Aspects of peroxisome evolution, uncoupling, carnitine shuttles, supercomplex formation, and missing neuronal fatty acid oxidation (FAO) are linked to reactive oxygen species (ROS) formation in respiratory chains. Oxidation of substrates with high FADH2/NADH (F/N) ratios (e.g., FAs) initiate ROS formation in Complex I due to insufficient availability of its electron acceptor (Q) and reverse electron transport from QH2, e.g., during FAO or glycerol‐3‐phosphate shuttle use. Here it is proposed that the Q‐cycle of Complex III contributes to enhanced ROS formation (...) going from low F/N ratio substrates (glucose) to high F/N substrates. This contribution is twofold: 1) Complex III uses Q as substrate, thus also competing with Complex I; 2) Complex III itself will produce more ROS under these conditions. I link this scenario to the universally observed Complex III dimerization. The Q‐cycle of Complex III thus again illustrates the tension between efficient ATP generation and endogenous ROS formation. This model can explain recent findings concerning succinate and ROS‐induced uncoupling. (shrink)

Depletion of mitochondrial endo/exonuclease G‐like (EXOG) in cultured neonatal cardiomyocytes stimulates mitochondrial oxygen consumption rate (OCR) and induces hypertrophy via reactive oxygen species (ROS). Here, we show that neurohormonal stress triggers cell death in endo/exonuclease G‐like‐depleted cells, and this is marked by a decrease in mitochondrial reserve capacity. Neurohormonal stimulation with phenylephrine (PE) did not have an additive effect on the hypertrophic response induced by endo/exonuclease G‐like depletion. Interestingly, PE‐induced atrial natriuretic peptide (ANP) gene expression was completely abolished in endo/exonuclease (...) G‐like‐depleted cells, suggesting a reverse signaling function of endo/exonuclease G‐like. Endo/exonuclease G‐like depletion initially resulted in increased mitochondrial OCR, but this declined upon PE stimulation. In particular, the reserve capacity of the mitochondrial respiratory chain and maximal respiration were the first indicators of perturbations in mitochondrial respiration, and these marked the subsequent decline in mitochondrial function. Although pathological stimulation accelerated these processes, prolonged EXOG depletion also resulted in a decline in mitochondrial function. At early stages of endo/exonuclease G‐like depletion, mitochondrial ROS production was increased, but this did not affect mitochondrial DNA (mtDNA) integrity. After prolonged depletion, ROS levels returned to control values, despite hyperpolarization of the mitochondrial membrane. The mitochondrial dysfunction finally resulted in cell death, which appears to be mainly a form of necrosis. In conclusion, endo/exonuclease G‐like plays an essential role in cardiomyocyte physiology. Loss of endo/exonuclease G‐like results in diminished adaptation to pathological stress. The decline in maximal respiration and reserve capacity is the first sign of mitochondrial dysfunction that determines subsequent cell death. (shrink)

