While outcomes for children with T-cell acute lymphoblastic leukemia have improved dramatically, survival rates for patients with relapsed/refractory disease remain dismal. Prior studies indicate that glucocorticoid resistance is more common than resistance to other chemotherapies at relapse. In addition, failure to clear peripheral blasts during a prednisone prophase correlates with an elevated risk of relapse in newly diagnosed patients. Here we show that intrinsic GC resistance is present at diagnosis in early thymic precursor T-ALLs as well as in a subset (...) of non-ETP T-ALLs. GC-resistant non-ETP T-ALLs are characterized by strong induction of JAK/STAT signaling in response to interleukin-7 stimulation. Removing IL7 or inhibiting JAK/STAT signaling sensitizes these T-ALLs, and a subset of ETP T-ALLs, to GCs. The combination of the GC dexamethasone and the JAK1/2 inhibitor ruxolitinib altered the balance between pro- and anti-apoptotic factors in samples with IL7-dependent GC resistance, but not in samples with IL7-independent GC resistance. Together, these data suggest that the addition of ruxolitinib or other inhibitors of IL7 receptor/JAK/STAT signaling may enhance the efficacy of GCs in a biologically defined subset of T-ALL.Leukemia advance online publication, 30 May 2017; doi:10.1038/leu.2017.136. SmithAt 471C4 in Plato’s Republic, the argument takes a sudden turn when Glaucon becomes impatient with all of the specific prescriptions Socrates has been making, and asks to return to the issue Socrates had earlier set aside—whether or not the city he was describing could ever be brought into being. In response to Glaucon’s impatient question, Socrates articulates his “third wave of (...) paradox” (472ag–7), namely, that the ills of the cities will never be ended until either philosophers become rulers or rulers become philosophers (473c11–d6). Glaucon immediately responds that such a view is likely to be greeted with violence and scorn (473e6–474a4), and so Socrates must hasten to explain his odd claim. His explanation, it turns out, is that true philosophers have an enormous cognitive advantage over non-philosophers—philosophers have and use knowledge (ἐπιστήμη), whereas non-philosophers have and use only opinion (δόξα).2 This distinction, between επιστήμη and δόξα, turns out: to be a distinction between two different cognitive powers (δυνάμεις—477d7–e3). And different powers, Plato clearly tells us, apply to or take as their objects (Plato says they are “ἐπί”) different things (477dl).In this paper, I shall argue that the relationship between the cognitive powers and their various objects has been fundamentally misunderstood, which has led scholars into one or more misinterpretations of important and explicit features of the text. BackgroundIn the Canadian Alliance for Healthy Hearts and Minds (CAHHM) cohort, participants underwent magnetic resonance imaging (MRI) of the brain, heart, and abdomen, that generated incidental findings (IFs). The approach to managing these unexpected results remain a complex issue. Our objectives were to describe the CAHHM policy for the management of IFs, to understand the impact of disclosing IFs to healthy research participants, and to reflect on the ethical obligations of researchers in future MRI studies.MethodsBetween 2013 and 2019, 8252 participants (...) (mean age 58 ± 9 years, 54% women) were recruited with a follow-up questionnaire administered to 909 participants (40% response rate) at 1-year. The CAHHM policy followed a restricted approach, whereby routine feedback on IFs was not provided. Only IFs of severe structural abnormalities were reported.ResultsSevere structural abnormalities occurred in 8.3% (95% confidence interval 7.7–8.9%) of participants, with the highest proportions found in the brain (4.2%) and abdomen (3.1%). The majority of participants (97%) informed of an IF reported no change in quality of life, with 3% of participants reporting that the knowledge of an IF negatively impacted their quality of life. Furthermore, 50% reported increased stress in learning about an IF, and in 95%, the discovery of an IF did not adversely impact his/her life insurance policy. Most participants (90%) would enrol in the study again and perceived the MRI scan to be beneficial, regardless of whether they were informed of IFs. While the implications of a restricted approach to IF management was perceived to be mostly positive, a degree of diagnostic misconception was present amongst participants, indicating the importance of a more thorough consent process to support participant autonomy.ConclusionThe management of IFs from research MRI scans remain a challenging issue, as participants may experience stress and a reduced quality of life when IFs are disclosed. The restricted approach to IF management in CAHHM demonstrated a fair fulfillment of the overarching ethical principles of respect for autonomy, concern for wellbeing, and justice. The objective of the study was to investigate the relationship between childhood IQ of parents and characteristics of their adult offspring. It was a prospective family cohort study linked to a mental ability survey of the parents and set in Renfrew and Paisley in Scotland. Participants were 1921-born men and