In normal human cells, a protein with a molecular weight of about 53,000 (p53) is present at a low level, detectable only by sensitive assays. When the cells are infected with some DNA tumour viruses or transformed by the same viruses, the amount of this protein increases dramatically, and much of it is found in a physical association with the virus‐coded protein that causes the transformation. The increase in amount and, incidentally, stability of p53 is not peculiar to virus‐transformed cells, (...) but also occurs in cells transformed by chemicals or radiation where there is no sign of any virus being involved (reviewed by Crawford).1 Cells in culture are not the same as cells in a patient, but these observations do raise questions about the possible involvement of p53 in human tumours. Numerous clinical, epidemiological and molecular findings link some types of Human Papillomaviruses (HPV) with cancer of the genital tract. They share a common pathway of transformation with a number of DNA tumour viruses, such as Adenovirus and SV40. Although all these viruses are termed ‘DNA tumour viruses’ and have similar in vitro transforming activities, Human Papillomavirus is the only one so far clearly involved in human cancer. Extensive studies on HPV E6 and E7 proteins have demonstrated their involvement in malignant (...) transformation. E6 and E7 bind the products of tumour suppressor genes, p53 and Rb1, respectively, modifying or inactivating their normal functions. The Rb1 and p53 genes are deleted or mutated in several cancers and both proteins regulate the transcription of genes involved in cell cycle progression control. The E6/p53 and E7/Rb1 interactions result in a deregulation of the cell cycle with loss of control of crucial cellular events, such as DNA replication, DNA repair and apoptosis. (shrink)

JenkinsIt is common to open a work regarding the merits of Mansfield Park by noting that Fanny Price is very difficult to like. Nietzsche might have described her as a "moral tarantula." 1 She sits, making negative moral judgments about the actions of others, while doing nothing herself. Fanny spends most of her time, literally, sitting or lying down. Austen describes her character (...) as "supine." Comparing Fanny with her sister Susan, Austen says that "Susan was only acting on the same truths, and pursuing the same system, which her [Fanny's] own judgment acknowledged, but which her more supine and yielding temper would have shrunk from asserting. Susan tried to be useful, where she could only have gone away and cried" (MP, p. 395). 2 And Fanny cries all of the time. She cries at least thirteen times over the course of the novel. So, there are two questions about the novel that need to be answered. Why did Austen create a heroine who is so clearly distasteful? Why do some critics and philosophers see Fanny as a moral ideal or cultural icon? Some (Alasdair MacIntyre, Gilbert Ryle, Lionel Trilling) argue that Fanny is so unappealing just because Austen wants to be didactic 3 and force us to focus on virtue. According to Lionel Trilling, for example, Mansfield Park has a different moral message from Pride and Prejudice. "Its praise is not for social freedom, but for social stasis. It takes full notice of spiritedness, vivacity, celerity and lightness, but only to reject them as having nothing to do with virtue and happiness, as being, indeed, deterrents to the good life." 4 Others think that the infelicities of the novel and its heroine make it a parody of traditional feminine values and conservative moral thought in general. 5 Here, I shall argue that a path between these two extreme ways of reading the novel is correct. I shall argue, using Alasdair MacIntyre's discussion as a starting [End Page 346] point, that Austen does not cast Fanny as a moral ideal. If Austen is Aristotelian (a common view among philosophers) Fanny cannot be an ideal. And, although I agree with Johnson's view that those who have read the novel as morally conservative are wrong, I do not think that the novel exhibits global skepticism about traditional standards for the beautiful/feminine. Austen condemns the idea that women should be passive dolls who do not think for themselves, but she does not parody the traditional idea that women should be sweet, gentle, and caring. In my view, Fanny's character constitutes a criticism of passivity, and of rigid adherence to moral rules.Alasdair MacIntyre's view is that Austen is an Aristotelian moralist with a Christian slant on the make-up of the virtues. He describes Fanny's character as an ideal of virtue. The Beginnings of Nietzsche's Theory of Language is concerned with the years 1865 through Winter/Spring 1870-71. Four texts of Nietzsche's, "Vom Ursprung der Sprache", "Zur Teleologie", "Zu Schopenhauer", and "Anschauung Notes", are