The autophagy initiation complex is brought about via a highly ordered and stepwise assembly process. Two crucial signaling molecules, mTORC1 and AMPK, orchestrate this assembly by phosphorylating/dephosphorylating autophagy‐related proteins. Activation of Atg1 followed by recruitment of both Atg9 vesicles and the PI3K complex I to the PAS (phagophore assembly site) are particularly crucial steps in its formation. Ypt1, a small Rab GTPase in yeast cells, also plays an essential role in the formation of the autophagy initiation complex through multiple regulatory (...) pathways. In this review, our primary focus is to discuss how signaling molecules initiate the assembly of the autophagy initiation complex, and highlight the significant roles of Ypt1 in this process. A popular argument against the unilateral appropriation of unowned resources maintains that such appropriation is impossible because it implies a power to unilaterally impose novel obligations on others—a power which people cannot have given that they are moral equals. However, Bas van der Vossen has recently argued that initial appropriation does not create obligations in this way; rather, it merely alters the empirical facts that, together with obligations, determine people’s practical moral requirements. This paper argues that van der Vossen is (...) mistaken. Specifically, it contends that the creation of obligations is accompanied by a distinctive kind of variation in the obliged party’s practical requirements across possible worlds. The paper proposes signaling theory as a framework supportive of a strategic CSR approach; maps extant research on signaling through CSR initiatives; offers a comprehensive assessment of the most diffused CSR initiatives and discusses their eligibility as signaling devices; and outlines a research agenda to further develop and test signaling theory in business ethics. ABSTRACT:Although the literature on multi-stakeholder initiatives for sustainability has grown in recent years, it is scattered across several academic fields, making it hard to ascertain how individual disciplines, such as business ethics, can further contribute to the debate. Based on an extensive review of the literature on certification and principle-based MSIs for sustainability, we show that the scholarly debate rests on three broad themes : theinputinto creating and governing MSIs; theinstitutionalizationof MSIs; and theimpactthat relevant initiatives create. While our discussion reveals (...) the theoretical underpinnings of the 3Is, it also shows that a number of research challenges related to business ethics remain unaddressed. We unpack these challenges and suggest how scholars can utilize theoretical insights in business ethics to push the boundaries of the field. The goal of this article is to differentiate initiation from indoctrination, and to return a positive significance to the notion of initiation, as a pedagogy that contributes not only to the perpetuation of a particular form of life or community, but that provides the next generation with means to advance that knowledge beyond its existing boundaries. When we conflate the terms ‘initiation’ and ‘indoctrination’ or only mark a minor difference between the two, we lose meaning. The explanatory and predictive power (...) of our statements is weakened by this failure to take seriously the difference between these two terms. By ignoring the progressive potential in initiation, and condemning that pedagogy as uncritical, educational theorists fail to recognize the intermediate steps that need to be taken in educating a student to be a creative and responsive thinker within a cultural, i.e. symbolic, context. Within the pedagogy of initiation crucial methods for teaching students to engage existing representations of truth, rules of practice, and principles of meaning-making are employed. This article draws upon case studies and theorists in anthropology to offer a description of initiation that holds progressive potential, and explains the possible relevance of initiatory pedagogy for multicultural education. Educationalists are currently engaging with Jacques Rancière’s thought on emancipation and equality. The focus of this paper is on what initiates the process that starts emancipation. With reference to teachers the question is: how do teachers become emancipated? This paper discusses how the teacher’s life is made ‘sensible’ and how sense is distributed in her life. Two stories are taken from Rancière’s own work, that of Ingrid Bergman and Joseph Jacotot, that give us an indication of the initiation process of (...) emancipation. This article examines initial-condition dependence and initial-condition uncertainty for climate projections and predictions. The first contribution is to provide a clear conceptual characterization of predictions and projections. Concerning initial-condition dependence, projections are often described as experiments that do not depend on initial conditions. Although prominent, this claim has not been scrutinized much and can be interpreted differently. If interpreted as the claim that projections are not based on estimates of the actual initial conditions of the world or that what makes (...) projections true are conditions in the world, this claim is true. However, it can also be interpreted as the claim that simulations used to obtain projections are independent of initial-condition ensembles. This article argues that evidence does not support this claim. Concerning initial-condition uncertainty, three kinds of initial-condition uncertainty are identified. The first is the uncertainty associated with the spread of the ensemble simulations. The second arises because the theoretical initial ensemble cannot be used in calculations and has to be approximated by finitely many initial states. The third uncertainty arises because it is unclear how long the model should be run to obtain potential initial conditions at pre-industrial times. A theoretical development is carried to establish fundamental results about rank-initial embeddings and automorphisms of countable non-standard models of set theory, with a keen eye for their sets of fixed points. These results are then combined into a “geometric technique” used to prove several results about countable non-standard models of set theory. In particular, back-and-forth constructions are carried out