Intensive research and development of electrophoresis methodology and instrumentation during past decades has resulted in unique methods widely implemented in bioanalysis. While two‐dimensional electrophoresis and denaturing polyacrylamide gel electrophoresis in sodium dodecylsulfate are still the most frequently used electrophoretic methods applied to analyses of proteins, new miniaturized capillary and microfluidic versions of electromigrational methods have been developed. High‐throughput electrophoretic instruments with hundreds of capillaries for parallel separations and laser‐induced fluorescence detection of labeled DNA strands have been (...) of key importance for the scientific and commercial success of the Human Genome Project. Another powerful method, capillary isoelectric focusing with pressurized and pH‐driven mobilization, provides efficient separations and on‐line sensitive detection of proteins, bacteria and viruses. Electrophoretic microfluidic devices can integrate single‐cell injection, cell lysis, separation of its components and fluorescence or mass spectrometry detection. It traces the historical development of gel electrophoresis from the mid-1940s to the mid-1960s, with careful attention to the interplay between technical developments and disciplinary shifts, especially the rise of molecular biology in this time-frame. Claiming that the early 1950s marked a decisive shift in the evolution of electrophoretic methods from moving boundary to zone electrophoresis, I reconstruct various trajectories in which scientists such as Oliver Smithies sought out the most desirable solid supporting medium for electrophoretic instrumentation. Biomolecular knowledge, I argue, emerged in part from this process of seeking the most appropriate supporting medium that allowed for discrete molecular separation and visualization. The early 1950s, therefore, marked not only an important turning point in the history of separation science, but also a transformative moment in the history of the life sciences as the growth of molecular biology depended in part on the epistemological access to the molecular realm available through these evolving technologies. This paper discusses two films by Harun Farocki – Ein Bild and Nicht löschbares Feuer – and relates them to the theoretical arguments of Mirzoeff’s counterhistory of visuality and Rancière’s emancipated spectator. The main thesis is that experimental, semantically unclear or incomplete image can be of purpose to the spectator in an epistemological sense. The paper suggests the use of notions capital image and capillary image for the purpose of understanding similarities and differences between historically divergent forms of image (...) production. The research concludes by reiterating the possibility of spectatorial emancipation from the dominant image in strategies of showing the structure of production of the image on the screen. The work seen in the image becomes the work of the spectator in trying to demystify the homogeneity of its semantic codes. (shrink)

Hyperbaric oxygen (HBO) therapy still lacks proper interpretations of its many actions. This hypothesis is based on reports of temporarily elevated peripheral vascular resistance (PVR) during HBO sessions. Besides that, during HBO sessions, hyperoxygenated tissues can reduce their perfusion so much that CO2 can accumulate in them. Tissue perfusion depends on vascular innervation and on the balance between systemic constrictors and local dilators. During an HBO session, increased tissue oxygen levels suppress dilatory mechanisms. Tissue hyperoxygenation increases PVR, suggesting that the (...) HBO action on an edematous tissue may be caused by an oxygen‐induced disbalance among Starling capillary forces. The presented hypothesis is that oxygen‐caused arteriolar constriction reduces the hydrostatic pressure in downstream capillaries. Thus, more tissue fluid is absorbed in vascular capillaries, under the condition that the plasma colloid osmotic pressure remains unaltered during the HBO session. Among several known mechanisms behind the HBO actions, the vasoconstriction has been listed as a therapeutic modality for the reduction of the tissue edema, for a crush injury, for burns (in an acute phase), and for the compartment syndrome. The Bell's palsy is among often listed indications for the HBO treatment, although evidence is poor and reports of randomized trials are scarce. (shrink)

