The progress of electronic health technologies and biobanks holds enormous promise for efficient research. Evidence shows that studies based on sharing and secondary use of data/samples have the potential to significantly advance medical knowledge. However, sharing of such resources for international collaboration is hampered by the lack of clarity about ethical and legal requirements for transfer of data and samples across international borders. Here, the International Clinical Trial Center Network reports the legal and ethical requirements governing data and sample exchange (...) across four continents. The most recurring requirement is ethical approval, whereas only in specific conditions approval of national health authorities is required. Informed consent is not required in all sharing situations. However, waiver of informed consent is only allowed in certain countries/regions and under certain circumstances. The current legal and ethical landscape appears to be very complex and under constant evolution. Regulations differ between countries/regions and are often incomplete, leading to uncertainty. With this work, ICN illuminates the unmet need for a single international collaborative framework to facilitate DSE. Harmonising requirements for global DSE will reduce inefficiency and waste in research. There are many challenges to realising this ambitious vision, including inconsistent terminology and definitions, and heterogeneous and dynamic legal constraints. Here, we identify areas of agreement and significant difference as a necessary first step towards facilitating international collaboration. We propose the establishment of a working group to continue the comparison across jurisdictions, create a standardised glossary and define a set of basic principles and fundamental requirements for DSE. Psychiatry is rapidly adopting digital phenotyping and artificial intelligence/machine learning tools to study mental illness based on tracking participants’ locations, online activity, phone and text message usage, heart rate, sleep, physical activity, and more. Existing ethical frameworks for return of individual research results (IRRs) are inadequate to guide researchers for when, if, and how to return this unprecedented number of potentially sensitive results about each participant’s real-world behavior. To address this gap, we convened an interdisciplinary expert working group, supported by (...) a National Institute of Mental Health grant. Building on established guidelines and the emerging norm of returning results in participant-centered research, we present a novel framework specific to the ethical, legal, and social implications of returning IRRs in digital phenotyping research. Our framework offers researchers, clinicians, and Institutional Review Boards (IRBs) urgently needed guidance, and the principles developed here in the context of psychiatry will be readily adaptable to other therapeutic areas. Cumulative evidence indicates that IL-6 production by HNSCC cells and/or stromal cells in the tumor microenvironment activates STAT3 and contributes to tumor progression and drug resistance. A library of 94,491 compounds from the Molecular Library Screening Center Network was screened for the ability to inhibit interleukin-6 -induced pSTAT3 activation. For contractual reasons, the primary high-content screening campaign was (...) conducted over several months in 3 distinct phases; 1,068 primary HCS actives remained after cytotoxic or fluorescent outliers were eliminated. One thousand one hundred eighty-seven compounds were cherry-picked for confirmation; actives identified in the primary HCS and compounds selected by a structural similarity search of the remaining MLSCN library using hits identified in phases I and II of the screen. Actives were confirmed in pSTAT3 IC50 assays, and an IFNγ-induced pSTAT1 activation assay was used to prioritize selective inhibitors of STAT3 activation that would not inhibit STAT1 tumor suppressor functions. Two hundred three concentration-dependent inhibitors of IL-6-induced pSTAT3 activation were identified and 89 of these also produced IC50s against IFN-γ-induced pSTAT1 activation. Forty-nine compounds met our hit criteria: they reproducibly inhibited IL-6-induced pSTAT3 activation by ≥70% at 20 μM; their pSTAT3 activation IC50s were ≤25 μM; they were ≥2-fold selective for pSTAT3 inhibition over pSTAT1 inhibition; a cross target query of PubChem indicated that they were not biologically promiscuous; and they were ≥90% pure. Twenty-six chemically tractable hits that passed filters for nuisance compounds and had acceptable drug-like and ADME-Tox properties by computational evaluation were purchased for characterization. The hit structures were distributed among 5 clusters and 8 singletons. Twenty-four compounds inhibited IL-6-induced pSTAT3 activation with IC50s ≤20 μM and 13 were ≥3-fold selective versus inhibition of pSTAT1 activation. Eighteen hits inhibited the growth of HNSCC cell lines with average IC50s ≤ 20 μM. Four chemical series were progressed into lead optimization: the guanidinoquinazolines, the triazolothiadiazines, the amino alcohols, and an oxazole-piperazine singleton. (shrink)

