We have synthesized a 582,970-base pair Mycoplasma genitalium genome. This synthetic genome, named M. genitalium JCVI-1.0, contains all the genes of wild-type M. genitalium G37 except MG408, which was disrupted by an antibiotic marker to block pathogenicity and to allow for selection. To identify the genome as synthetic, we inserted "watermarks" at intergenic sites known to tolerate transposon insertions. Overlapping "cassettes" of 5 to 7 kilobases (kb), assembled from chemically synthesized oligonucleotides, were joined by in vitro recombination to produce intermediate (...) assemblies of approximately 24 kb, 72 kb ("1/8 genome"), and 144 kb ("1/4 genome"), which were all cloned as bacterial artificial chromosomes in Escherichia coli. Most of these intermediate clones were sequenced, and clones of all four 1/4 genomes with the correct sequence were identified. The complete synthetic genome was assembled by transformation-associated recombination cloning in the yeast Saccharomyces cerevisiae, then isolated and sequenced. A clone with the correct sequence was identified. The methods described here will be generally useful for constructing large DNA molecules from chemically synthesized pieces and also from combinations of natural and synthetic DNA segments. 10.1126/science.1151721. In this note we shall show that if L is a balanced pseudocomplemented Ockham algebra then the set ${\fancyscript{I}_{k}(L)}$ of kernel ideals of L is a Heyting lattice that is isomorphic to the lattice of congruences on B(L) where ${B(L) = \{x^* | x \in L\}}$ . In particular, we show that ${\fancyscript{I}_{k}(L)}$ is boolean if and only if B(L) is finite, if and only if every kernel ideal of L is principal.

Objective: Work stress is one of the urgent public health problems, which has aroused wide attention. In addition, work stress also has a negative impact on the development of enterprises. This study has three purposes: to understand the current status of working stress among couriers, to examine the association between work stress, job satisfaction and sleep quality of Chinese couriers, and to verify the mediating role of psychological capital.Methods: A cross-sectional survey was conducted among 3000 couriers in Beijing of China (...) from January to December 2019 using cluster stratified random sampling. Descriptive statistical analysis was used to describe demographic characteristics of respondents. Pearson correlation analysis was used to analyze the correlation among work stress, job satisfaction, sleep quality, and psychological capital. The mediating analysis was calculated role of psychological capital.Results: A total of 2831 participants were included in this study. The average score of work stress was 2.49 ± 0.76. Work stress was negatively correlated with job satisfaction, sleep quality, and psychological capital, respectively. The direct effect of work stress on job satisfaction is –0.223, and the total effect of work stress on job satisfaction is –0.267. The a*b*c’ is positive, indicating the mediating role psychological capital has in explaining the relationship between couriers’ work stress and job satisfaction. Moreover, psychological capital plays a mediating role between work stress and sleep quality among couriers.Conclusion: Couriers face certain work stress and should be paid attention to and psychological capital can effectively alleviate the work stress of couriers, so measures can be taken to improve it, promoting healthy development of employees and organizations. Resumo: O estudo das ideias de Ibn Taymiyyah sobre o haram e o halal, no Islã, continua a ser relevante para a compreensão da jurisprudência islâmica, da ética, da identidade cultural e das questões contemporâneas. O objetivo do artigo é analisar as obras de Ibn Taymiyyah e identificar seus conceitos mais importantes para a comunidade muçulmana. Os métodos de análise das fatwas escritas por Ibn Taymiyyah e os métodos de interpretação e comparação dos conceitos de proibido e permitido, no Islã, (...) foram usados para atingir o objetivo do estudo. A principal conclusão da pesquisa é que Ibn Taymiyyah era conhecido por suas pesquisas em lei islâmica, teologia e moralidade. Ele tinha visões conservadoras e, devido às suas próprias ideias reformistas, foi repetidamente condenado. Apesar disso, ele criticou os ensinamentos tradicionais sobre o Islã e apoiou a necessidade de reformar alguns aspectos da prática islâmica. Ibn Taymiyyah atribuiu grande importância ao exame do Alcorão e da Sunnah do Profeta Muhammad, a fim de entender o que é proibido e permitido, em várias áreas da vida muçulmana. Ele enfatizou a importância de uma dieta saudável, a necessidade de seguir os princípios islâmicos em questões financeiras e a preservação da dignidade e do comportamento respeitoso, em todos os aspectos da vida. A indagação que motiva o diálogo Protágoras sobre a possibilidade ou não de se ensinar a virtude, perpassa também diversos outros diálogos platônicos. Na verdade, virtude e conhecimento são temas centrais para se pensar o ânthropos, a pólis, a ética, o ser e a razão, no contexto do corpus platônico. Em As Leis, última