A pathogenic connection between autoreactive T cells, fungal infection, and carcinogenesis has been demonstrated in studies of human autoimmune polyendocrinopathy-candidiasis-ectodermal dystrophy as well as in a mouse model in which kinase-dead Ikkα knock-in mice develop impaired central tolerance, autoreactive T cell–mediated autoimmunity, chronic fungal infection, and esophageal squamous cell carcinoma, which recapitulates APECED. IκB kinase α is one subunit of the IKK complex required for NF-κB activation. IKK/NF-κB is essential for central tolerance establishment by regulating the development of medullary (...) thymic epithelial cells that facilitate the deletion of autoreactive T cells in the thymus. In this review, we extensively discuss the pathogenic roles of inborn errors in the IKK/NF-κB loci in the phenotypically related diseases APECED, immune deficiency syndrome, and severe combined immunodeficiency; differentiate how IKK/NF-κB components, through mTEC, T cells/leukocytes, or epithelial cells, contribute to the pathogenesis of infectious diseases, autoimmunity, and cancer; and highlight the medical significance of IKK/NF-κB in these diseases. IKK/NF-κB regulates the expression of many genes that encode proteins involved in many crucial biological functions, such as immunity, tissue homeostasis, and fungal/bacterial/viral infections. Also, IKKα plays anti-tumor activities in many organs independently of NF-κB pathways. Thus, an inborn error in one of these gene loci can cause severe human diseases through these complicated mechanisms. (shrink)

I Norge er ikke surrogati tillatt, og myndighetene fraråder norske statsborgere å benytte seg av surrogati i utlandet. I denne artikkelen fokuserer vi på kommersiell gestational surrogati og stiller spørsmålet: Bør man tillate at norske statsborgere benytter seg av surrogati i India? De etiske problemstillingene rundt surrogati er mange og sammensatte og blir spesielt utfordrende når tjenesten tilbys i et land med store kulturelle og økonomiske forskjeller både internt og i forhold til Norge. Vi baserer analysen og drøftingen av dette (...) etisk utfordrende spørsmålet på Beauchamps og Childress sin veletablerte metodiske tilnærming innen biomedisinsk etikk. Vi anvender de fire allmennmoralske prinsipper: respekt for autonomi, velgjørenhet, ikke-skade og rettferdighet på sakskomplekset for å synliggjøre spenningene involvert i dette etiske dilemmaet. Med full bevissthet om at det ikke finnes noen lettvinte og omkostningsfrie løsninger på dilemmaer generelt og dette spesielt, konkluderer vi med at interessene til de berørte parter, og spesielt surrogatmødrenes, kan bli bedre ivaretatt om surrogati tillates under omfattende reguleringer. Dersom man velger å gjøre praksisen illegal, vil man også miste mulighetene til å påvirke prosessen og sikre rettighetene til de involverte partene.Nøkkelord: surrogati, Norge–India, utnyttelse, autonomi, regulering av prosessenEnglish summary: Should Norwegian citizens be permitted to use surrogacy in India?Surrogacy is not permitted in Norway, and the government strongly advises against Norwegian citizens travelling abroad to have children through the use of the surrogacy industry. In this article, we focus on commercial gestational surrogacy and debate the question: Should Norwegian citizens be permitted to use surrogacy in India? The ethical concerns regarding surrogacy are complex and are especially challenging when the service is offered in a country with big cultural and economic differences both internal and in comparison to Norway. We base our analysis of this ethical, challenging question on Beauchamp’s and Childress’s well-established approach within biomedical ethics. We apply the four principles of respect for autonomy, beneficence, nonmaleficence and justice to shed light on the conflicts of interests in this ethical dilemma. With full awareness that there are no simple and correct solutions to dilemmas in general and on this issue, especially, our conclusion is that the interests of the involved parties, and especially those of the surrogate mothers, might be better attended to if surrogacy is allowed with extensive regulations. If this practice is made illegal, the opportunity to influence the process and secure the rights of the involved parties is lost. (shrink)

