Bacteria use trans‐translation to rescue stalled ribosomes and target incomplete proteins for proteolysis. Despite similarities between tRNAs and transfer‐messenger RNA (tmRNA), the key molecule for trans‐translation, new structural and biochemical data show important differences between translation and trans‐translation at most steps of the pathways. tmRNA and its binding partner, SmpB, bind in the A site of the ribosome but do not trigger the same movements of nucleotides in the rRNA that are required for codon recognition by tRNA. tmRNA‐SmpB moves from (...) the A site to the P site of the ribosome without subunit rotation to generate hybrid states, and moves from the P site to a site outside the ribosome instead of to the E site. During catalysis, transpeptidation to tmRNA appears to require the ribosomal protein bL27, which is dispensable for translation, suggesting that this protein may be conserved in bacteria due to trans‐translation. These differences provide insights into the fundamental nature of trans‐translation, and provide targets for new antibiotics that may have decrease cross‐reactivity with eukaryotic ribosomes. Coronavirus disease 2019 is a fast-developing viral pandemic spreading across the globe. Due to lack of availability of proven medicines against COVID-19, physicians have resorted to treatments through large trials of investigational drugs with poor evidence or those used for similar diseases. Large trials randomize 100–500+ patients at multiple hospitals in different countries to either receive these drugs or standard treatment. In order to expedite the process, some regulatory agencies had also given permission to use drugs approved for other diseases, (...) despite a lack of evidence of efficacy in COVID-19. In this review, we highlight the potential ethical issues that should be addressed during the use of investigational drugs with little prior evidence as a treatment options during a pandemic. We discuss the impact of design of randomized clinical trials using LOTUS trial as an example and that of off-label use of drugs like chloroquine/hydroxychloroquine on the safety of patients during COVID-19. We conclude that the adaptive randomized clinical trial designs offer a flexible and efficient approach, enabling patients to quickly switch to successful treatments, while minimizing the number of patients on standard of care. Randomized clinical trial design should consider blinding of investigators and only representative patients who can provide consent should be included. We also conclude the emergency approvals of drugs should be carefully issued and off-label use should be restricted in pandemics. Streamlined regulatory guidelines for emergency drug use in a pandemic can also help in providing benefit and minimizing harm to patients in the future. (shrink)

ABSTRACTAdults use a variety of strategies to reason about fraction magnitudes, and this variability is adaptive. In two studies, we examined the relationships between mathematics anxiety, working memory, strategy variability and performance on two fraction tasks: fraction magnitude comparison and estimation. Adults with higher mathematics anxiety had lower accuracy on the comparison task and greater percentage absolute error on the estimation task. Unexpectedly, mathematics anxiety was not related to variable strategy use. However, variable strategy use was linked to more accurate (...) magnitude comparisons, especially among adults with lower working memory performance or those who use mathematics less frequently, as well as lower PAE on the estimation task. These findings shed light on the role of strategy variability in fraction problem solving and demonstrate a link between mathematics anxiety and fraction magnitude reasoning, a key predictor of general mathematics achievement. Earlier evidence suggests that besides humans, some species of mammals and birds demonstrate visual self-recognition, assessed by the controversial “mark” test. Whereas, there are high levels of inter-individual differences amongst a single species, some species such as macaques and pigeons which do not spontaneously demonstrate mirror self-recognition can be trained to do so. We were surprised to discover that despite being widely used as a model system for avian research, the performance of zebra finches on the mark test had not (...) been studied earlier. Additionally, we studied the behavioral responses of another species of passerine songbirds to a mirror and the MSR mark test. Although a small number of adult male zebra finches appeared to display heightened responses toward the mark while observing their reflections, we could not rule out the possibility that these were a part of general grooming rather than specific to the mark. Furthermore, none of the house crows demonstrated mark-directed behavior or increased self-exploratory behaviors when facing mirrors. Our study suggests that self-directed behaviors need to be tested more rigorously in adult male zebra finches while facing their reflections and these findings need to be replicated in a larger population, given the high degree of variability in mirror-directed behaviors. Eukaryotic genomes are packaged into nucleosomal chromatin, and genomic activity requires the precise localization of transcription factors, histone modifications and nucleosomes. Classic work described the progressive reassembly and maturation of bulk chromatin behind replication forks. More recent proteomics has detailed the molecular machines that accompany the replicative polymerase to promote rapid histone deposition onto the newly replicated DNA. However, localized chromatin features are transiently obliterated by DNA replication every S phase of the cell cycle. Genomic strategies now observe the rebuilding (...) of locus‐specific chromatin features, and reveal surprising delays in transcription factor binding behind replication forks. This implies that transient chromatin disorganization during replication is a central juncture for targeted transcription factor binding within genomes. We propose that transient occlusion of regulatory elements by disorganized nucleosomes during chromatin maturation enforces specificity of factor binding. (shrink)

