Co‐expression of two or more genes at the single‐cell level is usually associated with functional co‐regulation. While mRNA co‐expression—measured as the correlation in mRNA levels—can be influenced by both transcriptional and post‐transcriptional events, transcriptional regulation is typically considered dominant. We review and connect the literature describing transcriptional and post‐transcriptional regulation of co‐expression. To enhance our understanding, we integrate four datasets spanning single‐cell gene expression data, single‐cell promoter activity data and individual transcript half‐lives. Confirming expectations, we find that positive co‐expression necessitates (...) promoter coordination and similar mRNA half‐lives. Surprisingly, negative co‐expression is favored by differences in mRNA half‐lives, contrary to initial predictions from stochastic simulations. Notably, this association manifests specifically within clusters of genes. We further observe a striking compensation between promoter coordination and mRNA half‐lives, which additional stochastic simulations suggest might give rise to the observed co‐expression patterns. These findings raise intriguing questions about the functional advantages conferred by this compensation between distal kinetic steps. (shrink)

The investigation of sentimental fiction, however, has been sorting out the classifications that applied to a hodgepodge of works previously labeled as “sentimental novel” (“novela sentimental”). This label, once attached uncritically to a bunch of heterogeneous texts (...) that shared such intangibles as “loving focus” or “prevalence of psychological description over narratives,” has been applied more consistently nowaday, thanks in part to the scholarly efforts started in the 1980s like those of Martínez Jiménez and Muñoz Marquina (1982), and Martínez Latre (1989).1 Using some of these research efforts, Alan Deyermond has codified the genre accurately:Brevity; focus on psychological interest over external action; a tragic vision of love; autobiography (first-person narration or a narrator as a character) and the insertion of letters or poems (or, often, both) into the narration.[77]2Within this scholarly categorization, Regula Rohland de Langbehn has been pivotal in offering—first in 1986, and more recently in 1999—a threefold scheme in the development of the genre she labels as “sentimental novel”: a first group chronologically of the genre would be represented by Siervo libre de amor and Sátira de felice e infelice vida, both works marked by the influence of cancionero poetry—specially in the allegorical structuring and in the characterization of love, and by other subjects being limited to an abstract and general analysis of emotions. It was Antonio Cortijo Ocaña who, while editing a critical cluster in La Corónica on sentimental fiction, opened the door to some scholars such as Weissberger who voiced their skepticism over the generic nature of sentimental prose, a skepticism already present in an innovative volume published by Gwara and Gerli in 1997. This skeptical position was also supported by Cortijo Ocaña himself, who in 2001 published a monumental work in which, although using the generic term as a departure point, he ended up suggesting the difficulty of defining it, therefore jettisoning it. This position in turn was contested by Rohland de Langbehn, who in 2002 defended the validity of both the existence of a genre and its name novela sentimental, a position that has sparked a new round of replies in a La Corónica forum in 2003.In this debate, I would join today’s minority position on the existence of a sentimental genre, because, as I explained in my 2001 review of Rohland de Langbehn’s book and as I find convincingly argued in her 2002 contribution, there are internal traits within sentimental fictional texts that do not appear together in other sentimental generic texts [139]. The vast majority of research in nursing ethics over the last decade indicates that nurses may not be fully prepared to ‘deliver the good’ for their patients, or to contribute appropriately in the wider current health care climate. When suitable research projects were evaluated for this article, one key question emerged: if nurses are educationally better prepared than ever before to exercise their ethical decision-making skills, why does research still indicate that the expected practice-based improvements remain elusive? Hence, a number (...) of ideas gleaned from recent research about the current nature of nursing ethics, and especially teaching nursing ethics to student nurses, are analysed and critiqued in this article, which concludes with a cluster of ideas and conclusions based on that analysis. It is hoped that such a review may serve as a catalyst for nurse educators to re-examine their teaching practices with a view to enhancing good nursing practice through educational means. (shrink)