At profesjonsutøvere er ansvarlige og har ansvar knyttet til arbeidet de utfører, tar vi nærmest for gitt. Mitt sikte med denne artikkelen er å gi et innspill til nærere drøfting av hvordan det profesjonelle ansvaret kan forstås. I nyere profesjonsetiske kodekser knyttes ansvar først og fremst til konsekvenser av handlinger. Det innebærer at den profesjonelles ansvar forutsetter at det kan trekkes en linje fra en handlings konsekvens til den handlende. I komplekse organisasjoner kan imidlertid rekonstruksjon av veien fra konsekvens av (...) handling tilbake til den handlende vise seg vanskelig. Alternativt kan ansvar knyttes til spørsmål om handlingens samsvar med regler og prosedyrer. En slik tilnærming benevnes gjerne som «accountability». Disse to ulike tilnærmingene til ansvar har det til felles at det er handlingen som er i fokus – handlingens regelsamsvar eller dens konsekvenser, ikke en individuell dimensjon knyttet til den handlende som person. Den profesjonelle som selvstendig, tenkende og vurderende person blir perifer. Til videre drøfting av dette trekker jeg veksler på Hannah Arendts tenkning om ansvar. «To-i-én-dialog» og «tenkning» er sentrale elementer, og jeg vil peke på hennes bidrag som en mulig vei til en utdypet forståelse av det profesjonelle ansvarets individuelle dimensjon.Nøkkelord: profesjonelt ansvar, handlingens konsekvenser, accountability, to-i-én-dialog, tenkningEnglish summary: Individual responsibility and shallowness in professional workIt is almost taken for granted that professionals are responsible for the work they do. The scope of this article is to contribute to an evaluation of how we can understand this professional responsibility. Modern professional–ethical codes tie responsibility primarily to the consequences of acts. Thus the responsibility of a professional assumes that a connection can be drawn between the consequences of an act to the person who acted. However, in complex organizational structures, it can be difficult to find the path from consequence to actor. Another option is to look at responsibility as the product of an action's compliance with rules and procedures. Such an approach is often called «accountability». What these two different approaches to responsibility have in common is the focus on an action. It is the consequences of acts or compliance of rules that matter rather than the individual who performs the act. The idea of a professional as an autonomous, thinking and evaluating person becomes peripheral. The individual dimension of responsibility involves that the professional's responsibility goes beyond adherence to rules and procedures and what follows from tracking acts and consequences. I use Hannah Arendt's thoughts on the responsibility to facilitate further debate on this topic. A «two-in-one» dialogue and «thinking» are core elements in this, and I will point to her contribution as a possible path to a more in-depth understanding of the individual dimension of professional responsibility. An analysis of two heuristic strategies for the development of mechanistic models, illustrated with historical examples from the life sciences. In Discovering Complexity, William Bechtel and Robert Richardson examine two heuristics that guided the development of mechanistic models in the life sciences: decomposition and localization. Drawing on historical cases from disciplines including cell biology, cognitive neuroscience, and genetics, they identify a number of "choice points" that life scientists confront in developing mechanistic explanations and show how different choices result in divergent (...) explanatory models. Describing decomposition as the attempt to differentiate functional and structural components of a system and localization as the assignment of responsibility for specific functions to specific structures, Bechtel and Richardson examine the usefulness of these heuristics as well as their fallibility—the sometimes false assumption underlying them that nature is significantly decomposable and hierarchically organized. When Discovering Complexity was originally published in 1993, few philosophers of science perceived the centrality of seeking mechanisms to explain phenomena in biology, relying instead on the model of nomological explanation advanced by the logical positivists (a model Bechtel and Richardson found to be utterly inapplicable to the examples from the life sciences in their study). Since then, mechanism and mechanistic explanation have become widely discussed. In a substantive new introduction to this MIT Press edition of their book, Bechtel and Richardson examine both philosophical and scientific developments in research on mechanistic models since 1993. I denne artikkelen drøfter vi hvordan norske forbrukeres holdninger til genmodifisert mat har endret seg gjennom årene. Allerede da genteknologi ble etablert som et eget forskningsfelt og utviklingsområde på 1980-tallet, viste den norske opinionen stor skepsis. Den norske lovgivningen tidlig på 1990-tallet utmerket seg også som den mest restriktive i Europa. Dette bildet endret seg ikke mye i løpet av 1990-tallet, mens opinionen i mange europeiske land i disse årene kom mer på linje med den norske. I hele denne perioden (...) var både forbrukeropinionen og lovgivningen i USA langt mer aksepterende enn i Europa, noe som ga seg utslag i en handelskonflikt som toppet seg med EU sitt moratorium for utsetting av genmodifiserte planter i 1999. I denne artikkelen stiller vi spørsmålet om norske forbrukeres holdninger til genmodifisert mat har endret seg de siste årene. Artikkelen viser at mellom 2002 og 2007 er det en voksende gruppe av forbrukere som ser ut til å akseptere genmodifiserte matvarer hvis det fører til en helse- eller miljøgevinst. Men til tross for dette så er nordmenn fremdeles skeptiske til genmodifisert mat. Det kan tolkes slik at genmodifisert mat ikke har ført til den nytten som ansees nødvendig for å ta den risikoen det innebærer å gjøre et betydelig skifte i matseddel. Matpatriotisme og en kulturell konservatisme når det gjelder skifte i matvaner bidrar også til å forklare den dominerende vente-og-se-holdningen. (shrink)

