BackgroundAntipsychotic-induced weight gain is a contributing factor in the reduced life expectancy reported amongst people with psychotic disorders. CYP2D6 is a liver enzyme involved in the metabolism of many commonly used antipsychotic medications. We investigated if CYP2D6 genetic variation influenced weight or BMI among people taking antipsychotic treatment.MethodsWe conducted a systematic review and a random effects meta-analysis of publications in Pubmed, Embase, PsychInfo, and CENTRAAL that had BMI and/or weight measurements of patients on long-term antipsychotics by their CYP2D6-defined metabolic groups.ResultsTwelve (...) studies were included in the systematic review. All cohort studies suggested that the presence of reduced-function or non-functional alleles for CYP2D6 was associated with greater antipsychotic-induced weight gain, whereas most cross-sectional studies did not find any significant associations. Seventeen studies were included in the meta-analysis with clinical data of 2,041 patients, including 93 poor metabolizers, 633 intermediate metabolizers, 1,272 normal metabolizers, and 30 UMs. Overall, we did not find associations in any of the comparisons made. The estimated pooled standardized differences for the following comparisons were PM versus NM; weight = –0.07, BMI = 0.40. IM versus NM; weight = 0.09 and BMI = 0.09. UM versus EM; weight = 0.01 and BMI = –0.08.ConclusionOur systematic review of cohort studies suggested that CYP2D6 poor metabolizers have higher BMI than normal metabolizers, but the data of cross-sectional studies and the meta-analysis did not show this association. Although our review and meta-analysis constitutes one of the largest studies with comprehensively genotyped samples, the literature is still limited by small numbers of participants with genetic variants resulting in poor or UMs status. We need further studies with larger numbers of extreme metabolizers to establish its clinical utility in antipsychotic treatment. CYP2D6 is a key gene for personalized prescribing in mental health. One of the features of an encompassing account of language that Charles Taylor examines in Chapter Five, “The Figuring Dimension of Language,” ofThe Language Animalis a special kind of metaphor, which is rooted in the embodiment of humans. Their perspective-taking, their intuition of position in space, etc., provide ‘structural templates’ for thinking and leave their traces in their expressions. Taylor compares these metaphors with paradigms. My paper discusses the differences between the two. In his 1868 paper “Questions concerning Reality” (QR), an early version of his better-known paper “Questions Concerning Certain Faculties Claimed for Man” (QCF) of the same year (Fisch 1984: xxxviii), Charles S. Peirce quotes the unidentified Latin fragment Hoc loquor inde est (W 2:167). In QCF, the better-known essay first published in the second volume of the Journal of Speculative Philosophy and included in Peirce’s Collected Papers (CP) as well as in the first volume of the Essential Peirce, Peirce quotes (...) the same fragment again. Peirce's "Neglected Argument" uses more or less standard logical vocabulary, such as "argument," "retroduction," "premise," "conclusion," and "hypothesis." There cannot be any doubt, however, that the musement process as he characterizes it must be regarded as a semiotic process—that is, one that relates a sign to an object by way of forming an interpretant. As we try to show in this Special Issue: by understanding him literally and making use of his notion of moral maps – or, differently put, by ‘mapping’ Charles Taylor. As he is far too modest a person to reveal to us his own moral atlas, we have decided to seize the occasion of his 85th birthday (...) to ask several of his renowned colleagues, students and interlocutors to contribute to the reconstruction of such a map. In Hobbes's Theory of the Will, Jurgen Overhoff reveals the religious, ethical, and political consequences of Thomas Hobbes's doctrine of volition. The author gracefully describes how Hobbes's thought was governed by assumptions based firmly in Galilean natural philosophy and orthodox Protestant theology. Overhoff also demonstrates how his subject used materialist eschatology and an absolutist political theory to resolve the social and ethical predicaments that coincided with these assumptions. Finally, Overhoff provides a chronological study of the numerous philosophical, theological, religious and (...) political aspects of Hobbes's idea of the will and situates Hobbes's doctrine within the context of the most important responses and objections put forward by his critics. Soren Kierkegaard’s Lutheran existentialism represents a distinctively postmetaphysical philosophy of religion, focused in particular on a Christian vision of ethical authenticity. His philosophy continues to pose challenging questions for postmetaphysical philosophers in contemporary pluralistic settings. Focusing on specific works of Kierkegaard, this essay develops three such questions: (1) Can philosophy in a postmetaphysical vein still give advice for the pursuit of the good life, today’s diversity of life styles and values notwithstanding? (2) How can a postmetaphysical philosophy relate to the (...) ethical teachings and truth claims of religious doctrines? Methodenstreit in Economics. This essay offers an account of the Methodenstreit in economics between first Menger and Schmoller and later Max Weber and again Schmoller. It is argued that, for Schmoller, two issues were central; to use economics (widely conceived) as an instrument for economic policy and notably social policy: and to base the science empirically with all the modern methods available. In contrast, the Austrian position had a different view of economics as a science, seeing it more as a (...) system of ideas, which implies a radically different use of empirical evidence. (shrink)