Produtora de artefatos, a ciência química é inseparável da técnica, da tecnologia e da indústria, de maneira que sua identidade epistêmica é indissociável de sua história. Seu estilo caracteriza um valor profundo na cultura técnica do homo faber, do poder de agir sobre a natureza e de transformá-la através de manipulações e de rearranjos. ABSTRACT In this paper I will identify a "style of reasoning" that is proper to to Chemistry and to point to the dissemination of its products and concepts. My aim is to explain some elements that characterize the style of thinking and engaging in Chemistry in the second half of the 18th century, its techno-cultural capillary action. The shaping of a chemical style of reasoning originated from the constancy of the techno-epistemic sites in which chemical knowledge is formed. Chemistry is a laboratory science, eminently technical and operative, i.e., it deals with a techno-scientific knowledge stricto sensu. A producer of artifacts, chemical science is inseparable from technique, technology and industry, in such a way that its epistemic identity is inseparable from its history. Its style marks a profound value in the technical culture of the homo faber, the power of acting upon nature and transforming it through manipulations and rearrangements. (shrink)

The aim of this work is the reduction of the throttling noise when the capillary is used as a throttling device. Based on the theory of bubble dynamics, two-phase flow, and aerated supercavitation, four different sizes of aerated devices used in refrigerator refrigeration systems are designed. Throttling noise and the temperature and pressure of inlet and outlet of the capillary are measured under stable operation. To compare the noise suppression effects in different groups of experiments, we introduced the (...) cavitation number to analyze, revealed the principle of aerated supercavitation to suppress noise, and combined the results of Fluent simulations to get the relationship between the noise suppression effect and the aerated quality. The experimental results showed that the aerated device can obviously suppress the throttling noise of the capillary outlet, up to 2.63 dB, which provides a new way for reducing the capillary throttling noise. During replication, the topology of DNA changes continuously in response to well‐known activities of DNA helicases, polymerases, and topoisomerases. However, replisomes do not always progress at a constant speed and can slow‐down and even stall at precise sites. The way these changes in the rate of replisome progression affect DNA topology is not yet well understood. The interplay of DNA topology and replication in several cases where progression of replication forks reacts differently to changes in DNA topology ahead is discussed (...) here. It is proposed, there are at least two types of replication fork barriers: those that behave also as topological barriers and those that do not. A crucial part of the knowledge of molecular biologists is procedural knowledge, that is, knowledge of how to do things in laboratories. Procedural knowledge of molecular biologists involves both perceptual-motor skills and cognitive skills. We discuss such skills required in performing the most commonly used molecular biology techniques, namely, Polymerase Chain Reaction and gel electrophoresis. We argue that procedural knowledge involved in performing these techniques is more than just knowing their protocols. Creative exploration and experience are essential for the (...) acquisition of procedural knowledge in molecular biology. With enough experience, molecular biologists make intuitive judgments without recourse to analytical reasoning. We propose that procedural knowledge is intuitive recognition of the patterns of one's environment that are the most relevant for making a decision or acting appropriately. Finally, we argue that knowledge of molecular biologists requires an integration of procedural knowledge and propositional knowledge. (shrink)

High blood pressure is an important component of pre-eclampsia. The underlying mechanism of development of hypertension in pre-eclampsia is complicated and still remains obscure. Several theories have been advanced including endothelial dysfunction, uteroplacental insufficiency leading to generalized vasoconstriction, increased cardiac output, and sympathetic hyperactivity. Increased blood flow and pressure are thought to lead to capillary dilatation, which damages end-organ sites, leading to hypertension, proteinuria and edema. Additional theories have been put forward based on epidemiological research, implicating immunological and genetic (...) factors. None of these theories have been substantiated. Based on a review of literature this paper postulates that the initiating event for the development of preeclampsia is intermittent hypoxia associated with irregular breathing during sleep, hypoapnea, apnea,inadequate respiratory excursions during the waking hours and inadequate cardiopulmonary synchronization (abnormal sympatho-vagal balance). I aim to illustrate how the recommender systems of digital platforms create a particularly problematic kind of vulnerability in their users. Specifically, through theories of scaffolded cognition and scaffolded affectivity, I argue that a digital platform’s recommender