translated into English and interpreted from the perspective of Nietzsche's developing theory of language. An examination of the major influences of Schopenhauer, Kant, Eduard von Hartmann, and Frederick A. Lange are pursued. ;Theory, in this work, does not assume that it is possible to take a position of authority (...) or truth in the interpretation of texts, rather that interpretation of texts constitutes an attempt to uncover a discursive practice, to describe a discourse-object. The method used is Nietzsche's method of genealogy, which follows the formation of discourse, at once scattered, discontinuous, and regular and analyzes the process of rarefaction, consolidation and unification in that discourse. Genealogy is exegesis in Nietzsche's sense of it as rumination, as the slow, repetitive, gray activity of deciphering what is documented. Major genealogical nodes or elements of Nietzsche's beginning theory of language are used and reused in different contexts throughout the work, resulting in a cumulative effect which explores specific elements in Nietzsche's thinking about language. ;This work offers five major contributions to Nietzsche studies: a detailed introduction of the significant influence of Eduard von Hartmann upon Nietzsche's thinking; a comprehensive look at the influence of Lange's History of Materialism upon Nietzsche's beginning theory of language; a thorough analysis of "Zur Teleologie" and "Zu Schopenhauer" in the overall contexts of Nietzsche's thinking with regard to metaphysics and teleology; the gathering together and interpretation into a world view of the "Anschauung Notes"; and a close examination of Nietzsche's breaking away from Schopenhauer as it takes place before the publication of The Birth of Tragedy. ;Nietzsche's primary contributions to a theory of language in these beginning years are three: the adoption of the role of the unconscious instincts in the origins and ongoing production of language; the assertion that unconscious thought processes are a precondition for conscious thought; an examination of the relationships of language to epistemology, which results in the recognition of the limits of language as an instrument of truth, but a realization of the inestimable value and force of its symbolic and figurative aspects, aspects necessitated by the very nature of language itself. The present study concerns the issue of meaning in contemporary continental philosophy. In particular, it develops the two accounts of meaning offered by Heidegger and Levinas, each of whom presents us with a differing break from Husserl. As a first attempt to name the difference between these three thinkers, one could say that Husserl gives us an epistemological notion of meaning; whereas Heidegger gives us an ontological account, and Levinas gives us an ethical account. We will refine and reformulate this (...) division in relation to the issues of each chapter. ;We will thus see how both Heidegger and Levinas develop their separate notions in relation to Husserl's own revolutionary approach to the question of meaning. Whereas Husserl both replaced the term "experience" with the term "intuition" and gave a compelling account of the "prepredicative" roots of the meaningful, Heidegger advances the articulation of such a prepredicative realm of meaning even further than Husserl. He did so, however, in a way that ends up displacing Husserl's notion of intuition. This is the move from Husserl's account of phenomenological intuition to Heidegger's own notion of a more hermeneutic phenomenology . ;I will show how Heidegger articulates an account of meaning as that upon which any projection of understanding takes shape. Meaning becomes a quasi-transcendental, non-subjective notion of that which makes possible the disclosure of any "world." Levinas, on the other hand, offers an account of meaning---in intended opposition to Heidegger---as located primarily in the ethical exposure at the heart of language: in the "face-to-face" relation of Same and Other. He further develops his notion of meaning in terms of the notion of the saying and the said. Both Heidegger and Levinas give us a non-intentional account of the meaningful. For Heidegger it takes the form of a non-intentional background that makes possible both the "worldhood of the world" and the possibilities of Dasein . For Levinas, the meaningful is thought in terms of an interpersonal, quasi-ethical face to face relation between persons exposed to the vulnerability, obligation, and height of an "Other" in what he calls the "saying.". In the minds of many, Hox gene null mutant phenotypes have confirmed the direct role that these genes play in specifying the pattern of vertebrate embryos. The genes are envisaged as defining discrete spatial domains and, subsequently, conferring specific