Integration of knowledge concerning circadian rhythms, metabolic networks, and sleep‐wake cycles is imperative for unraveling the mysteries of biological cycles and their underlying mechanisms. During the last decade, enormous progress in circadian biology research has provided a plethora of new insights into the molecular architecture of circadian clocks. However, the recent identification of autonomous redox oscillations in cells has expanded our view of the clockwork beyond conventional transcription/translation feedback loop models, which have been dominant since the first circadian period mutants (...) were identified in fruit fly. Consequently, non‐transcriptional timekeeping mechanisms have been proposed, and the antioxidant peroxiredoxin proteins have been identified as conserved markers for 24‐hour rhythms. Here, we review recent advances in our understanding of interdependencies amongst circadian rhythms, sleep homeostasis, redox cycles, and other cellular metabolic networks. We speculate that systems‐level investigations implementing integrated multi‐omics approaches could provide novel mechanistic insights into the connectivity between daily cycles and metabolic systems. -/- . To examine whether anticipatory attention or expectancy is a cognitive process that is automatic or requires conscious control, we employed a paired-stimulus event-related potential paradigm during the transition to sleep. The slow negative ERP wave observed between two successive stimuli, the Contingent Negative Variation , reflects attention and expectancy to the second stimulus. Thirteen good sleepers were instructed to respond to the second stimulus in a pair during waking sessions. In a non-response paradigm modified for sleep, participants then fell asleep (...) while tones played. As expected, N1 decreased and P2 increased in amplitude systematically with the loss of consciousness at sleep onset; the CNV was increasingly more positive. Sleep onset latency was correlated with the amplitude of the CNV. The systematic attenuation of the CNV waveform at sleep onset and its absence in sleep indicates that anticipatory attention requires endogenous conscious control. (shrink)

Chronic pain is one of the most disabling medical conditions globally, yet, to date, we lack a satisfying theoretical framework for research and clinical practice. Over the prior decades, several frameworks have been presented with biopsychosocial models as the most promising. However, in translation to clinical practice, these models are often applied in an overly reductionist manner, leaving much to be desired. In particular, they often fail to characterize the complexities and dynamics of the lived experience of chronic pain. Recently, (...) an enactive, affordance-based approach has been proposed, opening up new ways to view chronic pain. This model characterizes how the persistence of pain alters a person’s field of affordances: the unfolding set of action possibilities that a person perceives as available to them. The affordance-based model provides a promising perspective on chronic pain as it allows for a systematic investigation of the interactive relation between patients and their environment, including characteristic alterations in the experience of their bodies and the space they inhabit. To help bridge the gap from philosophy to clinical practice, we unpack in this paper the core concepts of an affordance-based approach to chronic pain and their clinical implications, highlighting aspects that have so far received insufficient attention. We do so with an analogy to playing video games, as we consider such comparative illustration a useful tool to convey the complex concepts in an affordance-based model and further explore central aspects of the lived experience of chronic pain. The present review highlights an association between autism, Alzheimer disease , and fragile X syndrome . We propose a conceptual framework involving the amyloid-beta peptide , Abeta precursor protein , and fragile X mental retardation protein based on experimental evidence. The anabolic effect of the secreted alpha form of the amyloid-beta precursor protein may contribute to the state of brain overgrowth implicated in autism and FXS. Our previous report demonstrated that higher plasma sAPPalpha levels associate with more severe symptoms of (...) autism, including aggression. This molecular effect could contribute to intellectual disability due to repression of cell-cell adhesion, promotion of dense, long, thin dendritic spines, and the potential for disorganized brain structure as a result of disrupted neurogenesis and migration. At the molecular level, APP and FMRP are linked via the metabotropic glutamate receptor 5 . Specifically, mGluR5 activation releases FMRP repression of APP mRNA translation and stimulates sAPP secretion. The relatively lower sAPPalpha level in AD may contribute to AD symptoms that significantly contrast with those of FXS and autism. Low sAPPalpha and production of insoluble Abeta would favor a degenerative process, with the brain atrophy seen in AD. Treatment with mGluR antagonists may help repress APP mRNA translation and reduce secretion of sAPP in FXS and perhaps autism. (shrink)

In a continuation of the conversation with Fitch, Chomsky, and Hauser on the evolution of language, we examine their defense of the claim that the uniquely human, language-specific part of the language faculty (the “narrow language faculty”) consists only of recursion, and that this part cannot be considered an adaptation to communication. We argue that their characterization of the narrow language faculty is problematic for many reasons, including its dichotomization of cognitive capacities into those that are utterly unique and those (...) that are identical to nonlinguistic or nonhuman capacities, omitting capacities that may have been substantially modified during human evolution. We also question their dichotomy of the current utility versus original function of a trait, which omits traits that are adaptations for current use, and their dichotomy of humans and animals, which conflates similarity due to common function and similarity due to inheritance from a recent common ancestor. We show that recursion, though absent from other animals’ communications systems, is found in visual cognition, hence cannot be the sole evolutionary development that granted language to humans. Finally, we note that despite Fitch et al.’s denial, their view of language evolution is tied to Chomsky’s conception of language itself, which identifies combinatorial productivity with a core of “narrow syntax.” An alternative conception, in which combinatoriality is spread across words and constructions, has both empirical advantages and greater evolutionary plausibility. q 2005 Elsevier B.V. Article history: This article sketches the Parallel Architecture, an approach to the structure of grammar that Accepted 29 August 2006 contrasts with mainstream generative grammar (MGG) in that (a) it treats phonology, Available online 13 October 2006 syntax, and semantics as independent generative components whose structures are linked by interface rules; (b) it uses a parallel constraint-based formalism that is nondirectional; (c) Keywords: it treats words and rules alike as pieces of linguistic structure stored in long-term memory.