system is a cognitive and affective artifact that fulfills different functions for the platform’s users and its designers. While it acts as a content provider and facilitator of cognitive, affective and decision-making processes for users, it also provides a continuous and detailed amount of (...) information to platform designers regarding users’ cognitive and affective processes. This dynamic, I argue, engenders a kind of vulnerability in platform users, structuring a power imbalance between designers and users. This occurs because the recommender system can not only gather data on users’ cognitive and affective processes, but also affects them in an unprecedentedly economic and capillary manner. By examining one instance of ethically problematic practice from Facebook, I specifically argue that rather than being a tool for manipulating or exploiting people, digital platforms, especially by their underlying recommender systems, can single out and tamper with specific cognitive and affective processes as a tool specifically designed for mind invasion. I conclude by reflecting how the understanding of such AI systems as tools for mind invasion highlights some merits and shortcomings of the AI Act with regards to the protection of vulnerable people. Polyclonal antibodies raised against the human erythrocyte nucleoside transporter were used to investigate the distribution of the nucleoside transporters in the placenta. Immunoblots of brush-border membranes isolated from the human syncytiotrophoblast revealed a cross-reactive species that co-migrated with the erythrocyte nucleoside transporter as a broad band of apparent M 55,000. In contrast, no labelling was detected in basal membranes containing a similar number of equilibrative nucleoside transporters as assessed by nitrobenzylthioinosine -binding. The absence of cross-reactive epitopes in basal membranes and (...) their presence in brush-border membranes was confirmed by confocal immunofluorescence microscopy. The results suggest that at least two isoforms of the NBMPR-sensitive nucleoside transporter are present in the human placenta. The lumenal surfaces of fetal capillaries, small placental vessels and umbilical vein ware also strongly labelled by the antibody, a finding that suggests that the high fetal-placental adenosine uptake previously reported is due to endothelial transporters. For many years, there has been a gap in our capacity to study the structure and organization of chromosomal DNA molecules. The very small genomes of some viruses and bacteriophages (≤ 50,000 bp or 50 kb) are amenable to analysis by conventional gel electrophoresis, while the extremely large DNA molecules (> 100,000 kb) comprising the chromosomes of higher eukaryotes have been analysed under the light microscope, using a range of banding and in situ hybridization techniques. However, intact DNA molecules (...) with sizes between these two extremes have been largely inaccessible experimentally. This gap has recently been bridged with the development of two‐dimensional electrophoretic procedures that allow the separation and purification of chromosome‐sized DNA molecules ranging from ∼ 50 kb to several thousand kb.1–3 There are currently two variations of the technique in use: pulsed field gradient (PFG) gel electrophoresis1,2 and orthogonal‐field‐alteration gel electrophoresis (OFAGE).3 Both are based on a common principle, differing primarily in the geometry of the electrodes. Already, they have been employed to determine the approximate chromosome sizes and numbers for a variety of lower eukaryotes, including yeast and several protozoa.2–10 These ‘molecular karyotypes’ provide fundamental information about the genomic organization of each organism, and allow very rapid construction of linkage maps. Early in the work, we are asked to think with a practice, seemingly in a different register. An analyst finds herself accused of being an obstacle, of opposing the analysand’s wishes in life. In this context, we are asked to think about the blue of a painting in the room where the analysand waits (David-Ménard 2011, 34). [End Page 111]Given the criticisms of psychoanalysis elaborated in the work of Foucault and Deleuze, one might have expected these philosophers to be considered in Eloge in that stead.2 Or, one could imagine Foucault playing a role because of his genealogical analysis of the formation of the Oedipus complex.3Two years after the publication of Deleuze’s Anti-Oedipus, Foucault had, in Abnormal, characterized the desiring space of the family as stimulated in a capillary network of individualizing normalization (Foucault 2003, 257), linked by medicalized and expert concern of which parents became the auxiliaries. They are charged, and charge themselves, with the goals of family health, of preservative and defensive good parenting, and with the conduct of the child’s conduct. Foucault