In a recent issue of Nature Communications Ukleja and co‐workers reported a cryo‐EM 3D reconstruction of the Ccr4‐Not complex from Schizosaccharomyces pombe with an immunolocalization of the different subunits. The newly gained architectural knowledge provides cues to apprehend the functional diversity of this major eukaryotic regulator. Indeed, in the cytoplasm alone, Ccr4‐Not regulates translational repression, decapping and deadenylation, and the Not module additionally plays a positive role in translation. The spatial distribution of the subunits within the structure is compatible with (...) a model proposing that the Ccr4‐Not complex interacts with the 5′ and 3′ ends of target mRNAs, allowing different functional modules of the complex to act at different stages of the translation process, possibly within a circular constellation of the mRNA. This work opens new avenues, and reveals important gaps in our understanding regarding structure and mode of function of the Ccr4‐Not complex that need to be addressed in the future. (shrink)

The only RNA molecules known to be branched are circular structures with tails known as lariats that arise during nuclear pre‐mRNA splicing. Lariats accumulate within a large multicomponent particle called a spliceosome that forms upon the addition of unspliced mRNA to nuclear extracts. Recently an RNA molecule has been observed to catalyze branch formation. In this case a single intron of a yeast mitochondrial pre‐mRNA participates in a self‐splicing reaction that results in the accumulation of branched lariats (...) that are processed to correctly spliced exons.An enzyme highly specific for branch removal found in the same extracts that form branches during pre‐mRNA splicing can debranch RNA lariats to their linear forms without loss of nucleotides.The chemical synthesis of branched RNA has recently been achieved. High yields of sequence‐specific oligonucleotides are now available for the analysis of RNA splicing by techniques dependent on branch‐site recognition. (shrink)

Research on epistemic circularity has focused more on the problem of whether circular arguments are able to justify their conclusions rather than on the nature of circularity itself, and has kept a sharp delimitation from other types of circularities (logical, linguistic, and of definition). In this paper, I aim to move the focus toward the general concept of circularity, which I relate to the concept of ‘epistemic worry’. Through a brief overview on the known types of circularities, I show that (...) circularity has multiple natures (not only that associated with its type), and the linguistic aspect is essential in qualifying a construct as circular, which in turn raises the problem of genuineness of a circularity. Within an extensional approach, I suggest generalizing the meaning of the key epistemological terms referred to in the classical definition of epistemic circularity to also cover additional associated concepts specific to other theoretical disciplines beyond epistemology. The proposed concepts and ideas are reflected in a structural unificatory account of circularity in terms of epistemic determination, which raises a challenge to the relational accounts of determination in regard to the transitivity of the determination relation. Nuclear pre‐mRNAs must be precisely processed to give rise to mature cytoplasmic mRNAs. This maturation process, known as splicing, involves excision of intron sequences and ligation of the exon sequences. One of the major problems in understanding this process is how splice sites, the sequences which form the boundaries between introns and exons, can be accurately selected. A number of studies have defined conserved sequences within introns which were later shown to interact with small nuclear ribonucleoproteins (snRNPs). However, due to (...) the simplicity of these conserved sequences it has become clear that other elements must be involved and a number of studies have indicated the importance of secondary structures within pre‐mRNAs. Using various examples, we shall show that such structures can help to specify splice sites by modifying physical distances within introns or by being involved in the definition of exons and, lastly, that they can be part of the regulation of alternative splicing. The genetic code is a set of instructions that determine how the information in our genetic material is translated into amino acids. In general, it is universal for all organisms, from viruses and bacteria to humans. However, in the last few decades, exceptions to this rule have been identified both in pro‐ and eukaryotes. In this review, we discuss the 16 described alternative eukaryotic nuclear genetic codes and observe theories of their appearance in evolution. We consider possible molecular mechanisms that (...) allow codon reassignment. Most reassignments in nuclear genetic codes are observed for stop codons. Moreover, in several organisms, stop codons can simultaneously encode amino acids and serve as termination signals. In this case, the meaning of the codon is determined by the additional factors besides the triplets. A comprehensive review of various non‐standard coding events in the nuclear genomes provides a new insight into the translation mechanism in eukaryotes. (shrink)

