Melatonin is a hormone with complex roles in the pathogenesis of autoimmune disorders. Over the years, it has become clear that melatonin may exacerbate some autoimmune conditions, whereas it alleviates others such as multiple sclerosis. Multiple sclerosis is an autoimmune disorder characterized by a dysregulated immune response directed against the central nervous system. Indeed, the balance between pathogenic CD4+ T cells secreting IFN‐γ (TH1) or IL‐17 (TH17); and FoxP3+ regulatory T cells and IL‐10+ type 1 regulatory T cells (Tr1 cells) (...) is thought to play an important role in disease activity. Recent evidence suggests that melatonin ameliorates multiple sclerosis by controlling the balance between effector and regulatory cells, suggesting that melatonin‐triggered signaling pathways are potential targets for therapeutic intervention. Here, we review the available data on the effects of melatonin on immune processes relevant for MS and discuss its therapeutic potential. (shrink)

Why do we watch and like horror films? Despite a century of horror film-making and en-tertainment, little research has examined the human motivation to watch fictional horror and how horror film influences individuals’ behavioural, cognitive and emotional re-sponses. This review provides the first synthesis of the empirical literature on the psy-chology of horror film using multi-disciplinary research from psychology, psychotherapy, communication studies, development studies, clinical psychology, and media studies. The paper considers the motivations for people’s decision to watch horror, why (...) people enjoy horror, how individual differences influence responses to, and preference for, hor-ror film, how exposure to horror film changes behaviour, how horror film is designed to achieve its effects, why we fear and why we fear specific classes of stimuli, and how lik-ing for horror develops during childhood and adolescence. The literature suggests that (1) low empathy and fearfulness are associated with more enjoyment and desire to watch horror film but that specific dimensions of empathy are better predictors of people’s re-sponses than are others; (2) there is a positive relationship between sensation-seeking and horror enjoyment/preference but this relationship is not consistent; (3) men and boys prefer to watch, enjoy and seek our horror more than do women and girls; (4) women are more prone to disgust sensitivity or anxiety than are men and this may mediate the sex difference in the enjoyment of horror; (5) younger children are afraid of symbolic stimuli whereas older children become afraid of concrete or realistic stimuli; (6) in terms of coping with horror, physical coping strategies are more successful in younger children; priming with information about the feared object reduces fear and increases children’s enjoyment of frightening television and film. A number of limitations in the literature are identified, including the multifarious range of horror stimuli used in studies, disparities in methods, small sample sizes, and a lack of research on cross-cultural differences and similarities. In what follows I will discuss an example of the Duhem-Quine problem in which Pr(H A), Pr(A H), and Pr(OI +H& ?A) (where H is the hypothesis, A the auxiliary assumptions, and O the observational prediction) can be construed objectively; however, only some of those quantities are relevant to the analysis that I provide. The example involves medical diagnosis. The goal is to test the hypothesis that someone has tuberculosis; the auxiliary assumptions describe the er- ror characteristics of the test (...) procedure. Although it can make sense to talk about the objective probability that someone (randomly drawn from a given population) has tuberculosis and it also can make sense to talk about the objective probability that a test procedure has a certain set of error characteristics, neither of these quantities will enter into the analysis. The analysis proceeds entirely via likelihoods, what one needs to consider is just the probability of the observations conditional on four conjunctions of the form (?H & +A).' It is a special feature of the example that all four of these conjunctions are simple statistical hypotheses in the technical sense that each unambigu-ously confers a probability on the observations.'o After describing how the likelihood concept applies to the example concerning medical