Genome editing with engineered nucleases (GEENs) introduce site‐specific DNA double‐strand breaks (DSBs) and repairs DSBs via nonhomologous end‐joining (NHEJ) pathways that eventually create indels (insertions/deletions) in a genome. Whether the features of indels resulting from gene editing could be customized is asked. A review of the literature reveals how gene editing technologies via NHEJ pathways impact gene editing. The survey consolidates a body of literature that suggests that the type (insertion, deletion, and complex) and the approximate length of indel edits (...) can be somewhat customized with different GEENs and by manipulating the expression of key NHEJ genes. Structural data suggest that binding of GEENs to DNA may interfere with binding of key components of DNA repair complexes, favoring either classical‐ or alternative‐NHEJ. The hypotheses have some limitations, but if validated, will enable scientists to better control indel makeup, holding promise for basic science and clinical applications of gene editing. Delays in early language development are characteristic of young autistic children, and one of the most recognizable first concerns that motivate parents to seek a diagnostic evaluation for their child. Although early language abilities are one of the strongest predictors of long-term outcomes, there is still much to be understood about the role of language impairment in the heterogeneous phenotypic presentation of autism. Using a person-centered, Latent Profile Analysis, we first aimed to identify distinct patterns of language and social communication (...) ability in a clinic-based sample of 498 autistic children, ranging in age from 18 to 60 months. Next, a multinomial logistic regression analysis was implemented to examine sociodemographic and child-based developmental differences among the identified language and social communication profiles. Three clinically meaningful profiles were identified from parent-rated and clinician-administered measures: Profile 1 “Relatively Low Language and Social Communication Abilities,” Profile 2 “Relatively Elevated Language and Social Communication Abilities,” and Profile 3 “Informant Discrepant Language and Relatively Elevated Social Communication Abilities.” Overall, young autistic children from the lowest-resource households exhibited the lowest language and social communication abilities, and the lowest non-verbal problem-solving and fine-motor skills, along with more features of attention-deficit/hyperactivity disorder and atypical auditory processing. These findings highlight the need for effective community-based implementation strategies for young autistic children from low-resource households and underrepresented communities to improve access to individualized quality care. (...) 144 pp. Two very different books recently published by Continuum seem to complement each other in unexpected ways. Father Kennedy, Catholic Zen Teacher, Jesuit Priest, openly shares with us his own journey into unknowing and self-emptying. As he notes in his preface, "what I looked for in Zen was not a new faith, but a new way of being Catholic that grew out of my own lived experience..." (p. 13). David Hackett, now an associate professor of religion, freely shares his youthful search for genuine spiritual experience. His book is drawn from correspondence from the 1970s with Father Thomas Keating and the monks of the St. Joseph's Abbey during his two-year stay in Japan. Hackett says in his Prologue, "I became a Catholic through Zen meditation..." (p. 17). In both cases, rather than threaten or diminish their Christian faith, Zen practice served as a wonderful "can-opener," prying off lids of rigid ideas and concepts, allowing the depth of the spirit to be more deeply experienced.Part I of Zen Spirit, Christian Spirit, addressed primarily to the Christian reader, offers an excellent introduction into Buddhism, Zen, and the essential elements of practice. In this section Kennedy has reflected the questions Christians have asked during the years he taught at the Zen Community of New York. His teacher there, Bernard Tetsugen Glassman Roshi, clarifies in the foreword Father Kennedy's position as a lay Zen Teacher who remains a Catholic priest. Kennedy weaves a consistent thread of Christian contemplative teachings side by side with the Zen teachings, making very visible the fact that Zen reminds us, as do Eckhart, Merton, and many other contemplatives, that the highest point of our Christian mysticism is reached not in the experience that I know God or that I love God--not in any I-Thou experience--but in the experience that God lives in us (p. 36).The book has been structured according to the four stages of human growth: knowledge, love, purification, and union. These stages have been likened to the stages of alchemy: lead, quicksilver, sulphur, and gold--these four elements provide Kennedy with his titles for the four parts of his book. This very structure makes clear that the goal of both Christian mysticism and Zen practice is transformation of being. With a firm foundation of knowledge and teachings from both traditions, then it is time to actually enter the fire of practice itself. I am reminded of the stages of practice outlined in the ancient Chinese Zen ox-herding pictures. The herdsman sets out with some knowledge, longing for his True Nature. He sees traces--footprints in the sand--and a deep yearning, the quicksilver of desire and inspiration, begins to ache in his heart. As he begins to see and experience his own True Nature, he must pass through the fire of struggle, surrender, deep letting go, until finally both herdsman [End Page 263] and ox disappear, self-emptying is complete in Union. There are no shortcuts. Jesus and Buddha both have invited us to practice radical poverty. Students with learning disabilities (LDs) suffer from executive function deficits and impaired