Fluorescence microscopy is the primary tool for studying complex processes inside individual living cells. Technical advances in both molecular biology and microscopy have made it possible to image cells from many genetic and environmental backgrounds. These images contain a vast amount of information, which is often hidden behind various sources of noise, convoluted with other information and stochastic in nature. Accessing the desired biological information therefore requires new tools of computational image analysis and modeling. Here, we review some of the (...) recent advances in computational analysis of images obtained from fluorescence microscopy, focusing on bacterial systems. We emphasize techniques that are readily available to molecular and cell biologists but also point out examples where problem‐specific image analyses are necessary. Thus, image analysis is not only a toolkit to be applied to new images but also an integral part of the design and implementation of a microscopy experiment. (shrink)

Reuben Hersh confided to us that, about forty years ago, the late Paul Cohen predicted to him that at some unspecified point in the future, mathematicians would be replaced by computers. Rather than focus on computers replacing mathematicians, however, our aim is to consider the (im)possibility of human mathematicians being joined by “artificial mathematicians” in the proving practice—not just as a method of inquiry but as a fellow inquirer.

Understanding is a valued trait in any epistemic practice, scientific or not. Yet, when it comes to characterizing its nature, the notion has not received the philosophical attention it deserves. We have set ourselves three tasks in this paper. First, we defend the importance of this endeavor. Second, we consider and criticize a number of proposals to this effect. Third, we defend an alternative account, focusing on abilities as the proper mark of understanding.

From 2009 to 2013 the workplace innovation project “My Enterprise 2.0” was carried out in the region of Utrecht in the Netherlands in order to strengthen the workplace innovation capability of small and medium-sized enterprises. Participating enterprises completed a questionnaire regarding the “workplace innovativeness” of their company. A workplace innovation intervention was then implemented by some of the companies, while other companies chose not to take part. At the end of the project, a second questionnaire indicated that those companies that (...) implemented interventions had a significantly higher score with regard to overall workplace innovativeness. The companies without such interventions reported a small decrease. While the companies in the region had higher workplace innovativeness scores relative to a national reference group both before and after the project, the increase in the “workplace innovativeness” of the regional SMEs that experienced interventions suggests that the project proved beneficial to their continued “workplace innovativeness.” Moreover, these companies also reported positive effects on company performance, achieving company goals and improving labor productivity. While self?evaluation leads to valuable results in some schools, it appears that in other schools this is true only to a lesser extent or not at all. This raises the question of how differences in the results of self?evaluations can be explained. This study looks at to what extent the results of self?evaluation are determined by the way in which self?evaluation is conducted, by characteristics relating to the general functioning of the school and by the support which schools enjoy. One (...) thousand seven hundred and eighty?six school principals and team members from 96 schools (primary and secondary) were surveyed by means of a written questionnaire. The data collected were then analysed using multi?level structural equation modelling. The results provide strong empirical evidence that ?attitude with regard to self?evaluation?, ?self?evaluation as a policy action? and ?self?evaluation as an act of research? are powerful predictors of the quality of self?evaluations. Wilson & Sober's discussion of group selection is marred by the absence of plausible examples of human group-level behavioral adaptation. The trait of authoritarianism is one possible example of such an adaptation. It reduces within-group variance in reproductive success, manifests itself more strongly in response to group-level threat, and is found in a variety of cultures.