obra escrita pelo fundador da Academia, a relação conhecimento/virtude volta a ganhar destaque no processo de reelaboração de um novo projeto para salvar o homem e a (...) cidade de suas mazelas. A arte/ciência da justa medida indicada no Protágoras (357 b) será a chave para a viabilização do projeto legislativo, ético e político em As Leis. Será pela correta educação dos sentidos e da razão que se alcançará a virtude responsável pela condução da cidade livre, amiga de si e ponderada (Leis, 701 d). Por sua vez, a educação virtuosa é fruto do conhecimento assimilado pelos costumes, pelo aprendizado, pelo discernimento racional e pelo domínio das paixões, o que assegurará o devido equilíbrio mediante o amparo das leis e das instituições da pólis. Isto se pode constatar, especialmente, na leitura dos livros I, II,VI e VII de As Leis, embora tais questões estejam entrelaçadas no conjunto da obra. (shrink)

It is generally believed that moral reductionism is immune from notorious problems in moral metaphysics and epistemology, such as the problem of moral explanation – it is at least on this dimension that moral reductionism scores better than moral anti- reductionism. However, in this article I reject this popular view. First, I argue that moral reductionism fails to help vindicate the explanatory efficacy of moral properties because the reductionist solution is either circular or otiose. Second, I attempt to show that (...) a successful vindication, if any, of moral explanation requires moral-descriptive irreducibility. A disponibilidade de três versões dos comentários de João Duns Escoto para o livro das Sentenças de Pedro Lombardo, a saber, a Lectura, a Ordinatio e a Reportatio I-A, torna possível a reavaliação de interpretações contemporâneas sobre a teoria da distinção entre uma potência absoluta e outra ordenada apresentada por Escoto. Tomando como fio condutor o texto da Ordinatio e utilizando as outras versões na medida em que elas se esclarecem mutuamente, revisitaremos alguns tópicos defendidos por algumas daquelas interpretações: a (...) associação entre as duas potências e aquilo que é feito por direito e de fato, a compreensão que toma a potência absoluta e ordenada como a distinção um modelo operatório que envolve dois modos de ação e, finalmente, sua relação com a teoria das “possibilidades sincrônicas”: interpretações talvez não suficientemente precisas, como pretendemos mostrar. Também mostraremos que o texto da Reportatio I-B, atualmente sem uma edição contemporânea, pode ser útil na exemplificação de alguns argumentos da discussão em pauta. (shrink)

Seguiremos com a leitura da Crítica da razão prática, de Immanuel Kant, buscando apoio em dois eixos centrais: a) a formulação do imperativo categórico; b) a doutrina do fato da razão. A escolha desse percurso sustenta a posição de que, não obstante as inúmeras formulações dadas ao imperativo da moralidade ao longo da Fundamentação, haveria, nos termos da segunda Crítica, uma formulação mais clara e precisa desse princípio. Em relação à doutrina do fato da razão, a mesma será tratada em (...) dois sentidos: de um lado, a partir de uma tentativa de reconstrução das teses originais de Kant, com base no percurso textual do filósofo; de outro, mostrando em que medida essa doutrina representa uma mudança de posição em relação às impossibilidades encontradas na FMC. O objetivo, com efeito, será o de mostrar como Kant deixa de ocupar-se da tarefa de deduzir analiticamente o conceito de liberdade a partir do conceito de vontade, sendo, precisamente, o apelo ao fato da razão o traço distintivo dessa mudança. Disso se concluirá que não apenas a razão pura pode ser prática, mas que só ela, e não a razão restringida empiricamente, é incondicionalmente prática. De saída, falaremos da distinção entre vontade e arbítrio, ressaltando seu significado para a compreensão do conceito de autonomia face à antropologia e à natureza humana. (shrink)

The study of dialectical logic has a history of nearly one hundred years in China. It is significant for understanding both the growth of Chinese logic and the Sinicization of Marxism to review dialectical logic in the context of introducing the study of Marxism in China. Debates about dialectical logic in Chinese academic circles involve not only the problems of logic itself, but more importantly the understanding of Marxist philosophy. In the 1920s and 1930s, some Marxist researchers in China were (...) influenced by the philosophers and logicians of the former Soviet Union, and they came very close to equating dialectical logic with Marxist dialectics. They also identified the formal logic originating from ancient Greece as metaphysics, praising the former and criticizing the latter. In the upsurge of Marxist philosophy in the 1950s and 1960s, Chinese logicians carried out normal and formal academic research on dialectical logic. During the period from the end of 1970s to the end of 1990s, the research and teaching of dialectical logic in China reached its heyday, and logicians performed comprehensive and detailed research on some critical issues, including the basic laws of dialectical