Molecular interactions are at the origin of life. How molecules get at different locations in the cell and how they locate their partners is a major and partially unresolved question in biology that is paramount to signaling. Spatio‐temporal correlations of fluctuating fluorescently tagged molecules reveal how they move, interact, and bind in the different cellular compartments. Methods based on fluctuations represent a remarkable technical advancement in biological imaging. Here we discuss image analysis methods based on spatial and temporal correlation of (...) fluctuations, raster image correlation spectroscopy, number and brightness, and spatial cross‐correlations that give us information about how individual molecules move in cells and interact with partners at the single molecule level. These methods can be implemented with a standard laser scanning microscope and produce a cellular level spatio‐temporal map of molecular interactions. (shrink)

The development of artificial agents for social interaction pushes to enrich robots with social skills and knowledge about (local) social norms. One possibility is to distinguish the expressive and the functional orders during a human-robot interaction. The overarching aim of this work is to set a framework to make the artificial agent socially-competent beyond dyadic interaction – interaction in varying multi-party social situations – and beyond individual-based user personalization, thereby enlarging the current conception of “culturally-adaptive”. The core idea is to (...) provide the artificial agent with the capability to handle different kinds of interactional disruptions, and associated recovery strategies, in microsociology. The result is obtained by classifying functional and social disruptions, and by investigating the requirements a robot’s architecture should satisfy to exploit such knowledge. The paper also highlights how this level of competence is achieved by focusing on just three dimensions: (i) social capability, (ii) relational role, and (iii) proximity, leaving aside the further complexity of full-fledged human-human interactions. Without going into technical aspects, End-to-end Data-driven Architectures and Modular Architectures are discussed to evaluate the degree to which they can exploit this new set of social and cultural knowledge. Background Italy was the first European country to be involved with the COVID-19 pandemic. As a result, many healthcare professionals were deployed and suddenly faced end-of-life care management and its challenges. Aims To understand the experiences of palliative care professionals deployed in supporting emergency and critical care staff during the COVID-19 first and second pandemic waves. Research design A qualitative descriptive design was adopted, and in-depth interviews were used to investigate and analyse participants’ perceptions and points of view. Participants and (...) research context Twenty-four healthcare professionals (physicians, nurses, psychologists, physiotherapists, and spiritual support) from the most affected areas of Italy were recruited via the Italian society of palliative care and researchers’ network. Ethical considerations The University Institutional Board granted ethical approval. Participants gave written informed consent and agreed to be video-recorded. Findings The overarching theme highlighted participants’ experience supporting health professionals to negotiate ethical complexity in end-of-life care. Crucial topics that emerged within themes were: training emergency department professionals on ethical dimensions of palliative and end-of-life care, preserving dying patients’ dignity and developing ethical competence in managing end-of-life care. Conclusions Our study showed palliative care teams’ challenges in supporting health professionals’ ethical awareness in emergencies. However, while they highlighted their concerns in dealing with the emergency staff’s lack of ethical perspectives, they also reported the positive impact of an ethically-informed palliative care approach. Lastly, this study illuminates how palliative care professionals’ clinical and ethical competence might have assisted a cultural change in caring for dying patients during COVID-19 and future emergencies. The presence of Persian sources in Nietzsche’s Thus Spoke Zarathustra has been a topic of debate for decades. This paper summarizes the main results of a comparative study of Nietzsche and the ancient Persian scripture Avestā. In addition to several secondary sources Nietzsche repeatedly encountered, there is strong evidence that he read Johann Friedrich Kleuker’s German translation of the Avestā texts reconstructed by Abraham Hyacinthe Anquetil-Duperron. Analyzing Nietzsche’s diverse sources of Zoroastrianism as well as his knowledge of this religion, this (...) paper shows that many metaphors, references and claims in both Thus Spoke Zarathustra and in Nietzsche’s notebooks can be traced back to Zoroastrianism. BackgroundThe Authors have laid out an analysis of Italian COVID-19 confirmed data and fatality rates, pointing out how a dearth of health care resources in northern regions has resulted in hard, ethically challenging decisions in terms of granting patient access to intensive care units (ICU).Main textHaving to make such decisions certainly entails substantial difficulties, and that has led many health care professional to seek ethical guidance. The Italian Society of Anesthesia, Analgesia, Resuscitation and Intensive Care (SIAARTI) has attempted to meet (...) that growing need by a set of recommendations, applying “clinical soundness” as a beacon standard; that approach tends to prioritize patients with higher life expectancy, which could be characterized as a “moderately utilitarian” approach. Yet, such a selection has engendered daunting ethical quandaries. The authors believe it can only be warranted and acceptable if rooted in a transparent decision-making process and verifiable, reviewed criteria. Moreover, the authors have stressed how clinical experimentation in a pandemic setting is a subtext of great interest from an ethical perspective. In Italy, no drug therapy and trials were undertaken for COVID-19 patients for a rather long period of time. When the epidemic was already circulating, an intervention proved necessary on the system of administrative procedures, aimed at expediting the authorization and validation of protocols, then bogged down by bureaucracy. A new system has since been instituted by a government decree that was signed about one month after the first Covid-19 case was officially recorded in the country. Such a swift implementation, which took just a few weeks, is noteworthy and proves that clinical trials can be initiated in a timely fashion, even with a pandemic unfolding. The concerted, action of supportive care and RCTs is the only way to attain effective forms of treatments for COVID-19 and any other future outbreak.ConclusionsThe authors have arrived at the conclusion that the most effective and ethically sound response on the part of any national health care system would be to adequately reconfigure its organizational mechanisms, by making clinical trials and all related administrative procedures consistent with the current state of emergency. After a long time, people are still debating over “Cartesian-like Dualism” (CLD), i.e. towards the separation of “res-extensa” from “res-cogitans”. Since we suspect that this is due to a general attraction of mind towards the darkness of metaphysics, we have investigated the mental origin of this attraction. In human mind, we can envisage three different functional levels emerging one from the other like in a Matrioska; the three levels cause the arousal of as many forms of “dualisms”: 1) The 1st-level (...) corresponds to the “Dual-state” of mind, an archetypic dualism made of Unconscious Mind (UM, the domain of biophysical/biochemical signals) and Conscious Mind (CM, the domain of thoughts, images and sounds), genetically committed to reciprocal and fruitful interactions. 2) the 2nd-level corresponds to “Double-perspective” dualism; it emerges from CM’s splitting into two opposite perspectives: the 1st-order perspective (1PP) (subjective, emotional, Self-oriented, etc.) and the 3rd-order perspective (3PP) (rational, objective, Self-detached, etc.), that are in psychological contraposition. During individual growth, Ego-sense installs in CM with the illusion of possessing FW, and dominates 1PP. Conversely, 3PP is typically interested in scientific argumentations and investigations; so, 3PP confutes 1PP’s convictions on scien-tific bases (e.g. 1PP’s conviction that the binomial Ego-FW is real). Yet, on the one hand, 3PP hasn’t got a scientific alternative to explain “who we really are”, on the other hand, 1PP exhibits a great success in everyday life cognition. Then, we necessarily step over the dualism of the 3rd-level, “Cartesian-like Dualism” (CLD). The emotional origin of this CLD holds back 1PP’s trick to claim its supremacy over 3PP. (shrink)