prospective memory (PM). Yet the specificity of deficits associated with different types of LDs is still unclear. The object of the present research was to compare subgroups of students with different forms of LDs (<25th percentile) on executive function and PM. Students with a mathematics disability (MD,n= 30), reading disability (RD,n= 27), both (RDMD,n= 27), or neither (typically developing, TD,n= 30) were evaluated on a set of executive functioning (...) tasks (e.g., updating, inhibition, and shifting) and on PM. The results showed that students with MDs and RDMDs suffered from PM deficits. Among the subtypes of LDs, the deficit is different. The students with RDMDs showed a wide range of defects in PM, shifting, inhibition, and updating. In comparison, students with MDs experienced deficits in PM and shifting, while students with RDs experienced a deficit only in updating. For the RD group, the RDMD group and the TD group, updating, and shifting significantly predicted PM. For the MD group, only shifting significantly predicted PM performance, but PM deficits were not completely confined to shifting deficits. (shrink)

As historiographical studies on ancient China gradually move from the center to the margins of the public's field of vision, research on historiographical studies concerning ancient China have been undergoing some unusual changes. A truly considerable quantity of bamboo slip and silk manuscripts have either been discovered by archaeologists or accidentally unearthed in the last twenty years. Although these have been made public very slowly, even maddeningly so, the few of them that have appeared before the world in the course (...) of time have again and again both elated relatively sensitive historiographers and placed them in a quandary. They also have once and again confronted the system of ancient history that had been regarded as "final" with the prospect of utter collapse and the need to be rewritten. After the Mozuizi discoveries in Gansu in the 1950s, there were those of Mawangdui, Yinqueshan, and Shuihudi in the 1970s, and those of Shuanggudui, Bajiaolang, Zhangjiashan, and Baoshan in the 1980s; and it is said that there were astounding discoveries, though not yet made public, at Jingmen and Lianyungang in the 1990s. These "bamboo slip and silk manuscripts" that touch upon almost every aspect of the pre-Qin and Han areas of "the schools of the various masters," "number lore" (shu shu), "prescriptive techniques" (fang ji), "law," and "institutions" make us ask ourselves in astonishment: How many more mysteries of ancient China are still buried under the ground? Examining the nature of teachers’ emotions and how they are managed and regulated in the act of teaching is crucial to assess the quality of teacher’s instructions. Despite the essential role emotions play in teachers’ lives and instruction, research on teachers’ emotions has not paid much attention on teachers’ state emotions in the context of daily teaching. Significant portion of literature has described teachers’ emotions by foregrounding trait emotions through deductive methodological approaches. This paper explored elementary teachers’ state and trait (...) emotions while preparing for teaching and during teaching, reasons that underlie these emotions, and the relationship between their emotions and the quality of their mathematics instruction. Participants were seven elementary teachers working in the U.S. who participated in Holistic Individualized Coaching (HIC) professional development that consisted of 5 cycles of coaching over an year. For each coaching cycle pre-coaching conversation and post-coaching conversation data were collected regarding state emotions teachers felt in anticipation of teaching and during teaching retrospectively. Also, teachers responded adapted Teaching Emotion Scale (TES) that was aimed to measure trait emotions. In order to compare teachers’ emotions with instructional quality, coaching session were video recorded and analyzed to determine the quality of instruction. Findings of this study showed that teachers reported 6 categories of emotions (positive, negative, neutral, mixed-positive, mixed-negative, and mixed-complex), and often in non-typical ways (e.g., “not nervous”, “anxious but in a positive way”), and mixed emotions were the most dominant category. Also, teachers had more positive emotions anticipating teaching, than actually teaching the lesson, and there was no noticeable change of emotions across the 5 coaching cycles. The reason teachers felt mixed emotions reflected complex and context-specific nature of teaching, which was not articulated in the literature. There were no clear relationships between emotional experiences and instructional quality. This study allowed participants to freely describe their authentic, complex, overlapping, and ambiguous emotions in the context of active teaching, which contributes opening up the possibilities of diversifying teacher emotion research and shows the significance and usefulness of understanding teachers’ state emotions. Information unethical behavior is concerned with ethical behavioural conflicts in the use of information, information technologies, and information systems. This study examines the combination of locus of control and job insecurity as a joint moderator on the decision making process for information ethical behavioral intentions. A conceptual model is proposed to see the joint moderating role of LOC and JI. In the model, ethical behavioral intentions are influenced directly by ethical attitude, personal values, and perceived behavioural control. Simultaneously, personal values (...) also indirectly influence ethical behavioral intentions through the mediation of ethical attitude. The causal relationships are moderated by the joint moderator. Notably, the moderating effects were simultaneously examined using data from undergraduates