logic, which they tried to formalize. Since the beginning of the 21st century, dialectical logic has ceased to prosper and has faded from the core field of the discipline of logic, although some honorable scholars still persist in studying it. The automated valuation of benchmark land price plays an essential role in regulating land demand in Chinese real-estate market as the big data are currently accumulated rapidly. However, this problem becomes highly challenging due to the multidimension, large volume, and nonlinearity of the land price-influencing factors. In this paper, an effective data-driven automated valuation framework is proposed for valuing real estate assets by combining a GIS and neural network technologies. This framework can automatically obtain the values of spatial factors affecting (...) land price from GIS and generate training set data for training the neural network to identify the complex relationship between all kinds of factors and benchmark land prices. The effectiveness and universality of the framework is verified via the data of benchmark land prices in Wuhan. This study uses an integrated model of resource conservation theory and social learning theory to explore the antecedents of hotel interns’ perceptions of occupational stigma and to explore the mechanisms inherent to retention willingness. This study first manipulated relevant subjects’ experimental materials through a contextual experiment and used a one-way ANOVA to test the effects of competence stereotypes and occupational stereotypes on hotel interns’ stigma perceptions, respectively, and then used partial least squares structural equation modeling as a statistical tool and (...) the SmartPLS 3.0 program to validate the model of hotel interns’ occupational stigma perceptions-intention. The effects of both competence stereotypes and occupational stereotypes on hotel interns’ perceptions of occupational stigma were significant. The results of the partial least squares structural equation model showed that hotel interns’ perceptions of occupational stigma significantly contributed to emotional exhaustion and that emotional exhaustion significantly influenced hotel interns’ retention willingness, hotel interns’ perceptions of occupational stigma had a significant effect on their retention willingness, while the role of emotional exhaustion as a mediating variable and occupational commitment as a moderator. The inner psychological and behavioral linkage mechanisms of hotel interns’ occupational stigma perceptions and their retention willingness under COVID-19 were explored, and the resource dynamics operating mechanism and professional commitment were also confirmed. Point-of-interest recommendations are a popular form of personalized service in which users share their POI location and related content with their contacts in location-based social networks. The similarity and relatedness between users of the same POI type are frequently used for trajectory retrieval, but most of the existing works rely on the explicit characteristics from all users’ check-in records without considering individual activities. We propose a POI recommendation method that attempts to optimally recommend POI types to serve multiple users. The (...) proposed method aims to predict destination POIs of a user and search for similar users of the same regions of interest, thus optimizing the user acceptance rate for each recommendation. The proposed method also employs the variable-order Markov model to determine the distribution of a user’s POIs based on his or her travel histories in LBSNs. To further enhance the user’s experience, we also apply linear discriminant analysis to cluster the topics related to “Travel” and connect to users with social links or similar interests. The probability of POIs based on users’ historical trip data and interests in the same topics can be calculated. The system then provides a list of the recommended destination POIs ranked by their probabilities. We demonstrate that our work outperforms collaborative-filtering-based and other methods using two real-world datasets from New York City. Experimental results show that the proposed method is better than other models in terms of both accuracy and recall. The proposed POI recommendation algorithms can be deployed in certain online transportation systems and can serve over 100,000 users. (shrink)

