Transcription factor (TF) signaling regulates gene transcription and requires a complex network of proteins. This network includes co‐activators, co‐repressors, multiple TFs, histone‐modifying complexes, and the basal transcription machinery. It has been widely appreciated that pioneer factors, such as FoxA1 and GATA1, play an important role in opening closed chromatin regions, thereby allowing binding of a secondary factor. In this review we will focus on a newly proposed model wherein multiple TFs, such as steroid receptors (SRs), can function in a pioneering (...) role. This model, termed dynamic assisted loading, integrates data from widely divergent methodologies, including genome wide ChIP‐Seq, digital genomic footprinting, DHS‐Seq, live cell protein dynamics, and biochemical studies of ATP‐dependent remodeling complexes, to present a real time view of TF chromatin interactions. Under this view, many TFs can act as initiating factors for chromatin landscape programming. Furthermore, enhancer and promoter states are more accurately described as energy‐dependent, non‐equilibrium steady states. (shrink)

This work proposes an explanation of the Pioneer anomaly, the unmodelled and as yet unexplained blueshift detected in the microwave signal of the Pioneer 10 and other spaceships by Anderson et al. in 1998. What they observed is similar to the effect that would have either (i) an anomalous acceleration a P the ship towards the Sun, or (ii) an acceleration of the clocks a t =a P /c. The second alternative is investigated here, with a phenomenological model in which (...) the anomaly is an effect of the background gravitational potential Ψ (t) that pervades all the universe and is increasing because of the expansion. It is shown that 2a t =dΨ /dt=d 2τ clocks /dt 2, evaluated at present time t 0, where t and τ clocks are the coordinate time and the time measured by the atomic clocks, respectively. The result of a simple estimate gives the value a t ⋍ 1.8× 10−18 s−1, while Anderson et al. suggested a t =(2.9±0.4) × 10−18 s−1 on the basis of their observations. The calculation are performed near the Newtonian limit but in the frame of general relativity. (shrink)

In 1846 the Belgian scientist François Donny published a very important but still virtually unknown paper. Apparently the first publication on the ability of liquids to withstand tension, it appeared some four years before Marcelin Berthelot's famous paper on this property of liquids. It seems only fair to record the fact that, contrary to accepted belief, Donny, not Berthelot, was really the first pioneer in this field. In his paper Donny describes how he first observed by chance the fact that (...) a column of sulphuric acid in a manometer could sustain a small tension. This led him to investigate the matter further, and he found that a column of this acid could stand a tension of about a quarter of an atmosphere. From later experiments he concluded that a column of water could sustain at least one atmosphere of tension. The work described in Donny's paper is discussed and compared with the related work of Marcelin Berthelot and Osborne Reynolds which was published later in the nineteenth century. “Clinical ethics consultants” have been practicing in the United States for about 50 years. Most of the earliest consultants—the “pioneers”—were “outsiders” when they first appeared at patients' bedsides and in the clinic. However, if they were outsiders initially, they acclimated to the clinical setting and became “insiders” very quickly. Moreover, there was some tension between traditional academics and those doing applied ethics about whether there was sufficient “critical distance” for appropriate reflection about the complex medical ethics dilemmas of the (...) day if one were involved in the decision making. Again, the pioneers deflected concerns by identifying and instituting safeguards to assure professional objectivity in clinical ethics consultation services. One might suggest that in moving inside and establishing normative practices, the pioneer clinical ethics consultants anticipated adoption of their routines and professionalization of the field. Ptolemy of Lucca's sympathetic description of republican self-rule together with his unfavourable view of monarchy in the De regimine principum has led many scholars to categorize him as a pioneer of civic republicanism. The present study refutes this common opinion. It illuminates Ptolemy's theory of government through its relationship to the papalism he repeatedly expressed in several works. This study argues that Ptolemy's theory of government in the De regimine principum was inspired by his papalist convictions, and demonstrates how that (...) theory clearly reflected Ptolemy's papalist point of view, especially in the context of the feud between Pope Boniface VIII and the kings of Western Europe, in particular, Philip the Fair of France. Precise DNA replication is fundamental for genetic inheritance. In eukaryotes, replication initiates at multiple origins that are first “licensed” and subsequently “fired” to activate DNA synthesis. Despite the success in identifying origins with specific DNA motifs in Saccharomyces cerevisiae, no consensus sequence or sequences with a predictive value of replication origins have been recognized in metazoan genomes. Rather, epigenetic rules and chromatin structures are believed to play important roles in governing the selection and activation of replication origins. We propose that (...) replication initiation is facilitated by a group of sequence‐specific “replication pioneer factors,” which function to increase chromatin accessibility and foster a chromatin environment that is conducive to the loading of the prereplication complex. Dysregulation of the function of these factors may lead to gene duplication, genomic instability, and ultimately the occurrence of pathological conditions such as cancer. (shrink)

