Analysis of sites of newly integrated DNA in cellular genomes is important to several fields, but methods for analyzing and visualizing these datasets are still under development. Here, we describe tools for data analysis and visualization that take as input integration site data from our INSPIIRED pipeline. Paired-end sequencing allows inference of the numbers of transduced cells as well as the distributions of integration sites in target genomes. We present interactive heatmaps that allow comparison of distributions of integration sites to (...) genomic features and that support numerous user-defined statistical tests. To summarize integration site data from human gene therapy samples, we developed a reproducible report format that catalogs sample population structure, longitudinal dynamics, and integration frequency near cancer-associated genes. We also introduce a novel summary statistic, the UC50, which provides a single number summarizing possible clonal expansion. Using these tools, we characterize ongoing longitudinal characterization of a patient from the first trial to treat severe combined immunodeficiency-X1, showing successful reconstitution for 15 years accompanied by persistence of a cell clone with an integration site near the cancer-associated gene CCND2. The purpose of this research is to investigate the effectiveness of Digital Content Marketing on a Mixed Reality training platform environment with the consideration of online purchase intention through social media. E-commerce today encounters several common issues that cause customers to have reservations to purchase online. With the absence of physical contact points, customers often perceive more risks when making purchase decisions. Furthermore, online retailers often find it hard to engage customers and develop long-term relationships. In this research, a Structural (...) Equation Model is proposed to examine the efficacy of DCM from both immediate and long-term OPI. The results examine whether adopting DCM on an MR training platform environment through social media brings positive results in OPI. Empirical research was carried out through online questionnaires collected in 2021 and 2022. A total of 374 questionnaires were qualified for data analysis in this study, conducted with IBM SPSS and AMOS. The results imply that DCM is critical to stimulating both immediate and long-term OPI. The immediate OPI is positively affected by increasing perceived value through MR in DCM. Regarding the long-term OPI, increased customer engagement with DCM under MR environment can cultivate brand trust and significantly affect the long-term OPI. (shrink)

This book would be very important indeed if Mr. Spencer Brown had substantiated his claims “that the concept of probability used in statistical science is meaningless in its own terms” , and that confirming this is the only significance of experiments in psychical research. The six short introductory chapters need not be discussed here. It is in Chapters VII to IX that the author develops his thesis that the concept of randomness is self–contradictory, and the statistician's concept of probability consequently (...) meaningless. I shall examine what I take to be the central argument leading to this conclusion. This is developed from a distinction between “primary chance or randomness” and “secondary chance or randomness” . The former concept is to be applicable only to individual events and is to depend upon their “unexpectedness or unpredictability”; the latter concept, applicable only to a series as such, is denned as “possessing no discernible pattern” . The definition of “primary randomness” is amplified, but not clarified, on page 49: “An event is primarily random in so far as... one cannot be sure of its occurrence... The only relevant criterion is that we are able to guess ”. We are then told that primary randomness “admits of analysis in subjective terms”, since the same event may be predictable by one person but unpredictable by another; whereas secondary randomness is “a more objective concept”. I am grateful for Mr Bagger's thoughtful remarks, as well as those of Professors Cousins, Smith, Katz, and Gimello at a recent American Academy of Religion panel devoted to The Problem of Pure Consciousness . But I cannot help but be struck by the tone of Mr Bagger's notice. As one colleague communicated to me after reading the piece, this is one of the most gratuitously rude pieces he had ever seen! If our book is really as bad as all (...) this, it makes one wonder why Religious Studies , or Mr Bagger, would bother to give it such attention, and whether it indicates how deeply we have touched a nerve in the contemporary debate. Early commentaries on the Tractatus, such as Russell's introduction and Ramsey's review in Mind already noted and commented on Wittgenstein's peculiar views concerning nonsense and elucidation. Ramsey also complains that 'sentences apparently asserting such properties of objects are held by Mr Wittgenstein to be nonsense, but to stand in some obscure relation to something inexpressible'. However, there would not be 'the orthodox reading' of the Tractatus exemplified by these remarks, were it not for the emergence of the 'resolute reading', the (...) 'New Wittgensteinian', the 'therapeutic reading', or the 'post‐modernist interpretation', which challenge it. Resolute readings can be divided into 'strong' and 'weak' readings or, more or less equivalently, 'Jacobin' and 'Girondin' readings. The resolute reading proposes a new way of reading the Tractatus, and tries to give prominence to its distinctness and correctness by attacking the orthodox reading. Filing charges of scientific misconduct can be a risky and dangerous endeavor. This article presents rules of conduct to follow when considering whether to report perceived misconduct, and a set of step-by-step procedures for responsible whistleblowing that describe how to do so once the decision to report misconduct has been made. This advice is framed within the university setting, and may not apply fully in industrial settings.

Two cases of carcinoma of the stomach presenting during the same month and dying within four weeks of one another less than one year later are presented. One was treated symptomatically and the other received radical surgery. The care they received depended on decisions about diagnosis and treatment; the outcomes of these and the difficulties involved in evaluating monetary costs and quality of care are discussed in the light of recent interest in medical audit.