Objectives: This study aimed to provide a comprehensive understanding of all intervention for learning burnout by meta-analyzing their effects.Methods: Relevant studies that had been published up to September 18, 2020, were identified through a systematic search of the PubMed, Web of Science, the China National Knowledge Infrastructure, and Wan Fang databases. Eligible studies included randomized control trials of any learning burnout intervention conducted among students. The Jadad scale was used to evaluate the quality of the study. Random-effect meta-analyses, subgroup analyses, (...) meta-regression analysis, and sensitivity analysis were conducted. Funnel plots and Egger's tests were used to evaluate publication bias. Duval and Tweedie's non-parametric trim-and-fill method was used to adjust the effect of publication bias.Results: Of the 5,245 articles found, 39 met the inclusion criteria for the systematic review. There were 3,400 students, including 1,847 students in the intervention group and 1,553 in the control group. A meta-analysis of 44 studies showed that the interventions were effective. Subgroup analyses were conducted according to education, scales, intervention measures, and intervention time. The results showed that, compared with the control group, the learning burnout scores of undergraduates, secondary vocational students, and middle school students were significantly lower. Based on different scales, all interventions were also effective. The funnel plot was asymmetric and consistent with the results of Egger's test. The trim-and-fill method was used, and seven missing studies were merged to obtain a symmetric funnel plot.Conclusions: This meta-analysis indicated that learning burnout interventions are effective. The subgroup analyses showed that group counseling is the most widely used, exercise intervention is probably the most effective, and 8 weeks or more is the appropriate intervention time. An integrated intervention study based on the factors of learning burnout adds value. More studies are needed to supplement the results in the future. (shrink)

The target article provides an intermediate account of culture and freedom that is conceived to be curvilinear by treating economic development not as an adaptive outcome in response to climate but as a cause of culture parallel to climate. We argue that the extent of environmental variability, including climatic variability, affects cultural adaptation.

Within the same pathogen-stress framework as proposed by Fincher & Thornhill (F&T), we argue further that pathogen stress promotes matrilocal rather than patrilocal family ties which, in turn, slow down the process of modernity; and that pathogen stress promotes social learning or copying, including the adoption of foreign religions.

Moral education is a core component of ideological and political courses in primary and secondary schools and universities in China, and also an important part of contemporary Chinese Marxist educational theory and practice. In Chinese senior high schools, the main curriculum and platform for moral education is ideological and political courses. The Ideological and Political Curriculum Standards for General Senior High Schools (2017 Edition, 2020 Revised) issued by the Ministry of Education of the People’s Republic of China explicitly includes a (...) course on logical thinking in the ideological and political curriculum in general senior high schools, strengthening the importance of logic in moral education there. In this paper, the authors explain the necessity of intensifying logic training for contemporary Chinese teenagers, introduce and interpret the relevant contents in the Ideological and Political Curriculum Standards for General Senior High Schools (2017 Edition, 2020 Revised), and put forward some theoretical suggestions on how to conduct teaching on the subject of logic. O episcopado francês e a lei de Separação - Reflexão sobre um processo de reconhecimento ( The French episcopate and the Law of Separation -Reflection on a process of recognition - L`épiscopat français et la loi de Séparation - Retour sur un processus de reconnaissance). O “princípio da modernidade” – introduzido pela Revolução Francesa e ratificado por um conjunto de leis durante o século XIX – rompe com o tradicional “princípio do Estado confessional católico” e estabelece o regime de separação (...) entre a Igreja Católica e o Estado francês. Como a Igreja recebeu o regime de separação entre Igreja e Estado, constituído à sua revelia e, geralmente, contra ela? O artigo propõe-se a responder a pergunta, destacando dois pontos de vista: a) o da ruptura, segundo o qual a instituição católica, marcada pelo princípio do Estado cristão, nutre forte hostilidade contra o regime laico, ainda que estabelecendo com ele acordos práticos; b) e o da continuidade, sob o qual a Igreja acolhe e defende o princípio da “sã laicidade” e justifica a liberdade pública de consciência. Baseado em minuciosa leitura de documentos oficiais da Igreja e do episcopado francês, o artigo dialoga com os dois pontos de vista evocados, analisando seus termos e interpretando criticamente seus pressupostos e suas teses. Palavras-chave: Estado confessional; Estado laico; Laicidade; Episcopado francês.The "principle of modernity" - introduced by the French Revolution and ratified by a set of laws during the nineteenth century - breaks with the traditional "principle of the Catholic confessional state" and establishes the separation between the Catholic Church and the French State. How did the Church receive the system of separation of church and state, made in absentia and generally against the church? This paper seeks to answer that question, highlighting two points of view: a) the break, according to which the Catholic institution, marked by the principle of the Christian State, nourishes strong hostility against the secular