diagnosis, I will show how similar patterns can arise in the context of a second inferential framework-that of H. Background Over the last decade, the return of results (ROR) in precision medicine research (PMR) has become increasingly routine. Calls for individual rights to research results have extended the “duty to report” from clinically useful genetic information to traits and ancestry results. ROR has thus been reframed as inherently beneficial to research participants, without a needed focus on who benefits and how. This paper addresses this gap, particularly in the context of PMR aimed at increasing participant diversity, by providing investigator (...) and researcher perspectives on and questions about the assumed value of ROR in PMR.Methods Semi-structured interviews with a purposive sample of investigators and researchers across federally funded PMR studies in three national consortia, as well as observations of study activities, focused on how PM researchers conceptualize diversity and implement inclusive practices across research stages, including navigating ROR.Results Interviewees (1) validated the value of ROR as a benefit of PMR, while others (2) questioned the benefit of clinically actionable results to individuals in the absence of sufficient resources for translating findings into health care for diverse and disadvantaged populations; (3) expressed uncertainties in applying the presumed value of ROR as a benefit for non-clinical results; and (4) and debated when the promise of the value of ROR may undermine trust in PMR, and divert efforts to return value beyond ROR.Conclusions Conceptualizations of diversity and inclusion among PM researchers and investigators raise unique ethical questions where unexamined assumptions of the value of ROR inform study recruitment efforts to enroll minoritized and under-represented populations. A lack of consideration for resources and infrastructure necessary to translate ROR into actionable information may hinder trustworthy community-research relationships. Thus, we argue for a more intentional interrogation of ROR practices as an offer of benefit and for whom. (shrink)

Den moralfilosofiska traditionen har sina begränsningar. De frågor som stått i centrum för den moralfilosofiska diskussionen har ofta varit helt andra än de som människor i det praktiska livet har uppfattat som centrala moraliska problem. En viktig begränsning är att moralfilosofin, liksom beslutsteorin, nästan uteslutande har handlat om hur man hanterar välavgränsade problem där handlingsalternativen är givna. I det verkliga livet löser vi ofta moraliska problem genom att finna på nya handlingsalternativ, som inte fanns med från början.1 En annan begränsning (...) är att moralfilosofin till allra största delen har handlat om situationer där konsekvensen av de olika handlingsalternativen är välkänd. I det praktiska livet är det annorlunda. Vi vet sällan med säkerhet vad som kommer att bli följden av de olika alternativ som vi väljer mellan. Man kan också uttrycka detta så att moralfilosofin i huvudsak rör sig i en tänkt deterministisk värld, där man inte behöver räkna med någon slump. Detta är aningen märkligt mot bakgrund av det oerhörda genomslag som sannolikhetstänkande och på senare år även kaos har fått inom andra vetenskapsområden. Det finns naturligtvis förklaringar till detta moralfilosofins kvardröjande i en äldre världsbild. Redan i det deterministiska fallet uppstår svårhanterliga problem som man gärna ger sin odelade uppmärksamhet. Dessutom finns det en särskild disciplin, beslutsteorin, som behandlar beslut under risk och osäkerhet. Man har i allmänhet tänkt sig ett slags arbetsfördelning, där problemen med risk och osäkerhet utelämnas i moralfilosofin för att i stället hanteras inom beslutsteorin med rena rationalitetsprinciper (dvs. utan att moraliska principer är inblandade). Enligt min mening är denna arbetsfördelning inte hållbar. Betrakta t ex osäkerheten om hur klimatet kommer att förändras med nuvarande utsläppstrender för växthusgaser. Denna osäkerhet ger upphov inte enbart till ett problem om rationellt beslutsfattande utan också till moraliska problem.. (shrink)

Apart from the ethical and legal grounds, there are good practical rea-sons to design more effective counterter-rorism measures. Preventable harms to population health contribute to mistrust and instability and undermine the stated objectives of the intelligence services.