Cloud computing paradigm is growing rapidly, and it allows users to get services via the Internet as pay-per-use and it is convenient for developing, deploying, and accessing mobile applications. Currently, security is a requisite concern owning to the open and distributed nature of the cloud. Copious amounts of data are responsible for alluring hackers. Thus, developing efficacious IDS is an imperative task. This article analyzed four intrusion detection systems for the detection of attacks. Two standard benchmark datasets, namely, NSL-KDD and (...) UNSW-NB15, were used for the simulations. Additionally, this study highlights the proliferating challenges for the security of sensitive user data and gives useful recommendations to address the identified issues. Finally, the projected results show that the hybridization method with support vector machine classifier outperforms the existing techniques in the case of the datasets investigated. (shrink)

Spirituality-at-work is a new movement in the United States that links spirituality with business. It seeks to usher in a values-based environment wherein individuals can experience meaning and purpose in life by functioning holistically—as physical, intellectual, emotional and spiritual beings. Several examples are cited of companies actively working towards achieving such a climate, and their success in meeting social as well as fiscal responsibilities. The paper traces some of the undercurrents that have led to this new movement—rightsizing, labour force shortages, (...) entry of women, concerns for ecology and human collectivity, society's renewed interest in religion, and new thinking on the essence of leadership. The paper concludes with an examination of the likely future of the movement. (shrink)

The Sai Baba movement, one of the most widespread and popular modern South Asian religious movements, owes its origin to a saint, Sai Baba of Shirdi (d.1918), who was probably born around 1838. Through his successor, Sathya Sai Baba (b. 1926), the movement has become a transnational phenomenon in the late twentieth century and has also expanded the main centers of its charisma, including today Shirdi town in the Indian state of Maharashtra and Puttaparthi town in the neighboring state of (...) Andhra Pradesh. While most of the available literature is hagiographical in nature, some aspects of the movement have been studied - the figures of Shirdi Sai Baba and Sathya Sai Baba, the middle-class constituency of the Sathya Sai Baba movement, role of miracles, and the pedagogical role of movement, for instance. These studies are part of a growing interest in new religious and reformist movements in South Asia in the past century, their impacts on civil society and its institutions, and their relationships to the nation-state. Biobank-based research is not specifically addressed in Indian statutory law and therefore Indian Council for Medical Research guidelines are the primary regulators of biobank research in India. The guidelines allow for broad consent and for any level of identification of specimens. Although privacy is a fundamental right under the Indian Constitution, courts have limited this right when it conflicts with other rights or with the public interest. Furthermore, there is no established privacy test or actionable privacy right in the common (...) law of India. In order to facilitate biobank-based research, both of these lacunae should be addressed by statutory law specifically addressing biobanking and more directly addressing the accompanying privacy concerns. A biobank-specific law should be written with international guidelines in mind, but harmonization with other laws should not be attempted until after India has created a law addressing biobank research within the unique legal and cultural environment of India. Unknown to most Western psychologists, ancient Indian scriptures contain very rich, empirically derived psychological theories that are, however, intertwined with religious and philosophical content. This article represents our attempt to extract the psychological theory of cognition and consciousness from a prominent ancient Indian thought system: Samkhya-Yoga. We derive rather broad hypotheses from this approach that may complement and extend Western mainstream theorizing. These hypotheses address an ancient personality theory, the effects of practicing the applied part of Samkhya-Yoga on normal and (...) extraordinary cognition, as well as different ways of perceiving reality. We summarize empirical evidence collected in diverse fields of research that allows for making judgments about the hypotheses, and suggest more specific hypotheses to be examined in future research. We conclude that the existing evidence for the hypotheses is substantial but that there are still considerable gaps in theory and research to be filled. Theories of cognition contained in the ancient Indian systems have the potential to modify and complement existing Western mainstream accounts of cognition. In particular, they might serve as a basis for arriving at more comprehensive theories for several research areas that, so far, lack strong theoretical grounding, such as meditation research or research on aspects of consciousness. . Epochs present in the voiced speech are defined as time instants of significant excitation of the vocal tract system during the production of speech. Nonstationary nature of excitation source and vocal tract system makes accurate identification of epochs a difficult task. Most of the existing methods for epoch detection require prior knowledge of voiced regions and a rough estimation of pitch frequency. In this paper, we propose a novel method that relies on time-order representation based on short-time Fourier–Bessel series (...) expansion which can be employed on entire speech signal to detect epochs without any prior information. The proposed method automatically detects voiced regions in the speech signal by computing the marginal energy density with respect to time in the low frequency range from the energy distribution in the time-frequency plane. An estimate of pitch frequency for each detected voiced region is then obtained by computing the marginal energy density with respect to frequency in the LFR from the energy distribution in the time-frequency plane. Epochs are located for each detected voiced region as peaks in the derivative of the low pass filtered signal corresponding to falling edges of peak negative cycles in the LPF signal synthesized from TOR coefficients corresponding to LFR. Experimental results obtained by the proposed method on speech signals taken from the CMU-Arctic database are found to be promising. The proposed method detects epochs with high accuracy and reliability. (shrink)