describes the pre-twentieth century emergence of lay and expert preoccupation with the dangers of childhood masturbation. This produces an environment whose intensive scrutiny involves a close “folding of... parents’ bodies over... children’s bodies” (ibid., 265) in a saturated, hermetic environment stimulating oedipalized desire. This means one can offer a genealogical account of the conditions of formation of the latter. These include a responsibilization of parents, who control for the presence of abnormal sex, advised by new experts (lay, medical, pseudo-scientific) in collective crusades for health and life. Thus there is differentiation through minor gradations of abnormality. Even the specificity of the architectural spaces in which scrutiny is directed plays a role in the hollowing out of the child’s putatively interior psychic-sexual life, the intense object of “palpating” interest (Foucault 1978, 45) in new apparatuses of knowledge-power (Foucault 2003, 327).Because he presents this as the context in which the “nuclear and substantial family was gradually constituted” (Foucault 2003, 264), along with its oedipal configurations of desire, Foucault emphasizes the contingency of both. In Hiba Ra’uf’s Woman and Political Work, she argues that the family is the basic political unit of the Islamic community or nation (the umma). Her thesis is both feminist and Islamist, as she argues that the ‘private is political’. By drawing analogies between family and umma, family and caliphate, the personal and the political, the private and public, Ra’uf seeks to dismantle the oppositions of secular society, to challenge the division of society into discrete spheres. This entails an implicit (...) challenge to the secular state, but effected through the politics of the family. An Islamic family, she argues, is a powerful site for the transformation of socio-political institutions; a politics of the microcosmic with macrocosmic ramifications, effected through the very embodiment and practice of an Islamic ethos at a grassroots, capillary level. However, though Ra’uf contests liberal secularism’s division of spheres with feminist and Islamist critical methods, she reproduces some of its fundamental assumptions about the nature of the family: as the domain of religion, in opposition to the secular state; as rooting community, in opposition to the individualism of the citizen; as an ethics grounded in affect; and as an essentially feminine world. In making the family the sphere of Islamic politics, Ra’uf re-enacts secularism’s division of spheres, sacralizing the affective bonds of intimate relations and making the family the domain of religion. Furthermore, by emphasizing the family as the domain of women’s political work, she reinscribes the family as a feminine sphere, so that woman’s vocation is familial, as is her ethical disposition. Stereological techniques allow biologists to create quantitative, three-dimensional descriptions of biological structures from two- dimensional images of tissue viewed under the microscope. For example, they can accurately estimate the size of a particular organelle, the total length of a mass of capillaries, or the number of neurons or synapses in a particular region of the brain. This book provides a practical guide to designing and critically evaluating stereological studies of the nervous system and other tissues. Mitchell Jürgen Habermas's "colonization of the lifeworld" thesis (1987, 332-73) posits that many of society's pathologies are due to the tendency of institutions to convert social issues that ought to be sorted out by a debating citizenry into technical problems ripe for resolution by expert bureaucracies, thus pre-empting important public (...) discussion. Habermas has attempted to lay bare the pernicious effects of this colonization process in his analysis of public opinion polling, welfare policy, education, German reunification, immigration, and other social issues (see Habermas 1997, 1994, and 1970; Holub 1992). In each of these contexts, Habermas has publicly challenged the encroachment of scientistic modes of decision-making into spheres where joint communicative action by deliberating citizens would yield more appropriate and legitimate judgments. This critical impulse is also evident in Habermas's methodological reflections on the proper role of academic scholarship, where he has argued vigorously against attempts to graft "objectivating" methods of natural scientific inquiry onto research projects in the social sciences (1971, 304-17).From all of this, one might gather that Habermas's commitment to rolling back the influence of technical forms of reasoning is connected to some intrinsic quarrel he has with the natural sciences. Yet such a sweeping generalization is hard to sustain in light of the fact that Habermas does not oppose technical reasoning per se; he recognizes