We numerically solve the functional differential equations (FDEs) of 2-particle electrodynamics, using the full electrodynamic force obtained from the retarded Lienard–Wiechert potentials and the Lorentz force law. In contrast, the usual formulation uses only the Coulomb force (scalar potential), reducing the electrodynamic 2-body problem to a system of ordinary differential equations (ODEs). The ODE formulation is mathematically suspect since FDEs and ODEs are known to be incompatible; however, the Coulomb approximation to the full electrodynamic force has been believed to be (...) adequate for physics. We can now test this long-standing belief by comparing the FDE solution with the ODE solution, in the historically interesting case of the classical hydrogen atom. The solutions differ. A key qualitative difference is that the full force involves a ‘delay’ torque. Our existing code is inadequate to calculate the detailed interaction of the delay torque with radiative damping. However, a symbolic calculation provides conditions under which the delay torque approximately balances (3rd order) radiative damping. Thus, further investigations are required, and it was prematurely concluded that radiative damping makes the classical hydrogen atom unstable. Solutions of FDEs naturally exhibit an infinite spectrum of discrete frequencies. The conclusion is that (a) the Coulomb force is not a valid approximation to the full electrodynamic force, so that (b) the n-body interaction needs to be reformulated in various current contexts such as molecular dynamics. Resolving this mystery is of significance not only to philosophers and physicists but is also a very practical concern. Our perception of time shapes our values and way of life; it also mediates the interaction between science and religion both of which rest fundamentally on assumptions about the nature of time. C K Raju begins with a critical exposition of various time-beliefs, ranging (...) from the earliest times through Augustine, Newton and Einstein to Stephen Hawking and current notions of chaos and time travel. He traces the role of organised religion in subverting time beliefs for its political ends. The book points out how this resulted in a facile dichotomy between 'linear' and 'cyclic' time, thereby inaugurating a confusion which, according to the author, has handicapped Western thought ever since, eventually influencing the content of science itself. Thus, this book daringly asserts that physical theory, traditionally regarded as amoral and objective, has depended on cultural beliefs about time. The author points out that time beliefs are again being manipulated today as the credibility of science is being exploited to promote a picture of time and, hence, a pattern of human behaviour which is convenient to the agenda of globalisation of culture. The linkages between modern theology and this 'brave new physics' are traced against the wider context of the so-called 'clash of civilisations', and the attempts to remake the world order. The conclusions point to the need to de-theologise time. The author challenges Einstein's understanding of relativity theory and suggests that a 'tilt in the arrow of time', or a small tendency towards cyclicity, will help repair the prevalent confusion about time. A 'tilt' also enables a physics that permits both memory and creativity, so that purpose and spontaneous growth of order are returned to human life. The book ends with a vision of Man as Creator, surprising God. Extensive research in physics, the history of science, comparative religions, and sociology lend weight to the important and challenging conclusions reached by the author. Written as a rejoinder to Stephen Hawking's A Brief History of Time, this book goes much further and, unlike any previous book, it gives a critical exposition of various world religions-Buddhism, Christianity, Hinduism, Islam, and Jainism-while exploring their intricate links, through time beliefs, to current physics on the one hand, and to global political and economic trends, on the other. This book will appeal to scholars and laypersons equally. It will fascinate anyone who reads it and will teach its readers to question the unquestionable. (shrink)

Without any systematic data or evidence of a problem, or even a thoughtful analysis of costs and benefits, the application of the human participant review system within universities is overreaching at the same time that some risky experimentation on humans outside of universities is unregulated. This article questions the purpose, feasibility, and effectiveness of current IRB approaches to most "2 people talking" situations and proposes scaling back the regulatory system to increase respect accorded it by researchers and its ability to (...) protect human participants of research from real versus imagined harms. In too many cases, the focus is on form over ethical substance: counting what can be counted, rather than focusing instead on what counts. Some disciplines - oral history and journalism, for example - simply do not belong within the scope of institutional review board jurisdiction. Others, such as survey research, informational interviews, and informal interactions, call for a shift from centralized review to more departmentally based (i.e., rooted in disciplinary ethics) oversight, and clearer guidelines on what requires advance review as opposed to provision of post hoc complaint systems. In recent years philosophers have given a good deal of attention to imperatives. They have concerned themselves mainly with the logical grammar of sentences of this kind, that is to say their relations to each other and to interrogative and indicative sentences. Very often this topic has been raised in terms of the problem ‘Is imperative inference possible, and if so, what kind of inference is it?’. Many philosophers have contended that there are logically valid inferences that involve imperative sentences. (...) Against this it has been argued that no such inferences are possible. It has even been held that there are no such things as imperatives at all – regarded, that is, as types of expression logically sui generis and independent of indicative sentences. (shrink)

Current formal mathematics, being divorced from the empirical, is entirely a social construct, so that mathematical theorems are no more secure than the cultural belief in two-valued logic, incorrectly regarded as universal. Computer technology, by enhancing the ability to calculate, has put pressure on this social construct, since proof-oriented formal mathematics is awkward for computation, while computational mathematics is regarded as epistemo-logically insecure. Historically, a similar epistemological fissure between computational/practical Indian mathematics and formal/spiritual Western mathematics persisted for centuries, during a (...) dialogue of civilizations, when texts on "algorismus" and "infinitesimal" calculus were imported into Europe, enhancing the ability to calculate. It is argued here that this epistemological tension should be resolved by accepting mathematics as empirically based and fallible, and by revising accordingly the mathematics syllabus outlined by Plato.
