Regulation of microtubule (MT) dynamics is essential for many cellular processes, but the machinery that controls MT dynamics remains poorly understood. MT plus‐end tracking proteins (+TIPs) are a set of MT‐associated proteins that dynamically track growing MT ends and are uniquely positioned to govern MT dynamics. +TIPs associate with each other in a complex array of inter‐ and intra‐molecular interactions known as the “+TIP network.” Why do so many +TIPs bind to other +TIPs? Typical answers include the ideas that these (...) interactions localize proteins where they are needed, deliver proteins to the cortex, and/or create regulatory pathways. We propose an additional and more mechanistic hypothesis: that +TIPs bind each other to create a superstructure that promotes MT assembly by constraining the structural fluctuations of the MT tip, thus acting as a polymerization chaperone. Blindsight is the term commonly used to describe visually guided behaviour elicited by a stimulus falling within the scotoma (blind area) caused by a lesion of the striate cortex. Such is normally held to be unconscious and to be mediated by subcortical pathways involving the superior colliculus. Blindsight is of considerable theoretical importance since it suggests that destriate man is more like destriate monkey than had been previously believed and also because it supports the classical notion of two visual systems. It is also of potential clinical importance, since it has been claimed recently that systematic practice in blindsight can lead to the recovery of normal visual function in patients with cortical lesions. From a review of the literature it is concluded that all of the phenomena of blindsight can be attributed either to light scatter into unimpaired parts of the visual field or to residual vision resulting from spared striate cortex. The possible contribution f other factors is also considered. It is concluded that blindsight studies have generally failed to control for such nonblindsight interpretations partly because of poor methodology and partly because of difficulties in defining the term. In this work a calculation of the cluster decomposable formalism for relativistic scattering as developed by Lindesay, Markevich, Noyes, and Pastrana (LMNP) is made for an ultra-light quantum model. After highlighting areas of the theory vital for calculation, a description is made of the process to go from the general theory to an eigen-integral equation for bound state problems, and calculability is demonstrated. An ultra-light quantum exchange model is then developed to examine calculability.

Multigene delivery and expression systems are emerging as key technologies for many applications in contemporary biology. We have developed new methods for multigene delivery and expression in eukaryotic hosts for a variety of applications, including production of protein complexes for structural biology and drug development, provision of multicomponent protein biologics, and cell‐based assays. We implemented tandem recombineering to facilitate rapid generation of multicomponent gene expression constructs for efficient transformation of mammalian cells, resulting in homogenous cell populations. Analysis of multiple parameters (...) in living cells may require co‐expression of fluorescently tagged sensors simultaneously in a single cell, at defined and ideally controlled ratios. Our method enables such applications by overcoming currently limiting challenges.Here, we review recent multigene delivery and expression strategies and their exploitation in mammalian cells. We discuss applications in drug discovery assays, interaction studies, and biologics production, which may benefit in the future from our novel approach. (shrink)

The paper introduces the concept of B-Almost distributive fuzzy lattice in terms of its principal ideal fuzzy lattice. Necessary and sufficient conditions for an ADFL to become a B-ADFL are investigated. We also prove the equivalency of B-algebra and B-fuzzy algebra. In addition, we extend PSADL to PSADFL and prove that B-ADFL implies PSADFL.

In the early twentieth century, viruses had yet to be defined in a material way. Instead, they were known better by what they were not – not bacteria, not culturable, and not visible with a light microscope. As with the ill-defined “gene” of genetics, viruses were microbes whose nature had not been revealed. Some clarity arrived in 1929 when Francis O. Holmes, a scientist at the Boyce Thompson Institute for Plant Research reported that Tobacco mosaic virus could produce local (...) necrotic lesions on tobacco plants and that these lesions were in proportion to dilutions of the inoculum. Holmes’ method, the local lesion assay, provided the first evidence that viruses were discrete infectious particles, thus setting the stage for physicochemical studies of plant viruses. In a field where there are few eponymous methods or diseases, Holmes’ assay continues to be a useful tool for the study of plant viruses. TMV was a success because the local lesion assay “made the virus visible” and standardized the work of virology towards determining the nature of the virus. (shrink)