regime, although establishing practical agreements with such regime b) the point of view of continuity, under which the Church welcomes and embraces the principle of "healthy secularism" and justifies the public liberty of conscience. Based on careful reading of official documents of the Church and of the French episcopate, this article dialogues with the two mentioned points of view , analyses and interprets in a critical way their terms, assumptions and theses. Comment l’Église a-t-elle reçu le régime de séparation entre Église et État, institué malgré elle, et, généralement, contre elle? L’article se propose de répondre à cette question en soulignant deux points de vue: a) celui de la rupture qui fait que l’institution catholique, marquée par le principe de l’État chrétien, nourrit une forte hostilité envers le régime laïque, bien qu’elle passe des accords pratiques avec lui; b) celui de la continuité, qui fait que l’Église accueille et défend le principe de la “saine laïcité” et justifie la liberté publique de conscience. The aim of this study is to develop an integrative model linking the effect of authoritarian leadership and differential leadership on employee turnover intention, and further explore the moderating role of the in-group/out-group on the above-mentioned relationships. We collected a sample of 624 supervisor–subordinate dyads from 87 teams in Mainland China and Taiwan. We find that, at the individual level, authoritarian leadership is positively related with employee turnover intention, and the relationship will be enhanced especially when the subordinate is an (...) in-group member. Through hierarchical linear modeling analysis, we find a negative relationship between differential leadership and employee turnover intention, significantly moderated by the within-team mean degree and the within-team variance degree of in-group/out-group. The original decision criterion and method of the combined calculus, presented by D. Hilbert and W. Ackermann, and applied by later logicians, are illuminating, but also go seriously awry and lead the universality and preciseness of the combined calculus to be damaged. The main error is that they confuse the two levels of the combined calculus in the course of calculating. This paper aims to resolve the problem through dividing the levels of the combined calculus, introducing a mixed operation mode, (...) and therefore finding a normal-form decision method approximated by that in sentential logic. To improve the personalized service of cultural tourism, anticipatory calculation has become an essential technology in the content design of intelligence navigation system. Culture tourism, as a form of leisure activity, is being favored by an increasing number of people, which calls for further improvements in the cultural consumption experience. An important component of cultural tourism is for tourists to experience intangible cultural heritage projects with local characteristics. However, from the perspective of user needs and the content adaptive system, there (...) are few suitable intelligent navigation and user demand anticipatory systems for intangible cultural heritage content. Purple clay culture is one of the first batches of national intangible cultural heritage protection projects in China. Therefore, taking purple clay culture exhibition as an example, this paper attempts to analyze the personalized information demand of tourism consumption experience in intangible cultural heritage communication activities with affective computing and meaning-driven innovative design method, by taking the content design in the navigation system as the research object. This paper uses the theory of planned behavior to calculate the relationship between tourists’ attitude, experience behavior, and display information demand. The findings indicate two issues. First, tourists’ demand for the entertainment and leisure attributes of intangible cultural heritage is greater than the demand for educational function attributes. Second, the meaning elements of information can change tourists’ beliefs in intangible cultural heritage and affect their attitude and behavior toward such heritage. According to the research results, strengthening the meaning elements of specific group information can improve people’s cultural identity and tourism satisfaction. The research results provide the basis for the content design direction of future museum intelligent navigation systems. (shrink)