There have long been calls from industry for guidance in implementing strategies for sustainable development. The Circular Economy represents the most recent attempt to conceptualize the integration of economic activity and environmental wellbeing in a sustainable way. This set of ideas has been adopted by China as the basis of their economic development, escalating the concept in minds of western policymakers and NGOs. This paper traces the conceptualisations and origins of the Circular Economy, tracing its meanings, and exploring its antecedents (...) in economics and ecology, and discusses how the Circular Economy has been operationalized in business and policy. The paper finds that while the Circular Economy places emphasis on the redesign of processes and cycling of materials, which may contribute to more sustainable business models, it also encapsulates tensions and limitations. These include an absence of the social dimension inherent in sustainable development that limits its ethical dimensions, and some unintended consequences. This leads us to propose a revised definition of the Circular Economy as “an economic model wherein planning, resourcing, procurement, production and reprocessing are designed and managed, as both process and output, to maximize ecosystem functioning and human well-being”. Neuronal signalling involves multiple neuropeptides that are diverse in structure and function. Complex patterns of tissue‐specific expression arise from alternate RNA splicing of neuropeptide‐encoding gene transcripts. The pattern of expression and its role in cell signalling is diffecult to study at the level of single neurons in the complex vertebrate brain. However, in the model molluscan system, Lymnaea, it is possible to show that alternate mRNA expression of the FMRFamide gene is specific to single identified neurons. Two different transcripts (...) are expressed in a mutually exclusive manner in different neurons. Post‐translational processing of the two precursor proteins leads to completely distinct sets of neuropeptide transmitters. The function of these transmitter cocktails, resulting from alternate mRNA splicing, was studied physiologically in identified neurons forming part of a behaviourally important network regulating heartbeat. Circular economy thinking has become the subject of academic enquiry across several disciplines recently. Yet whilst its technical and business angles are more widely discussed, its philosophical underpinnings and socio-economic implications are insufficiently investigated. In this article, we aim to contribute to their understanding by uncovering the circular economy role in shaping a new vision, highlighting the social and economic dimensions of future imaginaries and the mechanisms that can enable them to bring about change in the social context. We believe (...) that defining the vision that the circular economy is contributing to shape is key to explain its conceptual framework and activities. Drawing on the concept of fictional expectations, we uncover one of the plausible social dimensions inherent to the circular economy thinking thereby opening up a new perspective on the current debate in the circular economy literature wherein authors, by contrast, are emphasising the lack of an explicit social dimension. Fictional expectations are introduced to refer to those imaginaries of the future that can catalyse social action in the present and counteract societal addictions, in which modern society seems to be trapped. We show how a circular economy inspired vision can be instrumental to the emergence of a fictional expectation that can provide therapies to the current societal addiction of wasteful production and consumption systems. This philosophical background allows us to provide, in conclusion, a new conceptualisation of the circular economy as a cognitive framework instrumental to the emergence of a future imaginary. The thesis of this paper is that philosophers are often too hasty in rejecting justifications because the argument that yields the justification is circular. Circularity is distinguished from vicious circularity and several examples are examined in which a proposed justification is circular in a precise sense, but not viciously circular. These include an observational procedure which could yield a velocity in excess of the velocity of light even though the impossibility of such velocities is assumed at a key step in (...) analyzing the data, and an argument that uses a specific argument form to show that that form is invalid. Is perception cognitively penetrable, and what are the epistemological consequences if it is? I address the latter of these two questions, partly by reference to recent work by Athanassios Raftopoulos and Susanna Seigel. Against the usual, circularity, readings of cognitive penetrability, I argue that cognitive penetration can be epistemically virtuous, when---and only when---it increases the reliability of perception.