to establish various generalizations and refinements of Friedman’s theorem on the existence of rank-initial embeddings between countable non-standard models of the fragment (...) \ + \-Separation of \; and Gaifman’s technique of iterated ultrapowers is employed to show that any countable model of \ can be elementarily rank-end-extended to models with well-behaved automorphisms whose sets of fixed points equal the original model. These theoretical developments are then utilized to prove various results relating self-embeddings, automorphisms, their sets of fixed points, strong rank-cuts, and set theories of different strengths. ABSTRACT:Global multi-stakeholder initiatives are important instruments that have the potential to improve the social and environmental sustainability of global supply chains. However, they often fail to comprehensively address the needs and interests of various supply-chain participants. While voluntary in nature, MSIs have most often been implemented through coercive approaches, resulting in friction among their participants and in systemic problems with decoupling. Additionally, in those cases in which deliberation was constrained between and amongst participants, collaborative approaches have often failed to materialize. (...) Our framework focuses on two key aspects of these breakdowns: assumptions about the orientation of MSI participants, and the deliberation processes that participants use to engage with each other to create these initiatives and sustain them over time. Drawing from stakeholder and deliberation theories, we revisit the concept of MSIs and show how their deliberative capacity may be enhanced in order to encourage participants to collaborate voluntarily. Initiating clinical ethics support in psychiatry and maintaining its continuity appear to be easy. This is contradicted by the observed delay or lack of CESiP, e.g. ethics consultation. On the basis of a published literature search and the discussion of practical experiences over 2.5 years 10 tasks and relating challenges of initiating and maintaining CESiP are formulated and illustrated by examples. Referral to experiences is grounded on the systematic documentation of ca. 100 CESiP activities. The tasks and challenges illustrate how (...) CESiP was initiated and maintained in child and adolescent, adult and forensic psychiatry. Each example is followed by a “rule of thumb” that was found useful in our centre. Discussion: Suggestions I–III are of organisational nature, IV–X have explicit ethical content concerning the ethos or professionalism of the ethics consultant and CES practice. The purpose of the study is to determine the impact of entrepreneurial initiative on the development of innovation activity of country enterprises. The theory of entrepreneurship by clarifying the essence of entrepreneurial initiative got further elaboration in the article. The methodical approach to the analysis and evaluation of the entrepreneurial initiative and its impact on the innovation activity of the enterprises, the results of which are the basis for further development of proposals to stimulate entrepreneurial initiative and development of their (...) innovation activity is suggested. The estimation of innovation activity of 1758 enterprises of Ukraine by calculating the integral indicator of innovation activity revealed its low level for innovation active enterprises. The method of one-dimensional linear regression has determined that an increase in the number of business structures per 10 thousand people of the country's population at a rate of 1.0 causes an increase in the indicator of innovation activity by 0.172. It is proved in the article that development of infrastructure of enterprenerial environment and creation of centers for entrepreneurship development act as an important mechanism of increasing innovation activity of enterprises of the country. We revisit the notion of initial sets by Xu and Cayrol 2016), i. e., non-empty minimal admissible sets in abstract argumentation frameworks. Initial sets are a simple concept for analysing conflicts in an abstract argumentation framework and to explain why certain arguments can be accepted. We contribute with new insights on the structure of initial sets and devise a simple non-deterministic construction principle for any admissible set, based on iterative selection of initial sets of the original framework and its induced (...) reducts. In particular, we characterise many existing admissibility-based semantics via this construction principle, thus providing a constructive explanation on the structure of extensions. We also investigate certain problems related to initial sets with respect to their computational complexity. (shrink)

Initial Judgment of Solvability (iJOS) is a metacognitive judgment that reflects solvers’ first impression as to whether a problem is solvable. We hypothesized that iJOS is inferred by combining prior expectations about the entire task with heuristic cues derived from each problem’s elements. In two experiments participants first provided quick iJOSs for all problems, then attempted to solve them. We manipulated expectations by changing the proportion of solvable problems conveyed to participants, 33%, 50%, or 66%, while the true proportion was (...) the 50% for all. In Experiment 1 we used the non-verbal Raven’s matrices and examined nameability as the element-based heuristic cue. Unsolvable matrices were generated by switching locations of elements in original Raven’s matrices. In Experiment 2 we used the verbal Compound Remote Associate (CRA) problems and examined word’s frequency in the language as the element-based heuristic cue. Unsolvable CRAs were random word triads from the same word pool. The results were consistent in suggesting that quick iJOS integrates prior expectations and experience-based heuristic cues. Notably, iJOS was predictive for the subsequent solving attempt only for Raven’s matrices. (shrink)

Abstract:While a ubiquitous phenomenon, initial public offerings (IPOs) have received no attention in the ethics literature. We provide an overview of a series of potential conflicts of interest that pervade the IPO process. We also report the results of an empirical assessment of IPOs and those elements that may inform a substantive moral hazard faced by key players in the period prior to and just after an IPO.