La construcción en madera es un oficio milenario que está en peligro de desaparición, y con ella uno de los elementos identitarios más importantes de las regiones costeras europeas. La escasa historiografía académica sobre los carpinteros de ribera como protagonistas es el principal motivo de este trabajo, que se adentra (...) en sus raíces, dando a conocer algunos de los aspectos más relevantes que definen e identifican la importancia del tema, como las principales actividades desarrolladas, su contextualización histórica, las técnicas, herramientas y la lengua franca marinera que compartían. Por ello, el fin último es proporcionar una fuente de referencia a quiénes apuesten, tanto de manera teórica como práctica, por la recuperación y la conservación de esta profesión ancestral sobre la que apenas existe información e investigación. Se concluye que la recuperación y gestión de este patrimonio, con vinculaciones amplias con otras actividades relacionadas, puede abrir perspectivas de consolidación patrimonial y económica en espacios marítimos poco desarrollados, ya que puede servir para sentar las bases de un futuro turismo cultural en el que estas técnicas sean protagonistas y sirvan para potenciar la identidad marítima en las regiones litorales. Many conserved eukaryotic traits, including apoptosis, two sexes, speciation and ageing, can be causally linked to a bioenergetic requirement for mitochondrial genes. Mitochondrial genes encode proteins involved in cell respiration, which interact closely with proteins encoded by nuclear genes. Functional respiration requires the coadaptation of mitochondrial and nuclear genes, despite divergent tempi and modes of evolution. Free‐radical signals emerge directly from the biophysics of mosaic respiratory chains encoded by two genomes prone to mismatch, with apoptosis being the default penalty (...) for compromised respiration. Selection for genomic matching is facilitated by two sexes, and optimizes fitness, adaptability and fertility in youth. Mismatches cause infertility, low fitness, hybrid breakdown, and potentially speciation. The dynamics of selection for mitonuclear function optimize fitness over generations, but the same selective processes also operate within generations, driving ageing and age‐related diseases. The ribosomal polypeptide tunnel exit is the site where a variety of factors interact with newly synthesized proteins to guide them through the early steps of their biogenesis. In mitochondrial ribosomes, this site has been considerably modified in the course of evolution. In contrast to all other translation systems, mitochondrial ribosomes are responsible for the synthesis of only a few hydrophobic membrane proteins that are essential subunits of the mitochondrial respiratory chain. Membrane insertion of these proteins occurs co‐translationally (...) and is connected to a sophisticated assembly process that not only includes the assembly of the different subunits but also the acquisition of redox co‐factors. Here, we describe how mitochondrial translation is organized in the context of respiratory chain assembly and speculate how alteration of the ribosomal tunnel exit might allow the establishment of a subset of specialized ribosomes that individually organize the early steps in the biogenesis of distinct mitochondrially‐encoded proteins. Consecutively, citations are analyzed. La Cátedra de Historia Naval, impulsada por la Armada Española y la Universidad de Murcia, tiene entre sus objetivos investigar y difundir la Historia y el Patrimonio Naval y Marítimo. Con objeto acercar el conocimiento científico y la información divulgativa versada en esta materia al usuario, nuestra institución no se limita a utilizar los canales clásicos de comunicación como pueden ser los artículos y las monografías, entre otros, sino que, en su apuesta por las Humanidades Digitales, hace un uso pro-activo (...) de un blog y de unos perfiles en las redes sociales que, aparentemente, nos hacen contactar con el lector virtual de manera eficaz. Con el objetivo de seguir mejorando, a través del presente trabajo se analizan qué temas han suscitado un mayor interés entre los lectores de nuestras herramientas sociales, a través del estudio de las publicaciones de nuestra bitácora 2.0 y del reflejo de éstas en nuestra página en Facebook. La respuesta nos ayudará a conocer mejor los intereses y necesidades de información de los usuarios y, consecuentemente, podremos seguir perfeccionando los puentes digitales que hemos tendido entre ellos y la Historia y del Patrimonio Naval y Marítimo. Over recent years, evidence has been accumulating in favour of the free radical theory of aging, first proposed by Harman. Despite this, an understanding of the mechanism by which cells might succumb to the effects of free radicals has proved elusive. This paper proposes such a mechanism, based on a previously unexplored hypothesis for the proliferation of mutant mitochondrial DNA: that mitochondria with reduced respiratory function, due to a mutation or deletion affecting the respiratory chain, suffer less (...) frequent lysosomal degradation, because they inflict free radical damage more slowly on their own membranes. Once such a mutation occurs in a mitochondrion of a non‐dividing cell, therefore, mitochondria carrying it will rapidly populate that cell, thereby destroying the cell's respiratory capability. The accumulation of cells that have undergone this transition results in aging at the organismal level. The consistency of the hypothesis with known facts is discussed, and technically feasible tests are suggested, of both the proposed mechanism and its overall contribution to mammalian aging. Recently, constructive neutral evolution has been touted as an important concept for the understanding of the emergence of cellular complexity. It has been invoked to help explain the development and retention of, amongst others, RNA splicing, RNA editing and ribosomal and mitochondrial respiratory chain complexity. The theory originated as a welcome explanation of isolated small scale cellular idiosyncrasies and as a reaction to ‘overselectionism’. Here I contend, that in its extended form, it has major conceptual problems, can not (...) explain observed patterns of complex processes, is too easily dismissive of alternative selectionist models, underestimates the creative force of complexity as such, and – if seen as a major evolutionary mechanism for all organisms – could stifle further thought regarding the evolution of highly complex biological processes. Since the human mitochondrial genome was characterised and sequenced in 1981(1), it has been viewed as the likely site of genetic diseases showing a maternal inheritance pattern and associated with defects of the respiratory chain, such as the mitochondrial myopathies (MMs)†(2). The properties that make it a candidate for the source of such conditions are that it encodes polypeptides involved in electron transport(3,4) and that it is maternally inherited(5). However, several of the mtDNA diseases only fulfill one or (...) other of these criteria: the first group of mtDNA diseases showed Only sporadic deletions(6,7), and the first point mutation in Leber's Hereditary Optic Neuropathy(8) (LHON) is not associated with a clear biochemical defect. Furthermore, it is now clear that both autosomal dominant(9) and probably recessive(10) nuclear genes can cause abnormalities of mtDNA. Each of these major groups will be considered in turn. Here, we review recent findings about nuclear gene defects in isolated OXPHOS enzyme complex deficiency. This information should help in identifying patients with mitochondrial disease and defining a biochemical and molecular basis of the disorder found in (...) each patient. Metabolic pathways underlying brain function remain largely unexplored during neurodevelopment, predominantly due to the lack of feasible techniques for use with awake infants. Broadband near-infrared spectroscopy provides the opportunity to explore the relationship between cerebral energy metabolism and blood oxygenation/haemodynamics through the measurement of changes in the oxidation state of mitochondrial respiratory chain enzyme cytochrome-c-oxidase alongside haemodynamic changes. We used a bNIRS system to measure ΔoxCCO and haemodynamics during functional activation in a group of 42 typically developing infants (...) aged between 4 and 7 months. bNIRS measurements were made over the right hemisphere over temporal, parietal and central cortical regions, in response to social and non-social visual and auditory stimuli. Both ΔoxCCO and Δ[HbO2] displayed larger activation for the social condition in comparison to the non-social condition. Integration of haemodynamic and metabolic signals revealed networks of stimulus-selective cortical regions that were not apparent from analysis of the individual bNIRS signals. These results provide the first spatially resolved measures of cerebral metabolic activity alongside haemodynamics during functional activation in infants. Measuring synchronised changes in metabolism and haemodynamics have the potential for uncovering the development of cortical specialisation in early infancy. (shrink)