in the MIS department of a college. The influences of the ethical attitude and personal values on ethical behavioral intentions are found to be similar for those with external locus of control and insecurity perception and those with internal locus of control and security perception. Furthermore, the influences of personal values on ethical attitude, and of perceived behavioural control on ethical behavioral intentions, are both greater for Controlists than Confusionists. Ugar and Malele write that generic machine learning (ML) technologies for mental health diagnosis would be challenging to implement in sub-Saharan Africa due to cultural specificities in how those conditions are diagnosed. For example, they say that in South Africa, the appearance of ‘schizophrenia’ might be understood as a type of spiritual possession, rather than a mental disorder caused by a brain dysfunction. Hence, a generic ML system is likely to ‘misdiagnose’ persons whose symptomatology matches that of schizophrenia in the (...) South African context. The authors thus claim that ‘a generic or universal design cannot be effective given the heterogeneity of value judgements in defining what mental health disorders are in different contexts’.1 Should we take this to mean that ML systems ‘should not be designed with a generalised perception of mental disorders,’1 as the authors suggest? On the contrary, my view is that generic ML can be useful, with the caveat that issues of cultural sensitivity may engender translational challenges in various national and cultural contexts. Yet, if it can be demonstrated that a generic ML system can reliably and accurately pick up on shared symptomatologies across cultures despite what any individual or community might believe about their actual causes, this aspect is surely the function and value of diagnostic ML which we should take to matter. Cultural gaps in aetiological understandings of mental health conditions make it all the more important to advance generic ML as a calibrating diagnostic tool. This is the first complete, one-volume English translation of the ancient Chinese text Xunzi, one of the most extensive, sophisticated, and elegant works in the tradition of Confucian thought. Through essays, poetry, dialogues, and anecdotes, the Xunzi articulates a Confucian perspective on ethics, politics, warfare, language, psychology, human nature, ritual, and music, among other topics. Aimed at general readers and students of Chinese thought, Eric Hutton’s translation makes the full text of this important work more accessible in English than ever (...) before. Named for its purported author, the Xunzi has long been neglected compared to works such as the Analects of Confucius and the Mencius. Yet interest in the Xunzi has grown in recent decades, and the text presents a much more systematic vision of the Confucian ideal than the fragmented sayings of Confucius and Mencius. In one famous, explicit contrast to them, the Xunzi argues that human nature is bad. However, it also allows that people can become good through rituals and institutions established by earlier sages. Indeed, the main purpose of the Xunzi is to urge people to become as good as possible, both for their own sakes and for the sake of peace and order in the world. In this edition, key terms are consistently translated to aid understanding and line numbers are provided for easy reference. The goal of the Salivaomics Knowledge Base (SKB) is to create a data management system and web resource constructed to support human salivaomics research. To maximize the utility of the SKB for retrieval, integration and analysis of data, we have developed the Saliva Ontology and SDxMart. For almost 700 years shoguns ruled Japan. These "gentleman warriors" developed a dedicated system of honor and strict guidlelines of behavior that Taira Shigesuke first brought together in his 16th century book—Bushido Shoshinshu. Present to a modern audience in clear, easy-to-read English, this new translation captures the majesty of the higher principles as well as the usefulness of the daily advice. From principles such as "a samurai should keep foremost in his mind the fact that he must die" to rules (...) regarding duty, personal obligation and even money management, this book sets out the guidelines that defined Japan's "way of the warrior." Honor and obligation are emphasised above all to the reader, who is taught that a samurai is expected to be indifferenet to pain, unquestioningly loyal, and an expert in his training. (shrink)

A tic disorder (TD) is a neurodevelopmental disorder characterized by tics, which are repetitive movements and/or vocalizations that occur due to aberrant sensory gating. Its pathophysiology involves dysfunction in multiple parts of the cortico-striato-thalamo-cortical circuits. Spontaneous brain activity during the resting state can be used to evaluate the baseline brain state, and it is associated with various aspects of behavior and cognitive processes. Theta–gamma coupling (TGC) is an emerging technique for examining how neural networks process information through interactions. However, the (...) resting-state TGC of patients with TD and its correlation with cognitive function have not yet been studied. We investigated the resting-state TGC of 13 patients with TD and compared it with that of 13 age-matched healthy children. The participants underwent resting-state electroencephalography with their eyes closed. At the global level, patients with TD showed a significantly lower resting-state TGC than healthy children. Resting-state TGC with the eyes closed was significantly negatively correlated with the attention quotient calculated for omission errors in a selective visual attention test. These findings indicate that the resting-state brain network, which is important for the attentional processing of visual information, is dysfunctional in patients with TD. Additionally, these findings support the view that TGC reflects information processing and signal interactions at the global level. Patients with TD may have difficulty gating irrelevant sensory information in the resting state while their eyes are closed. (shrink)