This article attacks “open systems” arguments that because constant conjunctions are not generally observed in the real world of open systems we should be highly skeptical that universal laws exist. This work differs from other critiques of open system arguments against laws of nature by not focusing on laws themselves, but rather on the inference from open systems. We argue that open system arguments fail for two related reasons; 1) because they cannot account for the “systems” central to their argument (...) (nor the implied systems labeled “exogenous factors” in relation to the system of interest) and 2) they are nomocentric, fixated on laws while ignoring initial and antecedent conditions that are able to account for systems and exogenous factors within a fundamentalist framework. There is a great deal that might be said about justice in property claims. The strategy that I shall employ focuses attention upon the initial acquisition of property -- the most sensitive and most interesting area of property theory. Every theory that discusses property claims favorably assumes that there is some justification for transforming previously unowned resources into property. It is often this assumption which has seemed, to one extent or another, to be vulnerable to attack by critics of particular (...) justifications of property. Nevertheless, this assumption is frequently left undefended by property theorists, and where it is defended, the defense is often remarkably weak. That some initial claim to property be defensible is required by any theory which holds that certain present distributions may be justified, that certain transfers of property are justified, or that restitution ought to be made for previous injustice in transfer or acquisition. This work aims at an initial description of prepositionless genitives in the Romance variety of San Marco in Lamis, spoken in the Southern Italian region of Apulia. The construction will be compared with other Romance, Semitic, Albanian, and Iranian varieties whereby the expression of possession is connected to the presence of D elements, or to morphology stemming from them. The paper deals, in particular, with the behaviour of the construction with elements such as definite and indefinite articles, demonstratives, proper names, (...) and with how pre-nominal adjectival modification of genitives and post-nominal adjectival modification of heads can only occur in the prepositional kind of the construction. This is also the case with demonstratives preceding heads and genitives in the form raised nominals. It will be seen that genitives are only interpreted as such when they are non-raised, i.e. when they are articled. The pre-genitival article is thus understood to be a pivotal element in the interpretation of the second DP as genitival. (shrink)

They were soon published in book form as Authority and the Individual. However, Russell started late in the year to write them, and manuscripts for the lectures show that he encountered (...) difficulty. Surviving in his archives is a false start on the concluding lecture, “How to Promote Initiative” (filed at  .). Lecture  had been called “Control and Initiative: Their Respective Spheres”. Lecture  was finally titled “Individual and Social Ethics”, but an early outline had it as “Principles of Reform”. In the false start Russell described how devolution of authority and individual initiative could be embodied in practice. He provided recipes on how to accomplish this in specific spheres of society: local government, industry, newspapers, books, and education. In replacing the nine leaves of manuscript, he had not come to disagree with them. Instead, as readers of what follows and the final lecture of the book will allow, he now engaged the topic at a higher level. Russell at this time was a friendly critic of the British Labour Party, and his devolutionary reformism is to be seen in that light. Yet his final text transcended politics and engaged his audience at an ethical level, treating of the freedom and duty of conscience, the justifiability of revolution, and life lived as an end. Takes up the questions of the initial bargaining position and the initial factor endowment, raised in Chs. 6 and 4 