One of the arguments advanced in favor of scientific realism is the 'miracle argument'. It says that for the anti-realist the predictive success of science appears as an utter miracle. This argument indeed has some prima facie plausibility, provided that it is sharpened by construing "predictive success" as prediction of previously unknown laws and the occurrence of a consilience of inductions. Still, the history of science teaches us that it is possible to arrive at predictive success in this sense by (...) employing radically non-referring theoretical mechanisms. The 'miracle argument' is thus unsound. Rather, the capacity of a theory to generate predictive success can be traced back to its "classificativity correspondence.". (shrink)

In the Anthropocene, the fact that human activity is enmeshed with the existence and actions of every kind of other being is inescapable. As a result, the planetary ecological crisis has brought forth an urgent need to rethink understandings of human action. One response holds that the transformations necessary to tackle today’s crises will emerge from the distinctive capacity of human beings to transcend their environment. Another school of thought calls for seeing action as composite, produced by distributed networks of (...) human and nonhuman agents. Yet the first of these is open to charges of human exceptionalism, while the second, according to its critics, lacks effective political traction. Martin Crowley argues that a new conception of political agency is necessary to break this impasse. Engaging with thinkers such as Bruno Latour, Bernard Stiegler, and Catherine Malabou, Crowley proposes an original account of agency as both distributed and decisive. Challenging the prevailing view of agency as exclusively human, he explores how a politics that incorporates nonhuman agency can intervene in the real world, examining timely issues such as climate-related migration and digital-algorithmic politics. To assess the merits and demerits of the content of Culp’s educational programme, the paper does three things: First, it discusses whether Culp’s defence against conceivable objections manages to effectively dispel the charge of cosmopolitan arrogance. Second, it spells out one implication of epistemic modesty, which Culp considers a core competence to be imparted by citizenship education. Third, it reflects upon the tricky task of motivating individuals to comply with the demands of justice. Taken together, the paper argues that Culp’s (...) case is impressively strong but nevertheless tends to suffer from a rationalist constriction. It does not leave sufficient room for tradition in private life and public reasoning. To achieve this, the article briefly reviews and critiques the main underpinnings of the concept from its origins and development in New Zealand, describes its sociocultural and sociopolitical elements, and provides an in-depth exploration of the key socioethical elements. Starting with Popper, social theorists across the board have acknowledged that traditions serve socially valuable functions. However, while traditions are usually understood as ‘living’ entities that come in overlapping varieties and evolve over time, the socially valuable functions attributed to tradition tend to presuppose invariability in ways of thinking and acting. Addressing this tension, this article provides a detailed analysis of the concept of tradition, and directs special attention to conceivable criteria for the authentic continuation of a tradition. It is (...) argued that the ways of thinking and acting that constitute the material of a tradition must – among faithful members of that tradition – stand in a relation of equivalence – not identity or similarity. The implications of this account concern our ability to decide (normatively) conflicts over authenticity among rival tradition branches as well as the role that traditions play in policy-making. I show that prospectism permits the decision-maker to make a series of choices she knows in advance will lead to a sure loss. I also argue that a theory that permits the decision-maker to make choices she knows in advance will lead to a sure loss should be rejected.

The thesis of social constructionism is that emotions are shaped by culture and society. I build on this insight to show that existing social constructionist views of emotions, while providing valid research methods, overly restrict the scope of the social constructionist agenda. The restriction is due to the ontological assumption that social construction is indissociable from language. In the first part, I describe the details of the influential social constructionist views of Averill and Harré. Drawing on recent theorizing in psychology, (...) I suggest that their fixation on language makes these approaches inadequate to the analysis of the social construction of human emotional experience. In the second part, I extend the argument to other species, suggesting that these social constructionist views are incapable of accommodating the fact, ascertained by primatologists, that animals have cultures, and that part of animal culture concerns the social molding of their emotions. I conclude that a reconstructed social constructionism should be regarded not as inimical to, but as part and parcel of, a nonreductive biology of emotions. (shrink)

