Drugs targeting a single TK/RTK in the treatment of solid cancers has not had the same success seen in blood cancers. This is, in part, due to acquired resistance in solid cancers arising from a range of mechanisms including the upregulation of compensatory RTK signalling. Rather than attempting to inhibit individual compensatory RTK—requiring knowledge of which RTKs are upregulated in any given tumour—strategies to universally inhibit signalling from multiple RTKs may represent an effective alternative. Endosomal trafficking of RTKs is a (...) common conduit that can regulate signalling from multiple RTKs simultaneously. As such, we posit that targeting endosomal trafficking—in particular, aberrant post‐translational modifications in cancers that contribute to dysregulated endosomal trafficking—could inhibit oncogenic signalling driven by multiple RTKs and pave the way for the development of a novel class of inhibitors that shift the trafficking of RTKs to inhibit tumour growth. Biomedical ontologies are emerging as critical tools in genomic and proteomic research where complex data in disparate resources need to be integrated. A number of ontologies exist that describe the properties that can be attributed to proteins; for example, protein functions are described by Gene Ontology, while human diseases are described by Disease Ontology. There is, however, a gap in the current set of ontologies—one that describes the protein entities themselves and their relationships. We have designed a PRotein Ontology (PRO) (...) to facilitate protein annotation and to guide new experiments. The components of PRO extend from the classification of proteins on the basis of evolutionary relationships to the representation of the multiple protein forms of a gene (products generated by genetic variation, alternative splicing, proteolytic cleavage, and other post-translational modification). PRO will allow the specification of relationships between PRO, GO and other OBO Foundry ontologies. Each of the main chapters in her book surveys a major world religion (Hinduism, Buddhism, Judaism, Christianity, Islam, and the Chinese moral tradition) through four lenses: a brief review of the tradition; the moral world of the religion; values, principles, or virtues in the tradition; and the religious tradition on a particular moral issue (e.g., Hinduism on abortion, Buddhism on wealth and poverty, Judaism on the environment, Christianity on war and peace, Islam on men and women, and China’s one-child [End Page 202] policy). The book also includes opening chapters that introduce students to the study of ethics and religious ethics; a final chapter on additional religious moral traditions (Sikhism, Jainism, Bahá’í, and Shinto); sidebars highlighting major religious figures, texts, or events (e.g., Gandhi, Thich Nhat Hanh, Elie Wiesel, the Charter of Medina, and the Cultural Revolution); and discussion questions, bibliography (including visual media), footnotes, a glossary, and an index.Although this book briefly discusses the discipline of comparative religious ethics and some of its descriptive and conceptual methods in its opening chapters, its broad scope and concise treatment mean that the actual task of comparing the similarities and differences in the ethics of the world religions is kept to a minimum. The few detailed comparisons that are made focus largely on Hindu, Buddhist, and Christian understandings of the concepts of reincarnation (29, 34, 64), dharma (18–19, 23–24, 34–35, 64), karma (18–19, 21–23, 35–36, 64), and ahimsa (25–27, 38, 70–71). While the main chapters are organized in a way that facilitates comparison, and discussion questions are included that invite students to observe similarities and differences, the book would be strengthened by giving greater attention to comparison throughout and adding a concluding chapter that summarizes its comparative content. It ends somewhat abruptly with a discussion of the Shinto moral tradition.Charles Mathewes, associate professor of religious studies at the University of Virginia, has written a book that, while limited to Judaism, Christianity, and Islam, achieves much greater comparative sophistication. The text is based on a course he has taught for more than ten years called “Religious Ethics and Moral Problems,” and it bears the marks of a seasoned teacher whose primary audience is advanced undergraduate students, both religious and secular. Mathewes writes from a Protestant Christian perspective “having a strong apocalyptic dimension which is focused on the next life,” acknowledging that Roman Catholics and Eastern Orthodox may find his views “odd or confused” (53). Part 1 discusses the relationship between God and morality and provides an overview of the ethics of each of the three Abrahamic religions. Parts 2 and 3 then examine not only the moral positions but “the arguments animating the traditions” (3) on a wide range of personal and social matters (e.g., friendship, sexuality, marriage and family, lying, forgiveness, capital punishment, war, and the environment). He breaks this task into two parts. In the first, Stiltner offers an introduction to the field of ethics in its personal, communal, and social dimensions. He discusses the three normative ethical theories most prominent in the West (deontological, consequentialist, and virtue ethics) and then applies these theories to a case study to examine the strengths and weakness of each. He then argues that, while applied ethicists often recommend employing all three approaches into the decision-making process, a person who wishes to make [End Page 198] formal ethical arguments must ultimately choose which framework to favor. In Stiltner's view, the framework of virtue ethics is best able to address the social and communal dimensions of ethics, which is the focus of the second part of the book.In the latter half of the first part, Stiltner describes the cultivation of virtue and examines specific categories—namely, virtues of care, respect, purpose, perceiving and reasoning, and acting and relating. The lynchpin between the first and second parts of the text is Stiltner's elucidation of human flourishing, in which he begins to link individual flourishing with the flourishing of communities. He draws heavily on Martha Nussbaum's capabilities approach to explain how individual flourishing can be enhanced or diminished by the communities of which one is a part. Finally, he argues that some groups perform as "mediating institutions" wherein "the pro-social influences of the virtues instilled in people in one communal context transfer to other contexts as the individuals participate in many associations," thereby influencing society as a whole (105).Following the primarily theoretical first part, the focus of the second part is how to apply those theoretical principles to actual communities. He describes his objective here as "recommend[ing] ethical principles and concrete practices that will help persons, groups, and societies to flourish" (131). He examines four specific types of mediating institutions—families, schools, work, and service—and follows the same pattern of analysis for each. He provides a brief sociological account of the group, then scrutinizes the ethical impact of the reciprocal relationships of