This book explains the relationship between intelligence and environmental complexity, and in so doing links philosophy of mind to more general issues about the relations between organisms and environments, and to the general pattern of 'externalist' explanations. The author provides a biological approach to the investigation of mind and cognition in nature. In particular he explores the idea that the function of cognition is to enable agents to deal with environmental complexity. The history of the idea in the work of (...) Dewey and Spencer is considered, as is the impact of recent evolutionary theory on our understanding of the place of mind in nature. (shrink)

In the Aristotelian tradition, the psychological difference between virtue and continence is commonly understood in terms of inner harmony versus inner conflict. Virtuous agents experience inner harmony between feeling and action because they do not care to do other than what their circumstances call for, whereas continent agents feel conflicted about doing what is called for because of competing concerns. Critics of this view argue, however, that when the circumstances require sacrificing something of genuine value, virtuous agents can indeed feel (...) conflicted about acting well. But if this is so, what differentiates virtuous from merely continent agency? This essay argues that the traditional distinction conflates two aspects of virtue as well as two species of continence. And distinguishing between them provides resources for making sense of the complex relationship between inner conflict and good moral agency. (shrink)

Complexity and Postmodernism explores the notion of complexity in the light of contemporary perspectives from philosophy and science. The book integrates insights from complexity and computational theory with the philosophical position of thinkers including Derrida and Lyotard. Paul Cilliers takes a critical stance towards the use of the analytical method as a tool to cope with complexity, and he rejects Searle's superficial contribution to the debate.

This article contends that the evolution toward interdisciplinary collaboration that we are witnessing in the sciences must also occur in the humanities to ensure their very survival. That is, humanists must be open to working with scientists and social scientists interested in similar research questions and vice versa. Digital humanities is a positive first step. Complexity science should be the next step. Even though much of the ground-breaking work in complexity science has been done in the natural sciences and mathematics, (...) it can, if critically adapted, provide the needed metaphor for a broad integration of disciplines, humanistic and otherwise. Given its almost a-disciplinary nature, a complexity approach to the research problems in the humanities necessarily breaks down silos. Moreover, it can restore and reframe the seamless intellectual fabric sought by researchers before the atomization of the various disciplines in the nineteenthcentury academy. (shrink)

Sad1 and UNC84 (SUN) and Klarsicht, ANC‐1, and Syne homology (KASH) proteins interact at the nuclear periphery to form the linker of nucleoskeleton and cytoskeleton (LINC) complex, spanning the nuclear envelope (NE) and connecting the cytoskeleton with the nuclear interior. It is now well‐documented that several cellular functions depend on LINC complex formation, including cell differentiation and migration. Intriguingly, recent studies suggest that SUN proteins participate in cellular processes where their association with KASH proteins may not be required. (...) Building on this recent research, we elaborate on the hypothesis that SUN proteins may perform LINC‐independent functions and discuss the modalities that may allow SUN proteins to function at the INM when they are not forming LINC complex. The complex systems approach to cognitive science invites a new understanding of extended cognitive systems. According to this understanding, extended cognitive systems are heterogenous, composed of brain, body, and niche, non-linearly coupled to one another. This view of cognitive systems, as non-linearly coupled brain–body–niche systems, promises conceptual and methodological advances. In this article we focus on two of these. First, the fundamental interdependence among brain, body, and niche makes it possible to explain extended cognition without invoking representations or computation. (...) Second, cognition and conscious experience can be understood as a single phenomenon, eliminating fruitless philosophical discussion of qualia and the so-called hard problem of consciousness. What we call “extended phenomenological-cognitive systems” are relational and dynamical entities, with interactions among heterogeneous parts at multiple spatial and temporal scales. (shrink)

We analyse the computational complexity of three problems in judgment aggregation: (1) computing a collective judgment from a profile of individual judgments (the winner determination problem); (2) deciding whether a given agent can influence the outcome of a judgment aggregation procedure in her favour by reporting insincere judgments (the strategic manipulation problem); and (3) deciding whether a given judgment aggregation scenario is guaranteed to result in a logically consistent outcome, independently from what the judgments supplied by the individuals are (the (...) problem of the safety of the agenda). We provide results both for specific aggregation procedures (the quota rules, the premisebased procedure, and a distance-based procedure) and for classes of aggregation procedures characterised in terms of fundamental axioms. This article introduces and defends the “pathological complexity thesis” as a hypothesis about the evolutionary origins of minimal consciousness, or sentience, that connects the study of animal consciousness closely with work in behavioral ecology and evolutionary biology. I argue that consciousness is an adaptive solution to a design problem that led to the extinction of complex multicellular animal life following the Avalon explosion and that was subsequently solved during the Cambrian explosion. This is the economic trade-off problem of having (...) to deal with a complex body with high degrees of freedom, what I call “pathological complexity.” By modeling the explosion of this computational complexity using the resources of state-based behavioral and life history theory we will be able to provide an evolutionary bottom-up framework to make sense of subjective experience and its function in nature by paying close attention to the ecological lifestyles of different animals. Unexpected discoveries in nonequilibrium physics and nonlinear dynamics are changing our understanding of complex phenomena. Recent research has revealed fundamental new properties of matter in far-from-equilibrium conditions, and the prevalence of instability-where small changes in initial conditions may lead to amplified effects.