that the daunting complexity of social life in late capitalism requiresthat certain "steering" tasks be delegated to systems that utilize largely instrumental logics to co-ordinate action. Likewise, he acknowledges that the disciplines of the natural sciences necessarily play important supporting roles in such steering projects.One normative presupposition of Habermas's colonization thesis is that there exists some proper boundary demarcating where the sphere of technical reasoning ends and the realm of communicative rationality begins. [End Page 1] On one side of this boundary, Habermas has provided many details on what he sees as the essential qualities of a properly functioning "public sphere," where "new social movements" continuously reweave the threads of communicative fabric holding society together (1996, 359-87), and where "historico-hermeneutic" academic study reflects on public life, sluicing insight back into the capillaries of democratic deliberations (1987, 374-403).What lies on the other side of the boundary is murkier. The proper role of natural scientific investigation has received relatively scant attention in Habermas's critical theory of society, and this has led to some confusion regarding his account of the natural sciences, as well as debate over whether this account has political purchase. One would think that sympathetic commentators, such as Helen Longino (who bases much of her own coherence theory of scientific truth on Habermas), would have reassuring things to say on this point. Yet her lukewarm assessment that, "in trying to clear a space for an autonomous social and critical theory, [Habermas] has ceded nature to the positivists" (1990, 202), raises questions about the flexibility and scope of Habermas's theory of communicative action as a basis for critique of scientific practice.One way to test Habermas's account of the natural sciences on this count is to put his views in conversation with science studies commentators who share a similar commitment to the idea that dialogue and argumentation are constitutive elements of scientific practice. Such an approach recasts the "ceding to positivism" question into a moment of controversy over the role of dialogue, not only as an essential motor driving the scientific enterprise, but also as a vehicle for democratic decision-making on issues related to the purpose and direction of scientific inquiry in society.Part one of this essay locates discursive norms embedded pragmatically in the notion of scientific objectivity. By bringing a modified version of Habermas's theory of communicative action into conversation with other approaches to science studies that foreground intersubjective dialogue as the key motor driving the scientific enterprise, I develop a foundation for robust criticism of systematically distorted scientific communication. On September 27, 2016 people across the world looked down at their buzzing phones to see the AP Alert: “Baby born with DNA from 3 people, first from new technique.” It was an announcement met with confusion by many, but one that polarized the scientific community almost instantly. Some celebrated the birth as an advancement that could help women with a family history of mitochondrial diseases prevent the transmission of the disease to future generations; others held it unethical, citing medical (...) tourism and consequences for the future of the therapy. The child in question was actually born a few months earlier on April 6, 2016, but the research was published a few months later in the October edition of Fertility and Sterility. The mother carries DNA that could have given the baby Leigh Syndrome, a severe neurological disorder characterized by psychomotor regression that typically results in death between ages two and three.[1] Also known as subacute necrotizing encephalomyelopathy, Leigh Syndrome is caused by genetic mutations in mitochondrial DNA, which causes defective oxidative phosphorylation. Because people with this condition cannot re-form ATP, “demyleniation, gliosis, necrosis, spongiosis, or capillary proliferation”[2] can occur, thereby producing bilateral lesions across the central nervous system. The 36-year-old mother previously had four miscarriages and successfully birthed two children, both of whom survived less than six years due to the syndrome. For religious reasons, the mother opted for Spindle Nuclear Transfer instead of Pronuclear Transfer,[3] which many religious organizations oppose because it entails the destruction of fertilized eggs.