Disagreement and debunking arguments threaten religious belief. In this paper, I draw attention to two types of propositions and show how they reveal new ways to respond to debunking arguments and disagreement. The first type of proposition is the epistemically self-promoting proposition, which, when justifiedly believed, gives one a reason to think that one reliably believes it. Such a proposition plays a key role in my argument that some religious believers can permissibly wield an epistemically circular argument in response to (...) certain debunking arguments. The second type of proposition is the epistemically others-demoting proposition, which, when justifiedly believed, gives one a reason to think that others are unreliable with respect to it. This paper explores what happens if we construe evidentialism as a thesis about the metaphysical grounds of justification. According to grounding evidentialism, facts about what a subject is justified in believing are grounded in facts about that subject’s evidence. At first blush, grounding evidentialism appears to enjoy advantages over a more traditional construal of evidentialism as a piece of conceptual analysis. However, appearances are deceiving. I argue that grounding evidentialists are unable to provide a satisfactory story about what grounds the (...) evidential facts, and that this provides good reason to reject grounding evidentialism. This paper criticizes the circularity reading of Frege's argument for the indefinability of truth. According to this reading, Frege is appealing to a sort of circularity in the argument. I argue that the circularity reading is interpretatively incorrect, or makes Frege's argument a non‐starter.

Six years ago, the Singer lab published a landmark paper which described how individual mRNA particles cross the nuclear pore complex in mammalian tissue culture cells. This involved the simultaneous imaging of mRNAs, each labeled by a large number of tethered fluorescent proteins and fluorescently tagged nuclear pore components. Now two groups have applied this technique to the budding yeast Saccharomyces cerevisiae. Their results indicate that in the course of nuclear export, mRNAs likely engage complexes that are present on (...) either side of the pore and that these interactions are modulated by proteins present in the messenger ribonucleoprotein (mRNP) complex. These findings lend support to the notion that just before and/or after the completion of nuclear export, mRNPs undergo one or more maturation steps that prepare the packaged mRNAs for translation. These results represent new and exciting insights into the mechanism of mRNA nuclear export. (shrink)

Messenger RNA is a flexible tool box that plays a key role in the dynamic regulation of gene expression. RNA modifications variegate the message conveyed by the mRNA. Similar to DNA and histone modifications, mRNA modifications are reversible and play a key role in the regulation of molecular events. Our understanding about the landscape of RNA modifications is still rudimentary in contrast to DNA and histone modifications. The major obstacle has been the lack of sensitive detection methods since (...) they are non-editing events. However, with the advent of next-generation sequencing techniques, RNA modifications are being identified precisely at single nucleotide resolution. In recent years, methylation at the N6 position of adenine has gained the attention of RNA biologists. The m6A modification has a set of writers, erasers, and readers. Here, we provide a summary of interesting facts, conflicting findings, and recent advances in the technical and functional aspects of the m6A epitranscriptome. The recent outburst of studies has brought the key role of mRNA N6-methyladenosine modification in post-transcriptional processing steps into the limelight. This review summarizes the current progress and the future prospective of m6A methylome. We addressed the debatable function of m6A modification and related modifiers in tumorigenesis and tumor progression. (shrink)

In a context of growing environmental challenges, circular economy (CE) business models appear necessary for business to contribute positively to the ecological transition. While platform business models have been identified as a new and promising model in CE, we still lack a fine-grained understanding of the critical capabilities involved in developing and scaling them. To fill this gap, we build on a single case study of Phenix, a French-based fast-growing start-up in the food industry, tackling the issue of food waste. (...) We identify three core managerial capabilities involved in the successful scaling of Phenix's business model. In addition to ecosystem orchestration capabilities, we identify two types of ambidextrous capabilities – forms of ambidexterity operating both at the organizational and at the institutional level. Our analysis highlights the importance of considering these capabilities in a bundle as they collectively contribute to the performance and scaling potential of the business model. We also call for an increased consideration of the role of institutional factors in shaping opportunities to design and scale profitable business models. (shrink)

Mainstream epistemology has typically taken for granted a traditional picture of the metaphysics of mind, according to which cognitive processes (e.g. memory storage and retrieval) play out entirely within the bounds of the skull and skin. But this simple ‘intracranial’ picture is falling in- creasingly out of step with contemporary thinking in the philosophy of mind and cognitive science. Likewise, though, proponents of active exter- nalist approaches to the mind—e.g. the hypothesis of extended cognitition (HEC)—have proceeded by and large without (...) asking what epistemological ramifications should arise once cognition is understood as criss-crossing the bounds of brain and world. This paper aims to motivate a puzzle that arises only once these two strands of thinking are brought in contact with one another. In particular, we want to first highlight a kind of con- dition of epistemological adequacy that should be accepted by proponents of extended cognition; once this condition is motivated, the remainder of the paper demonstrates how attempts to satisfy this condition seem to inevitably devolve into a novel kind of epistemic circularity. At the end of the day, proponents of extended cognition have a novel epistemological puzzle on their hands. (shrink)

