Short (“seed”) or extended base pairing between microRNAs (miRNAs) and their target RNAs enables post‐transcriptional silencing in many organisms. These interactions allow the computational prediction of potential targets. In model organisms, predicted targets are frequently validated experimentally; hence meaningful miRNA‐regulated processes are reported. However, in non‐models, these reports mostly rely on computational prediction alone. Many times, further bioinformatic analyses such as Gene Ontology (GO) enrichment are based on these in silico projections. Here such approaches are reviewed, their caveats are highlighted (...) and the ease of picking false targets from predicted lists is demonstrated. Discoveries that shed new light on how miRNAs evolved to regulate targets in various phyletic groups are discussed, in addition to the pitfalls of target identification in non‐model organisms. The goal is to prevent the misuse of bioinformatic tools, as they cannot bypass the biological understanding of miRNA–target regulation. (shrink)

This article offers an alternative reading of the Abbé Sieyès and the modern ‘nation-state’ problématique. I argue that the subject/object that is constituted in the early days of modernity is the incomplete society: an impossible-possibility ideal of congruency of population, authority and space. I suggest reading this ideal of congruency as a fantasy in that it offers a certain ‘fullness to come’, the promise of jouissance that can never be attained and is thus constantly re-envisioned and reinvoked. Drawing on discourse-analytical (...) and psychoanalytical tools I explain the logic of fantasy before analysing Sieyès’ What is the Third Estate?, as I show how he critiques and fragments the old model of the state and how his reading of the nation is fantasmatic, a continuous project towards impossible congruency. Authority and Estrangement addresses a set of questions about self-knowledge and seeks to answer them in the context of the broader differences between the first-person and third-person perspectives on oneself. Attention to these broader differences takes the discussion from epistemology to moral psychology, and seeks to relate some of the issues of contemporary philosophy of mind to the concerns with self-consciousness in post-Kantian thought.