This study explored LaRose and Eastin's model of media attendance, within a European context. It extended the uses and gratifications paradigm within the framework of social cognitive theory by instituting new operational measures of gratifications sought, reconstructed as outcome expectations. Although the model of media attendance offers some promising steps forward in measuring media selectivity and usage, and to some extent is applicable to another context of media use, the relative importance of outcome expectancies in explaining media usage and selectivity (...) is not fully supported. Despite the proliferation of research on ethical leadership, there remains a limited understanding of how specifically the assumingly moral component of this leadership style affects employee behavior. Taking an identity perspective, we integrate the ethical leadership literature with research on the dynamics of the moral self-concept to posit that ethical leadership will foster a sense of moral identity among employees, which then inspires followers to adopt more ethical actions, such as increased organization citizenship behavior. We further argue that these identity (...) effects should be more pronounced when leaders are perceived to be group prototypical, as their actions then speak louder to followers’ sense of identity. Two studies—a scenario experiment with 138 participants and a field study with 225 employees—provided support for our hypothesized moderated mediation model. Perceived ethical leadership positively affected OCB via followers’ moral identity but only under conditions of high perceived leader group prototypicality. We discuss how the identity pathway of ethical leadership can facilitate novel theorizing about moral transference. Our findings also suggest that, when hiring external ethical leaders or training internal managers, practitioners are well advised to consider that these individuals may only be effective in morally transforming followers when they are perceived as prototypical for the group. We investigated, by means of the Reverse Correlation Task, visual representations of the culturally dominating group of local people held by sojourners as a function of their degree of cross-cultural adaptation. In three studies, using three different methods with three independent samples of sojourners and seven independent samples of Portuguese and US-American raters, we gathered clear evidence that poor adaptation goes along with more negative representations of locals. This indicates that sojourner adaptation is reflected, at a social-cognitive level, in the (...) valence of outgroup representations. (shrink)

Cyclic adenosine monophosphate (cAMP) and cAMP‐dependent protein kinase A (PKA) are evolutionary conserved molecules with a well‐established position in the complex network of signal transduction pathways. cAMP/PKA‐mediated signaling pathways are implicated in many biological processes that cooperate in organ development including the motility, survival, proliferation and differentiation of epithelial cells. Cell surface polarity, here defined as the anisotropic organisation of cellular membranes, is a critical parameter for most of these processes. Changes in the activity of cAMP/PKA elicit a variety of (...) effects on intracellular membrane dynamics, including membrane sorting and trafficking. One of the most intriguing aspects of cAMP/PKA signaling is its evolutionary conserved abundance on the one hand and its precise spatial–temporal actions on the other. How do we ascribe subjective probability? In decision theory, this question is often addressed by representation theorems, going back to Ramsey (1926), which tell us how to define or measure subjective probability by observable preferences. However, standard representation theorems make strong rationality assumptions, in particular expected utility maximization. How do we ascribe subjective probability to agents which do not satisfy these strong rationality assumptions? Justification as Ignorance offers an original account of epistemic justification as both non-factive and luminous, vindicating core internalist intuitions without construing justification as an internal condition knowable by reflection alone. Sven Rosenkranz conceives of justification, in its doxastic and propositional varieties, as a kind of epistemic possibility of knowing and of being in a position to know. His account contrasts with recent alternative views that characterize justification in terms of the metaphysical possibility of knowing. Instead, he develops a suitable (...) non-normal multi-modal epistemic logic for knowledge and being in a position to know that respects the finding that these notions create hyperintensional contexts. He also defends his conception of justification against well-known anti-luminosity arguments, shows that the account allows for fruitful applications and principled solutions to the lottery and preface paradoxes, and provides a metaphysics of justification and its varying degrees of strength that is compatible with core assumptions of the knowledge-first approach and disjunctivist conceptions of mental states. The paper explores a structural account of propositional justification in terms of the notion of being in a position to know and negation. Combined with a non-normal logic for being in a position to know, the account allows for the derivation of plausible principles of justification. The account is neutral on whether justification is grounded in internally individuated mental states, and likewise on whether it is grounded in facts that are already accessible by introspection or reflection alone. To this extent, (...) it is compatible both with internalism and with externalism about justification. Even so, the account allows for the proof of principles that are commonly conceived to depend on an internalist conception of justification. The account likewise coheres both with epistemic contextualism and with its rejection, and is compatible both with the knowledge-first approach and with its rejection. Despite its neutrality on these issues, the account makes propositional justification luminous and so is controversial. However, it proves quite resilient in the light of recent anti-luminosity arguments. (shrink)