The ongoing coronavirus 2019 pandemic caused by the severe acute respiratory syndrome coronavirus 2 has resulted in a severe ramification on the global healthcare system, principally because of its easy transmission and the extended period of the virus survival on contaminated surfaces. With the advances in computer-aided diagnosis and artificial intelligence, this paper presents the application of deep learning and adversarial network for the automatic identification of COVID-19 pneumonia in computed tomography scans of the lungs. The complexity and time (...) limitation of the reverse transcription-polymerase chain reaction swab test makes it disadvantageous to depend solely on as COVID-19’s central diagnostic mechanism. Since CT imaging systems are of low cost and widely available, we demonstrate that the drawback of the RT-PCR can be alleviated with a faster, automated, and reduced contact diagnostic process via the use of a neural network model for the classification of infected and noninfected CT scans. In our proposed model, we explore the benefit of transfer learning as a means of resolving the problem of inadequate dataset and the importance of semisupervised generative adversarial network for the extraction of well-mapped features and generation of image data. Our experimental evaluation indicates that the proposed semisupervised model achieves reliable classification, taking advantage of the reflective loss distance between the real data sample space and the generated data. Respiratory rhythms sustain biological life, governing the homeostatic exchange of oxygen and carbon dioxide. Until recently, however, the influence of breathing on the brain has largely been overlooked. Yet new evidence demonstrates that the act of breathing exerts a substantive, rhythmic influence on perception, emotion, and cognition, largely through the direct modulation of neural oscillations. Here, we synthesize these findings to motivate a new predictive coding model of respiratory brain coupling, in which breathing rhythmically modulates both local and (...) global neural gain, to optimize cognitive and affective processing. Our model further explains how respiratory rhythms interact with the topology of the functional connectome, and we highlight key implications for the computational psychiatry of disordered respiratory and interoceptive inference. Recent findings regarding the cellular biology and immunology of BST‐2 (also known as tetherin) indicate that its function could be exploited as a universal replication inhibitor of enveloped respiratory viruses (e.g., influenza, respiratory syncytial virus, etc.). BST‐2 inhibits viral replication by preventing virus budding from the plasma membrane and by inducing an antiviral state in cells adjacent to infection via unique inflammatory signaling mechanisms. This review presents the first comprehensive summary of what is currently known about BST‐2 anti‐viral (...) function against respiratory viruses, how these viruses construct countermeasures to antagonize BST‐2, and how BST‐2 function might be targeted to develop therapies to treat respiratory virus infections. The authors address the current gaps in knowledge, including the need for mechanistic understanding of BST‐2 antagonism by respiratory viruses, that should be bridged to achieve that goal. (shrink)