Shortly before and during World War II many European psychoanalysts found refuge in South America, concentrated in Buenos Aires. Here, together with local professionals, they created a strong, creative and productive psychoanalytic movement that in turn gave birth to theoretical and clinical contributions that transformed psychoanalysis, psychology, medicine and culture in South America. _The Pioneers of Psychoanalysis in South America_ is a collection of those pioneers’ papers, and introduces the reader to a body of ideas and advancements, many (...) of which have had limited and piecemeal exposure within the psychoanalytic community in the rest of the world until now. Justification of theorems plays a vital role in any rational human activity. It is indispensable in science. The deductive method of justifying theorems is used in all sciences and it is the only method of justifying theorems in deductive disciplines. It is based on the notion of proof, thus it is a method of proving theorems. In the Warsaw School of Logic (WSL) – the famous branch of the Lvov-Warsaw School (LWS) – two types of the method: axiomatic deduction method (...) and natural deduction method were developed and practiced. In this paper, both of these methods are briefly discussed with an emphasis on their historical, groundbreaking significance for logic. The axiomatic method by means of rejection (proposed by Jan Łukasiewicz – a co-creator of the WSL), which is the method of the so-called rejection proof (rejection/refutation method) in logical systems and the proving method of generalized natural deduction, which is a hybrid deduction–refutation method of proving theorems, are also outlined in the paper. The author discusses their historical significance. This paper also contains a brief mention of the most significant results which the application of the discussed methods introduced into contemporary scientific research, not only logical one. We argue that in implanted neurotechnology research, participants and researchers experience what Henry Richardson has called “moral entanglement.” Participants partially entrust researchers with access to their brains and thus to information that would otherwise be private, leading to created intimacies and special obligations of beneficence for researchers and research funding agencies. One of these obligations, we argue, is about continued access to beneficial technology once a trial ends. We make the case for moral entanglement in this context through exploration of (...) participants’ vulnerability, uncompensated risks and burdens, depth of relationship with the research team, and dependence on researchers in implanted neurotechnology trials. The notion of a migration system is often invoked but it is rarely clearly defined or conceptualized. De Haas recently provided a powerful critique of the current literature highlighting some important flaws that recur through it. In particular, migration systems tend to be identified as fully formed entities, and there is no theorization as to how they come into being and how they break down. The internal dynamics which drive such changes are not examined. Such critiques of migration systems relate (...) to wider critiques of the concept of systems in the broader social science literature, where they are often presented as black boxes in which human agency is largely excluded. The challenge is how to theorize system dynamics in which the actions of people at one time contribute to the emergence of systemic linkages at a later time. This article focuses on the genesis of migration systems and the notion of pioneer migration. It draws attention both to the role of particular individuals, the pioneers, and also the more general activity of pioneering which is undertaken by many migrants. By disentangling different aspects of agency, it is possible to develop hypotheses about how the emergence of migrations systems is related to the nature of the agency exercised by different pioneers or pioneering activities in different contexts. In lieu of an abstract, here is a brief excerpt of the content:Rita Gross as Pioneer in the Study of Women and ReligionRosemary Radford RuetherRita Gross has been a pioneer in shaping both the theory and practice of women and religion and in Feminist theology. Her pathbreaking work in these fields has received insufficient recognition among both feminists and scholars of religion. This session at the 2010 AAR meeting devoted to her work is a small rectification of this neglect. Rita (...) has written more than 150 articles and written or edited around ten books. Three of these volumes—Beyond Androcentrism (1977), Feminism and Religion (1996), and A Garland of Feminist Reflections: Forty Years of Religion Explorations (2009)—span her writing career and exemplify her contributions to the field of women and religion.As Rita Gross makes clear, she went to the University of Chicago in the 1960s to explore the field of comparative religion, not to study women in religion explicitly. She was forced into this focus on women and has continued to return to this concern because of the resistance of the scholars in this field to paying attention to the ways in which half of humanity—women—participates in religion. Her concerns with women's roles in religion were shaped in response to the attitude of her mentors at the University of Chicago who insisted that what women did in religion was unimportant and was, in any case, covered by studying what men did in religion. One of these mentors ridiculed her by asking if she did not realize that the masculine is generic for humanness as a whole and so "includes the feminine." At most one might mention men's view of women. What women themselves think and do is irrelevant. At that time there were very few women in doctoral studies in religion at Chicago, and their teachers made clear that any concern with women as subjects of religion was a sure way to terminate their careers in the field.It was in responding to these kinds of messages from her teachers that Rita began to craft, in the late 1960s and 1970s, a fundamental shift in the methodology of religious studies. This would shape her decision to write her doctoral thesis on women