Although this is often thought to be vicious, externalist theories generally don't rule it out. For some philosophers, this is a reason to reject externalism. However, Michael Bergmann defends externalism by drawing on the tradition of common sense in two ways. First, he concedes that epistemically circular beliefs cannot answer a subject's doubts about her cognitive faculties. But, he argues, subjects (...) don't have such doubts, so epistemically circular beliefs are rarely called upon to play this role. Second, following Thomas Reid, Bergmann argues that we have noninferential, though epistemically circular, knowledge that our faculties are reliable. I argue, however, that Bergmann's view is undermined by doubts a subject should have and that there is no plausible explanation for how we can have noninferential knowledge that our faculties are reliable. Epistemically circular arguments have been receiving quite a bit of attention in the literature for the past decade or so. Often the goal is to determine whether reliabilists (or other foundationalists) are committed to the legitimacy of epistemically circular arguments. It is often assumed that epistemic circularity is objectionable, though sometimes reliabilists accept that their position entails the legitimacy of some epistemically circular arguments, and then go on to affirm that such arguments really are good ones. My goal in this (...) paper is to argue against the legitimacy of epistemically circular arguments. My strategy is to give a direct argument against the legitimacy of epistemically circular arguments, which rests on a principle of basis-relative safety, and then to argue that reliabilists do not have the resources to resist the argument. I argue that even if the premises of an epistemically circular argument enjoy reliabilist justification, the argument does not transmit that justification to its conclusion. The main goal of my argument is to show that epistemic circularity is always a bad thing, but it also has the positive consequence that reliabilists are freed from an awkward commitment to the legitimacy of some intuitively bad arguments. The aim of this paper is to provide a model that allows the representation and analysis of circularity in ethotic structures, i.e. in communication structures related to the speaker’s character and in particular, his credibility. The paper studies three types of cycles: in self-referential sentences, embedded testimony and ethotic begging the question. It is shown that standard models allow the reconstruction of the circularities only if those circular utterances are interpreted as ethotic arguments. Their alternative, assertive interpretation requires enriching the (...) existing models with a purely ethotic component related to the credibility of the performer of any (not necessarily argumentative) speech act. In this paper, I propose a critique of the naturalist version of the Desire-Based Reasons Model. I first set the scene by spelling out the connection between naturalism and the Model. After this, I introduce Christine Korsgaard’s circularity argument against what she calls the instrumental principle. Since Korsgaard’s targets, officially, were non-naturalist advocates of the principle, I show why and how the circularity charge can be extended to cover the naturalist Model. Once this is done, I go on to investigate (...) in some detail the different ways of responding to the circularity challenge. I argue that none of these responses succeed, at least not without serious costs to their advocates. I then end the paper with a brief summary and some concluding remarks. (shrink)

One of the gravest charges that has been brought against Hume’s essay “Of the Standard of Taste” is that of circularity. Hume is accused of defining good art in terms of “true judges,” and of defining true judges in terms of their ability to judge good art. First, I argue that Hume avoids circularity since he offers a way of identifying good art that is logically independent of the verdict of true judges. Second, I argue that this clarifies an enduring (...) puzzle in the scholarship on Hume’s essay: why he appears to offer not one but two standards of taste. Hume’s standard does not consist of general rules; however, Hume needs general rules to establish that some individuals’ tastes are more “delicate” than others’. (shrink)