respectively. We reject James Buchanan's identification of the initial bargaining position with the outcome of non‐cooperative interaction, and John Harsanyi's identification of it with the threat point. Instead, we argue that, as the basis of market and cooperative interaction, it must rule out all taking of advantage—bettering oneself through worsening the situation of others. We term this condition the ‘proviso’, deriving (...) it from Robert Nozick's interpretation of Locke's insistence that in acquisition, one leave ‘enough, and as good’ for others. We show how the proviso constrains directly maximizing behaviour, and we then demonstrate its role in introducing a structure of individual rights to person and property, rights that define each person's initial bargaining position and factor endowment. We show that the proviso satisfies our standard of impartiality, and why rational persons would accept the proviso, as part of a narrow interpretation of compliance with the requirements for cooperation. We show that cooperation in the absence of the proviso tends to be unstable, and to depend on ideologies, entrenched historical factors, and differential technologies, all of which undermine the equal rationality of persons on which an impartial morality depends. BackgroundDecision-making in initiating life-sustaining health technology is complex and often conducted at time-critical junctures in clinical care. Many of these decisions have profound, often irreversible, consequences for the child and family, as well as potential benefits for functioning, health and quality of life. Yet little is known about what influences these decisions. A systematic review of reasoning identified the range of reasons clinicians give in the literature when initiating technology dependence in a child, and as a result helps determine the (...) range of influences on these decisions.MethodsMedline, EMBASE, CINAHL, PsychINFO, Web of Science, ASSIA and Global Health Library databases were searched to identify all reasons given for the initiation of technology dependence in a child. Each reason was coded as a broad and narrow reason type, and whether it supported or rejected technology dependence.Results53 relevant papers were retained from 1604 publications, containing 116 broad reason types and 383 narrow reason types. These were grouped into broad thematic categories: clinical factors, quality of life factors, moral imperatives and duty and personal values; and whether they supported, rejected or described the initiation of technology dependence. The majority were conceptual or discussion papers, less than a third were empirical studies. Most discussed neonates and focused on end-of-life care.ConclusionsThere is a lack of empirical studies on this topic, scant knowledge about the experience of older children and their families in particular; and little written on choices made outside ‘end-of-life’ care. This review provides a sound basis for empirical research into the important influences on a child’s potential technology dependence. (shrink)

Initiation à la philosophie pour les non-philosophes en est le résultat. Mais bien loin d'être un simple ouvrage de vulgarisation ou d'introduction, c'est en réalité un véritable précipité des thèses les plus fondamentales de sa propre pensée qu'offre Althusser. Entre cent autres considérations, nulle part ailleurs (...) ne va-t-il aussi loin dans la distinction entre la philosophie et les autres pratiques ; nulle part non plus ne développe-t-il avec tant de détails le concept de "pratique" lui-même. Pourtant, parmi les nombreux apports cruciaux d'Althusser au "moment philosophique des années 1960", ce concept est peut-être celui qui a bénéficié du plus grand succès et été à la source du plus grand nombre de malentendus. Although debates continue, studies of cognition in infancy suggest that knowledge begins to emerge early in life and constitutes part of humans' innate endowment. Early-developing knowledge appears to be both domain-specific and task-specific, it appears to capture fundamental constraints on ecologically important classes of entities in the child's environment, and it appears to remain central to the commonsense knowledge systems of adults.

