Duchenne's muscular dystrophy (DMD), which affects 1/3500 live male births, involves a progressive degeneration of skeletal and cardiac muscle, leading to early death. The protein dystrophin is lacking in DMD and present, but defective, in the allelic, less severe, Becker muscular dystrophy and is also missing in the mdx mouse. Experiments on the mdx mouse have suggested two possible therapies for these myopathies. Implantation of normal muscle precursor cells (mpc) into mdx skeletal muscle leads to the conversion of dystrophin‐negative fibres (...) to ‐positive, with consequent improvement in muscle histology. Direct injectidn of dystrophin cDNA into skeletal or cardiac muscle also gives rise to dystrophin‐positive fibres. Although both appear promising, there are a number of questions to be answered and refinements to be made before either technique could be considered possible as treatments for myopathies in man. Genome‐wide identification of transcription factor binding sites with the ChIP‐seq method is an extremely important scientific endeavor − one that should ideally be performed for every transcription factor in as many cell types as possible. A major hurdle on the way to this goal is the necessity for a specific, ChIP‐grade antibody for each transcription factor of interest, which is often not available. Here, we describe CETCh‐seq, a recently published method utilizing genome engineering with the CRISPR/Cas9 system to circumvent the (...) need for a specific antibody. Using the CETCh‐seq method, targeted genomic editing results in an epitope‐tagged transcription factor, which is recognized by a well‐characterized, standard antibody, efficacious for ChIP‐seq. We have used CETCh‐seq in human cancer cell lines as well as mouse embryonic stem cells. We find that roughly 60% of transcription factors tagged using CETCh‐seq produce a high quality ChIP‐seq map, a significant improvement over traditional antibody‐based methods. (shrink)

Five appropriate roles of professional philosophers in environmental scholarship, policy and education are explored. Philosophers can and should function as social critics, as conceptual analysts, as “critical spectators” of science, as ethicists, and as educators. An example of the philosopher's role as social critic may be found in the analysis of the methods and presuppositions of public policy making. The philosopher as conceptual analyst can add clarity to environmental debates through analyses of such terms as “ecosystem”, “integrity”, “responsibiity” and “nature”. (...) “Meta-Science” is exemplified in an examination of the role of values in scientific investigation and in the scientist's contributions to public policy-making. The philosopher as environmental ethicist should seek a balanced and integrated view of both the naturalness of the human condition, and of the unique human quality of moral agency. Finally, all these contributions, and more, are combined in the philosopher's function as educator—first, of his student, and also of his profession, his academic colleagues, public policymakers, and the public at large. Future generations do not exist, and are not determinate in their make-up. The moral significance of future generations cannot be accounted for on the basis of a purely individualistic ethic. Yet future generations are morally significant. The Person-Affecting Principle, that (roughly) only acts which are likely to affect particular individuals are morally significant, must be augmented in such a way as to take into account the moral significance of Homo sapiens, a holistic entity which certainly does exist. The A. discusses conditions for the idea of obligations towards the dead, while simultaneously holding the view that dead people have ceased to exist in a morally relevant sense. He examines and rejects three «lateral» strategies (Callahan, Wellman, Partridge) that rest on a notion of obligations concerning, rather than towards the dead. He then goes on to scrutinize Feinberg's «frontal» strategy, that consists in defending the possibility of (pre)posthumous harms, and, as a result, of obligations towards the dead. The (...) A. rejects this standpoint also, especially because of the deterministic view on which it rests, and because of the coexistence it implies between post-mortem properties and ante-mortem harm. Both lateral and frontal strategies thus fail to justify obligations towards the dead. The A. then suggests that if we wish to stick to the latter, we might have to endorse the metaphysical claim that seems to be implied in it (i.e. that dead people do exist in some morally relevant sense), unless we are ready to abandon the idea that a moral obligation towards a person only makes sense if this person is in a position to be harmed. K. E. Boxer explores moral responsibility, and whether it is compatible with causal determinism. She suggests that to answer this question we must focus on responsibility in the sense of liability, and that an incompatibilist view may only be preserved on an understanding of the moral desert of punishment that many find morally problematic.

Al-Kindī, honoured as the 'philosopher of the Arabs', was the first philosopher of Islam. His pioneer philosophical writings engage with ideas that became available through the Graeco-Arabic translation movement. This volume makes his entire philosophical output-some two dozen works-available in English, most of them for the first time. The authors believe that Critical Thinking should engage students with issues of broader philosophical interest while they develop their skills in reasoning and argumentation. The text is informed throughout by philosophical theory concerning argument and communication—from Aristotle's recognition of the importance of evaluating argument in terms of its purpose to Habermas's developing of the concept of communicative rationality. Notwithstanding the remarkable achievements made by medical science over the last half of the twentieth century, there is a palpable sense that a strictly medical view of human health, that is one founded on modernist assumptions, has become problematic, if not counterproductive. In this study, I argue that as nursing continues to eagerly welcome and indeed champion medical epistemology in the form of knowledge transfer, evidence‐based practice, research utilization, outcomes‐based practice, quantifiable efficiency and effectiveness, it risks becoming little more than (...) a medical science addendum and indeed one that inherits the problems now facing contemporary medicine. The purpose of this study then is to attempt to resituate nursing as a discipline at work within an ontopolitical matrix of radical democratic pluralism. I begin by tracing a philosophical line from Kuhn’s paradigms to Bloor’s strong programme of Sociology of Scientific Knowledge. Following this, I attempt to explicate the thought of Bruno Latour as a philosophical alternative to Sociology of Scientific Knowledge. Next, I outline the radical pluralism of William Connolly in an effort to demonstrate its similarity to Latour’s philosophy and finally how such a position is germane to contemporary nursing and the reality of politics. I do this with reference to the controversial issue of illicit drug use and harm reduction. In effect, I argue that such an issue cannot be dealt with using scientific evidence alone, but rather requires a philosophy of advocacy, what I term democratic advocacy, that is capable of responding to the politics of suffering, which is to say suffering that results from identity/difference.