[4] In Pronuclear Transfer, both the parent and donor egg are fertilized. The parent’s nuclear material is then removed from the egg containing mutated mitochondria and inserted into the fertilized donor egg—from which the original nuclear material has been removed and destroyed. Spindle Nuclear Transfer, on the other hand, removes the mother’s nuclear material from the egg with unhealthy mitochondria, then implants it into a donor’s egg. The newly created egg is then fertilized with the father’s sperm. This Spindle Nuclear Transfer was performed in Mexico by a team of doctors led by Dr. John Zhang, the founder of the New Hope Fertility Center in New York City. In their report, the doctors outline that five oocytes were successfully reconstituted, four of which developed into blastocysts. Only one was euploid: containing 46 XY chromosomes. Researchers then biopsied that blastocyst and found that the transmission rate of maternal mtDNA was, “5.10 ± 1.11% and the heteroplasmy level for 8993T>G was 5.73%.”[5] This indicates that at the blastocyst stage, around five percent of the mitochondria are from the original maternal egg with the mutation for Leigh Syndrome. After birth, they biopsied different tissues and found that they had an average of less than 1.60 ± 0.92% of transmitted mtDNA from the original maternal egg. The baby is reportedly doing well and the team of doctors concluded that “[h]uman oocytes reconstituted by SNT are capable of producing a healthy live birth. SNT may provide a novel treatment option in minimizing pathogenic mtDNA transmission from mothers to their babies."[6] Even when considered theoretically, Spindle Nuclear Transfer is controversial. Many doctors believe that the risks at this stage of research are too great. Dr. Trevor Stammers, a bioethicist at St. Mary’s University in London, points out: “We do not yet have a clear picture of the interaction between nuclear DNA and mitochondria.”[7] Others hold that faulty mitochondrial DNA can still be transferred during the procedure. Still more argue this technology could create problems because of germline modification.[8] Dr. Paul Knoepfler, a cell biologist at the UC Davis School of Medicine, explains: “Since this is uncharted territory and the children born from this technology would have heritable genetic changes, there are also significant unknown risks to future generations.”[9] However, proponents counter these arguments. They say the benefits outweigh the risks, as “mitochondrial replacement techniques [like Spindle Nuclear Transfer] would eliminate maternal transmission of mitochondrial disease… allowing a woman with a family history of mitochondrial diseases to ensure her children would not be affected.”[10] Additionally, experts say that no symptoms will occur if less than 20% of the transferred mitochondrial DNA is faulty.[11] And while opponents see potential germline modification as a problem, advocates answer that this could stop a family history of mitochondrial disease, which they deem a more serious concern. In this case specifically, however, there are more issues at hand. While the team of doctors did eliminate the possibility of germline modification by selecting a male embryo, there are other ethical concerns. Some argue that this case could be described as “medical tourism.” While New Hope Medical Clinic does maintain a branch in Mexico, Dr. Zhang stated that Mexico has “no rules.”[12] It is a country with underdeveloped regulations, making it difficult to confirm that doctors adhere to widely-held medical and ethical standards. Furthermore, while it is highly unlikely that the child will develop Leigh’s Syndrome, Dr. Dietrich Egli of the New York Stem Cell Foundation says the 5% mitochondrial transfer rate indicates that the technique “was not carried out well.”[13] He points to studies of embryos where the rate of mtDNA transfer was almost ten times lower. Spindle Nuclear Transfer is currently legal in the UK,[14] and many are hoping to see it legalized in the US, although Congress currently prohibits the FDA from considering applications that would entail trials in people.[15] While Dr. Zhang’s work is arguably revolutionary, many wonder if the manner in which this study was performed—in Mexico and with a high mitochondrial transfer rate—will impact the technology’s future in the US. Last week, Dr. Zhang presented the report to scientists gathered in Salt Lake City, saying that while science isn’t a race, it is “in a sense, a race for the family to find a cure, to find hope.”