À la suite d’Ebeling, René Marlé s’était intéressé à cette question centrale de Bultmann « Quel sens cela a-t-il de parler de Dieu ? », au point d’en tirer le titre de son ouvrage sur Ebeling, Parler de Dieu aujourd’hui. La question ne porte pas sur le sens du mot Dieu ni sur la connaissance de Dieu, mais sur le sens du parler de Dieu : elle se situe donc sur le terrain de l’herméneutique , celui de l’analyse du langage.« (...) Si on veut parler de Dieu, répond Bultmann, on doit nécessairement parler de soi-même ». Il veut dire qu’on ne peut parler de Dieu qu’à partir de l’expérience personnelle de l’avoir rencontré, d’avoir été rencontré par sa Parole. Il n’interdit donc pas d’introduire le nom de Dieu dans le langage ; mais pas dans le langage objectivant propre à la condition spatio-temporelle, mais dans le parler existentiel, dont Heidegger a fait la théorie, qui est celui de l’expérience humaine et interpersonnelle, en tant que Dieu donne sens à ma vie. Grice, Michael Dummett, John McDowell, Sen develops his distinctive theory of meaning and understanding along with his theory of knowledge/knowing. In doing so he also addresses the larger issue regarding the nature of philosophical analysis. Güzellik ve sanat iç içe geçmiş iki kavramdır. Bir bakıma güzellik, sanatın özüdür denilebilir. Her sanat dalında olduğu gibi hat sanatında da güzeli arama arzusu ve eşsiz eserler ortaya koyma çabası, hattatları sabırla çalışmaya ve kendi sınırlarını zorlamaya sevk etmiştir. Bir kitap sanatı olan hüsn-i hattın kelime manasının “güzel yazı” olmasına mukabil, daha iyisini yazmak uğruna harcanan çaba yorucu ve zahmetlidir, ancak sabırla çalışan elbette ki karşılığını alır. Başka bir deyişle “Muhakkak ki Allah güzeldir, güzeli sever!” hadis-i şerifi ışığında bu (...) sanata gönül verenler, yazıyı daha muntazam yazma hususunda büyük ölçüde gayret gösterirler. Kişinin muntazam yazmayı başarması ise, sistematik çalışmasına bağlıdır. Yazıyı kaidelerine uygun ve ölçülü yazabilmek bıkmadan usanmadan, karamsarlığa kapılmadan çalışmakla mümkündür. Bu sebeple gerek öğrenme aşamasında ve gerekse sonrasında, harfler defalarca çalışma kâğıdına yazılarak el melekesi artırılır. Kelimeler ve harfler kâğıda kimi zaman noktalanıp ölçülendirilerek, kimi zaman da noktalanmadan yazılır. Talim amaçlı yapılan bu çalışmalar, başka bir deyişle karalamalar, hat sanatının temeli niteliğindedir. Arap alfabesiyle yazılan ve “karalama” adı verilen el alıştırmaları yazıyla uğraşan kişinin o an irticalen elinden çıkan harf veya kelimelerden oluşur ve çoğu zaman bir anlam ifade etmez. Bu çalışmanın amacı, karalamaların estetik unsurlarını ele almak ve zaman içerisinde uğradığı değişime mercek tutmaktır. Hattatların bu özgür yorumları gün geçtikçe daha fazla önemsenmekte ve hayranlık uyandırmaktadır. Harf bünyesini oluşturan kaidelerin dışında satır ve sınırlara bağlı kalmadan yazılan bu yazılar kendi içinde bir kompozisyon oluşturmakta ve çoğu zaman yazan kişiyi dahi hayrete düşürecek özgün çalışmalar ortaya çıkabilmektedir. Quelle est la nature et la fonction des sentiments dans l’activité de connaissance et, plus largement, dans l’existence humaine? Cette question, massive, n’a cessé d’inquiéter la philosophie, dont l’histoire paraît à bien des égards se confondre avec celle d’une opposition entre la lucidité de la raison et l’obscurité dangereuse des passions. Si le discours philosophique n’a jamais nié l’importance des sentiments, ni leur rôle dans l’ordonnancement général de la vie humaine, ce discours est cependant empreint d’ambiguïtés, comme l’est, déjà, le (...) statut du « cœur » dans la tripartition fonctionnelle de l’âme présentée par Platon : ni réductible à la droite raison, ni relégué dans la sphère des désirs qui nous empêchent de contempler les Idées, le sentiment est cet intermédiaire allié de la raison qui met en mouvement nos réactions morales. Cette ambiguïté se retrouve à l’âge classique, lorsque le discours philosophique découvre le fonctionnement des passions et cherche les modèles scientifiques susceptibles d’en rendre compte, hésitant entre la physiologie , la géométrie , la médecine et les sciences de la nature .En faisant de l’affectivité l’une des structures fondamentales de l’existence, la phénoménologie a replacé au centre du questionnement philosophique la problématique des sentiments. Mais comment décrire ces expériences singulières – celle de l’amour, de l’angoisse, du désespoir ou de la joie – sans les confondre avec des objets? Comment donner du sens aux affects sans adopter ce point de vue surplombant des sciences de la nature, qui confond le discours des causes et celui des raisons? Comment, en d’autres termes, continuer à parler des sentiments de manière raisonnée, tout en reconnaissant que les sentiments nous font autant que nous les connaissons – tout en continuant, par conséquent, à être réceptifs à leur pouvoir de nous ébranler? Sen’s capabilities approach offers a radical generalisation of the conventional approach to welfare economics. It has been highly influential in development and many researchers are now beginning to explore its implications for health care. This paper contributes to the emerging debate by discussing two