The pathogenic, Gram‐negative bacteria, Neisseria gon‐orrhoeae, Neisseria meningitidis and Haemophilus influenzae, secrete immunoglobulin A1 proteases into their extracellular surroundings. An extraordinary feature in the secretory pathway of these putative virulence factors is a self‐directed outer membrane transport step allowing the proteins to be secreted autonomously, even from foreign Gram‐negative host cells like Escherichia coli. Here we summarize recent achievements in the understanding of IgA protease outer membrane translocation.

Although AGM theory contraction (Alchourrón et al., 1985; Alchourrón and Makinson, 1985) occupies a central position in the literature on belief change, there is one aspect about it that has created a fair amount of controversy. It involves the inclusion of the postulate known as Recovery. As a result, a number of alternatives to AGM theory contraction have been proposed that do not always satisfy the Recovery postulate (Levi, 1991, 1998; Hansson and Olsson, 1995; Fermé, 1998; Fermé and Rodriguez, 1998; (...) Rott and Pagnucco, 1999). In this paper we present a new addition, systematic withdrawal, to the family of withdrawal operations, as they have become known. We define systematic withdrawal semantically, in terms of a set of preorders, and show that it can be characterised by a set of postulates. In a comparison of withdrawal operations we show that AGM contraction, systematic withdrawal and the severe withdrawal of Rott and Pagnucco (1999) are intimately connected by virtue of their definition in terms of sets of preorders. In a future paper it will be shown that this connection can be extended to include the epistemic entrenchment orderings of Gärdenfors (1988) and Gärdenfors and Makinson (1988) and the refined entrenchment orderings of Meyer et al. Epistemic entrenchment, as presented by Gärdenfors and Makinson (1988) and Gärdenfors (1988), is a formalisation of the intuition that, when forced to choose between two beliefs, an agent will giveup the less entrenched one. While their formalisation satisfactorilycaptures the intuitive notion of the entrenchment of beliefs in a number ofaspects, the requirement that all wffs be comparable has drawn criticismfrom various quarters. We define a set of refined versions of theirentrenchment orderings that are not subject to the same criticism, andinvestigate (...) the relationship between the refined entrenched orderings,the entrenchment orderings of Gärdenfors and Makinson, and AGM theorycontraction (Alchourrón et al., 1985). Research suggests that institutions affect the levels of corruption in a country. We take these arguments a step further and examine whether it is the presence of inclusive institutions and/or the credible and consistent implementation of institutions that matter, as regards corruption. We use a novel approach to theoretically conceptualise and empirically operationalise institutions along two analytically distinct dimensions: the nature of the institutions (the de jure dimension), and the extent to which they are credibly and consistently implemented over time (...) (the de facto dimension), using a panel dataset for 148 countries covering 2012 to 2018. We find that formal institutions are most effective in reducing corruption when the rules are credibly and consistently implemented. Furthermore, this effect appears to operate differently at different levels of national income. The nature of the formal institutions appears important across income levels, but particularly so at the upper middle income level, while the credible and consistent implementation of these institutions is primarily influential at upper middle income levels. Precision medicine and molecular systems medicine (MSM) are highly utilized and successful approaches to improve understanding, diagnosis, and treatment of many diseases from bench-to-bedside. Especially in the COVID-19 pandemic, molecular techniques and biotechnological innovation have proven to be of utmost importance for rapid developments in disease diagnostics and treatment, including DNA and RNA sequencing technology, treatment with drugs and natural products and vaccine development. The COVID-19 crisis, however, has also demonstrated the need for systemic thinking and transdisciplinarity and the limits (...) of MSM: the neglect of the bio-psycho-social systemic nature of humans and their context as the object of individual therapeutic and population-oriented interventions. COVID-19 illustrates how a medical problem requires a transdisciplinary approach in epidemiology, pathology, internal medicine, public health, environmental medicine, and socio-economic modeling. Regarding the need for conceptual integration of these different kinds of knowledge we suggest the application of general system theory (GST). This approach endorses an organism-centered view on health and disease, which according to Ludwig von Bertalanffy who was the founder of GST, we call Organismal Systems Medicine (OSM). We argue that systems science offers wider applications in the field of pathology and can contribute to an integrative systems medicine by (i) integration of evidence across functional and structural differentially scaled subsystems, (ii) conceptualization of complex multilevel systems, and (iii) suggesting mechanisms and non-linear relationships underlying the observed phenomena. We underline these points with a proposal on multi-level systems pathology including neurophysiology, endocrinology, immune system, genetics, and general metabolism. An integration of these areas is necessary to understand excess mortality rates and polypharmacological treatments. In the pandemic era this multi-level systems pathology is most important to assess potential vaccines, their effectiveness, short-, and long-time adverse effects. We further argue that these conceptual frameworks are not only valid in the COVID-19 era but also important to be integrated in a medicinal curriculum. (shrink)