Eukaryotic gene expression is extensively controlled at the level of mRNA stability and the mechanisms underlying this regulation are markedly different from their archaeal and bacterial counterparts. We propose that two such mechanisms, nonsense‐mediated decay (NMD) and motif‐specific transcript destabilization by CCCH‐type zinc finger RNA‐binding proteins, originated as a part of cellular defense against RNA pathogens. These branches of the mRNA turnover pathway might have been used by primeval eukaryotes alongside RNA interference to distinguish their own messages from (...) those of RNA viruses and retrotransposable elements. We further hypothesize that the subsequent advent of “professional” innate and adaptive immunity systems allowed NMD and the motif‐triggered mechanisms to be efficiently repurposed for regulation of endogenous cellular transcripts. This scenario explains the rapid emergence of archetypical mRNA destabilization pathways in eukaryotes and argues that other aspects of post‐transcriptional gene regulation in this lineage might have been derived through a similar exaptation route. -/- . Good informal arguments offer justification for their conclusions. They go wrong if the justifications double back, rendering the arguments circular. Circularity, however, is not necessarily a single property of an argument, but may depend on (a) whether the argument repeats an earlier claim, (b) whether the repetition occurs within the same line of justification, and (c) whether the claim is properly grounded in agreed‐upon information. The experiments reported here examine whether people take these factors into account in their judgments of (...) whether arguments are circular and whether they are reasonable. The results suggest that direct judgments of circularity depend heavily on repetition and structural role of claims, but only minimally on grounding. Judgments of reasonableness take repetition and grounding into account, but are relatively insensitive to structural role. (shrink)

This paper discusses some of the ways in which circular definitions and circular explanations entail or fail to entail infinite regresses. And since not all infinite regresses are vicious, a few criteria of viciousness are examined in order to determine when the entailment of a regress refutes a circular definition or a circular explanation.

The Fourth Industrial Revolution based on digitalization, the development of AI, robotics, big data, and increasing automation is dredging up older debates on the end of human work. This article contributes to this debate arguing that these changing circumstances represent an opportunity to advance a renewed consideration of human work. By emphasizing its most distinctively human dimensions, including gratuitousness, relationality, and meaningfulness, we propose the articulation of a social model that recognizes relational goods as a specific contribution of human work (...) in an attempt to overcome the state and the market's monopoly of public life built around the dichotomy between the private and the public. Such a social model is based on the concept of circular subsidiarity, which represents a departure from traditional notions of subsidiarity by promoting a dynamic and reciprocal interplay between three essential societal spheres: the state, the market, and organized civil society, all of which have their role to fulfill by providing security, efficiency, and relational goods. Ultimately, this article suggests that circular subsidiarity can lead to a more inclusive and equitable social model by acknowledging how relational goods not only humanize civil society but also sustain the functioning of both the market and the state. HEGEL says that "when enquiry is made as to the kind of predicate belonging to [a] subject, the act of judgement necessarily implies an underlying concept [Begriff]; but this concept is expressed only by the predicate." According to this, some concept of the subject must precede predication. This circularity can be formulated as follows: If the statement is the "factory" in which concepts are produced, how is it that the concepts precede the statement and are not merely produced within it (...) in their function as subjects? And, on the other hand, if the subject is not, prior to predication, a concept--if it has by itself no meaning--how can a predicate be matched to it? There would, in fact, be nothing to match. We are caught in a circular thesis that requires for its elucidation a fresh analysis of the relationship between concept and statement. (shrink)

Despite a library full of literature on miRNA biology, core issues relating to miRNA target detection, biological effect, and mode of action remain controversial. This essay proposes that the predominant mechanism of direct miRNA action is translational inhibition, whereas the bulk of miRNA effects are mRNA based. It explores several issues confounding miRNA target detection, and discusses their impact on the dominance of “miRNA seed” dogma and the exploration of non‐canonical binding sites. Finally, it makes comparisons between miRNA target (...) prediction and transcription factor binding prediction, and questions the value of characterizing miRNA binding sites based on which miRNA nucleotides are paired with an mRNA. The direct use of a physical law for the purpose of measurement creates a problem of circularity: the law needs to be empirically tested in order to ensure the reliability of measurement, but the testing requires that we already know the value of the quantity to be measured. This problem is discussed through some detailed examples of energy measurements in quantum physics; three major methods are analyzed in their interrelation, with a focus on the method of “material retardation.” It seems (...) that the only reasonable solution is to establish the law needed for the measurement by relying on an alternate method of measurement. This solution is also circular, since it amounts to letting different measurement methods justify each other. However, this circularity can be fruitful for the process of concept building; meaning can be created in a web of interconnected measurement methods. (shrink)

