Increasing interest has been paid to applications of fluorescence measurements to analyze physiological mechanisms in living cells. However, few studies have taken advantage of DNA quantification by fluorometry for dynamic assessment of chromatin organization as well as cell motion during the cell cycle. This approach involves both optimal conditions for DNA staining and cell tracking methods. In this context, this report describes a stoichiometric method for nuclear DNA specific staining, using the bisbenzimidazole dye Hoechst 33342 associated with verapamil, a calcium (...) membrane channel blocker. This method makes it possible to correlate variations of nuclear DNA content with cell motion in cells that are maintained alive. Motion measurement is the second goal of this paper and it explains the snake-spline method, and the associated cell following method. (shrink)

This position paper discusses emerging behavioral, social, and economic dynamics related to the COVID-19 pandemic and puts particular emphasis on two emerging issues: First, delayed effects (or second strikes) of pandemics caused by dread risk effects are discussed whereby two factors which might influence the existence of such effects are identified, namely the accessibility of (mis-)information and the effects of policy decisions on adaptive behavior. Second, the issue of individual preparedness to hazardous events is discussed. As events such as the (...) This article is based on German and Japanese sources and shows how around 1900 European psychiatric concepts and practices embedded themselves into emerging scientific Japanese discourses. The article argues that now forgotten German–Japanese exchanges in the field of psychiatric pathology, together with the historical development of psychiatric care, were central mechanisms for the establishment of a distinctly psychiatric discourse in Japan prior to its broad institutionalization. Three discursive strategies were key: Japanese and German experts from a range of medical fields (...) reinvented a body of traditions loosely related to actual pre-modern cultural practices; they engaged in comparative evaluations of psychiatric conditions; and, through the simple but effective transformation of specific concepts and termini at the margins of European psychiatry, these experts contributed to the transfer not only of a psychiatric discourse but also affected the power relations on a national and international scale as European psychiatry permeated into new territory, namely the Japanese landscape of emerging modern scientific disciplines. BackgroundBiobanking biospecimens and consent are common practice in paediatric research. We need to explore children and young people’s (CYP) knowledge and perspectives around the use of and consent to biobanking. This will ensure meaningful informed consent can be obtained and improve current consent procedures.MethodsWe designed a survey, in co-production with CYP, collecting demographic data, views on biobanking, and consent using three scenarios: 1) prospective consent, 2) deferred consent, and 3) reconsent and assent at age of capacity. The survey was disseminated (...) via the Young Person’s Advisory Group North England (YPAGne) and participating CYP’s secondary schools. Data were analysed using a qualitative thematic approach by three independent reviewers (including CYP) to identify common themes. Data triangulation occurred independently by a fourth reviewer.ResultsOne hundred two CYP completed the survey. Most were between 16–18 years (63.7%, N = 65) and female (66.7%, N = 68). 72.3% had no prior knowledge of biobanking (N = 73).Acceptability of prospective consent for biobanking was high (91.2%, N = 93) with common themes: ‘altruism’, ‘potential benefits outweigh individual risk’, 'frugality', and ‘(in)convenience’.Deferred consent was also deemed acceptable in the large majority (84.3%, N = 86), with common themes: ‘altruism’, ‘body integrity’ and ‘sample frugality’. 76.5% preferred to reconsent when cognitively mature enough to give assent (N = 78), even if parental consent was previously in place. 79.2% wanted to be informed if their biobanked biospecimen is reused (N = 80).ConclusionProspective and deferred consent acceptability for biobanking is high among CYP in the UK. Altruism, frugality, body integrity, and privacy are the most important themes. Clear communication and justification are paramount to obtain consent. Envy and Jealousy in Classical Athens examines the sensation, expression, and literary representation of envy and jealousy in Classical Athens.

As part of a European Erasmus Plus project entitled Death Education for Palliative Psychology, this study assessed the ways in which Master’s Degree students in psychology and the creative arts therapies self-rated their confidence and interest in death education and palliative and bereavement care. In five countries (Austria, Israel, Italy, Poland, Romania), 344 students completed an online questionnaire, and 37 students were interviewed to better understand their views, interest, and confidence. The results revealed some significant differences between countries, and showed (...) that older respondents with previous experience as formal caregivers for end-of-life clients showed greater interest in obtaining practical clinical competence in these fields. A mediation analysis indicated that students’ previous care experiences and past loss experiences were related to students’ current interest in death education and palliative and bereavement care through the mediation of their sense of confidence in this field. The qualitative findings identified five shared themes: life and death, learning about death, the psychological burden, personal experience and robust training, and four key training needs. Overall, students’ interest in studying and working with terminal illness and death are rooted in internal resources, a preliminary sense of confidence, but also external requirements. (shrink)