Historical Epistemology: On the Diversity and Change of Epistemic Values in Science. Historical epistemology involves the claim that the system of scientific knowledge is not determined by the observations but is also subject to epistemic requirements that may change in the historical process of doing research. As a result, the system of knowledge is path‐dependent in that its shape is contingent on epistemic choices made at certain historical points. I attempt to elaborate this approach by drawing attention to the double (...) role of epistemic values. First, such values create relations of significance and thereby contribute to directing research into certain avenues. Second, they are also important in the process of confirmation in that they entail that certain forms of agreement with the facts are superior and preferable to other such forms. Some epistemic orientations and reorientations can be reconstructed as arising from an interaction with nature, but others are based on commitments to the kind of knowledge we appreciate. The epistemic authority of science is created in large measure by rules of the scientific community that express how to deal with knowledge claims. (shrink)

The article explores epistemic and social conditions of the trustworthiness of scientific expertise. I claim that there are three kinds of conditions for the trustworthiness of scientific expertise. The first condition is epistemic and means that scientific knowledge enjoys high credibility. The second condition concerns the significance of scientific knowledge. It means that scientific generalizations are relevant for elucidating the particular cases that constitute the challenges for expert judgment. The third condition concerns the social processes involved in producing science-based recommendations. (...) This special issue is motivated by the observation that conservatism plays a marginal role in contemporary philosophy even though it appears to be of considerable importance in moral, social, and political reality. One reason for this neglect is that defenders of conservatism have often refrained from articulating their arguments in a language that is acceptable to and understandable by analytically -trained philosophers. The contributions of this special issue show that conservatism can profitably be approached from the point of view of (...) analytic philosophy. Many of them are indebted to Jerry Cohen’s (2012) seminal paper on conservative value, which develops a sophisticated justification of what Oakeshott (1991) called the disposition to be conservative. In this paper I respond to van de Poel’s claim that new technologies should be conceived as ongoing social experiments, which is an idea originally introduced by Schinzinger and Martin in the 1970s. I discuss and criticize three possible motivations for thinking of new technologies as ongoing social experiments.

Kant's theory of matter is reconstructed and his views about and impact on chemistry are studied. His early "monadological" conception is analyzed and compared to other dynamical approaches of the period. His later attempt to regard matter as a continuum and to derive some of its properties from the interaction of forces is reconstructed. His conception of chemistry is examined and compared to the notion of some chemists who were inspired by Kant's work.

It became obvious in classroom case discussions in a required MBA course, Corporate Social Responsibility and Business Ethics, that subjective opinion often prohibited complete and thorough case analyses. Over a two-year period an attempt was made to identify the parameters of situations involving corporate social responsibility in order to develop a methodology which would facilitate classroom learning. The model described in the following manuscript is the result of these efforts.

Moral deliberation involves a constant interplay among specific judgments, general moral values and principles, and background beliefs about the world. We may also construct broad moral theories to explain how our judgments and general commitments hang together, but there is little hope of settling on any one comprehensive moral theory.