In recent years, researchers and practitioners have increasingly paid attention to food waste, which is seen as highly unethical given its negative environmental and societal implications. Waste recovery is dependent on the creation of connections along the supply chain, so that actors with goods at risk of becoming waste can transfer them to those who may be able to use them as inputs or for their own consumption. Such waste recovery is, however, often hampered by what we call ‘circularity holes’, (...) i.e., missing linkages between waste generators and potential receivers. A new type of actor, the digital platform organization, has recently taken on a brokerage function to bridge circularity holes, particularly in the food supply chain. Yet, extant literature has overlooked this novel type of brokerage that exploits digital technology for the transfer and recovery of discarded resources between supply chain actors. Our study investigates this actor, conceptualized as a ‘circularity broker’, and thus unites network research and circular supply chain research. Focusing on the food supply chain, we adopt an interpretive inductive theory-building approach to uncover how platform organizations foster the recovery of waste by bridging circularity holes. We identify and explicate six brokerage roles, i.e., connecting, informing, protecting, mobilizing, integrating and measuring, and discuss them in relation to extant literature, highlighting novelties compared to earlier studies. The final section reflects on contributions, implications, limitations and areas for further research. (shrink)

Sometimes we get what seem to be good reasons for believing that we’ve misevaluated our evidence for a proposition P. In those cases, can we use our evidence for P itself to show that we haven’t misevaluated our evidence for P? I show why doing so appears to employ viciously circular reasoning. However, I then argue that this appearance is illusory in certain cases and that we sometimes can legitimately reason in that way. This claim sheds new light on the (...) nature of epistemic undermining and epistemic circularity. In addition, it has implications for the current debate about the epistemic significance of disagreement. An important and influential position in that debate says that disagreement with others dramatically undermines our justification for a wide range of our opinions (e.g., political, religious, moral, economic, and philosophical opinions). My view on undermining and circularity implies that this position on disagreement rests on a mistake. (shrink)

When one depends on a belief source in sustaining a belief that that very belief source is trustworthy, then that belief is an epistemically circular belief. A number of philosophers have objected to externalism in epistemology on the grounds that it commits one to thinking EC-beliefs can be justified, something they view as an unhappy consequence for externalism. In my 2004, I defend externalism against this sort of charge by explaining why this consequence needn’t be an unhappy one. In the (...) course of doing so, I appeal to what Thomas Reid calls ‘common sense’—a faculty or belief source by which we know noninferentially such things as that our faculties are trustworthy. In his 2006, Baron Reed raises what he takes to be serious objections to what I say about both epistemic circularity and common sense. In what follows, I’ll respond to his objections, explaining why I side with Reid against Reed. (shrink)

Sosa's work on epistemic circularity has significance beyond his own brand of virtue epistemology, with its characteristic distinction between animal and reflective knowledge. On the contrary, it demonstrates the necessity of embracing foundationalism and externalism in epistemology, while at the same time answering various charges (some perennial) against epistemology in general. This paper distinguishes six kinds of epistemic circularity that are discussed in Sosa's work: two virtuous, two vicious, and two benign. This dissertation is a study of some recent theories of truth. The theories fall into three groups: The Revision Theories, the context-sensitive theories, and the "Chrysippian theories". ;The "Chrysippian theories" are based on the intuition that pathologicalities arising from the concept of truth can be recognized and acknowledged with the concept of truth itself. Thus, from the pathologicality of the Liar, for example, we can conclude that the Liar is not true. This leads to immediate difficulties since the Liar claims (...) just that--that it is not true--and it would thus seem to be true. The Chrysippian theories are designed to avoid these difficulties while preserving the motivating intuition. Haim Gaifman's Pointer Semantics, and Brian Skyrms' Intensional Fixed-Point Theory of Truth are examples of such theories. They are exposited and criticized. ;The context-sensitive theories dealt with are Tyler Burge's Indexical Theory of Truth and the theory of Jon Barwise and John Etchemendy. These theories claim that an ambiguity due to a contextual parameter is the reason for the difficulties we encounter. Burge offers a theory of Tarskian levels coupled with a pragmatic theory that explains the assignment of levels to occurrences of 'true'. Barwise and Etchemendy make explicit a situational parameter in their account of propositions. They consider the ambiguity with respect to the situation a propositions is about a main source of the paradoxes. ;Finally, the Revision Theories of Truth of Anil Gupta and Nuel Belnap and two variations of this theory are considered. One is due to Aladdin Yaqub, the other, the "fully-varied theory", is due to Gupta and Belnap but remains largely unexplored. Revision theories are put forward as the most promising account of the difficulties. They are based on a definitional reading of all Tarski biconditionals. The circularity implied by such a reading is handled by Gupta's and Belnap's general theory of definitions. ;Yaqub's objections raised against the standard theory are examined, and his proposed solution is shown to be inadequate. The fully-varied Revision Theory is shown to provide a better solution. (shrink)

