Genetic variation in fitness is the fundamental prerequisite for adaptive evolutionary change. If there is no variation in survival and reproduction or if this variation has no genetic basis, then the composition of a population will not evolve over time. Consequently, the factors influencing genetic variation in fitness have received close attention from evolutionary biologists. One key factor is the mode of reproduction. Indeed, it has long been thought that sex enhances fitness variation and that this explains the ubiquity of (...) sexual reproduction among eukaryotes. Nevertheless, theoretical studies have demonstrated that sex need not always increase genetic variation in fitness. In particular, if fitness interactions among beneficial alleles (epistasis) are positive, sex can reduce genetic variance in fitness. Empirical data have been sorely needed to settle the issue of whether sex does enhance fitness variation. A recent flurry of studies1-4 has demonstrated that sex and recombination do dramatically increase genetic variation in fitness and consequently the rate of adaptive evolution. Interpreted in light of evolutionary theory, these studies rule out positive in these experiments epistasis as a major source of genetic associations. Securing equitable antibiotic access as an essential component for health system resilience and pandemic preparedness requires a systems perspective. This article discusses key components that need to be coordinated and paired with adequate financing and resources to ensure antibiotic effectiveness as a global public good, which should be central while discussing a new global agreement.

We estimate that 208,000 deep brain stimulation (DBS) devices have been implanted to address neurological and neuropsychiatric disorders worldwide. DBS Think Tank presenters pooled data and determined that DBS expanded in its scope and has been applied to multiple brain disorders in an effort to modulate neural circuitry. The DBS Think Tank was founded in 2012 providing a space where clinicians, engineers, researchers from industry and academia discuss current and emerging DBS technologies and logistical and ethical issues facing the field. (...) The emphasis is on cutting edge research and collaboration aimed to advance the DBS field. The Eighth Annual DBS Think Tank was held virtually on September 1 and 2, 2020 (Zoom Video Communications) due to restrictions related to the COVID-19 pandemic. The meeting focused on advances in: (1) optogenetics as a tool for comprehending neurobiology of diseases and on optogenetically-inspired DBS, (2) cutting edge of emerging DBS technologies, (3) ethical issues affecting DBS research and access to care, (4) neuromodulatory approaches for depression, (5) advancing novel hardware, software and imaging methodologies, (6) use of neurophysiological signals in adaptive neurostimulation, and (7) use of more advanced technologies to improve DBS clinical outcomes. There were 178 attendees who participated in a DBS Think Tank survey, which revealed the expansion of DBS into several indications such as obesity, post-traumatic stress disorder, addiction and Alzheimer’s disease. The INBIOSA project brings together a group of experts across many disciplines who believe that science requires a revolutionary transformative step in order to address many of the vexing challenges presented by the world. It is INBIOSA’s purpose to enable the focused collaboration of an interdisciplinary community of original thinkers. This paper sets out the case for support for this effort. The focus of the transformative research program proposal is biology-centric. We admit that biology to date has been more fact-oriented (...) and less theoretical than physics. However, the key leverageable idea is that careful extension of the science of living systems can be more effectively applied to some of our most vexing modern problems than the prevailing scheme, derived from abstractions in physics. While these have some universal application and demonstrate computational advantages, they are not theoretically mandated for the living. A new set of mathematical abstractions derived from biology can now be similarly extended. This is made possible by leveraging new formal tools to understand abstraction and enable computability. [The latter has a much expanded meaning in our context from the one known and used in computer science and biology today, that is "by rote algorithmic means", since it is not known if a living system is computable in this sense (Mossio et al., 2009).] Two major challenges constitute the effort. The first challenge is to design an original general system of abstractions within the biological domain. The initial issue is descriptive leading to the explanatory. There has not yet been a serious formal examination of the abstractions of the biological domain. What is used today is an amalgam; much is inherited from physics (via the bridging abstractions of chemistry) and there are many new abstractions from advances in mathematics (incentivized by the need for more capable computational analyses). Interspersed are abstractions, concepts and underlying assumptions “native” to