Engineers fine-tune the design of robot bodies for control purposes, however, a methodology or set of tools is largely absent, and optimization of morphology (shape, material properties of robot bodies, etc.) is lagging behind the development of controllers. This has become even more prominent with the advent of compliant, deformable or ”soft” bodies. These carry substantial potential regarding their exploitation for control—sometimes referred to as ”morphological computation”. In this article, we briefly review different notions of computation by physical systems and (...) propose the dynamical systems framework as the most useful in the context of describing and eventually designing the interactions of controllers and bodies. Then, we look at the pros and cons of simple vs. complex bodies, critically reviewing the attractive notion of ”soft” bodies automatically taking over control tasks. We address another key dimension of the design space—whether model-based control should be used and to what extent it is feasible to develop faithful models for different morphologies. (shrink)

I sin udmærkede kommentar til vores artikel «En etisk diskussion af screening for kræftsygdomme» beskriver Geir Hoff den udtalte mangel på evidens vedrørende nytteværdien af screeningsprogrammer for kræftsygdomme baseret på randomiserede studier. Ydermere fremhæver Geir Hoff misforholdet mellem den manglende evidens ved screening og de strenge krav, der er til evidensen i den farmaceutiske industri. Dette er en velkommen kritik, pga. en udtalt ukritisk og uvidenskabelig tilgang til anvendelse af screening for denne eller hin sygdom eller risikofaktor.

The consensus among evolutionists seems to be that the morphological complexity of organisms increases in evolution, although almost no empirical evidence for such a trend exists. Most studies of complexity have been theoretical, and the few empirical studies have not, with the exception of certain recent ones, been especially rigorous; reviews are presented of both the theoretical and empirical literature. The paucity of evidence raises the question of what sustains the consensus, and a number of suggestions are offered, including the (...) possibility that certain cultural and/or perceptual biases are at work. In addition, a shift in emphasis from theoretical to empirical inquiry is recommended for the study of complexity, and guidelines for future empirical studies are proposed. (shrink)

How does the complex nature of ecological systems affect ecologists' ability to study them? This Element argues that ecological systems are complex in a rather special way: they are causally heterogeneous. The author presents an updated philosophical account with an optimistic outlook of the methods and status of ecological research.

The paper discusses the epistemological basis for how you know what you are doing intentionally (and why). In particular, it challenges and ulimately rejects the claim made by Sarah K. Paul that such knowledge has an inferential basis.

Weighing complex sets of evidence (i.e., from multiple disciplines and often divergent in implications) is increasingly central to properly informed decision-making. Determining “where the weight of evidence lies” is essential both for making maximal use of available evidence and figuring out what to make of such evidence. Weighing evidence in this sense requires an approach that can handle a wide range of evidential sources (completeness), that can combine the evidence with rigor, and that can do so in a way (...) other experts can assess and critique (transparency). But the democratic context in need of weight-of-evidence analysis also places additional constraints on the process, including communicability of the process to the general public, the need for an approach that can be used across a broad range of contexts (scope), and timeliness of process (practicality). I will compare qualitative and quantitative approaches with respect to both traditional epistemic criteria and criteria that arise from the democratic context, and argue that a qualitative explanatory approach can best meet the criteria and elucidate how to utilize the other approaches. This should not be surprising, as the approach I argue for is the one that most closely tracks general scientific reasoning. (shrink)

Standard semantic theories predict that non-deictic readings for complex demonstratives should be much more widely available than they in fact are. If such readings are the result of a lexical ambiguity, as Kaplan (1977) and others suggest, we should expect them to be available wherever a definite description can be used. The same prediction follows from ‘hidden argument’ theories like the ones described by King (2001) and Elbourne (2005). Wolter (2006), however, has shown that complex demonstratives admit non-deictic (...) interpretations only when a precise set of structural constrains are met. In this paper, I argue that Wolter’s results, properly understood, upend the philosophical status quo. They fatally undermine the ambiguity theory and demand a fundamental rethinking of the hidden argument approach. (shrink)