Die unuberschaubare Anzahl an Moglichkeiten in Ausbildung, Studium, Beruf, aber auch Partnerschaft und Lebensformen fuhren zu der Einsicht, dass Identitat sich immer neu konstituieren muss. Tatsachlich ist Identitat das, was mich als unverwechselbares Ich korperlich und geistig - und das heisst: leiblich - konstituiert. Zugleich entzieht sie sich mir und muss immer wieder neu errungen werden. Dies gilt gerade dann, wenn fruhere Ideale zusammenbrechen oder das zuvor Selbstverstandliche nicht mehr (...) tragt. Identitat und Identitatsbildung betreffen den Einzelnen ebenso wie politische oder religiose Gemeinschaften. In Zeiten, in denen vieles in Bewegung ist, ist Identitat eng verbunden mit der Frage nach den gemeinsamen Werten - das, wofur es sich zu leben lohnt und was wir an die nachste Generation weitergeben mochten. Besondere Beachtung verdienen in diesem Kontext phanomenologische Ansatze, die den leiblichen, sinnlichen und affektiven Dimensionen des Lebens Rechnung tragen. Generalisations of theory change involving operations on arbitrary sets ofwffs instead of on belief sets (i.e., sets closed under a consequencerelation), have become known as base change. In one view, a base should bethought of as providing more structure to its generated belief set, whichmeans that it can be employed to determine the theory contraction operationassociated with a base contraction operation. In this paper we follow suchan approach as the first step in defining infobase change. We think of an infobase (...) as a finite set of wffs consisting of independently obtainedbits of information. Taking AGM theory change (Alchourrón et al. 1985) as the general framework, we present a method that uses the structure of aninfobase B to obtain an AGM theory contraction operation for contractingthe belief set Cn(B). Both the infobase and the obtained theory contraction operation then play a role in constructing a unique infobasecontraction operation. Infobase revision is defined in terms of an analogueof the Levi Identity, and it is shown that the associated theory revisionoperation satisfies the AGM postulates for revision. Because every infobaseis associated with a unique infobase contraction and revision operation, the method also allows for iterated base change. (shrink)

In 1756 Joseph Black published a new theory of the nature of lime, one that is now viewed as essentially correct. Black's theory was not immediately accepted, and a competing theory, published in 1764 by Johann Meyer, was widely preferred to Black's for some years. In this paper, probability theory is used to show that, and why, some of Black's evidence made his theory more probable than Meyer's.

Episodic memory has been analyzed in a number of different ways in both philosophy and psychology, and most controversy has centered on its self-referential,autonoeticcharacter. Here, we offer a comprehensive characterization of episodic memory in representational terms and propose a novel functional account on this basis. We argue that episodic memory should be understood as a distinctive epistemic attitude taken toward an event simulation. In this view, episodic memory has a metarepresentational format and should not be equated with beliefs about the (...) past. Instead, empirical findings suggest that the contents of human episodic memory are often constructed in the service of the explicit justification of such beliefs. Existing accounts of episodic memory function that have focused on explaining its constructive character through its role in future-oriented mental time travel do justice neither to its capacity to ground veridical beliefs about the past nor to its representational format. We provide an account of the metarepresentational structure of episodic memory in terms of its role in communicative interaction. The generative nature of recollection allows us to represent and communicate the reasons why we hold certain beliefs about the past. In this process, autonoesis corresponds to the capacity to determine when and how to assert epistemic authority in making claims about the past. A domain where such claims are indispensable are human social engagements. Such engagements commonly require the justification of entitlements and obligations, which is often possible only by explicit reference to specific past events. (shrink)