examples of such applications: first, at the individual decision making level, namely the right to die, and second, at the social choice level. For the first application, which draws on Nussbaum’s list of capabilities, it is argued (...) that many capabilities are ambiguously or indirectly related to the right to die, but the ability to form a concept of the good life and plan one’s own life provides a direct justification for such a right. In the second application, the focus is specifically on healthcare rationing and it is argued that, although not committed to age based rationing, the capabilities approach provides a more natural justification of age related access to health care than the fair innings argument, which is often used to justify the alleged ageism inherent in quality adjusted life years maximisation. The emergence of the two great late modern crises—economic and environmental—has prompted calls for a return to Marx. This article describes a Marxian account of the 2008 economic crisis relating it to the phenomena of job polarization, de-industrialization, the decline of the middle class, and political populism in Europe and elsewhere. These are argued to spring from political mobilization due to certain kinds of capability deprivations as understood in Amartya Sen’s capability approach. The article demonstrates the continued relevance of Marx (...) for philosophy of the social sciences as well as for a better understanding of the future challenge of maintaining societal stability in the West. ABSTRACT: Many of the results of Timothy Williamson's Knowledge and Its Limits depend upon his argument that many, if not all, of our mental states fail to be luminous in the sense that if we are in them, then we are in a position to know that we are in them. The purpose of this article is to show that his argument is unsound. I conclude by distinguishing between partial and total luminosity, and by arguing that even if mental states (...) are not totally luminous, they are at least partially so.RÉSUMÉ: Plusieurs des résultats du livre de Timothy Williamson, Knowledge and Its Limits dépendent de son argument à l'effet que la plupart, sinon tous nos états mentaux ne sont pas lumineux au sens où tout en se trouvant dans un tel état, nous sommes en position, de savoir que tel est notre état. Cet article a pour objectif de démontrer que ce raisonnement ne tient pas. Les dérives politiques, au XIXe siècle, du thème du Royaume de Dieu et des attentes messianiques, et les désillusions qui en résultent aujourd’hui, invitent à mieux souligner le caractère théologal de l'espérance chrétienne en lien avec la foi au Dieu créateur.Confesser la création, en effet, désacralise la nature et donc aussi l'histoire et renvoie ainsi la fin ultime au-delà des aspirations aux satisfactions d’ordre mondain. La logique de l'incarnation également, tout en inscrivant notre action dans le monde, projette notre vérité (...) au-delà de lui, en Dieu seul. La fin ultime est pascale : en lien avec l'humain et le monde, mais en rupture avec l'histoire et la nature, et non en prolongement ni en comblement de l’une ni de l'autre. La matrice apocalyptique disjoint le Règne de Dieu de l’ordre de ce monde. L'ultime habite et transforme le présent, mais il n’est pas le terme du déroulement du temps.Le chrétien est institué témoin du Règne de Dieu sur ce monde, « théâtre de sa gloire » : c'est pourquoi son espérance est hétérogène à ce monde. En tant que vertu théologale, elle n'oublie pas de faire mémoire du passé pour se disposer à recevoir le futur, mais elle ne fait pas du futur qui vient de Dieu l'accomplissement d'un sens de l'histoire, pas plus que l’éternité ne se situe sur la ligne du temps. The political deviations in the 19th century of the theme of the Kingdom of God and of Messianic expectations, and the disillusions that are the result today, invite us to underline better the theological character of Christian hope connected with faith in a creator God.To avow the creation, indeed, desacralizes nature and, therefore, also history, thereby sending the ultimate end beyond aspirations to satisfactions of a worldly kind. The logic of the Incarnation, while writing our action in the world, also projects our truth beyond it, in God alone. The ultimate end is paschal : in connection with the human and the world, but in rupture with history and nature, and not in prolonging nor in filling up of the one nor of the other. The apocalyptic matrix disconnects the Kingdom of God from the nature of this world. The ultimate inhabits and transforms the present, but it is not the end of the passing of time.The Christian is made witness to the Kingdom of God in this world, « theatre of his glory » : that is why his hope is heterogeneous to this world. As a theological virtue it does not fail to remember the past in order to dispose itself to receive the future. But hope does not make of the future that comes from God the accomplishment of a sense of history, no more than eternity is situated on the time-line. (shrink)