It is not limited to the study of humble objects but is rather concerned with all of the undeniably aesthetic experiences that arise when one contemplates objects or performs acts that are outside the traditional realm of aesthetics. It is concerned with the nature of the relationship between subject and object. In the past thirty years environmental ethics has emerged as one of the most vibrant and exciting areas of applied philosophy. Several journals and hundreds of books testify to its growing importance inside and outside philosophical circles. But with all of this scholarly output, it is arguably the case that environmental ethics is not living up to its promise of providing a philosophical contribution to the resolution of environmental problems. Institutional corruption is a normative concept of growing importance that embodies the systemic dependencies and informal practices that distort an institution’s societal mission. An extensive range of studies and lawsuits already documents strategies by which pharmaceutical companies hide, ignore, or misrepresent evidence about new drugs; distort the medical literature; and misrepresent products to prescribing physicians. We focus on the consequences for patients: millions of adverse reactions. After defining institutional corruption, we focus on evidence that it lies behind the epidemic of (...) harms and the paucity of benefits. (shrink)

Over the past 35 years, patients have suffered from a largely hidden epidemic of side effects from drugs that usually have few offsetting benefits. The pharmaceutical industry has corrupted the practice of medicine through its influence over what drugs are developed, how they are tested, and how medical knowledge is created. Since 1906, heavy commercial influence has compromised congressional legislation to protect the public from unsafe drugs. The authorization of user fees in 1992 has turned drug companies into the FDA's (...) prime clients, deepening the regulatory and cultural capture of the agency. Industry has demanded shorter average review times and, with less time to thoroughly review evidence, increased hospitalizations and deaths have resulted. Meeting the needs of the drug companies has taken priority over meeting the needs of patients. Unless this corruption of regulatory intent is reversed, the situation will continue to deteriorate. We offer practical suggestions including: separating the funding of clinical trials from their conduct, analysis, and publication; independent FDA leadership; full public funding for all FDA activities; measures to discourage R&D on drugs with few, if any, new clinical benefits; and the creation of a National Drug Safety Board. In this paper, we introduce the notion of intuitionistic fuzzy PMS-subalgebras under homomorphism and Cartesian product and investigate several properties. We study the homomorphic image and inverse image of the intuitionistic fuzzy PMS-subalgebras of a PMS-algebra, which are also intuitionistic fuzzy PMS-subalgebras of a PMS-algebra, and find some other interesting results. Furthermore, we also prove that the Cartesian product of intuitionistic fuzzy PMS-subalgebras is again an intuitionistic fuzzy PMS-subalgebra and characterize it in terms of its level sets. Finally, we consider (...) the strongest intuitionistic fuzzy PMS-relations on an intuitionistic fuzzy set in a PMS-algebra and demonstrate that an intuitionistic fuzzy PMS-relation on an intuitionistic fuzzy set in a PMS-algebra is an intuitionistic fuzzy PMS-subalgebra if and only if the corresponding intuitionistic fuzzy set in a PMS-algebra is an intuitionistic fuzzy PMS-subalgebra of a PMS-algebra. The question has become a perennial one in environmental ethics, as well as in allied fields in environmental history, sociology, and politics. And just when we think it is settled—as many did after William Cronon's famous deconstruction of wilderness in (...) 1995—and it feels safe to go back into the metaphysical waters of environmental philosophy, such issues pop up again to send us running.What follows extends this debate in the context of Eric Katz's well-known work in environmental ethics. Brought together here are Katz and two critics sympathetic to his positions, Wayne Ouderkirk and Ned Hettinger. While the occasion for the exchange was the publication of Katz's book, Nature as Subject: Human Obligation and Natural Community (1997), which collects his papers from 1979 to 1996, the focus quickly became somewhat narrower, centering on possible problems with Katz's ontological dualism between natural and artifactual entities.Katz has exerted a strong influence on the development of environmental ethics, particularly through his persistent arguments for a distinction between individualist and holist approaches to obligations to the nonhuman natural world and his metaphysical and moral critique of attempts [End Page 102] by humans to create, repair, or restore nature. While these positions continue to be on display here, there is also something more. In this exchange Katz throws down the gauntlet on his nature-culture dualism, admitting at the start that the accusation is true. Like the Protestants in Monty Python's The Meaning of Life, he is a dualist—and fiercely proud of it! Despite whatever misgivings I or others may have about this position, I believe that in making this claim, Katz, similar to David Chalmers's open embrace and resuscitation of dualism in philosophy of mind, has done us a great service. For it is arguably true that many environmental philosophers, along with other academics, have settled into a complacent "labeling as argument" pattern. If we can label a position as "dualist" or "essentialist," then our work is done. But surely not every form of dualism is necessarily wrong, even if, as Ouderkirk reminds us in his reading of Val Plumwood's work, the logic of many forms of dualism has tended toward a pernicious pattern. Arguments are needed in both directions. If nature-culture dualism is mistaken or wrongheaded, or leads to unfortunate conclusions, then we must know why; if dualism is the only way to make sense of the hurdles presented to us in coming to a better understanding of the value of nature and culture then we must know why as well. And further, because many in the field have assumed a nature-culture dualism without admitting to it, we can admire that Katz chooses to "fess up" and finally put the issue squarely on the table.If this is the road we are heading down with this particular exchange, then we are off to a good start. Both critics of Katz do a thorough job of pressing home their arguments, rather than only pointing accusatory fingers. Ned Hettinger, in "The Problem of Finding a Positive Role for Humans in the Natural World," avoids using the term "dualism," but nonetheless argues that Katz has a tendency to "rigidly dichotomize the natural/artifactual distinction." Additionally, Hettinger argues that Katz tends to overlook his own admission that the differences between nature and artifacts admit to degrees rather than a strict dualism. Hettinger focuses the bulk of his critique on the unfortunate upshot of such a position: given such a distinction, there is little if any place for a positive role for human intervention in nature, or for a philosophical contribution to the formation of better environmental policies.Wayne Ouderkirk, in "Katz's Problematic Dualism and its 'Seismic' Effects on His Theory," is sympathetic with Katz's intuitions that some distinction is needed between artifacts and natural entities. There are many ways to describe cities. As a physical environment, more so than many other environments, they are at least an extension of our present intentions. But cities are not confined to the moment. Built spaces are also in conversation with the past and oriented toward the future as physical manifestations of our values and priorities. But even with all of the ways we have to describe cities we do not normally think of them as in any way akin (...) to the “natural” environment. City and country, nature and culture, are opposed. We move through cities differently, with a different set of values, whether articulated or not. Consider just one small example: Even the most jaded urban dweller may hesitate to sully the environment around him when he perceives it to be something other than a product of the human community. Though we have all seen trash in a national park, we suspect (or at least hope) that there is a kind of hesitancy that occurs with a person wondering what to do with her candy wrappers on a trail in Yosemite. On the other hand, a visitor to my home, Greenwich Village, will usually not think twice about tossing his cigarette butts on the ground as he walks toward his next destination. (shrink)

