Paradox is a logical phenomenon. Usually, it is produced in type theory, on a type Ω of “truth values”. A formula Ψ (i.e., a term of type Ω) is presented, such that Ψ↔¬Ψ (with negation as a term¬∶Ω→Ω)-whereupon everything can be proved: In Sect. 1 we describe a general pattern which many constructions of the formula Ψ follow: for example, the well known arguments of Cantor, Russell, and Gödel. The structure uncovered behind these paradoxes is generalized in Sect. 2. This (...) allows us to show that Reynolds' [R] construction of a typeA ≃℘℘A in polymorphic λ-calculus cannot be extended, as conjectured, to give a fixed point ofevery variable type derived from the exponentiation: for some (contravariant) types, such a fixed point causes a paradox.Pursueing the idea that $$\frac{{{\text{type theory}}}}{{{\text{categorical interpretation}}}} = \frac{{{\text{(propositional) logic}}}}{{{\text{Lindebaum algebra}}}}$$ the language of categories appears here as a natural medium for logical structures. It allows us to abstract from the specific predicates that appear in particular paradoxes, and to display the underlying constructions in “pure state”. The essential role of cartesian closed categories in this context has been pointed out in [L]. The paradoxes studied here remain within the limits of the cartesian closed structure of types, as sketched in this Lawvere's seminal paper — and do not depend on any logical operations on the type Ω. The apolipoprotein E gene plays an important role in the pathogenesis of Alzheimer's disease , and amyloid plaque comprised mostly of the amyloid-beta peptide ) is one of the major hallmarks of AD. However, the relationship between these two important molecules is poorly understood. We examined how A treatment affects APOE expression in cultured cells and tested the role of the transcription factor NF-B in APOE gene regulation. To delineate NF-B's role, we have characterized a 1098 nucleotide segment containing the (...) 5'-flanking region of the human APOE gene . Sequence analysis of this region suggests the presence of two potential NF-B elements. To demonstrate promoter activity, the region was cloned upstream of a promoterless luciferase gene. This segment was able to drive expression of luciferase in transient transfections of human fetal glial cells. Promoter activity was stimulated twofold by A treatment. Pretreatment with double-stranded DNA decoy oligonucleotides against NF-B reduced A stimulation. Deletion and mutagenetic analyses demonstrated that the distal NF-B element was functional and showed a strong DNA-protein complex band in gel shift analysis, similar to that from control NF-B consensus element. An anti-inflammatory and anti-NF-B drug, sodium salicylate, significantly blocked A-induced APOE promoter function. Our data provide evidence that upregulation of APOE by A in astroglial cells is mediated by an NF-B-element present in the 5'-flanking region of the APOE gene. (shrink)

ABSTRACTPeople high in social anxiety experience fear of social situations due to the likelihood of social evaluation. Whereas happy faces are generally processed very quickly, this effect is impaired by high social anxiety. Mouth regions are implicated during emotional face processing, therefore differences in mouth salience might affect how social anxiety relates to emotional face discrimination. We designed an emotional facial expression recognition task to reveal how varying levels of sub-clinical social anxiety related to the discrimination of happy and fearful (...) faces, and of happy and angry faces. We also categorised the facial expressions by the salience of the mouth region. In a sample of 90 participants higher social anxiety was associated with a reduced happy face reaction time advantage. What responsibilities, if any, do we have towards our genetic offspring, before or after birth and perhaps even before creation, merely by virtue of the genetic link? What claims, if any, arise from the mere genetic parental relation? Should society through its legal arrangements allow 'fatherless' or 'motherless' children to be born, as the current law on medically assisted reproduction involving gamete donation in some legal systems does? Does the possibility of establishing genetic parentage with practical certainty necessitate reform of (...) current legal regimes of parenthood? And what limits, if any, should we set on parental procreative choices in the interests of future children, particularly with regard to genetic engineering and related techniques? These are the questions explored in this book by some of the foremost legal, bioethical and biomedical thinkers. Assembled with a view to assisting the reader to reflect critically on the ongoing social experiment which medically assisted reproduction is today, the essays in this collection highlight what are - and what else might in the nearby future become - possible reproductive options and respond to the difficulties we encounter in assessing these practices and possibilities from our traditional ethical vantage points. The focus on translational research in clinical trials has the potential to generate clinically relevant genetic data that could have importance to patients. This raises challenging questions about communicating relevant genetic research results to individual patients. An exploratory pharmacogenetic analysis was conducted in the international ovarian cancer phase III trial, AGO-OVAR 16, which found that patients with clinically important germ-line BRCA1/2 mutations had improved progression-free survival prognosis. Mechanisms to communicate BRCA results were evaluated, because these findings may be beneficial to (...) patients and their families. Communicating individual BRCA results was not anticipated during clinical trial design. Consequently, options were not available for patients to indicate their preference for receiving their individual results when they signed pharmacogenetic informed consent. Differences in local requirements, clinical practice, and opinion regarding the ethical aspects of how to convey genetic results to patients are all potential barriers to returning individual BRCA results to patients. Communicating the aggregate BRCA result from this study provided clinical investigators with a mechanism to disseminate the overall study finding to patients while taking individual circumstances, local guidelines and clinical practice into account. This study illustrates the importance of increasing the clarity and scope of informed consent and the need for patient engagement to ensure clinical trial participants can indicate their preference regarding receipt of potentially important individual pharmacogenetic results. For quite some time now philosophers have stressed the need to distinguish between explanatory (motivating) reasons and justifying (good) reasons. The distinction is often illustrated with an example of someone doing something that is intended to strike the reader or listener, at least at the outset, as incomprehensible. The story of Abraham on Mount Moriah, who decided to sacrifice his son, Isaac, illustrates this pattern. Killing one’s own child is a horrific thing to do, and it is hard to understand (...) what would drive a parent to do such a thing. But once we are informed that Abraham actually believed God had told him to do so, we can see (allegedly) that there is at least an explanation of why he decided to kill his son. In the eyes of most people the planned act remains awful. But once we assume that Abraham was mistaken (say, because the best explanation in this case need not postulate the existence of God), we can see that his decision to kill his son is not a response to a normative good reason.1 So in a few lines we have outlined two reason notions: explanatory and normative (good) reasons. Cases like this afford an intuitive grasp of the distinction between explanatory and normative reasons — the difference between what explains Abraham’s motivation and behaviour and the good or bad reasons that apply to him. However, more recently (see e.g. Dancy 2000) the picture such cases present has been supplemented, or perhaps even corrected. There is a further feature of the Abrahamic story that needs to be teased out — one that gives a finer-grained understanding of what is going on than that provided by talk of the agent’s explanatory reasons. I share this view, and so what I will be doing in this paper is mainly to underline the need to dig a bit deeper. Motivation, as I shall argue, comes in different forms . (shrink)