segmental identities on cells undergoing differentiation along the antero‐posterior axis. However, several aspects of the observed mutant phenotypes are inconsistent with this view. These include: the appearance of other, unexpected transformations along the dorsal axis; the occurrence of mirror‐image duplications; and the (...) development of anomalies outside the established domains of normal Hox gene expression. In this paper, Hox gene disruptions are shown to elicit regeneration‐like responses in tissues confronted with discontinuities in axial identity. The polarities and orientations of transformed segments which emerge as a consequence of this response obey the rules of distal transformation and intercalary regeneration. In addition, the incidence of periodic anomalies suggests that the initial steps of Hox‐mediated patterning occurs in Hensen's node. As gastrulation proceeds, mesoderm cell cycle kinetics impose constraints upon subsequent cellular differentiation. This results in the delayed manifestation of transformations along the antero‐posterior axis. Finally, a paradigm is sketched in which temporal, rather than spatial axial determinants direct differentiation. Specific, testable predictions are made about the role of Hox genes in the establishment of segmental identity. (shrink)

According to Russellianism (or Millianism), the two sentences ‘Ralph believes George Eliot is a novelist’ and ‘Ralph believes Mary Ann Evans is a novelist’ cannot diverge in truth-value, since they express the same proposition. The problem for the Russellian (or Millian) is that a puzzle of Kaplan’s seems to show that they can diverge in truth-value and that therefore, since the Russellian holds that they express the same proposition, the Russellian view is contradictory. I argue that the standard Russellian appeal (...) to “ways of thinking” or “propositional guises” is not necessary to solve the puzzle. Rather than this retrograde concession to Fregeanism, appeal should be made to second-order belief. The puzzle is solved, and the contradiction avoided, by maintaining that both sentences are indeed true in addition to the sentence ‘Ralph (mistakenly) believes that he does not believe Mary Ann Evans/George Eliot is a novelist’. Surveys Western literature and thought to reveal the evolution of the ideals of sincerity and authenticity.

This seminal work by political philosopher C.B. Macpherson was first published by the Clarendon Press in 1962, and remains of key importance to the study of liberal-democratic theory half-a-century later. In it, Macpherson argues that the chief difficulty of the notion of individualism that underpins classical liberalism lies in what he calls its "possessive quality" - "its conception of the individual as essentially the proprietor of his own person or capacities, owing nothing to society for them." Under such a conception, (...) the essence of humanity becomes freedom from dependence on the wills of others; society is little more than a system of economic relations; and political society becomes a means of safeguarding private property and the system of economic relations rooted in property. As the New Statesman declared: "It is rare for a book to change the intellectual landscape. The existence of object-dependent thoughts has been doubted on the grounds that reference to such thoughts is unnecessary or 'redundant' in the psychological explanation of intentional action. This paper argues to the contrary that reference to object-dependent thoughts is necessary to the proper psychological explanation of intentional action upon objects. Section I sets out the argument for the alleged explanatory redundancy of object-dependent thoughts; an argument which turns on the coherence of an alternative 'dual-component' model of explanation. Section II rebuts (...) this argument by showing the dual-component model to be incoherent precisely because of its exclusion of object-dependent thoughts. Section III concludes with a conjecture about the further possible significance of object-dependent thoughts for the prediction of action. (shrink)