in Australian Aboriginal religion, to edit a group of essays in her 1977 volume, Beyond Androcentrism, as well as doing pioneering work with Nancy Falk in the volume Unspoken Worlds, where they demonstrated the transformative understanding of religions that opens up when scholars focused on women's roles.This methodological shift, which Rita Gross has returned to again and again [End Page 75] through her writings, she called the movement from androcentrism to androgyny. Androcentrism as a bias in religious studies, or any other field, means quite simply the assumption that the male (usually the elite male) is the normative human being. To understand the "essence" of the human in any field it is sufficient to study what this elite male thinks and does. For Rita this is not only an injustice to women, but it also means turning a blind eye to at least half of what is actually going on in this or any other field. In other words, what is wrong about androcentrism as a way of doing study in religion is that it gives one a partial and distorted picture of reality. It is bad empiricism.The shift to androgyny means really looking equally at what women are doing as subjects of religion, not simply adding information about what men think of women. It is not that this information is totally absent or impossible to get. Often it is right there in the primary data, but is ignored by the scholars because of their androcentric bias. Bringing in women's activities and viewpoints as equal does not merely add a missing corner to the picture; it means transforming the whole picture. What men are thinking and doing is relativized and put in a larger context. For example, in understanding Australian Aboriginal religion one discovers not only that are women excluded from men's key rituals, but also that men are excluded from women's key rituals. Power-law city-size distributions are a statistical regularity researched in many countries and urban systems. In this history of science treatise we reconsider Felix Auerbach’s paper published in 1913. We reviewed his analysis and found (i) that a constant absolute concentration, as introduced by him, is equivalent to a power-law distribution with exponent $$\approx 1$$ ≈ 1, (ii) that Auerbach describes this equivalence, and (iii) that Auerbach also pioneered the empirical analysis of city-size distributions across countries, regions, and time periods. We (...) further investigate his legacy as reflected in citations and find that important follow-up work, e.g. by Lotka (Elements of physical biology. Williams & Wilkins Company, Baltimore, 1925) and Zipf (Human behavior and the principle of least effort: an introduction to human ecology, Martino Publishing, Manfield Centre, CT (2012), 1949), does give proper reference to his discovery—but others do not. For example, only approximately 20% of city-related works citing Zipf (1949) also cite Auerbach (Petermanns Geogr Mitteilungen 59(74):74–76, 1913). To our best knowledge, Lotka (1925) was the first to describe the power-law rank-size rule as it is analyzed today. Saibante (Metron Rivista Internazionale di Statistica 7(2):53–99, 1928), building on Auerbach and Lotka, investigated the power-law rank-size rule across countries, regions, and time periods. Zipf’s achievement was to embed these findings in his monumental 1949 book. We suggest that the use of “Auerbach–Lotka–Zipf law” (or “ALZ-law”) is more appropriate than “Zipf’s law for cities”, which also avoids confusion with Zipf’s law for word frequency. Single-country approaches to the study of Buddhism miss the crucial significance of international networks in the making of modern Buddhism, in a period when the material basis for such networks had been transformed. Southeast Asia in particular acted as a dynamic crossroads in this period enabling the emergence of a ?global Buddhism? not controlled by any single sect, while India and Japan both played unexpectedly significant roles in this crossroads. A key element of this process was the encounter between Asian (...) Buddhist networks and western would-be Buddhists. Those involved, however, were often marginal - ?creative failures? in many cases - whose stories enable us to think this history in a more diverse way than is often done. In other cases as isolated figures they could pave the way for the ?mainstreaming? of new forms of Buddhism by established actors in later decades. This article introduces the special issue of Contemporary Buddhism entitled ?A Buddhist crossroads: pioneer European Buddhists and globalizing Asian networks 1860?1960?. The research described in this issue often raises other methodological questions of representativity and significance, while posing important challenges around collaborative research and the use of new technologies. Arakawa and Gins have been fomenting revolution for a long time. In the last twenty years their attention has turned more and more towards architecture and urban planning as a way of reforming our bodily existence. Their proposals enter daily life rather than staying in the isolated sphere of the museum or gallery. These constructions are to be lived in, not contemplated. Will daily life then blunt or sharpen Arakawa and Gins's power to educate and revise our "architectural bodies"?

Around 3 decades ago, Jayant Narlikar & Halton Arp argued on possible variable mass hypothesis cosmology (VMH). In the meantime, the Earth expansion problem has attracted great interest, and recent study gives geodetic evidence that the Earth has been expanding, at least over the recent several decades. Therefore, in the present article discusses some interesting effects related to varying G, but here we argue that instead of varying G we can think of varying mass (M). Among other things we discuss (...) receding planets from the Sun, calculation of Pioneer spacecraft anomaly as proposed by B.G. Sidharth, and also possible slight change of LAGEOS Satellite. Further observation to verify or refute this conjecture is recommended, especially based on geological evidence of expanding earth. (shrink)