biology and distinct from the mechanical language of physics and computation as we know them. A pressing agenda should be to single out the most concrete and at the same time the most fundamental process-units in biology and to recruit them into the descriptive domain. Therefore, the first challenge is to build a coherent formal system of abstractions and operations that is truly native to living systems. Nothing will be thrown away, but many common methods will be philosophically recast, just as in physics relativity subsumed and reinterpreted Newtonian mechanics. -/- This step is required because we need a comprehensible, formal system to apply in many domains. Emphasis should be placed on the distinction between multi-perspective analysis and synthesis and on what could be the basic terms or tools needed. The second challenge is relatively simple: the actual application of this set of biology-centric ways and means to cross-disciplinary problems. In its early stages, this will seem to be a “new science”. This White Paper sets out the case of continuing support of Information and Communication Technology (ICT) for transformative research in biology and information processing centered on paradigm changes in the epistemological, ontological, mathematical and computational bases of the science of living systems. Today, curiously, living systems cannot be said to be anything more than dissipative structures organized internally by genetic information. There is not anything substantially different from abiotic systems other than the empirical nature of their robustness. We believe that there are other new and unique properties and patterns comprehensible at this bio-logical level. The report lays out a fundamental set of approaches to articulate these properties and patterns, and is composed as follows. -/- Sections 1 through 4 (preamble, introduction, motivation and major biomathematical problems) are incipient. Section 5 describes the issues affecting Integral Biomathics and Section 6 -- the aspects of the Grand Challenge we face with this project. Section 7 contemplates the effort to formalize a General Theory of Living Systems (GTLS) from what we have today. The goal is to have a formal system, equivalent to that which exists in the physics community. Here we define how to perceive the role of time in biology. Section 8 describes the initial efforts to apply this general theory of living systems in many domains, with special emphasis on crossdisciplinary problems and multiple domains spanning both “hard” and “soft” sciences. The expected result is a coherent collection of integrated mathematical techniques. Section 9 discusses the first two test cases, project proposals, of our approach. They are designed to demonstrate the ability of our approach to address “wicked problems” which span across physics, chemistry, biology, societies and societal dynamics. The solutions require integrated measurable results at multiple levels known as “grand challenges” to existing methods. Finally, Section 10 adheres to an appeal for action, advocating the necessity for further long-term support of the INBIOSA program. -/- The report is concluded with preliminary non-exclusive list of challenging research themes to address, as well as required administrative actions. The efforts described in the ten sections of this White Paper will proceed concurrently. Collectively, they describe a program that can be managed and measured as it progresses. (shrink)

Dissipative structures consisting of a few macrovariables arise out of a sea of reversible microvariables. Unexpected residual effects of the massive underlying reversibility, on the macrolevel, cannot therefore be excluded. In the age of molecular-dynamics simulations, explicit dissipative structures like excitable systems (“explicit observers”) can be generated in a computer from first reversible principles. A class of classical, 1-D Hamiltonian systems of chaotic type is considered which has the asset that the trajectorial behavior in phase space can be understood geometrically. (...) If, as natural, the number of particle types is much smaller than that of particles, the Gibbs symmetry must be taken into account. The permutation invariance drastically changes the behavior in phase space (quasi-periodization). The explicit observer becomes effectively reversible on a short time scale. In consequence, his ability to measure microscopic motions is suspended in a characteristic fashion. Unlike quantum mechanics whose “holistic” nature cannot be transcended, the present holistic (internal-interface) effects—mimicking the former to some extent—can be understood fully in principle. İnsanın kendini bir hiçliğin önünde durmuş ve gerçek bir olabilmeye davet edilmiş bulduğu ruh hali olarak korku içinde bulunuş, ölümle karşılaştırıldığında insanın en uç korkusuna da işaret ediyor (...) olmalıdır. Die gesamte „Lebensstimmung“ einer Person hängt von der Gesamtheit der Lebenslagenteile ab, von der Morbidität ebenso wie von der Wohnung. Ob man die Lebens-Stimmung in „positive“ und „negative“ Elemente zerlegen kann, die man einzelnen Elementen der Lebenslage zuordnet, ist eine offene Frage.Die