Due to the large-scale mining of mineral resources, the surface strata in many areas have collapsed and even developed into deformation and subsidence. Based on the conventional geotechnical deformation monitoring technology, the surface deformation prediction and disaster caused by the conventional geotechnical deformation monitoring technology can be avoided. In this paper, based on GIS, combined with the analysis of surface settlement, the surface settlement, slope value, curvature deformation value, horizontal displacement, and corresponding horizontal deformation formula are comprehensively analyzed and studied. In order to make the surface deformation visualization and realize the high approximation processing with the subsidence area, this paper innovatively introduces the 3D visualization settlement data analysis and realizes the calculation result analysis and corresponding retrieval multiple visualization in the subsidence area depth prediction process. On this basis, the prediction data of subsidence area can be optimized and analyzed so as to master the efficient and accurate law of surface deformation. In the experimental part, a mine is taken as the experimental object, and the 3D visual monitoring processing is carried out. In this paper, the main method to prevent electromagnetic leakage of the computer system is the source suppression method. The source suppression method solves the problem fundamentally by reducing or eliminating the emission of computer electromagnetic leakage source. In the actual experiment, the effect of this method is very effective. The experimental results show that the prediction results and corresponding early warning accuracy of the system are significantly improved compared with the traditional scheme, which provides practical significance for disaster prevention and corresponding environmental impact assessment. The article explores Chinese rural migrants’ perspective on work and their relations with each other and with the Chinese state by drawing upon the ethnographical study of a group of rural home renovators in Beijing in the 1990s. The rural renovators were dubbed “guerrilla” workers because of their physical mobility, irregular employment, and unregistered status. After considering the novelty of guerrilla workfare in China, the article demonstrates the bifurcation of migrants’ social networks along the lines of work and everyday association, (...) locates the politics of worker-state interaction at the place of their everyday living, and explores their understanding of work that is remarkably devoid of nostalgia for state socialism. This paper proposes an adaptive neural network control approach for a direct-current system with full state constraints. To guarantee that state constraints always remain in the asymmetric time-varying constraint regions, the asymmetric time-varying Barrier Lyapunov Function is employed to structure an adaptive NN controller. As we all know that the constant constraint is only a special case of the time-varying constraint, hence, the proposed control method is more general for dealing with constraint problem as compared with the existing works on (...) DC systems. As far as we know, this system is the first studied situations with time-varying constraints. Using Lyapunov analysis, all signals in the closed-loop system are proved to be bounded and the constraints are not violated. The pandemic has catalysed to hasten the wider use of virtual hearings in international arbitration. However, the promotion of virtual hearings in international commercial dispute resolution was more complex than commonly thought due to the highlighted concerns of cybersecurity and breach of confidentiality in arbitration. The worries against the wide use of virtual hearings cannot stand because technological innovations can largely improve and solve this. However, virtual arbitration hearings may not be common post-COVID times. Technology shapes how people behave, interact, (...) grow, and develop in their relationships with others and wider communities. Yet greater immersion in the digital world undoubtedly creates new challenges and can adversely affect human-to-human interactions. There is very little scientific study on the psychological impacts of virtual hearings on arbitrators, witnesses and counsels. It is too early to assess its effectiveness from the user's perspective until the much-needed scientific data is released. Virtual hearings are unlikely to replace in-person ones necessary for more complex and high-value disputes requiring greater interaction and personal connection. Strategically, international arbitration is a private initiative-orientated, flexible, and market-driven dispute resolution mechanism. The parties are best positioned to choose the hearing format after balancing off. (shrink)

The Exclusion Argument has afflicted non-reductionists for decades. In this article, I attempt to show that emergentism—the view that mental entities can downwardly cause physical entities in a non-overdetermining way—is the most plausible approach to solving the exclusion problem. The emergentist approach is largely absent in contemporary philosophy of mind, because emergentism rejects the Causal Closure of Physics, a doctrine embraced by almost all physicalists. This article, however, challenges the consensus on causal closure and defends a physicalist version of emergentism. (...) On closer examination, I find that neither physical evidence nor physicalist considerations can satisfactorily support causal closure. Moreover, I argue that two competing approaches that accept causal closure, compatibilism and autonomism, involve ad hoc postulations of mental causation. Therefore, all things considered, emergentism is the best option available to non-reductive physicalists. (shrink)