women who took part in the Scottish Mental Survey in 1932 and the Renfrew/Paisley study in the 1970s, and whose offspring took part in the Midspan Family study in 1996. There (...) were 286 offspring from 179 families. Parental IQ was related to some, but not all characteristics of offspring. Greater parental IQ was associated with taller offspring. Parental IQ was inversely related to number of cigarettes smoked by offspring. Higher parental IQ was associated with better education, offspring social class and offspring deprivation category. There were no significant relationships between parental IQ and offspring systolic blood pressure, diastolic blood pressure, cholesterol, glucose, lung function, weight, body mass index, waist hip ratio, housing, alcohol consumption, marital status, car use and exercise. Structural equation modelling showed parental IQ associated with offspring education directly and mediated via parental social class. Offspring education was associated with offspring smoking and social class. The smoking finding may have implications for targeting of health education. (shrink)

This analogy is presented explicitly and in great detail in the famous ‘similes (...) of light,’ the Sun, Divided Line, and Cave passages of Books VI and VII. How many words is a bilingual 2-year-old supposed to know or say in each of her languages? Speech and language therapists or researchers lack the tools to answer this question, because several factors have an impact on bilingual language skills: gender, amount of exposure, mode of acquisition, socio-economic status and the distance between L1 and L2. Unfortunately, these factors are usually studied separately, making it difficult to evaluate their weight on a unique measure of vocabulary. The present study measures the (...) contribution of the following factors to the vocabulary scores of bilingual toddlers: i) gender; ii) sibling ranking; iii) relative amount of exposure to each language; iv) mode of exposure; v) SES; vi) linguistic distance; vii) language spoken between parents. Close to the child’s second birthday, parents of 278 UK-based bilinguals completed successively: a 100-word version of the Oxford-CDI, the CDI in the child’s Additional Language, a family questionnaire, and the Language Exposure Questionnaire. Thirty-six British-English-AL pairs were considered, with languages contrasted on a second-language-learning scale : for example, Dutch and French are close to British-English, while Polish or Cantonese are more distant. Data from the corpus were included in two mixed-effect models, one with the English scores in comprehension as the dependent variable, and the other with production scores. The seven factors listed above were included as predictors. The amount of English exposure was the strongest predictor of comprehension scores = 9.35, p <.005, β = 0.02, t = 3.08, p <.005), followed by the language that parents speak between themselves = 14.94, p <.001, β = 1.37, t = 3.76, p <.0005), linguistic distance = 6.92, p <.01, β = -0.74, t = -2.66, p <.01) and age = 4.86, p <.05, β = 0.55, t = 2.17, p <.05). In production, gender = 13.57, p <.0005, β = -0.91, t = -03.72, p <.0005), amount of exposure to English = 13.57, p <.0005, β = -0.91, t = -03.72, p <.0005), the language that parents speak between themselves = 11.85, p <.005, β = 1.09, t = 3.41, p <.001), and the mother’s occupation = 4.51, p <.05, β = 0.63, t = 2.13, p <.05) were the significant predictors. The more English parents use to address one another, the more English words the child says and understands. This surprising result could be simply explained by the fact that parents who speak English together are also more likely to speak English to their child. The main results of this study is that linguistic distance is a powerful predictor of toddlers’ vocabulary in English, with children learning two close languages growing their vocabulary faster than those learning distant languages. This intervention queries whether subjecting feminist jurisprudence to a methodological transfiguration toward pragrammatology (Derrida, in Smith and Kerrigan 1984) holds liberating potential for f (be it ‘woman’, ‘female’ or ‘feminine’) as both a signifier and a subject to be read with lucidity and nuance in feminist justice claims. I examine feminist jurisprudence’s methodological challenges in the settler colonial context of Alaska with respect to the murders of Della Brown (in 2000) and Mindy Schloss (in 2007). I advance my case (...) for pragrammatology as a critical methodology for feminist jurisprudence by subjecting the case(s) of Brown and Schloss to a more lucid (re)reading in two related and intersecting moves. First, I trace and deconstruct the citations of Brown and Schloss’ legal narratives to expose violence in the systemisation of f as a power and legal relation (Ahmed in Soc Legal Stud 4:55–73, 1995). I then examine the paradoxical nature of f justice claims which necessitate a pragrammatological approach. I conclude by advancing counsel on how we might move within paradox. (shrink)