wissenschaftliche Praxis geht am besten von „Lebenslagenphysiognomien“ (Lebenslagensilhouetten) aus, die durch (...) verschiedenartig gemessene und skalierte Grössen gebildet werden. Wenn man die Lebenslagen einzelner Personen oder ganzer Länder auch nicht stets „skalierend“ zu vergleichen vermag, so kann man sie doch immer beschreiben. Geht man daran, „Lebenslagenkataster“ (Lebenslageninventare) verschiedener Völker und Zeiten aufzustellen, so muss man beachten, dass die Lebensstimmung eines Menschen nicht etwa schon durch die Garten fläche gekennzeichnet wird, die ihm zur Verfügung steht ; man muss angeben, über wieviel Freizeit er verfügt, um die Gartenfläche zu benutzen.Man kann Gesellschaftsordnungen durch die von ihnen „produzierten“ Lebenslagen ln Verbindung mit einer umfassenden „Naturalrechnung“ kennzeichnen. Réservons provisoirement la question.Si nous envisageons la méthode scientifique, le point de départ le meilleur serait la notion de „physiognomie (silhouette) de vie“. On les dessinerait à l'aide de grandeurs diversement mesurées et graduées.Certes, il ne serait guère possible de comparer les „situations de vie“ de personnes ou de peuples entiers en utilisant une seule échelle de graduation, tout au moins est-il possible de les décrire. Si l'on tente de dresser un „cadastre (inventaire) de situations de vie“ de différents peuples en différentes époques, il ne faut pas oublier que le „climat“ dans lequel vit un individu n'est pas assez déterminé par la surface du jardin qui lui appartient, il faut aussi préciser le loisir dont il dispose pour le cultiver.On peut caractériser les différents régimes sociaux par les situations de vie qu'ils produisent, à condition de combiner cette analyse avec un calcul en nature aussi étendu que possible. Le calcul en argent peut-il remplacer dans une certaine mesure le calcul en nature? With Ernst Christoph von Manteuffel and Luise Adelgunde Victorie Gottsched, two people come into contact with each other who are unique in the early Enlightenment. After relinquishing his high political functions, Imperial Count Manteuffel devoted his energy to promoting enlightened ideas characteristic of the Leibniz-Wolff tradition. In the field of tension between the bourgeoisie, the nobility and the Enlightenment, his work has its own weight. The exceptional position of L. A. V. Gottsched among female authors in the second third of (...) the 18 th century has always been recognized. In her dealings with Manteuffel, her position as a satirist, as yet barely perceived, also unfolds, and her contributions touch the nerve of the politically explosive conflicts between Wolffianism and traditional theological positions. The Gottschedin-Manteuffel correspondence shows two sovereign actors connected in mutual respect. (shrink)

Certain palaeontological observations, of which some examples are described do not fit in either a Lamarckian or Darwinian view of evolution. Both of these mechanistic theories assume small character-changes in the end stages of ontogeny, which gradually accumulate and are supposed thus to have given rise to all evolutionary progress. Palaeontology, on the other hand, teaches that the fundamental qualitative changes in the morphological plan have taken place in a saltatory manner in more or less young ontogenetic stages . Only (...) after the morphological plan has been built in a phylogenetic phase of explosive, early ontogenetic changing of type follows a phase of stable, continous evolution. It is shown that, theoretically, no other kind of fundamental change in the organisation is possible and that many, otherwise inexplicable, phenomena, the lack of transition forms, the question of ancestral stocks, the ideas of the biology of organisms as a “whole” etc., find their explanation by this means.Il y a certaines observations paléontologiques, dont l'auteur donne plusieurs exemples, et qu'on ne pourrait mettre d'accord avec un aspect larmarckienne ou darwinienne. Ce sont là des essais mécaniques d'explication, qui comptent avec de petites modifications de caractères dans les derniers stades de l'ontogénèse qui se totalisent peu à peu et devraient ainsi avoir occasionné tout progrès phylogénétique. Par contre, la paléontologie enseigne que les transformations qualitatives profondes des plans de constructions ont évolué par bonds dans les stades ontogénétiques plus ou moins juvéniles . C'est seulement après que les types ont été formés dans une phase phylogénétique de transformation explosive et juvénile, qu'une phase d'une évolution stable et continue vient s'y ajouter. On démontre que théoriquement un autre procédé d'un changement profond d'organisation est absolument impossible et que de cette manière un grand nombre de phénomènes par ailleurs incompréhensibles trouvent de la sorte leurs explications: par exemple l'absence des formes transitoires, la question des formes de racines, les conceptions de la biologie du „tout” de l'organisme etc. (shrink)