persons to groups and groups to society. He then discusses a case study that highlights some of the challenges in each particular institution and what resources are available to individuals and institutions that could overcome those challenges.Stiltner's text is an excellent introduction to the field of virtue ethics for an undergraduate audience. One particular strength is his discussion of many contemporary voices from the fields of ethics, philosophy, and sociology—from Nussbaum to Robert Putnam, his reliance on fields outside of ethical theory proper strengthens and broadens his analysis significantly. In addition, he provides ample resources along the way for the reader. Each chapter begins with a boxed overview of the chapter and ends with questions for review and discussion as well as resources for further research. The text does not specifically address the spiritual dimension of ethical development or the role of religion as a mediating institution; while this is not necessarily a weakness, one should know this is not specifically a Christian ethical analysis. Kyte succeeds in creating a text that overcomes these deficiencies. He uses common language and examples while remaining rigorous and fully explaining the common ethical theories.He opens with a chapter debunking thirteen misconceptions about ethics, such as "Ethics Is 'Nothing but Words,'" "Ethics Is Just a Matter of Opinion," and "Ethics Is Separate from Religion." This chapter should set the tone for a class that doesn't allow sloppy thinking or the reduction of ethics to feelings. The chapter concludes by defining ethics as common sense, meaning "the ability to find shared meaning in the words we use together to make sense of our lives" (51).The second chapter examines four ways of ethical thinking: truth, thinking in terms of the facts of the situation; consequences, thinking in terms of the results of an action; fairness, thinking in terms of equality and consistency; and character, thinking in terms of people's motivations, character traits, or both. He then uses practical problems to illustrate how, by using these four ways of ethical thinking, a conversation working toward an agreed-on action can take place. Chapter 3, "Truth," on its own would be an excellent text for any course that has "critical thinking" as a learning outcome. He covers moral responsibility, bias, self-deception, and power with succinct but powerful illustrations from life. He then moves on to discernment, which includes a discussion of good reading, evaluating sources of information, and evaluating arguments, including examples of the most common fallacies. His examples are from diverse sources such as Mark Twain's Life on the Mississippi, E. M. Forster's A Passage to India, the Bible, Francis Bacon, and situations in ordinary life. This gives richness to the text and presents these concepts in an accessible and interesting way.The next three chapters, "Consequences," "Fairness," and "Character," cover the main features of utilitarianism, deontological ethics, and virtue ethics [End Page 191] in good detail, with summaries about major figures associated with these theories: Jeremy Bentham, John Stuart Mill, Epictetus, Immanuel Kant, Aristotle, and St. Thomas Aquinas. These give the students biographical information as well as the significance of each thinker's work, explaining the key features, strengths, and weaknesses of their theories. The chapter on character has a very helpful discussion of the cardinal and theological virtues. Each chapter has suggestions for further reading.The appendix covers how to use this four-way method in group decision-making and in writing case studies or issues analyses, helpfully including an example of each. This book is written by a gifted teacher who draws students into his subject, shows them its relevance, and helps them develop the confidence and skills they need to take their part in the ethical conversations that will occur throughout their lives. Each text on its own provides a very good initial evaluation of contemporary conversations in health care ethics. Together they provide a thorough foundation for the study of health care ethics that encourages further exploration of topics in the field. They both provide an excellent overview for undergraduate students as well as anyone interested in the field of medical morality.Health Care Ethics is especially helpful for those entering the field of ethics in general and the more particular realm of medical morality. User-friendly, [End Page 211] the book is accessible to beginners while also being relevant to those already well-versed in medical ethics. The book provides case studies culled from the experiences of the authors throughout their careers in the practice of medical ethics. Perhaps even more useful, however, are the other exercises in which readers are given the opportunity to actively engage the text in order to discover and more fully articulate their own moral stance. Although Health Care Ethics approaches the topic from a theological perspective, using scriptural passages as well as broader religious themes and contexts, the book is accessible to those outside of the religious arena as well.The first three chapters lay the framework for general theological and ethical inquiry while the later chapters discuss more specific topics in applied health care ethics. The opening chapters by Michael Panicola are thought-provoking, especially since they encourage readers to participate in exercises to determine how they would respond to particular scenarios in their personal decision making. For example, after laying out the classic ethical theories, readers are asked to take a “quiz” by responding to multiple-choice questions to determine how they would respond to various ethical situations. The exercise demonstrates that moral discernment that relies on one ethical theory alone or one single approach to ethics is too narrow and confining for authentic dialogue in ethics. Another exercise calls upon readers to examine their fundamental bases for personal decision making by asking them to create a list of what they think are the ten things that constitute human flourishing and then narrow that list down to the top two. In addition to being engaging, these exercises help readers establish their personal moral stance by asking them to appropriate foundational norms in the process of moral discernment to the case studies throughout the book. They also help readers apply these norms to the specific topics in health care ethics presented in the later chapters.The authors of Health Care Ethics clearly identify human flourishing as the center and goal for all ethical inquiry. Demonstrating that “human flourishing” has different connotations for different people, they also argue that certain norms can be agreed upon to establish a framework for all conversations regarding moral decision making in health care. They especially emphasize a concern for the optimization of human flourishing in final chapters of the text. Chapters 4–12 provide detailed examinations of current topics in medical ethics. Each chapter includes case studies that raise questions concerning social ethics as well as health care morality, employing sources from law, sociology, philosophy, and theology.