In the present paper we address the old question of respiratory planning in speech production. We recast the problem in terms of speakers’ communicative goals and propose that speakers try to minimise respiratory effort in line with H&H theory. We analyze respiratory cycles coinciding with no speech (i.e. silence), short verbal feedback expressions (SFE’s) as well as longer vocalisations in terms of parameters of the respiratory cycle and find little evidence for respiratory planning in feedback (...) production. We also investigate timing of speech and SFEs in the exhalation and contrast it with nods. We find that while speech is strongly tied to the exhalation onset, SFEs are distributed much more uniformly throughout the exhalation and are often produced on residual air. Given that nods, which do not have any respiratory constraints, tend to be more frequent towards the end of an exhalation, we propose a mechanism whereby respiratory patterns are determined by the trade-off between speakers’ communicative goals and respiratory constraints. This article reports two studies investigating the relationship between emotional feelings and respiration. In the first study, participants were asked to produce an emotion of either joy, anger, fear or sadness and to describe the breathing pattern that fit best with the generated emotion. Results revealed that breathing patterns reported during voluntary production of emotion were (a) comparable to those objectively recorded in psychophysiological experiments on emotion arousal, (b) consistently similar across individuals, and (c) clearly differentiated among joy, anger, fear, (...) and sadness. A second study used breathing instructions based on Study 1's results to investigate the impact of the manipulation of respiration on emotional feeling state. A cover story was used so that participants could not guess the actual purpose of the study. This manipulation produced significant emotional feeling states that were differentiated according to the type of breathing pattern. The implications of these findings for emotion theories based on peripheral feedback and for emotion regulation are discussed. (shrink)

The history of productivity of the κ-chain condition in partial orders, topological spaces, or Boolean algebras is surveyed, and its connection to the set-theoretic notion of a weakly compact cardinal is highlighted. Then, it is proved that for every regular cardinal κ > א1, the principle □ is equivalent to the existence of a certain strong coloring c : [κ]2 → κ for which the family of fibers T is a nonspecial κ-Aronszajn tree. Eyal et al have recently argued that researchers should consider conducting severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) human challenge studies to hasten vaccine development. We have conducted (J. L.) and overseen (L. D.) human challenge studies and agree that they can be useful in developing anti-infective agents. We also agree that adults can autonomously choose to undergo risks with no prospect of direct benefit to themselves. However, we disagree that SARS-CoV-2 challenge studies are ethically appropriate at this time, (...) for 3 reasons: (1) current scientific knowledge of SARS-CoV-2 infection is insufficient to manage risks; (2) autonomous decision making, while necessary, does not override concerns about risk; and (3) undertaking challenge studies now would imperil confidence in the research enterprise, potentially undermining the global response to the coronavirus disease 2019 (COVID-19) pandemic. There is a growing consensus that math anxiety highly correlates with trait anxiety and that the emotional component elicited by math anxiety affects math performance. Yet few studies have examined the impact of “specific math anxiety” on math performance and the underlying physiological and affective mechanism. The present study examines the mediation effect of heart rate variability—an affective measurement indexed by respiratory sinus arrhythmia —in the relationship between specific math anxiety and arithmetic speed. A total of 386 junior high (...) school students completed a self-reported questionnaire to measure their anxiety level. Among this sample, 29 individuals with specific math anxiety, 29 with specific reading anxiety, 24 with specific trait anxiety, and 22 controls were selected to participate in an arithmetic task and a reading task while RSA was recorded when they performed the tasks. Results revealed that individuals with specific math anxiety showed lower RSA and longer reaction time than the other three groups in the arithmetic task. Regression and mediation analyses further revealed that RSA mediated the relation between specific math anxiety and arithmetic speed. The present study provides the first account of evidence for the affective hypothesis of specific math anxiety and suggests that affective responses may be an important mechanism underlying the detrimental effect of specific math anxiety on math performance. Graphical AbstractTesting for respiratory viruses and SARS-CoV-2 in clinical and epidemiological settings has contrasting purposes and utility. Symptomatic patients are best tested with respiratory virus panels to establish the pathogen and guide personalized treatment. Asymptomatic patients are tested for a single infectious pathogen to establish carrier status and guide containment.