We show that in the lattice E Π of Π 0 1 classes there are initial segments [ $\emptyset$ , P] = L(P) which are not Boolean algebras, but which have a decidable theory. In fact, we will construct for any finite distributive lattice L which satisfies the dual of the usual reduction property a Π 0 1 class P such that L is isomorphic to the lattice L(P)*, which is L(P), modulo finite differences. For the 2-element lattice, we obtain (...) a minimal class, first constructed by Cenzer, Downey, Jockusch and Shore in 1993. For the simplest new Π 0 1 class P constructed, P has a single, non-computable limit point and L(P)* has three elements, corresponding to $\emptyset$ , P and a minimal class P $_0 \subset$ P. The element corresponding to P 0 has no complement in the lattice. On the other hand, the theory of L(P) is shown to be decidable. A Π 0 1 class P is said to be decidable if it is the set of paths through a computable tree with no dead ends. We show that if P is decidable and has only finitely many limit points, then L(P)* is always a Boolean algebra. We show that if P is a decidable Π 0 1 class and L(P) is not a Boolean algebra, then the theory of L(P)interprets the theory of arithmetic and is therefore undecidable. (shrink)

Dung’s abstract argumentation provides us with a general framework to deal with argumentation, non-monotonic reasoning and logic programming. For the extension-based semantics, one of the basic principles is I-maximality which is in particular related with the notion of skeptical justification. Another one is directionality which can be employed for the study of dynamics of argumentation. In the old days, material egalitarians tended to favor equality of outcome advantage, on some suitable conception of advantage (happiness, resources, etc.). Under the influence of Dworkin’s seminal articles on equality[i], contemporary material egalitarians have tended to favor equality of brute luck advantage—on the grounds that this permits people to be held appropriately accountable for the benefits and burdens of their choices. I shall argue, however, that a plausible conception of egalitarian justice requires neither that brute luck advantage always be (...) equalized nor that people always bear the full cost of their voluntary choices. Instead, justice requires that initial opportunities for advantage be equalized—roughly along the lines suggested by Arneson and Cohen.[ii] Brute luck egalitarianism and initial opportunity egalitarianism are fairly similar in motivation, and as a result they have not been adequately distinguished. Once the two views are more clearly contrasted, equality of opportunity for advantage will, I claim, be seen to be a more plausible conception of equality. (shrink)

Since the introduction of drugs to prevent vertical transmission of HIV, the purpose of and approach to HIV testing of pregnant women has increasingly become an area of major controversy. In recent years, many strategies to increase the uptake of HIV testing have focused on offering HIV tests to women in pregnancy-related services. New global guidance issued by the World Health Organization (WHO) and the Joint United Nations Programme on HIV/AIDS (UNAIDS) specifically notes these services as an entry point for (...) provider-initiated HIV testing and counseling (PITC). The guidance constitutes a useful first step towards a framework within which PITC sensitive to health, human rights and ethical concerns can be provided to pregnant women in health facilities. However, a number of issues will require further attention as implementation moves forward. It is incumbent on all those involved in the scale up of PITC to ensure that it promotes long-term connection with relevant health services and does not result simply in increased testing with no concrete benefits being accrued by the women being tested. Within health services, this will require significant attention to informed consent, pre- and post-test counseling, patient confidentiality, referrals and access to appropriate services, as well as reduction of stigma and discrimination. Beyond health services, efforts will be needed to address larger societal, legal, policy and contextual issues. The health and human rights of pregnant women must be a primary consideration in how HIV testing is implemented; they can benefit greatly from PITC but only if it is carried out appropriately. Following the Second Summit on Human Gene Editing in Hong Kong in 2018, where the birth of two girls with germline genome editing was revealed, the need for a responsible pathway to the clinical application of human germline genome editing has been repeatedly emphasised. This paper aims to contribute to the ongoing discussion on research ethics issues in germline genome editing by exploring key issues related to the initial applications of CRISPR in reproductive medicine. Following an overview of the current (...) discussion on bringing germline genome editing into clinical practice, we outline the specific challenges associated with such interventions and the features that distinguish them from conventional clinical testing of new medical treatments. We then review proposed ethical requirements for initial heritable genome editing, such as the absence of reasonable alternatives, the existence of sufficient and reliable preclinical data, appropriate informed consent, requirements related to safety, and long-term follow-up. In 2016 the Chinese government initiated a nationwide campaign aiming to eliminate poverty in China by 2020. Over 20% of listed firms in China have made significant contributions to the campaign. Using hand-collected data on listed firms’ contributions to the campaign and multivariate analyses, we examine whether managers’ and politicians’ personal incentives play an important role in firms’ contributions to the campaign. The results show that firms are more likely to contribute if they are state-owned and managers are appointed by (...) governments, if managers have a higher risk of being targeted in a concurrent anti-corruption campaign, if the political leaders in their province are new or intend to seek promotion, and if managers or directors have experienced poverty in their early life. The results suggest that it is important to consider managers’ and politicians’ personal incentives in CSR activities that could have a grand social impact. (shrink)