In an article which appeared a few years ago, entitled ‘God's Death’ , A.D. Smith launched one of the most interesting of recent attacks on the traditional doctrine of the Incarnation. Focusing on the death of Christ, he claimed to demonstrate the logical impossibility of Jesus having been both human and divine. Each of the premises of his argument was said to be a commitment of orthodox theology. He thus presented his reasoning as displaying an internal incoherence in that (...) way of thinking about divinity, humanity, and the person of Christ. The argument was basically quite simple: According to Christian theology and in concurrence with general thought on the matter, we must hold that human death involves the possibility of annihilation. As a man, Jesus of Nazareth faced and underwent a human death. He thus faced the possibility of annihilation. But orthodox theologians hold God to be of such an ontological status that no divine being could even possibly be annihilated. So no divine person could die a human death. From this follows the impossibility of the traditional claim that the Second Person of the divine Trinity became a man, lived a human life, and died a human death for us and our salvation. The qualitative difference between God and man is such as to render incarnational christology an incoherent theological stance. (shrink)

Pp. xii q- 185) D~Sring has assembled the first complete collection of textual fragments concerning the Megarian philosophers of the fourth and third centuries B.c., together with a commentary. The fragments are divided by the author into four groups, each centered around one of the better-known figures of the school: (...) Euclid, Eubulides, Diodorus Cronus, Stilpo. Evidences concerning lesser-known personalities are included under the figure to whose "circle" they belonged. A fifth group of texts appears in an Anhang devoted to the sophist Bryson (often mistakenly associated with the Megarians) and his student Polyxenus. Divisions in the commentary parallel those in the text. The author also adds a stemma summarizing his conclusions about teacher-pupil relationships within the school, and there is a selective bibliography and a SteUenregister. An index, which in view of the work's organization would be helpful, is not included. Simply as a collection of texts, this is a major addition to the scholarship in its field; with the author's commentary, it is the most important work on the Megarians since Kurt von Fritz's Real-Encyclopiidie article.1 D~Sringpresents convincing evidence that some widely disseminated opinions about the school have little or no foundation. Some of his alternative views have important consequences, and not only for the history of the Megarians. DSring rejects the dates customarily assigned to the school's founder Euclid (ca. 450ca. 380), pointing out that there is actually no good evidence for them (pp. 73-74). His own figures (ca. 435-ca. 365) rely on the evidence of Plato's Theaetetus to give the year 369 as a terminus post quem for Euclid's death. The first consequence of this chronology is that the story of Euclid stealing into Athens disguised as a woman in order to avoid the anti-Megarian decree of 432 must be rejected; Dfiring feels that it is probably one of a family of tales elaborated by later writers "urn die Macht der Philosophie zu demonstrieren " (p. 74). A more important consequence is in store when this chronology is combined with a reassessment of the evidence relating to Euclid's philosophical position. D6ring rejects the traditional description of Euclid as a "neo-Eleatic" who attempted to combine a strict Parmenidean monism with Socrates' ethical views by identifying the "One" with the "Good." Together with the revision of Euclid's dates, this would appear to eliminate decisively Kr~imer's thesis that Euclid originated the "Gleichsetzung ontologischer und axiologischer Momente''e fundamental to Platonic metaphysics. First, Euclid was only a few years older than Plato (not "at least two decades," as KrOner requires); second, there is no evidence whatever that he had studied the Eleatic doctrines before meeting Socrates; and finally, the purported "Gleichsetzung" appears to be rather the work of doxographers (pp. 87-89). When we add DSring's skeptical assessment of the story of Plato's Megarian period (p. 76), there remains little evidence to support the theory that Euclid was the ultimate source of Plato'S metaphysics. The Native American Church is examined as an illustrative example in the political anthropology of consciousness. Specific attention is paid to the Supreme Court's ignoring of accepted research on this tradition and its sacrament, Peyote, in the case of Employment Division of Oregon v. Smith. An anthropological reaction to the Smith decision is constructed, focusing on ethnographic findings regarding Peyote that contradict the Supreme Court's ethnocentric assumptions. This paper argues that Peyote's Schedule I status is not supported by (...) the ethnographic findings. Peyote, as used in the Native American Church, is recognized as safe and therapeutic. It is also argued that the Supreme Court's rationale for denying Peyotist religious freedom is not supported by the ethnographic findings nor by legal precedent. Not only is the Native American's right to use Peyote a matter of freedom of religion, it also involves other rights such as the right to raise one's children in one's own