À moins de définir la justice ou le droit de telle manière qu’il existe entre eux un rapport intrinsèque parce qu’on les a déjà conçus comme indiscernables, le rapport entre eux ne peut être que contingent d’un point de vue conceptuel et ontologique. Puisque la justice au sens matériel constitue toujours une conception particulière de la distribution des biens au sens le plus large, la réalisation d’une telle conception est en revanche toujours subordonnée à la technicité du droit qui la (...) concrétise en réglant les comportements sous des conditions non idéales, faisant intervenir sous certaines conditions, entre autres éléments, des actes de violence. Si le droit est de ce point indifférent par rapport à la justice, toute théorie de la justice doit tenir compte des contraintes de sa réalisation juridique. (shrink)

La juridicisation croissante de notre vie publique et le fonctionnement de l'Etat de droit ont rendu nécessaire de reposer la question des modes de raisonnement fondant désormais les décisions judiciaires, de plus en plus importantes, de plus en plus nombreuses, qui régissent notre activité quotidienne. L'enjeu n'en est pas négligeable. Au-delà d'une vision "technologique" du droit - le droit, une technique, un outil... -, c'est aussi, et de manière peut-être plus inquiétante, une vision plus politique qui en est souvent suggérée (...) : la mort annoncée du droit comme technique autonome, capable de résister au politique - simple habillage, habile camouflage, d'une décision dont la motivation ne serait jamais - ne pourrait jamais être - mise en échec, orientée, ou commandée par les contraintes du droit. Entre positivisme et réalisme, l'oscillation risque d'être catastrophique. Entre ces deux abîmes, cet ouvrage tente d'ouvrir les voies d'une réflexion rigoureuse et scientifique sur les moyens intellectuels les plus nécessaires à l'affirmation - et à l'affermissement - de l'Etat de droit. Autour du Manifeste de Vienne se trouvent reunis des textes fondateurs ecrits autour de 1929. Leurs auteurs: Carnap, Hahn, Neurath, Schlick l'ame du Cercle de Vienne, et Waismann plus proche de Wittgenstein, temoignent d'un courant philosophique constituant aujourd'hui la tradition analytique de source continentale a la fois empiriste et logique. Forme de maniere informelle a Vienne, au coeur de l'Europe, le Cercle reunissait des savants de differentes branches qui voulaient se donner une philosophie susceptible d' unifier leurs vues. Echo aux (...) reves d'unite de la science, cette philosophie en exprime aussi le renversement complet. Veritable Discours de la methode, mais d'esprit anti-cartesien, elle entend reconstruire la science de maniere a evincer la speculation metaphysique, le but etant qu'aucun enonce ne soit laisse a l'exterieur de cette grande architecture de symboles s'il etait pourvu de sens. La declaration de guerre contre la metaphysique est un geste de militance a une epoque riche en differents manifestes. Incomprehensible en dehors du contexte socio-politique viennois de ces annees, elle porte aussi un programme de reconstruction rationnelle qui, au dela du langage de la science, devait toucher tous les secteurs de la vie. Plus complexe qu'il n'y parait, ce programme contient, en reference a Wittgenstein, son prestigieux representant a cote d'Einstein et Russell, une mise en garde contre differentes formes d'irrationalismes, qui est d'actualite. Plus précisément, il s’agit d’une critique de l’ « architectonique” de la différenciation discussionnelle, menée à propos du rapport entre principe de discussion, principe moral, principe du droit et principe démocratique. Mon point de vue résulte de la position de l’éthique de la discussion comme discipline de base de la philosophie pratique, dans la perspective d’une fondation pragmatique-transcendantale ultime. Une « architectonique » alternative (...) de la différenciation discussionnelle du point de vue de l’éthique de la discussion est proposée.The following essay takes a critical stand to Habermas’ book Faktizität und Geltung of 1992 ; more precisely expressed : my stance is critical with regard to Habermas’ new « architectonics » of discourse-differentiation concerning the relationship between « discourse-principle », « principle of morality », « principle of law », and « principle of democracy ». My critique represents the view of a « transcendentalpragmatic ultimate foundation of discourse-ethics » . An alternative « architectonics » of discourse differentiation in the sense of discourse-ethics is suggested. (shrink)

Le besoin d'une ethique globale a son origine dans la situation humaine comme telle (l'hominisation nous a en effet eloignes de la securite naturelle des instincts animaux et nous a jetes dans l'etat de liberte) et specialement dans la situation presente de l'humanite. Celle-ci se caracterise par un accroissement extreme des defis externes a notre co-responsabilite. Ces defis sont poses par les resultats de la science naturelle, de la technologie, de la politique et de l'economie tandis que semblent bien marquer (...) les ressources internes de la raison ethique. Le present livre tente de montrer que l'approche pragmatico-transcendantale de l'ethique de la discussion est en mesure de reconstruire la genese de cette situation et de fournir une reponse rationnelle tant aux defis externes qu'aux defis internes auxquels une ethique globale est confrontee.