The aim of this study was to test red - green dichromats' ability to discriminate between illuminant and surface-reflectance changes in natural scenes. Stimuli were simulations of natural scenes presented on a colour monitor with 10-bit resolution per gun. The natural scenes were obtained with a fast hyperspectral imaging system. Six different scenes (including rocks, foliage, and buildings) were tested. In each trial, two images were presented in sequence, each for 1 s, with no interval. The images differed in the (...) phase of daylight on the scene: first with correlated colour temperature 25 000 K, then 6 700 K. The spectral reflectance of a region in the second image was changed randomly, consistent with a local change in daylight. The observer's task was to decide whether a particular surface in the successive images was the same (pure illuminant change) or different (illuminant change with a surface-reflectance change). The performance of four deuteranopes and five protanopes varied considerably across the scenes tested, with mean colour constancy indices (± 1 SEM) of 0.48 (± 0.13) and 0.19 (± 0.10), respectively (1.0 representing ideal performance). Deuteranopes seemed less disadvantaged than protanopes, performing close to normal with some scenes. (shrink)

Presentists and Growing Block theorists appeal to ‘powerful intuitions’ when they defend their respective conceptions of time . Eternalists are prepared to go some length towards ‘reconciling’ the view from nowhen with at least some of these intuitions, or try to explain them away . Unaided intuitions may in fact underdetermine any particular metaphysical choice. One set of intuitions about time seems to have been (...) neglected, though, in spite of its power. These intuitions are the mirror image of those supporting the Growing Block theory , and they are modelled on a pre-reflective understanding of our own life. According to the intuitions, only the future and the present are real; the past isn’t. The present is the constantly eroding edge of the future, which thus shrinks incessantly. There is less and less future as the present proceeds. The obvious intuitive model of this view of time is our life; from which every day eats up a little bit. Smaller scale models are available in everyday life. As you are reading this text, you have less and less to read. You know it. You know that at some point you’ll finish, or stop, reading it. But a model is not enough to make a grand philosophical claim, and indeed the very existence of the model is the ground for a negative claim: a picture, it may be said, is holding you captive. The Future Shrinker has two tasks. The first is to show that the intuitive picture supporting the claim is robust and does not make implausible epistemological claims. Le livre-testament d'un des plus grands écologues français qui nous offre un plaidoyer limpide et convaincant pour nous aider à nous réconcilier avec la nature avec notre nature et à nous construire ainsi un autre avenir. L'homme fait partie de la nature, au même titre que les autres mammifères, les insectes, les plantes... Pendant des milliards d'années, tous ces organismes ont évolué, se sont adaptés, sont parfois entrés en compétition les uns avec les autres, et surtout ont coopéré. Là se (...) trouve la raison principale du remarquable succès du monde vivant, comme le montrent de nombreux exemples concret : si les tensions et les conflits existent bien entre les espèces, la nature est surtout un lieu d'échanges, d'entraide, de coopération. Mais l'emprise grandissante d'une espèce particulière, la nôtre, bouleverse cet équilibre subtil : l'homme est devenu une menace pour la planète, ses habitants et lui-même. Pour autant, il n'est pas trop tard pour redresser le cap! C'est en reconnaissant que nous faisons tous partie d'une même communauté, en développant une nouvelle solidarité écologique, en nous réconciliant avec le monde vivant que nous pourrons le sauver : réconcilions-nous avec la nature, car c'est notre nature!