Since Socrates, and through Descartes to the present day, the problems of self-knowledge have been central to philosophy's understanding of itself. Today the idea of ''first-person authority''--the claim of a distinctive relation each person has toward his or her own mental life--has been challenged from a number of directions, to the point where many doubt the person bears any distinctive relation to his or her own mental life, let alone a privileged one. In Authority and Estrangement, Richard Moran argues (...) for a reconception of the first-person and its claims. Indeed, he writes, a more thorough repudiation of the idea of privileged inner observation leads to a deeper appreciation of the systematic differences between self-knowledge and the knowledge of others, differences that are both irreducible and constitutive of the very concept and life of the person. Masterfully blending philosophy of mind and moral psychology, Moran develops a view of self-knowledge that concentrates on the self as agent rather than spectator. He argues that while each person does speak for his own thought and feeling with a distinctive authority, that very authority is tied just as much to the disprivileging of the first-person, to its specific possibilities of alienation. Drawing on certain themes from Wittgenstein, Sartre, and others, the book explores the extent to which what we say about ourselves is a matter of discovery or of creation, the difficulties and limitations in being ''objective'' toward ourselves, and the conflicting demands of realism about oneself and responsibility for oneself. The notion of ‘bipolar’ or ‘second‐personal’ normativity is often illustrated by such situations as that of one person addressing a complaint to another, or asserting some right, or claiming some authority. This paper argues that the presence of speech acts of various kinds in the development of the idea of the ‘second‐personal’ is not accidental. Through development of a notion of ‘illocutionary authority’ I seek to show a role for the ‘second‐personal’ in ordinary testimony, despite Darwall's argument that the notion (...) of the ‘second‐personal’ marks a divide between practical and theoretical reason. The concept of habit enfolds an enormous richness and diversity of meanings. According to Husserl, habit, along with association, memory, and so on, belongs to the very essence of the psychic.1 Husserl even speaks of an overall genetic “phenomenology of habitualities”. In this paper, as an initial attempt to explicate the complexity of phenomenological treatments of habit, want to trace Husserl’s conception of habit as it emerged in his mature genetic phenomenology, in order to highlight his enormous and neglected original (...) contribution in this area. I shall show that Husserl was by no means limited to a Cartesian intellectualist explication of habitual action (as commentators such as Bourdieu and Dreyfus have claimed), but attempted to characterize its complexity across the range of human individual, sub-personal, personal, social and collective experience. Habit, as we shall see, for Husserl, is intimately involved in the constitution of meaningfulness (Husserl’s Sinnhaftigkeit) and forms of sense (Sinnesgestalten) at all levels, from the level of perceptual experience, through the formation of the ego, to the development of society, history and tradition, indeed to our whole sense of the harmonious course of worldly life and to the genetic constitution of worldhood as such. Habituality, furthermore, is a key structural principle in the genetic constitution of the transcendental ego itself, as it unfolds as a concrete living and acting person in an intersubjective, cultural and historical world. It is undeniable that the assumption of sincerity is important to assertion, and that assertion is central to the transmission of beliefs through human testimony. Discussions of testimony, however, often assume that the epistemic importance of sincerity to testimony is that of a guarantee of access to the actual beliefs of the speaker. Other things being equal, we would do as well or better if we had some kind of unmediated access to the beliefs of the other person, without the (...) risks involved in the overt act of speaking, and the assumption of sincerity in speech is the closest we can come to this access. Contrary to this picture, I argue that sincerity has a quite different epistemic role to play in testimony than that of an indicator of the speaker’s beliefs. The epistemology of testimony requires reference to the speaker as agent, and not just the speaker’s beliefs, as well as a sense of ‘expression of belief’ that links it to the specifically addressive relation to another person. I’m very grateful for the attention given to my book by all the commentators, and their various and thoughtful responses have helped me in many ways. Several related issues are raised by the comments of Heal and Reginster, and to avoid repetition I will discuss them together here. Both of them raise questions about the scope and authority of rationality over a person’s beliefs and other attitudes, and ask what is supposed to be wrong with adopting what I describe as (...) a spectator’s point of view on oneself, and whether this stance by itself involves the evasion of rational responsibility for one’s attitudes. They also, in their different ways, provide searching discussions of the ‘rakehell’ case from Chapter 5, where several of these issues come together, so I’d like to respond to those parts of their comments together as well. This thesis is a study of the philosophical system of a little-studied, but important medieval thinker, John Scottus Eriugena , concentrating on his Periphyseon . ;I argue that Eriugena's system of nature must be approached through an investigation of his epistemology and general philosophy of mind. Instead of beginning with his fourfold classification of Nature, as most commentators have done, I begin with Eriugena's concept of the mind and its dialectical operations , and continue with an examination of his anthropology (...) , and his concept of self-knowledge , before turning to his relativist ontology of being and non-being . Chapter Seven examines Eriugena's fourfold division of Nature in the light of the earlier chapters. ;Against some recent commentators, I argue that Eriugena should not be interpreted solely from within the framework of the Latin-Augustinian metaphysical tradition of the early Middle Ages, that it was his sympathy for Greek Christian Neoplatonism which led him to develop an idealist philosophical outlook . Eriugena synthesises the Greek Eastern and the Latin Western traditions into a profound and original philosophical system. ;I show further that Eriugena "deconstructs" Latin metaphysical realism in favour of his own "me-ontology" or "meta-ontology," inspired by the Greeks. He produces the most detailed account of non-being between Plato's Parmenides and the phenomenologies of nothingness of Heidegger and Sartre. ;The summit of Eriugena's system is infinite Non-being, or infinite self-identical subjectivity. This philosophy bears strong similarities to post-Kantian critical philosophy, especially the Idealism of Fichte, Schelling and Hegel. My interpretation thus agrees with the nineteenth-century Idealist reading of Eriugena. At the same time, it suggests the implications of Eriugena's idealism for the interpretation of certain modern philosophies. (shrink)

Living Nonviolently: Language for Resisting Violence proposes distinctions of language that effectively address issues of force, power, aggressiveness, violence and war. No other book provides such a consistent language for living nonviolently through examples drawn from nonhuman animals, human infancy, personal transactions, domestic politics, and international conflicts.

In this paper I discuss Edmund Husserl’s phenomenological account of the constitution of the social world, in relation to some phenomenological contributions to the constitution of sociality found in Husserl’s students and followers, including Heidegger, Gurwitsch, Walther, Otaka, and Schutz. Heidegger is often seen as being the first to highlight explicitly human existence as Mitsein and In-der-Welt-Sein, but it is now clear from the Husserliana publications that, in his private research manuscripts especially during his Freiburg years, Husserl employs many of (...) the terms associated with Heidegger, e.g. Mitwelt, Weltlichkeit, Alltäglichkeit, Zeitlichkeit, and Geschichtlichkeit, and had detailed discussions of various forms of social constitution. It is clear that Husserl and Heidegger were exploring these themes in dialogue with one another, and that Husserl, in fact, has a rich phenomenology of sociality that is worth exploring in its own right. Philosophy and Kafka is a collection of original essays interrogating the relationship of literature and philosophy. The essays either discuss specific philosophical commentaries on Kafka’s work, consider the possible relevance of certain philosophical outlooks for examining Kafka’s writings, or examine Kafka’s writings in terms of a specific philosophical theme, such as communication and subjectivity, language and meaning, knowledge and truth, the human/animal divide, justice, and freedom.