Smell 'sensations' are among the most mysterious of conscious experiences, and have been cited in defense of the thesis that the character of perceptual experience is independent of the physical events that seem to give rise to it. Here we review the scientific literature on olfaction, and we argue that olfaction has a distinctive profile in relation to the other modalities, on four counts: in the physical nature of the stimulus, in the sensorimotor interactions that characterize its use, in the (...) structure of its intramodal distinctions and in the functional role that it plays in people's behaviour. We present two thought experiments in which we detail what would be involved in transforming sounds into smells, and also smells into colours. Through these thought-experiments, we argue that the experiential character of smell derives precisely from the structural features of olfaction, and that an embodied account of olfactory phenomenology is called for. This paper presents the development and validation of a new scale to measure consequentialism and formalism that is well grounded in prior research. The Ethical Standards of Judgement Questionnaire is validated via six studies. Measurement items are developed in the first three studies, which also confirm the need (...) to eliminate a unidimensional measure and evaluate these frameworks separately. The fourth study addresses discriminant validity and the two remaining studies provide insight into how consequentialism and formalism predict the degree to which behaviors are deemed acceptable by individuals in the context of consumer beliefs and religious beliefs. In this paper, I explore the question of how the costs of undertaking an important type of climate change mitigation should be shared amongst states seeking an environmentally effective and equitable response to global climate change. While much of the normative literature on climate mitigation has focused on burden sharing within the context of reductions in emissions of greenhouse gas, I explore the question of how the costs of protecting tropical forests in order to harness their climate mitigation potential should (...) be distributed amongst developing and developed states. In response to this question, I outline and defend a ‘beneficiary pays’ account of forestry mitigation burden sharing that requires affluent states to finance measures supporting avoided deforestation while less affluent states, within whose territory these forests tend to be located, implement these measures. The normative basis for this account, I argue, is a principle of ‘unjust enrichment’ according to which developed states must bear much of the cost of avoided deforestation for its climate mitigation potential because of the huge economic benefits their citizens have accumulated from productive activities that have contributed to climate change. This book draws together the latest work from scholars around the world using subjective well-being data to understand and compare well-being across countries and cultures. Starting from many different vantage points, the authors reached a consensus that many measures of subjective well-being, ranging from life evaluations through emotional states, based on memories and current evaluations, merit broader collection and analysis. Using data from the Gallup World Poll, the World Values Survey, and other internationally comparable surveys, the authors document wide divergences (...) among countries in all measures of subjective well-being, The international differences are greater for life evaluations than for emotions. Despite the well-documented differences in the ways in which subjective evaluations change through time and across cultures, the bulk of the very large international differences in life evaluations are due to differences in life circumstances rather than differences in the way these differences are evaluated. Traditional interpretations of quantum theory in terms of wave function collapse are particularly unappealing when considering the universe as a whole, where there is no clean separation between classical observer and quantum system and where the description is inherently relativistic. As an alternative, the consistent histories approach provides an attractive “no collapse” interpretation of quantum physics. Consistent histories can also be linked to path-integral formulations that may be readily generalized to the relativistic case. A previous paper described how, in such (...) a relativistic spacetime path formalism, the quantum history of the universe could be considered to be an eigenstate of the measurements made within it. However, two important topics were not addressed in detail there: a model of measurement processes in the context of quantum histories in spacetime and a justification for why the probabilities for each possible cosmological eigenstate should follow Born’s rule. The present paper addresses these topics by showing how Zurek’s concepts of einselection and envariance can be applied in the context of relativistic spacetime and quantum histories. The result is a model of systems and subsystems within the universe and their interaction with each other and their environment. (shrink)

