Understanding the mechanisms and the time and spatial evolution of penumbra following an ischemic stroke is crucially important for developing therapeutics aimed at preventing this area from evolving towards infarction. To help in integrating the available data, we decided to build a formal model. We first collected and categorised the major available evidence from animal models and human observations and summarized this knowledge in a flow-chart with the potential key components of an evolving stroke. Components were grouped in ten sub-models (...) that could be modelled and tested independently: the sub-models of tissue reactions, ionic movements, oedema development, glutamate excitotoxicity, spreading depression, NO synthesis, inflammation, necrosis, apoptosis, and reperfusion. Then, we figured out markers, identified mediators and chose the level of complexity to model these sub-models. We first applied this integrative approach to build a model based on cytotoxic oedema development following a stroke. Although this model includes only three sub-models and would need to integrate more mechanisms in each of these sub-models, the characteristics and the time and spatial evolution of penumbra obtained by simulation are qualitatively and, to some extent, quantitatively consistent with those observed using medical imaging after a permanent occlusion or after an occlusion followed by a reperfusion. In publications in 1914 and 1918, Einstein claimed that his new theory of gravity in some sense relativizes the rotation of a body with respect to the distant stars and the acceleration of the traveler with respect to the stay-at-home in the twin paradox. What he showed was that phenomena seen as inertial effects in a space-time coordinate system in which the non-accelerating body is at rest can be seen as a combination of inertial and gravitational effects in a space-time (...) coordinate system in which the accelerating body is at rest. Two different relativity principles play a role in these accounts: the relativity of non-uniform motion, in the weak sense that the laws of physics are the same in the two space-time coordinate systems involved; what Einstein in 1920 called the relativity of the gravitational field, the notion that there is a unified inertio-gravitational field that splits differently into inertial and gravitational components in different coordinate systems. I provide a detailed reconstruction of Einstein's rather sketchy accounts of the twins and the bucket and examine the role of these two relativity principles. I argue that we can hold on to but that is either false or trivial. (shrink)