Mitochondrial bioenergetics plays a key role in multiple basic cellular processes, such as energy production, nucleotide biosynthesis, and iron metabolism. It is an essential system for animals' life and death (apoptosis) and it is required for embryo development. This, in conjunction with its being subjected to adaptive processes in multiple species and its gene products being involved in the formation of reproductive barriers in animals, raises the possibility that mitochondrial bioenergetics could be a candidate genetic mechanism of speciation. Here, we (...) discuss genetic and biochemical evidence for the possible involvement of this unique system, encoded by two genomes (the mitochondrial and nuclear genomes), that differ by an order of magnitude in their mutation rates in processes leading to speciation events. BackgroundInformed consent is a key element of ethical clinical research. Addicted population may be at risk for impaired consent capacity. However, very little research has focused on their comprehension of consent forms. The aim of this study is to assess the capacity of addicted individuals to provide consent to research.Methods53 subjects with DSM-5 diagnoses of a Substance Use Disorder and 50 non psychiatric comparison subjects participated in the survey from December 2014 to March 2015. This cross-sectional study was carried out (...) at a community-based Outpatient Treatment Center and at an urban-located Health Centre in Spain. A binary judgment of capacity/incapacity was made guided by the MacArthur Competence Assessment Tool for Clinical Research and a clinical interview. Demographics and clinical characteristics were assessed by cases notes and the Mini-Mental State Examination, the Global Assessment Functional Scale and the Clinical Global Impression Scale.ResultsNPCs performed the best on the MacCAT–CR, and patients with SUD had the worst performance, particularly on the Understanding and Appreciation subscales. 32.7 % SUD people lacked research-related decisional capacity. There were no statistically significant differences between the groups in terms of capacity to consent to research.ConclusionsThe findings of our study provide evidence that a large proportion of individuals with SUD had decisional capacity for consent to research. It is therefore inappropriate to draw conclusions about capacity to make research decisions on the basis of a SUD diagnosis. In the absence of advanced cognitive impairment, acute withdrawal or intoxication, we should assume that addicted persons possess decision-making capacity. Thus, the view that people with SUD would ipso facto lose decision-making power for research consent is flawed and stigmatizing. (shrink)

In his published texts and lectures on moral philosophy, Kant repeatedly singles out gratitude for discussion. Nevertheless, puzzles about the derivation, content, and nature of this duty remain. This paper seeks to solve some of these puzzles. Centrally, I argue that it is essential to attend to a distinction that Kant makes between well-wishing benevolence (Wohlwollen) and active beneficence (Wohlthun) on the part of a benefactor. On the Kantian account, I argue, a different type of gratitude is owed in response (...) to each. With this distinction in place, I argue further that we can construct what many have thought to be missing from Kant’s discussion of gratitude – an argument explicating the precise contradiction generated by a maxim of ingratitude. Thus, De Memoria et Reminiscentia treats about memory, which is common to man and perfect animals, and reminiscence, which is proper to man.

Saint Thomas Aquinas explains that in the science of mobiles, when descending to the living, each one of the species of the animals and plants are attended specifying what is proper to each of them, and here is where De Sensu et Sensato is found. Aquinas enumerates the diverse types of living beings according to the parts of soul and enlists the books that have a relation with them, deducing that De Sensu et Sensato corresponds to the exterior act that (...) occurs when feeling. In his later works, Merleau-Ponty proposes the notion of ‘the flesh’ (la chair) as a new ‘element’, as he put it, in his ontological monism designed to overcome the legacy of Cartesian dualism with its bifurcation of all things into matter or spirit. Most Merleau-Ponty commentators recognise that Merleau-Ponty's notion of ‘flesh’ is inspired by Edmund Husserl's conceptions of ‘lived body’ (Leib) and ‘vivacity’ or ‘liveliness’ (Leiblichkeit). But it is not always recognised that, for Merleau-Ponty, the constitution of the world (...) of perception, the problem of embodiment or incarnation, is at the very same time one with the problem of the experience of others in what Husserl called Einfühlung or Fremderfahung and indeed one with the problem of the constitution of the commonly shared world ‘for all’. As Merleau-Ponty put it in his late essay ‘The Philosopher and His Shadow’ in Signs, ‘the problem of Einfühlung, like that of my incarnation, opens on the meditation of sensible being, or, if you prefer, it betakes itself there’. In other words, the problem of the apprehension of the other is part of the overall apprehension of the transcendent world. In this paper I want to meditate on the relations between embodiment, experience of others, and experience of the world in Merleau-Ponty's philosophy. I will take particular note, as in the title of this presentation, of the claim made by Merleau-Ponty in The Visible and the Invisible that ‘there is no brute world, only an elaborated world’ (il n'y a pas de monde brut, il n'y a qu'un monde élaboré).