Formal theories of mental representation have receded from the importance they had in the early days of cognitive science. I argue that such theories are crucial in any mental domain, not just for their own sake, but to guide experimental inquiry, as well as to integrate the domain into the mind as a whole. To illustrate the criteria of adequacy for theories of mental representation, I compare two theoretical approaches to language: classical generative grammar (Chomsky, 1965, 1981, 1995) and the (...) parallel architecture (Jackendoff, 1997, 2002). The grounds for comparison include (a) the internal coherence of the theory across phonology, syntax, and semantics; (b) the relation of language to other mental faculties; (c) the relationship between grammar and lexicon; (d) relevance to theories of language processing; and (e) the possibility of languages with little or no syntax. Social conflicts of ever widening scope have in recent years emphasized the importance of the problem of the relation of facts and values. This problem has received increasing attention from researchers and theorists in both the physical and social sciences. A number of interesting but by no means compatible solutions have been proposed.Perhaps the simplest and most striking is the position of Carnap, Russell, and others, that value sentences, such as “A ought not to kill B” or “Killing is evil” (...) are merely commands or expressions of wish and, as such, are emotional ejaculations or expletives but not statements which have any theoretical sense or enter into science. On this view the problem of the relation of fact and value statements is meaningless, or at least is not worth considering further. (shrink)

In his classic 1936 essay "On the Concept of Logical Consequence", Alfred Tarski used the notion of satisfaction to give a semantic characterization of the logical properties. Tarski is generally credited with introducing the model-theoretic characterization of the logical properties familiar to us today. However, in his book, The Concept of Logical Consequence, Etchemendy argues that Tarski's account is inadequate for quite a number of reasons, and is actually incompatible with the standard model-theoretic account. Many of his criticisms are meant (...) to apply to the model-theoretic account as well. In this paper, I discuss the following four critical charges that Etchemendy makes against Tarski and his account of the logical properties: (1) (a) Tarski's account of logical consequence diverges from the standard model-theoretic account at points where the latter account gets it right. (b) Tarski's account cannot be brought into line with the model-theoretic account, because the two are fundamentally incompatible. (2) There are simple counterexamples (enumerated by Etchemendy) which show that Tarski's account is wrong. (3) Tarski committed a modal fallacy when arguing that his account captures our pre-theoretical concept of logical consequence, and so obscured an essential weakness of the account. (4) Tarski's account depends on there being a distinction between the "logical terms" and the "non-logical terms" of a language, but (according to Etchemendy) there are very simple (even first-order) languages for which no such distinction can be made. Etchemendy's critique raises historical and philosophical questions about important foundational work. However, Etchemendy is mistaken about each of these central criticisms. In the course of justifying that claim, I give a sustained explication and defense of Tarski's account. Moreover, since I will argue that Tarski's account and the model theoretic account really do come to the same thing, my subsequent defense of Tarski's account against Etchemendy's other attacks doubles as a defense against criticisms that would apply equally to the familiar model-theoretic account of the logical properties. Maximization theory, which is borrowed from economics, provides techniques for predicing the behavior of animals - including humans. A theoretical behavioral space is constructed in which each point represents a given combination of various behavioral alternatives. With two alternatives - behavior A and behavior B - each point within the space represents a certain amount of time spent performing behavior A and a certain amount of time spent performing behavior B. A particular environmental situation can be described as a constraint (...) on available points (a circumscribed area) within the space. Maximization theory assumes that animals always choose the available point with the highest numerical value. The task of maximization theory is to assign to points in the behavioral space values that remain constant across various environmental situations; as those situations change, the point actually chosen is always the one with the highest assigned value. The two differ in many aspects of structure and function, and, with the exception of the metrical grid, all aspects they share appear to be instances of more general capacities.

The aim of the paper is twofold: i) to give a logically explicit formulation of a slight generalization of Quine's master argument about de re modality—an argument which imposes important constraints on modal semantics, ii) to briefly present my favored account of modal locutions (especially locutions of the de re metaphysical flavor) and show how it successfully copes with Quine's argument. Though Quine made this argument so many years ago, it is still widely misunderstood, and so careful attention to detail (...) seems warranted. From what I have seen, philosophers' attitudes towards Quine's master argument fall into two kinds: i) there are those that think that the argument has no force, because it is based on some mistake (usually, something about definite descriptions), and ii) there are those that think that the argument poses some insuperable barrier to any kind of de re modality. Neither of these attitudes is justified. So, I hope to make plain along the way that a) the original version of Quine's argument is sound, b) there is a version of this same basic argument which imposes very definite constraints on any proposed account of de re "metaphysical" modality in particular, an c) there is an account that satisfies these constraints. Part 1 of the paper is concerned with laying out and discussing three versions of Quine's argument, in the service of establishing points (a) and (b). -/- In Part 2 of the paper, I briefly sketch what I take to be a very promising, and also very Quinean account of de re modality -- one that respects the constraint on modal semantics that Quine's argument reveals and comports well with the few positive remarks Quine makes, for example, in Word and Object regarding our use of modal locutions. This will put us in a position to see that the proposed account does not fly in the face of Quine's master argument. (shrink)