Cet article propose les éléments d’un cadre théorique permettant d’analyser des situations sociales comportant une part d’imprévisibilité. S’intéresser à des ruptures, des changements soudains, c’est en partie revenir sur de vieux tabous de la sociologie et plus généralement des sciences sociales : la contingence, l’événement, l’imprévisible. Une solution possible réside dans la définition précise de ce qui est considéré comme imprévisible et dans la prise en compte de différents niveaux de temporalité, à condition d’accepter l’idée que les temps « courts (...) » peuvent parfois influer sur les temps « longs ».In this article I propose the elements of a theoretical framework that makes it possible to analyse social situations with a part of unpredictability. Being interested in ruptures, unexpected changes, is for a part going back to some old taboos of sociology, and more generally, of social sciences : contingency, events, unpredictability. One possible solution consists in a precise definition of what is considered as unpredictable and in taking into account different levels of temporality, providing to accept the idea that « short » time can sometimes have an influence on « long » time. Le débat sur l'interprétation de la mécanique quantique est, aujourd'hui, sensiblement différent de ce qu'il était dans la période de «fondation» de cette théorie. Cette modification tient à deux causes : l'ancrage des conceptions quantiques dans la pensée des physiciens, favorisé par l'utilisation systématique et fructueuse de la théorie quantique en physique atomique et subatomique, d'une part et, d'autre part, les développements théoriques et expérimentaux survenus au cours des vingt dernières années, qui ont amené à considérer comme des faits physiques (...) des énoncés qui paraissaient naguère relever davantage de l'interprétation, avec liberté d'options, voire de la spéculation. Quelle est la signification des énoncés et des concepts théoriques de la physique quantique ? et comment celle-ci s'est-elle modifiée ? Qu'entend-on par «interprétation» ? Telles sont les questions qui servent de guide à ce travail. Nous nous proposons de clarifier ce qui, dans les questions de signification, relève respectivement de la physique (essentiellement les contenus conceptuels et théoriques), et de perspectives philosophiques. Annoncé à grand fracas, le décryptage do génome humain devait nous révéler le secret ultime de la vie et ouvrir la voie à de nouvelles thérapies miracles. Espoirs déçus : à l'ère de la post-génomique, les secrets du vivant sont maintenant recherchés dans les théories de la complexité, dans la convergence des efforts des biologistes, des physiciens et des mathématiciens. Comment comprendre la signification de cette succession rapide d'objectifs apparemment différents, de cette alternance d'espoirs et de désillusions? Dans ce livre (...) novateur, Michel Morange propose une clé pour rendre compte de ces difficultés, et de beaucoup d'autres analogues touchant toutes les branches de la biologie. Les annonces sensationnelles reflètent l'espoir toujours déçu qu'une explication unique pourrait suffire. Or les faits biologiques - comme ceux relevant de bien d'autres disciplines scientifiques -ne peuvent être expliqués par un principe d'intelligibilité unique. Exemples à l'appui et de façon très pédagogique, Michel Morange montre pourquoi des explications différentes doivent être articulées pour décrire le fonctionnement des macromolécules aussi bien que l'évolution humaine ou le développement des cancers. Admettre une idée aussi simple n'est pas évident, car tout scientifique a été formé à privilégier un principe d'intelligibilité particulier. L'articulation entre explications différentes est pourtant indispensable pour le progrès des connaissances ; elle est aussi une exigence éthique ; elle est requise pour que la science conserve sa place dans nos sociétés. Dans la Grèce ancienne comme dans l'Antiquité romaine, plusieurs mots sont utilisés pour désigner le peuple, mais leur signification, positive ou négative, peut varier en fonction du point de vue de l'utilisateur. Comme la nôtre, la culture gréco-romaine distingue le peuple comme masse capricieuse et le peuple en tant qu'entité souveraine à laquelle se réfèrent les orateurs et les hommes politiques. Dans tous les cas, il faut se garder d'extrapoler à partir d'un seul texte. La tendance à déprécier le peuple (...) est une réalité à toutes les époques comme la polarisation entre riches et pauvres , mais dans la Rome impériale, cette dernière prend des proportions inédites. Si, depuis l'époque de César, il n'a jamais été mauvais de se réclamer du peuple contre l'aristocratie, il y a lieu de s'interroger sur le pouvoir réel dont jouissait le populus.In Ancient Greece and Rome, several words were used to refer to the people, but their signification, positive or negative, might vary with the point of view of the user. As in our own culture, there was a distinction in the Greco-Roman culture between the capricious mass and the people as a sovereign entity to which the orators and the politicians always referred. In any case, care must be taken not to extrapolate from a single text. The tendency to depreciate the people is a reality during all the periods of polarization between the rich and the poor , but in Imperial Rome this trend grew unusually. While it has always been good to claim one’s identification and side with the people against the aristocracy since Caesar’s time, one has every reason to question the real power of the populus. Quel que soit l'angle sous laquelle on l'aborde, l'innovation apparaît comme une activité collective qui implique des collaborations, des échanges, des circulations de toutes sortes. C'est aussi une activité située, puisque l'on dispose à présent d'un ensemble de résultats empiriques qui convergent sur l'existence d'« effets de proximité spatiale », c'est-à-dire sur le fait que ces collaborations, échanges et circulations sont plus fréquentes lorsque les partenaires sont proches dans l'espace géographique, en particulier lorsqu'ils sont situés dans la même métropole ou (...) le même bassin d'emploi. Diverses explications de ces effets de proximité spatiale ont été proposées, qui mettent en jeu des conceptions variables des rapports entre la sphère économique et la vie sociale d'ensemble, des contraintes qui s'exercent sur les collaborations et les échanges, de la dynamique même de l'innovation. Ainsi dans le cas des relations science-industrie par exemple, de nombreux auteurs attribuent les effets de proximité aux contraintes sur les échanges de savoirs tacites (les savoirs et savoir faire scientifiques non publiés ou non codifiables parce que trop nouveaux), qui exigeraient des échanges en face à face, et donc une proximité forte entre les entreprises et les laboratoires. Mucoidy and cytotoxicity arise from two independent modifications of the phenotype of the bacterium Pseudomonas aeruginosa that contribute to the mortality and morbidity of cystic fibrosis. We show that, even though the transcriptional regulatory networks controlling both processes are quite different from a molecular or mechanistic point of view, they may be identical from a dynamic point of view: epigenesis may in both cases be the cause of the acquisition of these new phenotypes. This was highlighted by the identity of (...) formal graphs modelling these networks. A mathematical framework based on formal methods from computer science was defined and implemented with a software environment. It allows an easy and rigorous validation and certification of these models and of the experimental methods that can be proposed to falsify or validate the underlying hypothesis. (shrink)

The conviction, due to previous failures, that bacteriology and darwinism were incompatible, has postponed the application of molecular phylogenesis to bacteria. But once introduced, this new field has led to a profound revolution of this science. A stable classification of the bacteria is at last possible; a new domain, the Archae, as distant from the Bacteria as from the Eukarya, has been discovered; noncultivable new species can be identified from the environment. It may even be possible to unravel the pathway (...) of early evolution after the common ancestor. However, controversies on this subject are already disregarded by the new paradigm. (shrink)

The mechanisms by which bacteria adapt to changes in their environment involve transcriptional regulation in which a transcriptional regulator responds to signal(s) from the environment and regulates (positively or negatively) the expression of several genes or operons. Some of these regulators exert a positive feedback on their own expression. This is a necessary (although not sufficient) condition for the occurrence of multistationarity. One biological consequence of multistationarity may be epigenetic modifications, a hypothesis unusual to microbiologists, in spite of some well-known (...) epigenetic modifications in bacteria. We propose here that the occurrence of mucoidy in the opportunistic pathogen Pseudomonas aeruginosa, which is currently attributed to mutations only, may also be an epigenetic modification. A theoretical approach using a generalised logical analysis lends credit to this hypothesis and suggests experiments to ascertain it. (shrink)

The transformations undergone by research and science in the name of the so called "knowledge economy" cover the decisions of scientific policies and the "management" of research, and also the meaning of scientific activities (devoted to innovation) and even more fundamentally the very structure of the sciences (transformed to technosciences). The science that is contributing to capitalist competitiveness (and to the current economic crisis) is not the same as that which would be able to contribute "to the conception and democratic (...) carrying out of another form of globalization and another European project". However, this is not self-evident, and it needs to be thought about since it is not simply a matter of returning to the science of the 20th century that opened the way to technoscience. (shrink)