SummaryIn 1968, a prospective study was started in collaboration with the Family Planning Association to try to provide a balanced view of the beneficial and harmful effects of different methods of contraception. This investigation is now in progress at seventeen clinics and over 17,000 women are under observation. At the time of recruitment, all these women were married white British subjects, aged 25–39 years, who voluntarily agreed to participate. Fifty-six per cent were using oral contraceptives, 25% were using a diaphragm (...) and 19% were using an intrauterine device. During follow-up each woman is questioned at return visits to the clinic and a record of pregnancies and their outcome, hospital referrals, changes in contraceptive methods and the results of cervical smears, is accumulated. Women who default are sent a postal questionnaire and, if this is not returned, are telephoned or visited in their homes to collect the necessary information.So far, data obtained during 56,000 woman-years of observation are available for analysis. Follow-up has been maintained with an annual lapse rate of about 0.3%due to withdrawal of co-operation or loss of contact; adherence to the method of contraception in use at recruitment has been reasonably good, and the reporting of pregnancies and hospital admissions appears to have been both reliable and unbiased.The present data include only 24 deaths, so the mortality associated with different contraceptive methods cannot yet be estimated. With regard to morbidity, however, our preliminary results closely resemble those obtained in the prospective study carried out by the Royal College of General Practitioners and in the principal retrospective studies carried out in Britain and the United States. Women who used oral contraceptives at the start of the study experienced adeficiencyof hospital referrals for cancer, benign lesions of the breast, menstrual disorders other than amenorrhoea, duodenal ulcer, and retention cysts of the ovary; they showed anexcessof referrals for cerebrovascular disease, cervical erosion, skin disorders, self-poisoning, migraine, venous thrombosis and embolism, hayfever, gallbladder disease, amenorrhoea, and sterility. Women who used a diaphragm showed adeficiencyof hospital referrals for carcinoma-in-situ and dysplastic lesions of the cervix uteri and accidental injury; and anexcessof referrals for haemorrhoids and cystitis. Women who used an IUD experienced anexcessof hospital referrals for anaemia, varicose veins and salpingitis. About half of these differences were predicted from other studies while some suggestive evidence already existed for a further five. Of the remaining six, some probably reflect the influence of selective factors or chance.Multiple pregnancies, stillbirths, malformations, the sex ratio and birthweight showed no consistent relationship to method of contraception. The outcome of unplanned pregnancies occurring in women using an IUD, however, was remarkably unfavourable both in terms of ectopic gestation and miscarriage.Clear evidence was found of impairment of fertility after discontinuation of oral contraceptives. Whether this is likely to lead to permanent sterility in some women is uncertain.The study has provided data on the efficacy of a wide variety of contraceptive methods. In general the failure rates are in keeping with those obtained in other large-scale studies, save that those for the diaphragm and the sheath are much lower than those usually quoted.The available evidence does not yet allow a final balance to be struck between the benefits and risks associated with the new methods of contraception that have become widely used during the last two decades. It seems clear, however, that there are no material risks associated with the use of the diaphragm apart from the risk of pregnancy and that there may be some unintended benefits. (shrink)

We study generalizations of Dodd parameters and establish their fine structural properties in Jensen extender models with λ-indexing. These properties are one of the key tools in various combinatorial constructions, such as constructions of square sequences and morasses.

A close investigation of Wittgenstein’s MS 104, which contains the so-called Prototractatus, has shown that the manuscript originally contained an additional folio that was later cut out and is now missing. The content of this missing folio could be partly reconstructed by a faint inverse imprint that it has left behind on page 2. The paper discusses the consequences of this discovery for the interpretation of the beginning and early formation of the Prototractatus, including the introduction and role of the (...) numbering system. Martin F. Meyer untersucht die Entwicklung des lebenswissenschaftlichen Denkens von den frühsten Anfängen bis zur Geburt der wissenschaftlichen Biologie bei Aristoteles. Whether or not we adopt some form of physicalism in our thinking about the psychology of humans and other organisms we all believe that a mind is something that comes into being, changes, develops and decays. The correlation of the development and then later the decay of our mental powers with changes in the brain post-dates our belief that the mental realm is as much an area where things ebb and flow, come to be and pass away, as is the (...) physical. Even ancient authors who hold to the indestructibility of the soul allow that it changes in a host of ways. It is very odd, then, to discover that Aristotle appears to deny all this and feels impelled to treat the soul as unchanging and motionless. I propose to explore here exactly what Aristotle meant by saying the soul does not move and why he said it. ZusammenfassungIm vorliegenden Artikel wird das von historischer Seite wenig erforschte Ereignis der Magdalenenflut von 1342 unter Einbeziehung administrativer Überlieferung, vor allem aus Urkunden und Rechnungsbüchern, und durch Heranziehung naturwissenschaftlicher Proxydaten, insbesondere aus Baumring-Niederschlagsrekonstruktionen, neu ausgeleuchtet. So gelingt es nicht nur, einen wesentlich differenzierteren Verlauf einer insgesamt zweijährigen Hochwasserkatastrophe mit peaks im Februar und Juli 1342, aber auch im Juli 1343 zu gewinnen, sondern auch mehrmonatige Trockenphasen im März bis Juni 1342 erstmals zu fassen, die den (auch in Schriftquellen nachgewiesenen) Erosionseffekt (...) der Starkregenfälle im Juli 1342 besser erklären als bislang. Dabei wird klar, dass 1342/43 ein multifaktorieller compound event vorliegt, der erst durch die Koinzidenz natürlicher Extremereignisse mit sozio-ökonomischen Faktoren zur Katastrophe führt. Die bewusste Ausweitung des Quellenspektrums lässt beispielsweise erstmals eine überregionale Teuerung bei Lebensmitteln im Nachklang der Flutereignisse feststellen, die aber wesentlich im zufälligen Auftreten des Hochwassers noch vor der Getreideernte begründet liegt. Der Fokus des Beitrags liegt auf infrastrukturellen Anpassungsmaßnahmen, etwa veränderten Brückendesigns (am Bsp. der Balduinbrücke in Koblenz) und großer Wasserbaumaßnahmen (am Beispiel des Donaudurchstichs von Oberalteich), oder normativen Reaktionen wie den Würzburger Polizeisätzen 1342/43 und der Errichtung von Deichbaugemeinschaften am Niederrhein, die zumindest als indirekte Reaktionen auf die Hochwasserereignisse verstanden werden müssen. Diese technisch-normativen Adaptionsvorgänge können einerseits im Luhmann’schen Sinn als Umweltbeobachtungen zweiter Ordnung verstanden werden, andererseits erläutern sie die Bedeutung von natürlichen Extremereignissen als Taktgeber für vormoderne Infrastrukturentwicklung aufgrund ihrer als kritisch erlebten, ungenügenden Ausgestaltung oder gar ihres völligen Fehlens. Nordalpin sehen wir nach 1342/43 im Bereich des Hochwasserschutzes zaghafte Anfänge obrigkeitlicher Daseinsvorsorge jenseits lokaler Zuständigkeiten. (shrink)