Several key areas in modeling the cardiovascular and respiratory control systems are reviewed and examples are given which reflect the research state of the art in these areas. Attention is given to the interrelated issues of data collection, experimental design, and model application including model development and analysis. Examples are given of current clinical problems which can be examined via modeling, and important issues related to model adaptation to the clinical setting.

Panenmentalist possibilities are individual pure possibilities existing independently of any mind, actual reality, and possible-world conception. These possibilities are a priori accessible to our intellect and imagination. In this paper, I attempt to shed some panenmentalist light on the discovery of chemical branched chain reactions and its implications on biology and cancer research. I also examine the case of the Belousov–Zhabotinsky reaction which, at first, was believed to be impossible. Finally, I proceed to examine through a panenmentalist lens Szilard’s (...) discovery of the possibility of physical chain reactions and Mullis’s discovery of a polymerase chain reaction. All these major actual discoveries clearly depend on the discovery of the relevant panenmentalist possibilities and the ways in which they relate to one another. (shrink)

The rationality of scientific concept formation in theory transitions, challenged by the thesis of semantic incommensurability, can be restored by theChains of Meaning approach to concept formation. According to this approach, concepts of different, succeeding theories may be identified with respect to referential meaning, in spite of grave diversity of the mathematical structures characterizing them in their respective theories. The criterion of referential identity for concepts is that they meet a relation ofsemantic embedding, i.e. that the embedding concept can be (...) substituted by the embedded one in classical limit situations. Three case studies from contemporary physics theories will be used to show that the Chains of Meaning approach not only yields meaning comparisons for already established concepts (as for Newtonian and Schwarzschild mass) but is also well suited to characterize actual scientific strategies of concept formation in yet open cases such as black hole entropy or relativistic thermodynamics. Contemplative practices, such as meditation and yoga, are increasingly popular among the general public and as topics of research. Beneficial effects associated with these practices have been found on physical health, mental health, and cognitive performance. However, studies and theories that clarify the underlying mechanisms are lacking or scarce. This theoretical review aims to address and compensate this scarcity. We will show that various contemplative activities have in common that breathing is regulated or attentively guided. This respiratory discipline in (...) turn could parsimoniously explain the physical and mental benefits of contemplative activities through changes in autonomic balance. We propose a neurophysiological model that explains how these specific respiration styles could operate, by phasically and tonically stimulating the vagal nerve: respiratory vagal nerve stimulation. The vagal nerve, as a proponent of the parasympathetic nervous system, is the prime candidate in explaining the effects of contemplative practices on health, mental health, and cognition. The Chain of Change is the first full-scale philosophical commentary devoted to Aristotle's Physics VII, in which Aristotle argues for the existence of a first, unmoved cosmic mover. This study systematically considers the major issues of the book, and argues for the fundamental importance of Physics VII in our understanding of Aristotelian cosmology and natural science. Physics VII is extant in two versions, and therefore poses special editorial problems. For this reason one of the features of Dr. Wardy's study (...) is the provision of an improved text and translation in both versions. The author's comprehensive comparison of their merits, philosophical and philological, has a significant bearing on our understanding of the nature and evolution of the Aristotelian corpus. The second part of the book is devoted to critical examination of the argument, including one of the most elaborate and challenging in the entire Aristotelian corpus. Throughout, the author concentrates on those points where Aristotle diverges most sharply and provocatively from contemporary presumptions in philosophy and natural science. (shrink)

