former's Proceedings and the latter's Reports remain (one hopes) on the shelves for those who know to look for them. A step in the opposite direction has recently been taken by the Grolier Club, which has not had such a tradition (except for two biographical volumes in 1959 and 2000): its Gazette now includes a section of memorials in each issue. This is a welcome move, but one has to regret that so little scholarly use is made of this whole body of biographical detail and reminiscence. (shrink)

This is the second volume in the Library of Living Philosophers which has appeared in German; it follows the familiar pattern of the other volumes in this series. The long autobiographical sketch sets the scene for a philosophy asserting that it deals with a reality which, ultimately, cannot be known unequivocally but must be realized in the philosopher's existence and which, accordingly, always presents reality in a definite historical perspective. The articles are introduced by an illuminating explanation of some (...) of the key terms in Jasper's work. Some of these are discussed in critical detail in the next six articles. Four further pieces place Jaspers in his historical situation, although not with that detail and completeness which Jaspers' historicism would seem to demand. These papers deal with Jaspers' relation to Max Weber, Judaism, Nietzsche and Kierkegaard. A final, rather extensive section explores the relations between Jaspers' central position and such peripheral philosophical doctrines as political philosophy, aesthetics, criticism, the philosophies of history and religion. This volume is particularly successful in mirroring the philosophy which it tries to clarify and celebrate, for it brings to light select aspects of a philosophy which maintains that reality can only be illuminated in part, but can never be known as a whole. Appropriate to the philosophy which tends to identify the philosopher's knowing with his being, Jaspers' final reply to his critics not only makes his ideas clearer but reveals a person of impressive human stature.--R. The length of participant information sheets for research and difficulties in their comprehension have been a cause of increasing concern. We aimed to examine the information sheets in research proposals submitted to an Australian HREC in one year, comparing the results with national recommendations and published data. Information sheets in all 86 research submissions were analysed using available software. The work of Flesch was used for Reading Ease or Readability and that of Flesch and Kincaid for the level of education (...) required for comprehension, the Reading Grade Level. The mean length of 86 information sheets was 3110 words; many had more than 5000 words. Using the Flesch scale of 0 to 100, with 0 meaning most difficult and 100 very easy to read, the mean readability level was 47. The mean length of education needed to easily grasp the information was 11.6 years, equivalent to senior secondary school. Information sheets in research projects submitted to an HREC were often too long to be read in a reasonable time and too difficult to be easily understood. Recommended standards for information sheets were infrequently met. (shrink)

The purpose of Donald Winch’s "historiographic revision" is to show that most recent interpretations of Smith have distorted his meaning because they have misread the intention of Smith’s work, treating it either as the first great justification of the nascent liberal capitalist polity, or as such a justification infiltrated by intimations of the Marxian notion of alienation. In Winch’s view, either account of Smith’s project is misleading by virtue of imposing nineteenth-century perspectives and categories upon "what is quintessentially a work (...) of the eighteenth century." The problem, then, is to determine the historically appropriate context or problematic within which the complex and at times paradoxical thought of the Wealth of Nations, the Theory of Moral Sentiments, and the Lectures on Jurisprudence can reasonably be situated. As Winch notes, his study is an application of the contextualist methodological principles of Quentin Skinner and J. G. A. Pocock, and is similar in design to recent works on Locke by John Dunn and on Hume by Duncan Forbes. (shrink)

This paper introduces a new, expanded range of relevant cognitive psychological research on collaborative recall and social memory to the philosophical debate on extended and distributed cognition. We start by examining the case for extended cognition based on the complementarity of inner and outer resources, by which neural, bodily, social, and environmental resources with disparate but complementary properties are integrated into hybrid cognitive systems, transforming or augmenting the nature of remembering or decision-making. Adams and Aizawa, noting this distinctive complementarity argument, (...) say that they agree with it completely: but they describe it as “a non-revolutionary approach” which leaves “the cognitive psychology of memory as the study of processes that take place, essentially without exception, within nervous systems.” In response, we carve out, on distinct conceptual and empirical grounds, a rich middle ground between internalist forms of cognitivism and radical anti-cognitivism. Drawing both on extended cognition literature and on Sterelny’s account of the “scaffolded mind” (this issue), we develop a multidimensional framework for understanding varying relations between agents and external resources, both technological and social. On this basis we argue that, independent of any more “revolutionary” metaphysical claims about the partial constitution of cognitive processes by external resources, a thesis of scaffolded or distributed cognition can substantially influence or transform explanatory practice in cognitive science. Critics also cite various empirical results as evidence against the idea that remembering can extend beyond skull and skin. We respond with a more principled, representative survey of the scientific psychology of memory, focussing in particular on robust recent empirical traditions for the study of collaborative recall and transactive social memory. We describe our own empirical research on socially distributed remembering, aimed at identifying conditions for mnemonic emergence in collaborative groups. Philosophical debates about extended, embedded, and distributed cognition can thus make richer, mutually beneficial contact with independently motivated research programs in the cognitive psychology of memory.