An Essay on Metaphysics is one of the finest works of the great Oxford philosopher R. G. Collingwood : in it he considers the nature of philosophy, especially of metaphysics, and puts forward his original and influential theories of absolute presuppositions, causation, and the logic of question and answer. Three fascinating unpublished pieces by Collingwood have been added for this revised edition: they illuminate and amplify the ideas of the Essay, to which they are closely related. The editor Rex (...) class='Hi'>Martin contributes a substantial introduction telling the story of the composition of all these works, discussing their major themes, and setting them in the context of Collingwood's philosophy as a whole. Rawdon Wilson's "On Character" raised a great many questions, and I should like to deal with lesser matters before going on to those of more consequence. He has found in my work the Fallacy of Novelistic Presumption. To commit this unnatural act is to assume "that the novel possesses a history that is independent of other modes of fiction and that it may be discussed independently of the history of literature." Let me say at the outset that I am not (...) trying to frame a restrictive definition of the novel. Novels are whatever most critics agree to call novels, and if I speak of "the novel" I can only hope that the phrase will be taken as convenient shorthand rather than an attempt to define an essence. And of course the novel has a history of its own, just as the state of Connecticut has a history even as it remains one of the fifty states, just as literature has a history although it is only one of the arts or institutions of our culture. Mr. Wilson wants a theory of characters that "will necessarily account for - go to the heart of - all instances of character, symbolic, allegorical, naturalistic, whether in the novel, in epic, in romance, in drama, or in lyric." To that I can only reply with E.M. Forster's sentence: "We must exclude someone from our gathering, or we shall be left with nothing." In this essay Martin Price, Thomas E. Donnelley Professor of English at Yale University, responds to Rawdon Wilson's "On Character" which raised objections to Price's "People of the Book: Character in Forster's A Passage to India". The subtlety of the novel lies in its unrelieved tension of flesh and spirit, exclusion and invitation, the social self and the deeper impersonal self. At one extreme are the caricatures caught in the social grid - the Turtons and Burtons. At the other are the characters who slip out of the meshes of social responsibility through despair or obliviousness. We move from the elaborate rituals of Anglo-Indian to Mau, where the only aspects of life we are shown are ecstasy (...) and neglect. Where does the mind rest? The difficulty with looking at reality directly is that reality will tend to dissolve: "not now, not here, not to be apprehended except when it is unattainable." Transcendence dehumanizes, the deeper self is a source rather than a habitation, we cannot see the unseen. We only glimpse it through paradox, violence, or farce; and each of these contributes something to Forster's conception of character. Martin Price, Thomas E.