Episodic representations can be entertained either as “remembered” or “imagined”—as outcomes of experience or as simulations of such experience. Here, we argue that this feature is the product of a dedicated cognitive function: the metacognitive capacity to determine the mnemicity of mental event simulations. We argue that mnemicity attribution should be distinguished from other metacognitive operations (such as reality monitoring) and propose that this attribution is a “cognitive gadget”—a distinctively human ability made possible by cultural learning. Cultural learning is a (...) type of social learning in which traits are inherited through social interaction. In the case of mnemicity, one culturally learns to discriminate metacognitive “feelings of remembering” from other perceptual, emotional, action-related, and metacognitive feelings; to interpret feelings of remembering as indicators of memory rather than imagination; and to broadcast the interpreted feelings in culture- and context-specific ways, such as “I was there” or “I witnessed it myself.” We review evidence from the literature on memory development and scaffolding, metacognitive learning and teaching, as well as cross-cultural psychology in support of this view before pointing out various open questions about the nature and development of mnemicity highlighted by our account. In this essay, Johannes Giesinger comments on the current philosophical debate on educational justice. He observes that while authors like Elizabeth Anderson and Debra Satz develop a so-called adequacy view of educational justice, Harry Brighouse and Adam Swift defend an egalitarian principle. Giesinger focuses his analysis on the main objection that is formulated, from an egalitarian perspective, against the adequacy view: that it neglects the problem of securing fair opportunities in the competition for social rewards. Giesinger meets this objection (...) by expressing two basic theses: First, he argues that Brighouse and Swift themselves fail to give an adequate account of fair competition; and, second, he shows that the adequacy view provides the theoretical resources to face this problem. In this paper, we explore the extent to which issues of simulation model validation take on novel characteristics when the models in question become particularly complex. Our central claim is that complex simulation models in general, and global models of climate in particular, face a form of confirmation holism. This holism, moreover, makes analytic understanding of complex models of climate either extremely difficult or even impossible. We argue that this supports a position we call convergence skepticism: the belief that the (...) existence of a plurality of different models making a plurality of different forecasts of future climate is likely to be a persistent feature of global climate science. Several promising approaches have been developed to represent conscious experience in terms of mathematical spaces and structures. What is missing, however, is an explicit definition of what a ‘mathematical structure of conscious experience’ is. Here, we propose such a definition. This definition provides a link between the abstract formal entities of mathematics and the concreta of conscious experience; it complements recent approaches that study quality spaces, qualia spaces, or phenomenal spaces; and it provides a general method to identify and investigate (...) structures of conscious experience. We hope that ultimately this work provides a basis for developing a common formal language to study consciousness. (shrink)

The goal of the present article is to contribute to the epistemology and methodology of computer simulations. The central thesis is that the process of simulation modeling takes the form of an explorative cooperation between experimenting and modeling. This characteristic mode of modeling turns simulations into autonomous mediators in a specific way; namely, it makes it possible for the phenomena and the data to exert a direct influence on the model. Anhand einer klinischen Vignette wird ausgeführt, dass das psychoanalytische Geschehen sich in mehreren Dimensionen entfaltet. Drei solcher Dimensionen – als Bedeutung (Erkenntnis), Berührung (Kontakt) und Bewegung (Ereignis) bezeichnet – werden beschrieben und deren dimensionale Charakteristik auf die Sinnesqualitäten des Sehens, des Berührungssinnes und des Hörens bezogen. Es wird gezeigt, dass sie Raum und Zeit auf je eigene Weise konstituieren und somit einander inkommensurable, durch keine logische oder dialektische Operation in eine Einheit überführbare Aprioritäten darstellen. Mit deren Unvereinbarkeit ist auch auf (...) Grenzen theoretisch-begrifflicher Bemächtigung verwiesen, womit die Frage nach der Reichweite metapsychologischer Begriffe, etwa des Begriffs des Unbewussten, aufgeworfen ist. Von hier aus wird in einem Ausblick nach der Begegnung als organisierendem Phantasma der analytischen Situation gefragt und – auf dem Weg einer Erörterung des Begegnungsbegriffs bei Martin Buber – versucht, Begegnung dimensional näher zu bestimmen. In recent years, the construct of mindfulness has gained growing attention in psychological research. However, little is known about the effects of mindfulness on interpersonal interactions and social relationships at work. Addressing this gap, the purpose of this study was to investigate the role of mindfulness in leader-follower-relationships. Building on prior research, we hypothesize that leaders’ mindfulness is reflected in a specific communication style (“mindfulness in communication”), which is positively related to followers’ satisfaction with their leaders. We used nested survey (...) data from 34 leaders and 98 followers from various organizations and tested mediation hypotheses using hierarchical linear modeling. Our hypotheses were confirmed by our data. Leaders’ self-reported mindfulness showed a positive relationship with several aspects of followers’ satisfaction. This relationship was fully mediated by leaders’ mindfulness in communication as perceived by their followers. Our findings emphasize the potential value of mindfulness in workplace settings. They provide empirical evidence for a positive link between leaders’ dispositional mindfulness and the wellbeing of their followers, indicating that mindfulness is not solely an individual resource but also fosters interpersonal skills. By examining leaders’ mindfulness in communication as an explanatory process, we created additional clarification about how leaders’ mindfulness relates to followers’ perceptions. Moreover, by suggesting mindfulness in communication as a behavioral correlate of mindfulness, we offered a promising starting point for indirect measures of mindfulness. (shrink)