culture and the right to be treated using a culturally relevant therapeutic modality. With the exception of three articles, all of the pieces collected here by Ballard and Scott appeared in the Winter, 1970 issue of The Southern Journal of Philosophy commemorating Heidegger’s 80th birthday. The opening essay by Poeggeler, "Heidegger Today," masterfully reviews the state of Heideggerian scholarship, sketching the direction which Heidegger’s interpretations have taken, and outlining his own unitary view of Heidegger’s development. This is followed by an interesting essay from the Heidegger critic Karl Löwith who, after some revealing personal (...) recollections about Heidegger, takes up the question of the relationship of Dasein, which stands out from and transcends nature, with the natural world, a question Heidegger himself omits. There is also a close exposition by Joseph Kockelmans of the all-important "Time and Being" lecture of 1962 and of its relationship to Being and Time. Hans-Georg Gadamer contributes a philosophical essay—not a piece of Heideggerian scholarship—on the nature of "empty" time, i.e., the temporal project which we hope to "fill up," and of the "transition" into fulfillment. Editor Scott gives an account of Dasein in Being and Time by an analogy with Leibniz’s monad as a self-originating, purposeful unit of activity. Theodore Kisiel differentiates the mathematical a priori, described by Heidegger as the root conception of modern science, from Heidegger’s own "hermeneutical" a priori. There are three studies of language: Volmann-Schluck discusses language and myth, while John Sallis studies how language and the reversal are intertwined in Heidegger’s thinking; Don Ihde differentiates "existential" phenomenology as a phenomenology of perception from "hermeneutic" phenomenology as a phenomenology of meaning, signification, and language. In addition to essays by A. V. Schoenborn, F. J. Smith, and Edward Ballard, there is also a concluding contribution by Jean Beaufret who offers a portrait of Heidegger "as seen from France."—J.D.C. One of the traditional ways mathematical ideas and even new areas of mathematics are created is from experiments. One of the best-known examples is that of the Fermat hypothesis, which was conjectured by Fermat in his attempts to find integer solutions for the famous Fermat equation. This hypothesis led to the creation of a whole field of knowledge, but it was proved only after several hundred years. This book, based on the author's lectures, presents several new directions of mathematical research. (...) All of these directions are based on numerical experiments conducted by the author, which led to new hypotheses that currently remain open, i.e., are neither proved nor disproved. The hypotheses range from geometry and topology (statistics of plane curves and smooth functions) to combinatorics (combinatorial complexity and random permutations) to algebra and number theory (continuous fractions and Galois groups). For each subject, the author describes the problem and presents numerical results that led him to a particular conjecture. In the majority of cases there is an indication of how the readers can approach the formulated conjectures (at least by conducting more numerical experiments). Written in Arnold's unique style, the book is intended for a wide range of mathematicians, from high school students interested in exploring unusual areas of mathematics on their own, to college and graduate students, to researchers interested in gaining a new, somewhat nontraditional perspective on doing mathematics. In the interest of fostering a greater awareness and appreciation of mathematics and its connections to other disciplines and everyday life, MSRI and the AMS are publishing books in the Mathematical Circles Library series as a service to young people, their parents and teachers, and the mathematics profession. In the present article the author studies and reviews the essential scientific opinions and discourse on the understanding of the term and phenomena of Cybernetics. The article is the author's vision of the concept of cybernetic epistemology based on its progressive methodological features. The main idea of the understanding of the information warfare category as a complex social system in the context of cybernetic epistemology is also developed in the present research. The concept of complexity theory has not yet been (...) applied in the study of ontological characteristics of the "information warfare" phenomenon so far. This kind of analyzing method allows us to highlight the true essential characteristics of "information warfare". There are recursiveness and spontaneous nature among them. This kind of epistemology methods and their possible and proper application in Cybernetics were analyzed and reviewed in the present paper. Such an analyzing confirmed the relevance of this research for the formalization of the world’s complex phenomena. It also determined the possibility of constructive generalization of knowledge and research data. However, this knowledge should be gained in the process of complex formalization of the studied phenomena characteristics and the maximum permissible changes and transformation in their conditions and forms.