Discussion of ecological restoration in environmental ethics has tended to center on issues about the nature and character of the values that may or may not be produced by restored landscapes. In this paper we shift the philosophical discussion to another set of issues: the social and political context in which restorations are performed. We offer first an evaluation of the political issues in the practice of restoration in general and second an assessment of the political context into which restoration (...) is moving. The former focuses on the inherent participatory capacity at the heart of restoration; the latter is concerned with the commodified (primarily in the United States) and nationalized (primarily in Canada) uses to which restoration is being put. By comparing these two areas of inquiry we provide a foundation for a critical assessment of the politics of restoration based on the politics in restoration. (shrink)

J. Baird Callicott has thrown down the gauntlet once again in the monism?pluralism debate in environmental ethics. In a recent article he argues that his ?communitarianism? (combined with a limited intertheoretic pluralism) is sufficient to get the advantages of pluralism advocated by his critics, while at the same time retaining the framework of moral monism. Callicott's attempt to set the record straight on the monism?pluralism debate has once again derailed us from answering the most important question in this discussion: how (...) do we achieve a compatibilism among ethical theories which will inform better environmental practices? But if Callicott got it wrong, then who is getting it right? Arne Naess, whose work has heretofore been excluded from the mainstream discussion of this issue, has all along understood the heart of the monism?pluralism question. This paper updates the current state of the monism?pluralism debate, provides an answer to Callicott's latest challenge, and advances the thesis that all involved in this argument would do well to take a look at what Naess has to say on this issue. Empathy is the combined ability to interpret the emotional states of others and experience resultant, related emotions. The relation between prefrontal electroencephalographic asymmetry and emotion in children is well known. The association between positive emotion (assessed via parent report), empathy (measured via observation), and second-by-second brain electrical activity (recorded during a pleasurable task) was investigated using a sample of one hundred twenty-eight 6- to 10-year-old children. Contentment related to increasing left frontopolar activation (p < .05). Empathic concern and positive empathy (...) related to increasing right frontopolar activation (ps < .05). A second form of positive empathy related to increasing left dorsolateral activation (p < .05). This suggests that positive affect and (negative and positive) empathy both relate to changes in prefrontal activity during a pleasurable task. (shrink)