Sven Bernecker presents an analysis of the concept of propositional (or factual) memory, and examines a number of metaphysical and epistemological issues crucial to the understanding of memory. -/- Bernecker argues that memory, unlike knowledge, implies neither belief nor justification. There are instances where memory, though hitting the mark of truth, succeeds in an epistemically defective way. This book shows that, contrary to received wisdom in epistemology, memory not only preserves epistemic features generated by other epistemic sources but also (...) functions as a source of justification and knowledge. According to the causal theory of memory argued for in this book, the dependence of memory states on past representations supports counterfactuals of the form: if the subject hadn't represented a given proposition in the past he wouldn't represent it in the present. The book argues for a version of content externalism whereupon the individuation of memory contents depends on relations the subject bears to his past physical or social environment. Moreover, Bernecker shows that memory doesn't require identity, but only similarity, of past and present attitudes and contents. Although it has often been claimed that all the information contained in second‐order probabilities can be contained in first‐order probabilities, no practical recipe for the elimination of second‐order probabilities without loss of information seems to have been presented. Here, such an elimination method is introduced for repeatable events. However, its application comes at the price of losses in cognitive realism. In spite of their technical eliminability, second‐order probabilities are useful because they can provide models of important features of the world (...) that are cognitively more plausible than those that can be obtained with single‐level probabilities. Recent studies on neural pathways in a broad spectrum of vertebrates suggest that, in addition to migration and an increase in the number of certain select neurons, a significant aspect of neural evolution is a “parcellation” (segregation-isolation) process that involves the loss of selected connections by the new aggregates. A similar process occurs during ontogenetic development. These findings suggest that in many neuronal systems axons do not invade unknown territories during evolutionary or ontogenetic development but follow in their ancestors' paths (...) to their ancestral targets; if the connection is later lost, it reflects the specialization of the circuitry.The pattern of interspecific variability suggests (1) that overlap of circuits is a more common feature in primitive (generalized) than in specialized brain organizations and (2) that most projections, such as the retinal, thalamotelencephalic, corticotectal, and tectal efferent ones, were bilateral in the primitive condition. Specialization of these systems in some vertebrate groups has involved the selective loss of connections, resulting in greater isolation of functions. The parcellation process may also play an important role in cell diversification.The parcellation process as described here is thought to be one of several underlying mechanisms of evolutionary and ontogenetic differentiation. While 4E approaches to cognition are increasingly introduced in educational contexts, little has been said about how 4E commitments can inform pedagogy aimed at fostering ethical competencies. Here, we evaluate a 4E-inspired ethics exercise that we developed at a technical university to enliven the moral imagination of engineering students. Our students participated in an interactive tinkering workshop, during which they materially redesigned a healthcare artifact. The aim of the workshop was twofold. Firstly, we wanted students to experience how material choices (...) at the levels of design and functionality can enable morally significant reimaginings of the affordances commonly associated with existing artifacts. We term this type of reimagining world-directed moral imagination. Secondly, through the design process, we wanted students to robustly place themselves in the lived embodied perspectives of (potential) users of their selected artifacts. We term this person-directed moral imagination. While student testimonies about the exercise indicate that both their world-directed and person-directed moral imagination were enlivened, we note that the fostering of robust person-directed moral imagination proved challenging. Using 4E insights, we diagnose this challenge and ask how it might be overcome. To this end, we engage extensively with a recent 4E-informed critique of person-directed moral imagination, raised by Clavel Vázquez and Clavel-Vázquez (2023). They argue that person-directed moral imagination is profoundly limited, if not fundamentally misguided, particularly when exercised in contexts marked by emphatic embodied situated difference between the imaginer and the imagined. Building upon insights from both the 4E field and testimonies from critical disability studies, we argue that, while their critique is valuable, it ultimately goes too far. We conclude that a 4E approach can take on board recent 4E warnings regarding the limits of person-directed moral imagination while contributing positively to the development of moral imagination in engineering ethics education. Bernard Williams argued that historical and philosophical inquiry were importantly linked in a number of ways. This introductory chapter distinguishes four different connections he identified between philosophy and history. (1) He believed that philosophy could not ignore its own history in the way that science can. (2) He thought that when engaging with philosophy’s history primarily to produce history, one still had to draw on philosophy. (3) Even doing history of philosophy philosophically, i.e. primarily to produce philosophy, required a keen (...) sense of how historically distant from us past philosophers were, on his view, because the point of reading them was to confront something different from the present. (4) He held that systematic philosophy itself needed to be done historically, engaging not necessarily with its own history, but with that of the concepts it sought to understand. Ockhamism implies that future contingents may be true, their historical contingency notwithstanding. It is thus opposed to both the Peircean view according to which all future contingents are false, and Supervaluationist Indeterminism according to which all future contingents are neither true nor false. The paper seeks to defend Ockhamism against two charges: the charge that it cannot meet the requirement that truths be grounded in reality, and the charge that it proves incompatible with objective indeterminism about the future. In each (...) case, the defence draws on the idea that certain truths are truths only courtesy of others and of what makes the latter true. After introduction of the Ockhamist view, its competitors and implications, a suitable definition of grounded truth is being devised that both is faithful to the spirit of the grounding-requirement and allows the Ockhamist to heed that requirement quite comfortably. Then two senses in which the future might be open are being introduced, indeterminacy as failure of predetermination by past and present facts, and indeterminacy as failure of entailment by past and present truths. It is argued that while openness in the former sense, but not in the latter sense, coheres with the Ockhamist view, it is only openness in the former sense that matters for objective indeterminism. Many ethicists writing about automated systems attribute agency to these systems. Not only that; they seemingly attribute an autonomous or independent form of agency to these machines. This leads some ethicists to worry about responsibility-gaps and retribution-gaps in cases where automated systems harm or kill human beings. In this paper, I consider what sorts of agency it makes sense to attribute to most current forms of automated systems, in particular automated cars and military robots. I argue that whereas it indeed (...) makes sense to attribute different forms of fairly sophisticated agency to these machines, we ought not to regard them as acting on their own, independently of any human beings. Rather, the right way to understand the agency exercised by these machines is in terms of human–robot collaborations, where the humans involved initiate, supervise, and manage the agency of their robotic collaborators. This means, I argue, that there is much less room for justified worries about responsibility-gaps and retribution-gaps than many ethicists think. (shrink)