Arming uninhabited vehicles (UVs) is an increasing trend. Widespread deployment can bring dangers for arms-control agreements and international humanitarian law (IHL). Armed UVs can destabilise the situation between potential opponents. Smaller systems can be used for terrorism. Using a systematic definition existing international regulation of armed UVs in the fields of arms control, export control and transparency measures is reviewed; these partly include armed UVs, but leave large gaps. For preventive arms control a general prohibition of armed UVs would be (...) best. If that is unattainable, several measures should be taken. An explicit prohibition of autonomous attack, that is without a human decision, should be added to IHL. Concerning armed UVs remotely controlled by a human soldier, recommendations differ according to type or mission. New kinds of uninhabited nuclear-weapon carriers should be banned. Space weapons should be prohibited in general. UVs smaller than 0.2–0.5 m should be banned. Bigger remotely controlled armed UVs not equipped with weapons of mass destruction should be subject to numerical limitations in various categories. For these the Treaty on Conventional Armed Forces in Europe is an important precedent. (shrink)

In a case study Kuhn's morphology of scientific revolutions is put to the test in confronting it with the contemporary developments in physics. It is shown in detail, that Kuhn's scheme is not compatible with the situation in physics today.

In _The New Order and Last Orientation,_ Eric Voegelin explores two distinctly different yet equally important aspects of modernity. He begins by offering a vivid account of the political situation in seventeenth-century Europe after the decline of the church and the passing of the empire. Voegelin shows how the intellectual and political disorder of the period was met by such seemingly disparate responses as Grotius's theory of natural right, Hobbes's _Leviathan,_ the role of the Fronde in the formation of the (...) French national state, Spinoza's _Tractatus Theologico-Politicus,_ and Locke's _Second Treatise,_ the blueprint of a modern middle-class society. By putting these responses and the thought of Montesquieu, Hume, and others in the context of the birth pains of the national state and the emergence of a new self-understanding of man, Voegelin achieves a brilliant mixture of political history and profound philosophical analysis. Voegelin's verdict of modernity is pronounced most powerfully in the opening part of "Last Orientation," in the chapter entitled "Phenomenalism." His discussion of the intellectual confusion underlying the modern project of scientistic phenomenalism is the most original criticism leveled against modernity to date. It is at the same time the first step toward a recovery of reality through philosophy conceived as a science of substance in the spirit of Giordano Bruno. Voegelin's first example of such an effort at recovering reality is the chapter on Schelling, one of the spiritual realists who has not been affected by the prevailing rationalist or reductionist creeds that are part of the modern disorder. Schelling's indirect yet powerful influence on Schopenhauer, Nietzsche, and Freud more than justifies Voegelin's interest in his philosophy and character, even though Voegelin would later distance himself from some of Schelling's positions. The volume's concluding chapter, "Nietzsche and Pascal," applies the understanding gained from the study of Schelling to the thought of the most powerful critic of the age, Nietzsche. Nietzsche's self-avowed affinity with Pascal provides the key to an analysis of the strengths and weaknesses of his thought and reaffirms the connection that links the beginning of modernity with its most recent crises and the efforts to overcome them.