Within the ambit of modernity, "to be" a "person" means "to have" "a body." But what exactly do we mean when we say: ‘I have a body’? Who or what is this ‘I’ that ‘has’ ‘a body’ anyway? And how and why does this ‘having’, this possessing, of ‘a body’ confer legal and psychological personhood on us? Does such bodily possession necessarily define a mode of ‘self ownership’? Is distinguishing between the notions of ‘being an organism’, or even ‘being alive’, (...) and ‘having a body’ just a matter of semantics? Or are there pragmatic, bio-political consequences to the ways we construe our vitality and embodiment? Can thinking about ‘the body’ as a (or even, the) matter of ‘self possession’ tell us something interesting about how ‘personhood’ informs our lives? After all, what is ‘the nature’ of ‘the body’, if it can be possessed in such a manner? François Laruelle's writing on Non-Photography is examined from its ontological condition to its desired form of a unity derived from the work of Kant, discussing precisely how the logic of transcendence and the ontology of immanence central to Laruelle's theory impact on how the photographic image is incontrovertibly involved with Kant's paradox of appearance and reality. In a context of burgeoning technoscience, which lays bare the meaning of Non-Photography for the seemingly impossible reversion to actual photography, the article goes on (...) to consider Photo-Fiction in the context of the Real of science now yoked to an economy of technical models. But by foreclosing the Kantian Real by fractal‐virtual ratio, does not this science reify the Real of Identity and displace real photography with a photography in the Real? The article thus questions how Laruelle's' thesis hangs together in terms of its cause and the contingency of effect that infuses Laruelle's radical aesthetic. (shrink)

If, as the title of this book suggests, the state of Tractatus commentary has at times recently resembled something close to a state of war, then it has most of all resembled a war of attrition. Against this background, Roger White's "Throwing the Baby Out with the Ladder" makes for refreshing reading. To be sure, White repeats some of the familiar misconceptions of what resolute readers do or must claim that have marred the debate over the adequacies or inadequacies of (...) such an approach to the Tractatus (TLP). But he also introduces some novel and interesting lines of criticism that merit serious attention. Foremost among the latter is White's treatment (in Section III of his paper) of three engaging examples that he sees as making trouble for resolute readers, and for their opposition to the standard idea that the lesson of the Tractatrrs could consist in its communicating, and our grasping, ineffable insights by way of its nonsense-sentences. Public debates on reality television often address the display of emotion and immoral conduct. Television scholars have recently proposed that while reality television offers its audience an opportunity to learn valuable lessons, they rarely address the issue of the morality of the genre. In this contribution, we analyze the display of emotion and immoral conduct in the Dutch reality show The Golden Cage. Reality television is viewed as constituting a ‘moral laboratory’. The question guiding our research revolved around the kind (...) of exercise this moral laboratory provides for. A dramaturgical study of the display of actions and emotions as part of participant “projects” was used to answer this question. Results indicate that moral issues and emotions were continuously at stake, and immoral conduct appeared to be coupled with pleasurable emotions. It is concluded that the laboratory of The Golden Cage is at least equally suited for exercising autonomy and identity performance under competitive pressure as it is for the simulation of moral conflicts. ABSTRACT In order to explore what it means to pursue philosophical investigations for theological reasons, this paper argues that Ludwig Wittgenstein continues and corrects Pseudo-Denys’ project in The Divine Names. I first argue that The Divine Names should be interpreted as attempting to render human thought transparent to the divine by relativizing our concepts. The success of this project is compromised because the concept of ‘unity’ is not relativized. I then develop the claim that Wittgenstein does relativize unity in a (...) similar way and for similar religious reasons to Pseudo-Denys. As such, he can be read as continuing and correcting the Pseudo-Dionysian project. I conclude by reflecting on several of this argument’s implications for the relationship between philosophy and Christian systematic theology. (shrink)

Under the normal assumptions of quantum field theory, Haag’s theorem states that any field unitarily equivalent to a free field must itself be a free field. Unfortunately, the derivation of the Dyson series perturbation expansion relies on the use of the interaction picture, in which the interacting field is unitarily equivalent to the free field but must still account for interactions. Thus, the traditional perturbative derivation of the scattering matrix in quantum field theory is mathematically ill defined. Nevertheless, perturbative quantum (...) field theory is currently the only practical approach for addressing scattering for realistic interactions, and it has been spectacularly successful in making empirical predictions. This paper explains this success by showing that Haag’s Theorem can be avoided when quantum field theory is formulated using an invariant, fifth path parameter in addition to the usual four position parameters, such that the Dyson perturbation expansion for the scattering matrix can still be reproduced. As a result, the parameterized formalism provides a consistent foundation for the interpretation of quantum field theory as used in practice and, perhaps, for better dealing with other mathematical issues. No one doubts that for Kierkegaard's definition of Christian faith one should look to the "Concluding Unscientific Postscript." The contention of this paper is that within the Postscript, most have looked in the wrong place. The well-known definition that is usually cited is actually a definition of Socratic or religious faith, and the definition of specifically Christian faith, given a few pages later, represents an existential intensification, which moves from an 'objective uncertainty' to an 'objective absurdity'.