The present study examines the development of complex sentences with non-finite clause combining with particular focus on clause chaining, in narratives of 40 Turkish-speaking 4- to 11-year-olds and six adults elicited by a wordless picture book. Results show a gradual increase by age in the variety of clauses combined, the length of the complex sentences and their frequency of use. Clause chains formed with converbal clauses are the earliest and most frequent type of clause combinations, already present in 4-year-olds’ complex (...) sentences with 1-non-finite clause. Older children’s and adults’ 2- or 3-non-finite clause complex sentences consist of some combinations of adverbial, complement, relative and converbal clauses. Developmentally, clause chains establish first, aspectual-temporal continuity, then temporal-causal continuity. Sentence-internal and cross-sentence-boundary referential continuities are present early, from age 4 onwards. These findings are discussed in terms of the demands of narrative organization as well as the syntactic and semantic complexity of the clause combination devices in Turkish. (shrink)

This article examines whether the involvement of stakeholders in the design of corporate codes of conduct leads to a higher implementation likelihood of the code. The empirical focus is on Occupational Safety and Health (OSH). The article compares the inclusion of OSH issues in the codes of conduct of 30 companies involved in International Framework Agreements (IFAs), agreed upon by trade unions and multinational enterprises, with those of a benchmark sample of 38 leading Multinational Enterprises in comparable industries. It is (...) found that codes of the IFA group have a higher implementation likelihood in OSH than the codes of the benchmark group. Further, European firms, culturally more used to stakeholder involvement, score higher than their US and Japanese competitors, and hence are more capable of addressing the safety and health issues in international supply chains. The implementation likelihood of codes seems closely related to the type of corporate CSR approach. (shrink)

This paper proposes an extensionalist analysis of computer simulations (CSs). It puts the emphasis not on languages nor on models, but on symbols, on their extensions, and on their various ways of referring. It shows that chains of reference of symbols in CSs are multiple and of different kinds. As they are distinct and diverse, these chains enable different kinds of remoteness of reference and different kinds of validation for CSs. Although some methodological papers have already underlined the role of (...) these various relationships of reference in CSs and of cross-validations, this diversity is still overlooked in the epistemological literature on CSs. As a consequence, a particular outcome of this analytical view is an ability to classify existing epistemological theses on CSs according to what their authors choose to select and put at the forefront: either the extensions of symbols, or the symbol-types, or the symbol-tokens, or the internal denotational hierarchies of the CS or the reference of these hierarchies to external denotational hierarchies. Through the adoption of this extensionalist view, it also becomes possible to explain more precisely the reasons why some complete reduction of CSs to classical epistemic paradigms such as “experiment” or “theoretical argument” remains doubtful. On this last point, in particular, this paper is in agreement with what many epistemologists already have acknowledged. (shrink)

As a number of high profile companies have found to their cost, corporate reputations can be significantly affected by firms' management of sustainability issue, including those that are outside their direct control, such as the environmental and social impacts of their supply networks. This paper begins by examining the relationship between corporate social responsibility, reputation, and supply network conditions. It then looks at the effectiveness of one tool for managing supply network sustainability issues, ethical sourcing codes of conduct, by examining (...) how the characteristics of three supply networks branded clothes, DIY wood products and branded confectionary affects the implementation ethical sourcing codes of conduct. It ends by setting out conclusions on why implementation of such codes has been so much more successful in some sectors than others and recommendations on effective approaches to managing sustainability issues in supply networks.