Does remembering require a causal process connecting the past representation to its subsequent recall and, if so, what is the nature of the causal process? Of what kind are the primary intentional objects of memory states? How do we know that our memory experiences portray things the way they happened in the past? Given that our memory is not only a passive device for reproducing thoughts but also an active device (...) for processing stored thoughts, when are thoughts sufficiently similar to be memory-related? The Metaphysics of Memory defends a version of the causal theory of memory, argues for direct realism about memory, proposes an externalist response to skepticism about memory knowledge, and develops a contextualist account of the factivity constraint on memory. Responding to the critical comments by the symposiasts Julien Dutant, Niccolò Rossi, Martin Smith, Daniel Waxman, and Yiwen Zhan, I further elaborate, refine, and defend the account of epistemic justification I advanced in my book Justification as Ignorance (Oxford: Oxford University Press, 2021). Central issues tackled here include logical omniscience and non-normal epistemic logics, the luminosity of epistemic justification, methods for telling what one is in no position to know, and the relation between doxastic and propositional justification.

We show that Bell inequalities can be violated in the macroscopic world. The macroworld violation is illustrated using an example involving connected vessels of water. We show that whether the violation of inequalities occurs in the microworld or the macroworld, it is the identification of nonidentical events that plays a crucial role. Specifically, we prove that if nonidentical events are consistently differentiated, Bell-type Pitowsky inequalities are no longer violated, even for Bohm's example of two entangled spin 1/2 quantum particles. We (...) show how Bell inequalities can be violated in cognition, specifically in the relationship between abstract concepts and specific instances of these concepts. This supports the hypothesis that genuine quantum structure exists in the mind. We introduce a model where the amount of nonlocality and the degree of quantum uncertainty are parameterized, and demonstrate that increasing nonlocality increases the degree of violation, while increasing quantum uncertainty decreases the degree of violation.