Le livre d’Albert Jacquard, « Dieu ? », fait voir à quel point le langage du credo est devenu inintelligible pour des esprits contemporains. Il oblige à s’interroger sur le sens d’une récitation non comprise, à reconnaître la légitimité de textes plus parlants, mais aussi à contester les limites d’une approche purement objectivante du réel. Cela admis, il reste que c’est l’interprétation du contenu même de la foi qui divise aujourd’hui les croyants les plus sincères, alors que leur apport au (...) débat public semble mieux accueilli que naguère. Selon un lieu commun, la géographie médiévale se réduirait à une vaste parenthèse ouverte entre la fin du siècle d'or de l'Empire romain et la renaissance des grandes découvertes du XV° siècle. Mais quelle réalité s'agit-il de représenter ? Car toute représentation est tributaire, non seulement de conceptions particulières, mais aussi de moyens techniques qui ont présidé à son élaboration. Dans cet article, le lien entre la réalité et ses images, qui organise habituellement l'histoire de la cartographie, sera relâché au (...) bénéfice des représentations comme un système de signes appelé à prendre sens dans une production et une réception . L'enquête médiévale révèle l'attention des contemporains à la complexité de ces relations et représentations.According to common understanding, medieval geography would be reduced to a vast parenthesis, open between the end of the golden century of the Roman Empire and the renaissance of the great discoveries of the 15th century. But what is the reality being represented ?For every representation is tributary, not only of particular conceptions, but also of the technical capacities that preceded its elaboration. In this article, the connection between reality and its images, which habitually organized the history of cartography, will be set aside for the benefit of representations as a system, of signs called to have meaning in a production and a reception . L'article explore le sens que peut avoir le symbole du Règne de Dieu aujourd'hui dans le contexte asiatique. Après une courte histoire du symbole, l'article présente six défis que l'Asie pose à son usage en un discours théologique en contexte asiatique. La seconde partie passe en revue l'usage fait du symbole du Règne de Dieu par certains théologiens asiatiques contemporains: les théologiens Tissa Balasuriya et Aloysius Pieris du Sri Lanka, les théologiens Coréens du minjung, et le théologien presbytérien du Taiwan (...) Choan-Seng Song. En se basant sur les oeuvres de ces théologiens et à la suite de la théologie de la libération, la dernière partie essaie de montrer comment les six défis posés plus tôt peuvent être résolus dans la perspective de la philosophie confucianiste. C'est le travail du Centre A.J. Festugiere (...) de Paris X-Nanterre sur les entrecroisements et reprises entre philosophie et religion qui est a l'origine des etudes rassemblees en cet ouvrage. Que nous voyions apparaitre aujourd'hui une nouvelle forme de repentir publique convie a mieux distinguer deux modeles de constitution de soi qui se reconnaissent en ce que l'accent porte tantot sur l'etre et tantot sur le temps. La theorie de la signification s'est d'abord construite a partir d'une presupposition selon laquelle les mots postulent les choses. Mais le sens d'un enonce se revele bien souvent a partir (...) de la negation d'une telle relation, car l'usage que l'homme fait de son langage repose frequemment sur la liberte qu'il experimente a partir des mots de recreer un imaginaire qui s'oppose aux donnees du reel. C'est dans l'espace historique et dans celui, actuel, des theories linguistiques qu'il convient de proposer une interpretation des relations complexes de l'homme avec son langage. Le probleme du sens est actuellement au cur des preoccupations de la linguistique. Cet ouvrage de reflexion s'efforce d'eclairer le probleme du sens a la fois dans ses aspects diachroniques et synchroniques, tout en faisant siens les cadres theoriques herites de Saussure et de Guillaume. La demarche suivie bouscule bien des barrieres et se limite volontairement a l'esquisse d'une theorie de la signification integrant les dimensions convergentes d'une systematique de la langue, de la pragmatique du discours et d'une ontogenie du sens qui se construit d'abord dans le champ clos de toute conscience individuelle. La cle de toute theorie linguistique, depassant la langue saisie comme un objet, se trouve dans une approche neurolinguistique qui replace le sujet parlant au cur du debat. Si cet ouvrage heurte quelques principes et peut susciter de violentes reactions, sinon des contestations, conduisant a poser les problemes sous des jours nouveaux, il aura atteint son but essentiel. This book explores a different approach to poverty, one suggested by the notion of capabilities emphasized by Amartya Sen and Martha Nussbaum. In the spirit of the capabilities approach, the book argues that poverty refers not to a lack of things but to the lack of the ability to live life in a particular way. Avoiding poverty, then, means having the capacity and opportunity for creative living. The authors argue that the capacity to do skilled work plays a particularly important role in creative living, and suggest that the development of the ability to do skilled work is a vital part of solving the problem of poverty.