[16] Only time will tell if this study set back other families racing and hoping for a cure. The replicon hypothesis, first proposed in 1963 by Jacob and Brenner(1), states that DNA replication is controlled at sites called origins. Replication origins have been well studied in prokaryotes. However, the study of eukaryotic chromosomal origins has lagged behind, because until recently there has been no method for reliably determining the identity and location of origins from eukaryotic chromosomes. Here, we review a technique we developed with the yeast Saccharomyces cerevisiae that allows both the mapping of replication origins and an (...) assessment of their activity. Two‐dimensional agarose gel electrophoresis and Southern hybridization with total genomic DNA are used to determine whether a particular restriction fragment acquires the branched structure diagnostic of replication initiation. The technique has been used to localize origins in yeast chromosomes and assess their initiation efficiency. In some cases, origin activation is dependent upon the surrounding context. The recent 2014 Nobel Prize in chemistry honored an era of discoveries and technical advancements in the field of super‐resolution microscopy. However, the applications of diffraction‐unlimited imaging in biology have a long road ahead and persistently engage scientists with new challenges. Some of the bottlenecks that restrain the dissemination of super‐resolution techniques are tangible, and include the limited performance of affinity probes and the yet not capillary diffusion of imaging setups. Likewise, super‐resolution microscopy has introduced new paradigms in the (...) design of projects that require imaging with nanometer‐resolution and in the interpretation of biological images. Besides structural or morphological characterization, super‐resolution imaging is quickly expanding towards interaction mapping, multiple target detection and live imaging. Here we review the recent progress of biologists employing super‐resolution imaging, some pitfalls, implications and new trends, with the purpose of animating the field and spurring future developments. The paper retraces the early reception of La Mettrie’s work L’Homme machine in Germany. It focuses in particular on the role played by Wolff and by Manteuffel’s “Society of Alethiophiles” in promoting a scandalous image of La Mettrie’s work by reducing it to the old formula of mechanistic materialism à la Hobbes. By defending Wolff’s interpretation, Manteuffel contributed discretely but actively to the capillary dissemination of such an image, which was to become very soon the dominant one. By means (...) of their collaboration Wolff and Manteuffel didn’t only introduce a deep and long-lasting misunderstanding of La Mettrie’s materialism in German philosophy; they additionally even vetoed any possible appreciation of his peculiar idea of active matter. Following A. V.Hill, those formulae are developed in a mathematical research which are of interest in the field of physiological diffusion processes in cylindrical objects.At first, an introduction into the problem is given. Then a development follows of the mathematical methods which are suitable for the integration of the partial differential equation of the diffusion.The special part contains the investigation of problems which are importance for the physiology of respiration. Thus the query is answered for the concentration of a substance (...) in dependance of time and place for the diffusion in tissue cylinders taking into consideration a constant rate of consumption, as well as the query of the temporal alteration of the average concentration. The cylinder fed from the outside and the one fed from a central capillary must hereby be specially treated according to the boundary conditions. The developed equations are explained by special examples and numerical lists. Further, the steady state of supply of tissue cylinders is investigated in case that apart from the radial also an axial diffusion component has to be counted with. The numerical results from the developed formulae are in good agreement with the values found by experiments.Suivant A. Puis on a étudié la provisionnement stationnaire des cylindres cellulaires en cas que outre la composante de diffusion radiale, il faut tenir compte à une composante de diffusion axiale . Concrete is one of the most intelligent and widely utilized man-made materials in the construction industry. Despite this, even high-quality concrete is susceptible to porosity, which reduces its serviceability period. Furthermore, there is an increasing need to increase longevity due to environmental exposure such as soil moisture, corrosive outside elements, or structural defects forming in the surface of concrete. The use of crystalline admixtures in concrete is one of the many approaches to reducing these risks. When crystalline admixtures come into (...) contact with water, they form thin crystals that fill pores, capillaries, and micro fractures, as a result making concrete a self-sealing material. When the concrete has dried, the crystalline particles remain dormant until they come into contact with additional water, which causes them to crystallize once more. This research aims to analyze and compare the material properties of commonly used concrete and concrete where crystalline waterproofing is present. Furthermore, experiments are conducted to evaluate each of the concrete samples: compressive strength, water permeability, and flexure strength. As a result, demonstrating the benefits or negative aspects of the use of crystalline admixture in the early stages of concrete is important. It is not yet defined, whether this is the future of cutting-edge concrete and the impact that it will have in the Albanian building market. (shrink)