Brandom's "inferentialism"—his theory that contentfulness consists in being governed by inferential norms—proves dubiously compatible with his own deflationary approach to intentional objectivity. This is because a deflationist argument, adapted from the case of truth to that of correct inference, undermines the criterion of adequacy Brandom employs in motivating inferentialism. Once that constraint is abandoned, moreover, the very constitutive-explanatory availability of Brandom's inferential norms becomes suspect. Yet Brandom intertwines inferentialism with a separate explanatory project, one that in explaining the pragmatic significance (...) of meaning-attributions does yield a convincing construal of the claim that the concept of meaning is normative. Deflationists about truth seek to undermine debates about the nature of truth by arguing that the truth predicate is merely a device that allows us to express a certain kind of generality. I argue that a parallel approach is available in the case of logical consequence. Just as deflationism about truth offers an alternative to accounts of truth's nature in terms of correspondence or justification, deflationism about consequence promises an alternative to model-theoretic or proof-theoretic accounts of consequence's nature. I then (...) argue, against considerations put forward by Field and Beall, that Curry's paradox no more rules out deflationism about consequence than the liar paradox rules out deflationism about truth. Innocence is a notion that can prove controversial. Claims of innocence typically support not imposing burdens on the innocent when their conduct is relevantly unobjectionable. This paper examines innocence in the context of violent conflict between states or groups. Many thinkers about the morality of such violence want to establish a principle that would protect innocent civilians. Yet the common view in just war theory does not affirm the moral innocence of civilians. Most contemporary metaphysicians are sceptical about the reality of familiar objects such as dogs and trees, people and desks, cells and stars. They prefer an ontology of the spatially tiny or temporally tiny. Tiny microparticles 'dog-wise arranged' explain the appearance, they say, that there are dogs; microparticles obeying microphysics collectively cause anything that a baseball appears to cause; temporal stages collectively sustain the illusion of enduring objects that persist across changes. Crawford L. Elder argues that all such attempts to (...) 'explain away' familiar objects project downwards, onto the tiny entities, structures and features of familiar objects themselves. He contends that sceptical metaphysicians are thus employing shadows of familiar objects, while denying that the entities which cast those shadows really exist. The starting point of his argument is that ontology should operate under empirical load -- that is, it should give special weight to the objects and properties that we treat as real in our best predictions and explanations of what happens in the world. Elder calls this presumption "mildly controversial" because it entails that arguments are needed for certain widely assumed positions such as "mereological universalism". Elder begins by defending realism about essentialness. He then defends this view of familiar objects against causal exclusion arguments and worries about vagueness. Finally, he argues that many of the objects in which common sense believes really exist, including artifacts and biological devices shaped by natural selection, and that we too exist, as products of natural selection. This paper examines two explanations Sellars gives, at successive stages of his career, of how semantic vocabulary lets us relate linguistic expressions to extra-linguistic reality. Despite their differences, both explanations reveal a distinctive pragmatist approach. According to Sellars, we do not use semantic vocabulary to describe language-world relations. Rather, our taking language to relate to the world is implicit in the moves licensed by our semantic assertions. I argue that Sellars's discussions of the function of semantic vocabulary point to an (...) overlooked position regarding the relation between the concepts of meaning and truth. According to him, the function of meaning ascriptions cannot be explained independently of the function of truth ascriptions. That is because the function of meaning ascriptions essentially involves licensing claims about the world when combined with truth ascriptions. According to relativist accounts of discourse about, e.g., epistemic possibility and matters of taste, the truth of propositions must be relativized to nonstandard parameters. This paper argues that the central thrust of such accounts should be understood independently of relative truth, in terms of a perspectival account of assertoric force. My point of departure is a stripped-down version of Brandom’s analysis of the normative structure of discursive practice. By generalizing that structure, I make room for an analogue of the “assessment (...) sensitivity” MacFarlane characterizes in terms of relative truth. I argue that my reformulation supplies a stronger rationale for the most distinctive feature of MacFarlane’s brand of relativism, its account of when speakers ought to retract assertions. Furthermore, I show that the view usually regarded as a “moderate” alternative to MacFarlane’s “radical” relativism requires the more radical deviation from an absolutist account of assertoric force.