Despite growing evidence to the contrary, researchers continue to posit causal links between cannabis, crime, psychosis, and violence. These spurious connections are rooted in history and fueled decades of structural limitations that shaped how researchers studied cannabis. Until recently, research in this area was explicitly funded to link cannabis use and harm and ignore any potential benefits. Post-prohibition cannabis research has failed to replicate the dire findings of the past. This article outlines how the history of controlling cannabis research has (...) led to various harms, injustices, and ethical complications. We compare commonly cited research from both the prohibition and post-prohibition eras and argue that many popular claims about the dangers of cannabis are the result of ethical lapses by researchers, journals, and funders. We propose researchers in this area adopt a duty of care in cannabis research going forward. This would oblige individual researchers to establish robust research designs, employ careful analytic strategies, and acknowledge limitations in more detail. This duty involves the institutional recognition by funders, journals, and others that cannabis research has been deliberately misconstrued to criminalize, stigmatize, and pathologize. (shrink)

To what extent does the common claim that childhood is ‘socially constructed’ affect the ethical debate on the ‘intrinsic’ and ‘special’ goods of childhood? Philosophers have referred to this kind of goods in their critique of overly adult-centred and future-oriented conceptions of childhood. The view that some goods are child-specific, in the sense that they are only good for children, not for adults, seems to presuppose an understanding of what children ‘are’, and how they differ from adults. However, if the (...) social-constructionist view is accepted, it cannot be assumed that childhood is a given. This essay claims that the social-constructionist understanding of childhood does not undermine the debate on the moral status and the goods of childhood, but that nevertheless important lessons can be drawn from the insight that childhood, as we know it, is not a natural and universal phenomenon. We claim that adjustable parameters play a crucial role in building and applying simulation models. We analyze that role and illustrate our findings using examples from equations of state in thermodynamics. In building simulation models, two types of experiments, namely, simulation and classical experiments, interact in a feedback loop, in which model parameters are adjusted. A critical discussion of how adjustable parameters function shows that they are boon and bane of simulation. They help to enlarge the scope of simulation far (...) beyond what can be determined by theoretical knowledge, but at the same time undercut the epistemic value of simulation models. (shrink)

This article discusses the educational significance of the moral demand for respect. In Ethics and Education, Richard Peters presents a conception of educational respect that was recently taken up by Krassimir Stojanov. This article responds to both Peters' and Stojanov's contributions and proposes another understanding of educational respect: to respect children is to treat them in a way that enables them to see themselves as persons endowed with dignity; that is, as having the equal standing to make claims on others.

Integrated Information Theory is one of the leading models of consciousness. It aims to describe both the quality and quantity of the conscious experience of a physical system, such as the brain, in a particular state. In this contribution, we propound the mathematical structure of the theory, separating the essentials from auxiliary formal tools. We provide a definition of a generalized IIT which has IIT 3.0 of Tononi et al., as well as the Quantum IIT introduced by Zanardi et al. (...) as special cases. This provides an axiomatic definition of the theory which may serve as the starting point for future formal investigations and as an introduction suitable for researchers with a formal background. Do any nonhuman animals have hedonically valenced experiences not directly caused by stimuli in their current environment? Do they, like us humans, experience anticipated or previously experienced pains and pleasures as respectively painful and pleasurable? We review evidence from comparative neuroscience about hippocampus-dependent simulation in relation to this question. Hippocampal sharp-wave ripples and theta oscillations have been found to instantiate previous and anticipated experiences. These hippocampal activations coordinate with neural reward and fear centers as well as sensory and cortical areas (...) in ways that are associated with conscious episodic mental imagery in humans. Moreover, such hippocampal “re- and preplay” has been found to contribute to instrumental decision making, the learning of value representations, and the delay of rewards in rats. The functional and structural features of hippocampal simulation are highly conserved across mammals. This evidence makes it reasonable to assume that internally triggered experiences of hedonic valence (IHVs) are pervasive across (at least) all mammals. This conclusion has important welfare implications. Most prominently, IHVs act as a kind of “welfare multiplier” through which the welfare impacts of any given experience of pain or pleasure are increased through each future retrieval. However, IHVs also have practical implications for welfare assessment and cause prioritization. (shrink)

This essay starts from the observation that the issue of talent, in relation to the problem of distributive justice, can be approached from two different angles. First, it is common to discuss the justificatory function of talent, that is, its role in the justification of educational or social inequalities. In addition, however, this essay proposes to look at practices of talent ascription and their causal role in the distribution of educational prospects. These practices tend to exacerbate educational inequalities due to (...) social background, especially when they are tied to selective educational settings. There is reason to avoid talent-based selection in education, in order to contain the negative effects of practices of talent ascription. (shrink)

In response to the commentaries, we clarify and defend our characterization of both the nature and function of episodic memory. Regarding the nature of episodic memory, we extend the distinction between event and episodic memory and discuss the relational role of episodic memory. We also address arguments against our characterization of autonoesis and argue that, while self-referential, it needs to be distinguished from an agentive notion of self. Regarding the function of episodic memory, we review arguments about the relation between (...) future mental time travel and memory veridicality; clarify the relation between autonoesis, veridicality, and confidence; and finally discuss the role of episodic memory in diachronic commitments.