It has long been thought that gene expression is tightly regulated in multicellular eukaryotes, so that expression profiles match functional profiles. This conception emerged from the assumption that gene activity is synonymous with gene function. This paradigm was first challenged by comparative protein electrophoresis studies showing extensive differences in expression patterns among related species. The paradigm is now being challenged by evolutionary transcriptomics using microarray technologies. Most gene expression profiles display features that lack any obvious functional significance. The so‐called (...) “ectopic” expression refers to the expression of genes at times and locations where the target gene is not known to have a function. However, ectopic expression might be associated with genuine function even if this function is not essential or has yet to be ascertained. Alternatively, ectopic expression might come about as a superfluous by‐product of regulatory systems, which would call for a revision of prevailing ideas about the specificity of gene regulation. We herein review available evidence for ectopic expression and the hypotheses proposed for its origin and evolution. We propose that ectopic expression must be regarded as part of an integrated phenotypic whole. It seems likely that ectopic expression represents a leak in the evolution of regulatory systems, but one that is endowed with considerable evolutionary possibilities. Until quite recently, recombinant DNA technology was not able to deal with DNA molecules larger than 20–40 kb. This is a serious limitation for the study of mammalian, and in particular human genomes whose total length is approx. 3 × 106 kb, since the best resolution of genetic and chromosomal analysis is usually the rough equivalent of 1000–5000 kb. Three recently developed methods promise to bridge this gap: pulsed field gel electrophoresis, which can analyze megabase‐sized DNA fragments; cloning in (...) yeast, which can clone and propagate DNA fragments of several hundred kb; and jumping libraries, which allow ‘jumping’ over large distances along the chromosome. Background: Increasing resistance of Pseudomonas aeruginosa to ciprofloxacin in ICU/burn units has created a problem in the treatment of infections caused by this microorganism. -/- Methods: Fifty P. aeruginosa strains were isolated from burn patients hospitalized in the Kerman Hospital during May 1999-April 2000 and were tested for in-vitro sensitivity to different antibiotics by disc diffusion breakpoint assay. The isolates were subjected to minimum inhibitory concentration (MIC) test by agar dilution method. Existence of the plasmids was also investigated in the (...) isolates. -/- Results: Thirty-four patients infected with ciprofloxacin strains showed MIC of 8 mg/ml [p<0.001]. Sixteen patients were infected with sensitive strains exhibiting MIC range of 0.0125-0.125 ± 0.033 mg/ml. The isolates were also also resistant to other antibiotics [p<0.001]. Plasmid isolation and agarose gel electrophoresis (0.7%) revealed three plasmid bands in strains 8 and 16, and one band in strain 35. -/- Conclusion: The emergence of ciprofloxacin resistance of P. aeruginosa in burn patients is alarming since this antibiotic has only recently been introduced onto the market in Iran. One important observation was that some isolates exhibited cross resistance to other antibiotics. Furthermore, some strains were carriers of plasmids which might have acted as the potential source of acquired resistance in the hospital setting. -/- . (shrink)

In order to accomplish the transition from avascular to vascular growth, solid tumours secrete a diffusible substance known as tumour angiogenesis factor (TAF) into the surrounding tissue. Endothelial cells which form the lining of neighbouring blood vessels respond to this chemotactic stimulus in a well-ordered sequence of events comprising, at minimum, of a degradation of their basement membrane, migration and proliferation. Capillary sprouts are formed which migrate towards the tumour eventually penetrating it and permitting vascular growth to take place. (...) It is during this stage of growth that the insidious process of invasion of surrounding tissues can and does take place. A model mechanism for angiogenesis is presented which includes the diffusion of the TAF into the surrounding host tissue and the response of the endothelial cells to the chemotactic stimulus. Numerical simulations of the model are shown to compare very well with experimental observations. The subsequent vascular growth of the tumour is discussed with regard to a classical reaction-diffusion pre-pattern model. (shrink)