The Iowa Gambling Task (IGT) simulates uncertain gains and losses in real life situations and thus is a good measure of uncertain decision making. The role of working memory (WM) in IGT performance still remains unclear. The present study aimed to examine the effect of WM on IGT performance. Three groups of participants matched on gender ratio were randomly assigned to no WM load, low WM load, and high WM load conditions. Initially the three groups did not show significant difference (...) in WM capacity. They finished a modified version of IGT and then their implicit learning effect and explicit cognition on IGT were assessed. Results indicated a linear increasing trend of IGT performance among high WM load, low WM load and no WM load groups; participants in the no WM load and low WM load groups revealed implicit learning effect, while participants in the high WM load group did not; all participants showed explicit cognition on IGT to the same level. These results suggested that participants in the high WM load group showed good explicit cognition to IGT but showed poor performance. This pattern is similar to frontal patients. Further studies should be conducted to explore this issue. (shrink)

BackgroundTo explore how self-disclosure leads to post-traumatic growth in adults who have experienced traumatic events, this study identified the relationship between self-disclosure and post-traumatic growth in Korean adults. We examined a parallel multiple mediating model for this relationship.MethodsParticipants were 318 Korean male and female adult participants aged 20 years or older who had experienced trauma. We measured deliberate rumination, positive social responses, and the meaning of life as mediating variables.ResultsThe results revealed that the study variables positively correlated with PTG. Self-disclosure (...) was positively correlated with deliberate rumination, positive social responses, and meaning of life. In the multiple mediating model, deliberate rumination, positive social responses, and meaning of life mediated the relationship between self-disclosure and PTG.ConclusionSelf-disclosure, deliberate rumination, positive social responses, and meaning of life play an important role in the growth of adults who have experienced traumatic events. The findings of this study should provide valuable information for future research and for mental health professionals who want to promote the PTG of their clients. (shrink)

BackgroundFatigue is an important factor for the safety of ships. In order to alleviate fatigue of the seafarers, the STCW Convention has made many regulations on the working time of seafarers. At present, if a crew member takes only one day off at home before returning to work on the ship, the working time on the ship must be re-calculated again. If the time spent at home is not sufficient to allow the crew to recover, the regulations of only stipulating (...) the working time, not stipulating the home vacation time, cannot guarantee the crew’s fatigue been well controlled. The aim of present study is to explore the relationship between vacation schedule and fatigue of the seafarers.MethodsIn present study, a simplified stress scale developed by the Ministry of Labor of Japan has been used as a measurement tool. The method of stratified sampling was adopted. Data collection mainly came from domestic ocean-going seafarers. Analysis was conducted using the Cross analysis and hierarchical multiple regression analysis methods.ResultsWe found that there was no difference between crew members of different positions in terms of average vacation time and on-board service time. The length of last vacation time and this service time for seafarers of different positions showed obvious differences. The rank has a significant effect on the length of the last vacation and the length of this service time. Also, the results showed that there was a significant negative correlation between the duration of vacation and overall fatigue, while there was a significant positive correlation between the length of service time on board and overall fatigue.ConclusionThe results indicated that a reasonable vacation schedule was crucial for the relief of the seafarers’ fatigue, and also played a positive role in the state of working on the ship again. Industry developed -- thanks in part to the development of physics -- in ways other than Marx had suspected. The use of modern physics, leading to cybernetics and automation, gradually changed the workers from forces of production to supervisory engineers. Were one to undertake today an extrapolation like that which Marx carried out, one would have to see as highly probable the disappearance of the very working class that Marx saw as carrying out the world-revolution. A general game player automatically learns to play arbitrary new games solely by being told their rules. For this purpose games are specified in the general Game Description Language (GDL), a variant of Datalog with function symbols that uses a few game-specific keywords. A recent extension of basic GDL allows the description of nondeterministic games with any number of players who may have incomplete, asymmetric information. In this paper, we analyse the epistemic structure and expressiveness of this language in terms (...) of modal epistemic logic and prove two main results: (1) The operational semantics of GDL entails that the situation at any stage of a game can be characterised by a multi-agent epistemic (i.e., S5-) model; (2) GDL is sufficiently expressive to model any situation that can be described by a (finite) multi-agent epistemic model.