While the school of analytical philosophy of education is often credited for providing Philosophy of Education with a sense of identity, purpose and academic respect and for marking out a legitimate field in which philosophy of education could lay claim to funds and positions in competition with sociologists and psychologists, it has not escaped falling prey to criticism from different schools of thoughts. Whereas analytical traditions propound the idea of liberal education as value free, neutral and irrespective of the context (...) in which the concepts are embedded, Marxist school of thought criticizes the tradition for serving the propaganda of liberal education. Analytical philosophy of education, according to Marxists, is the educational representative of a liberal rationalist world view which has political, economic, historical and cultural manifestations. The paper presents a Marxist critique of the School of Analytical Philosophy of Education as discussed by Michael R. Making transformative services such as healthcare accessible to low-income consumers is an ethical challenge of vital importance to marketers. However, most low-income consumers across the world are excluded from the market for such transformative services because of financial constraints arising from poverty. In this paper, instead of focusing on the micro-interplay between firms and consumers, we examine the macro-interplay among firms, consumers, and public policy in addressing the ethical challenge of market inclusion at the base of the pyramid. Specifically, we (...) examine how the Vietnam government used a policy of free and universal health insurance for children under the age of six as a means of lowering affordability barriers and fostering market inclusion in the healthcare market. Overnight in 2005, all children under the age of six living anywhere in Vietnam became eligible for free health insurance. Using this policy intervention as a natural experiment, we compare market inclusion outcomes of children under the age of six with older children who were ineligible before and after the program was implemented. We show that lowering affordability barriers through public policy increases access to target services, increases consumers’ overall out-of-pocket spending, and increases access to complementary services. By adopting a macromarketing lens, this study makes a strong case for collaboration among firms, governments, and communities in addressing the ethical challenge of system-wide market inclusion in base-of-the-pyramid markets. However, the FDA was increasingly criticized during the Bush administration for politicizing decisions such as rejecting an application to grant the “morning after pill,” known as Plan B, over-the-counter status (...) for women of all ages. After an atypical approval process, the FDA approved OTC Plan B use only for women over the age of 18. Because some groups perceive the morning after pill as a form of abortion, the politically appointed leadership of the FDA was suspected of dramatically departing from normal FDA procedures in order to ensure that the application would not be approved.