Upshot: The target article defends the fundamental role of circularity for systems sciences and the necessity to develop a conceptual and methodological approach to it. The concept of circularity, however, is multifarious, and two of the main challenges in this respect are to provide distinctions between different forms of circularities and explore in detail the roles they play in organizations. In his recent book, Reasons and Persons, Derek Parfit propounds a version of the psychological criterion of personal identity.1 According to the variant he adopts, the numerical identity through time of persons consists in non-branching psychological continuity no matter how it is caused. One traditional objection to a view of this sort is that it is circular, since psychological continuity presupposes personal identity. Although Parfit frequently denies the importance of personal identity, he considers his own psychological account of identity important (...) enough to attempt to defend it against the charge of circularity. The aim of this paper is to argue that Parfit's attempted defence is unsuccessful because it ultimately rests on an analysis of the unity of consciousness that is itself either circular or otherwise inadequate. In this article, we address why our management of the economy, community and business has led to global warming and we discuss the importance of worldviews, ontology, epistemology and axiology in the search for alternative paths of development. We do this by focusing on the concept of Circular Economy. Circular Economy is often presented as a solution to the problems of a globalized economy in the form of over-exploitation of resources, climate change and pollution of the environment. Within the mainstream (...) economics paradigm, the aim is how to increase the effectiveness of resource extraction and utilization in order to maintain growth. We contrast this with the paradigm of Ecological Economics where the goal of the economy is to achieve and secure a higher quality of life. Circular Economy often side steps the deep ontological and epistemological questions we need to answer if we are to address the complex and interrelated environmental, economic and social problems we face today. This can be a limiting factor in the search for creative and long-term solutions. However, within the Circular Economy discourse, we find strands that critically engage with the foundations of mainstream Economics and so, innovative solutions for the society of the future are a possibility. We argue that to bring about a lasting solution to the interconnected social, economic and environmental problems, Circular Economy must engage with the ontological, epistemological and axiological foundations of mainstream economics. To further this argument, we apply Imre Lakatos’ ‘research program’ model to philosophy of science as our framework of analysis and draw lessons for business and communities. (shrink)

In this paper I defend epistemic circularity by arguing that the “No Self-Support” principle (NSS) is false. This principle, ultimately due to Fumerton ( 1995 ), states that one cannot acquire a justified belief in the reliability of a source of belief by trusting that very source. I argue that NSS has the skeptical consequence that the trustworthiness of all of our sources ultimately depends upon the trustworthiness of certain fundamental sources – sources that we cannot justifiably believe to be (...) reliable. This is a problem, I claim, because if the trustworthiness of all of our sources depends upon sources that we should not believe to be reliable, then a reflective individual should not trust any of his sources at all. The hidden cost of rejecting epistemic circularity is thus the unacceptable skeptical thesis that reflective individuals like you and I have no justified beliefs whatsoever. (shrink)

The neuromuscular junction has been used for several decades as an excellent model system to examine the cellular and molecular events involved in the formation and maintenance of a differentiated chemical synapse. In this context, several laboratories have focused their efforts over the last 15 years on the important contribution of transcriptional mechanisms to the regulation of the development and plasticity of the postsynaptic apparatus in muscle fibers. Converging lines of evidence now indicate that post‐transcriptional events, operating at the level (...) of mRNA stability and targeting, are likely to also play key roles at the neuromuscular junction. Here, we present the recent findings highlighting the role of these additional molecular events and extend our review to include data showing that post‐transcriptional events are also important in the control of the expression of genes encoding synaptic proteins in muscle cells placed under different conditions. Finally, we discuss the possibility that mis‐regulation of post‐transcriptional events can occur in certain neuromuscular diseases and cause abnormalities of the neuromuscular junction.