A distinct group of receptors including DCC, UNC5, RET and Ptc1 is known to function in ligand‐dependent neuronal growth and differentiation or axon guidance. Acting as “dependence receptors”, they may also regulate neuronal cell survival by inducing apoptosis in the absence of cognate ligand. Receptor‐initiated apoptosis requires proteolytic (caspase) cleavage and exposure of a pro‐apoptotic region in the cytoplasmic domains of the receptors. In contrast, classical apoptosis induced by growth factor or cytokine deprivation involves loss of survival signaling without receptor (...) cleavage. DCC, UNC5, RET and Ptc1 are downregulated or mutated in diverse cancers, and show properties characteristic of tumor suppressors, consistent with their ability to promote neuronal cell death. Dysfunctional dependence receptors have been linked to the loss of specific neurons in certain inherited and neurodegenerative diseases. Dependence receptor‐initiated apoptosis represents a novel paradigm for the controlled removal of specific cells during neural development and elimination of malignant cells that have strayed beyond regions of ligand availability. Peer interaction has been found to be conducive to learning in many settings. Knowledge co-construction has been proposed as one explanatory mechanism. However, KCC is a theoretical construct that is too abstract to guide the development of instructional software that can support peer interaction. In this study, we present an extensive analysis of a corpus of peer dialogs that we collected in the domain of introductory Computer Science. We show that the notion of task initiative shifts correlates with both KCC (...) and learning. Speakers take task initiative when they contribute new content that advances problem solving and that is not invited by their partner; if initiative shifts between the partners, it indicates they both contribute to problem solving. We found that task initiative shifts occur more frequently within KCC episodes than outside. In addition, task initiative shifts within KCC episodes correlate with learning for low pre-testers, and total task initiative shifts correlate with learning for high pre-testers. As recognizing task initiative shifts does not require as much deep knowledge as recognizing KCC, task initiative shifts as an indicator of productive collaboration are potentially easier to model in instructional software that simulates a peer. ABSTRACTThis article studies multi-stakeholder initiatives as spaces for both deliberation and contestation between constituencies with competing discourses and disputed values, beliefs, and preferences. We review different theoretical perspectives on MSIs, which see them mainly as spaces to find solutions to market problems, as spaces of conflict and bargaining, or as spaces of consensus. In contrast, we build on a contestatory deliberative perspective, which gives equal value to both contestation and consensus. We identify four types of internal contestation which can be (...) present in MSIs—procedural, inclusiveness, epistemic, and ultimate-goal—and argue that embracing contestation and engaging in ongoing revision of provisional agreements, criteria, and goals can enhance the democratic quality of MSIs. Finally, we explore the implications of this perspective for theorizing about the democratic quality in MSIs and about the role of corporations in transnational governance. (shrink)

A rich literature in bioethics argues against the use of anonymous gamete donation in the name of the ‘interest in knowing one’s genetic origins’. This interest stems from medical as well as psychosocial and identity reasons. The term ‘genealogical bewilderment’ has been coined to express the predicament of those deprived of access to information about their origins. Another rich body of literature in bioethics discusses arguments for and against posthumous-assisted reproduction (PAR), with a recent focus on PAR that is initiated (...) by the parents of a deceased man (rather than his partner). This paper revisits arguments against PAR, in light of arguments regarding the interest in knowing one’s genetic origins. Limiting the discussion to the specific context of parent-initiated PAR (PIPAR), we argue that the use of cryopreserved sperm from a deceased identifiable man in the context of PIPAR may be ethically preferable when compared with the use of anonymous donor sperm, since it allowsgenealogical certainty, that is, giving the prospective child access to information about the identity, life story and ancestry of the genetic progenitor as well asgenealogical embeddedness, that is, close relationship with extended family members. There is a need to rapidly assess the impact of new technology initiatives on the Counter Improvised Explosive Device battle in Iraq and Afghanistan. The immediate challenge is the need for rapid decisions, and a lack of engineering test data to support the assessment. The rapid assessment methodology exploits available information to build a probabilistic model that provides an explicit executable representation of the initiative’s likely impact. The model is used to provide a consistent, explicit, explanation to decision makers on (...) the likely impact of the initiative. Sensitivity analysis on the model provides analytic information to support development of informative test plans. (shrink)