Responds to Leiter's naturalist/realist approach to jurisprudence - particularly his claim that such an approach implies exclusive positivism. Considers analogy with naturalized epistemology. "With regard to the first step the realists were anti-foundationalists in the sense that they 'denied that legal reasons justify a unique decision: the legal reasons underdetermine the decision '. The second step, the replacement suggests that instead of a justificatory account of adjudication, i.e. some prescription as to how judges should decide cases, the reaslists provided an (...) empirical description o fhow judges actually decide cases." Notes that positivists are generally commited to a moderate foundationalism by their acceptance of a RoR. "[T]he analogy with the epistemological anti-foundationalist thesis would suggest that foundationalism in law is committed to the view that if X is a legal norm, it is a legal norm in virtue of inference from self-validating foundations. There is nothing in this view that commits the legal foundationalist to the view that legal norms necessarily cover the entire practical domain, and therefor provide a determinate answer to every practical question...Now, if the anti-foundationl step fails, then we need not, indeed must not, take the second step, because if we take it, we will lsoe something of great importance for our understanding of law. If legal norms qua reasons are part of what determines the outcome of cases, then in order to understand how legal decisions are reached, we cannot disregard them." If Leiter rejects this moderate foundationalism, then it is in tension with his other claim that the realists were positivists. To understand how Dewey, one of the century's foremost philosophers of education, understood the concept of hope, Stephen Fishman begins with theoretical questions like: What is hope? What are its objects? How can hope foster a new understanding of democracy and social justice? The book's second half is a classroom (...) study that mir-rors in practice what Fishman explores in theory, as Lucille McCarthy observes Fishman's undergraduate students reading the theorists. Artificiell intelligens (AI) är ett ungt forskningsområde där många av de grundläggande problemen förefaller att vara av filosofisk art.1 Ämnet har sina filosofiska rötter dels i traditionen från Leibniz, Frege, Russell och Hilbert, som strävar efter att formalisera principerna för exakt tänkande, dels i den klassiska mekanismen: idén att människan är en maskin och att det mänskliga tänkandet är en mekanisk process. Som en första approximation kan vi säga att AI är det vetenskapliga studiet av hur man konstruerar och bygger (...) maskiner (datorer) som är intelligenta (eller uppvisar intelligent beteende). Det är lätt att se att denna forskning aktualiserar frågor av filosofiskt slag. Hit hör problem som rör tänkandets natur samt förhållandet mellan medvetandet och kroppen. Inom AI ses ofta datorns arbetssätt som en modell för människans kognitiva processer och man föreställer sig kanske att hjärnan är ett slags dator. Eftersom hjärnan liknas vid en dator och en dator tänks arbeta rent mekaniskt i enlighet med ett givet program (en given algoritm), så föreställer man sig att samma sak bör gälla för hjärnan. Analogin hjärna-dator leder till föreställningen att tänkande inte är något annat än en dators mekaniska manipulerande av symboler i enlighet med givna formella regler. Denna uppfattning om tänkandets natur förefaller samtidigt problematisk: mekanisk manipulation av symboler förutsätter ingen förståelse av symbolernas innebörd; genuint tänkande, å andra sidan, tycks förutsätta sådan förståelse. Ibland verkar också analogin hjärna-dator i den motsatta riktningen. Eftersom hjärnan liknas vid en dator och hjärnan har en mental dimension, så föreställer man sig kanske att samma sak måste gälla för vissa datorer: synen på hjärnan som en dator leder till tanken att det borde vara möjligt att konstruera datorer som bokstavligen tänker, känner och förnimmer. Man föreställer sig kanske att man skall kunna konstruera ett artificiellt medvetande. Vi är alla förtrogna med datorer som (i en viss mening) kan lösa förelagda uppgifter på ett intelligent sätt, men följer det därav att dessa datorer kan tänka? Inte tänker väl en schackspelande dator på schack? Hur intelligent dagens datorer än beter sig, så verkar det ändå inte rimligt att säga att de har tankar, känslor eller upplevelser. Men kan man utesluta att vi någon gång i framtiden kommer att kunna konstruera datorer som verkligen förstår, är medvetna, och tänker? Dessa frågor rymmer många begreppsliga oklarheter. Vad är intelligens, tänkande, förståelse, medvetande? Hur förhåller sig intelligens och tänkande till medvetandets fenomenella sida: upplevelser och känslor? Om vad slags ting är det meningsfullt att säga att de har intelligens, medvetande, mentala tillstånd, etc.? Är det meningsfullt att säga detta om en hjärna, en maskin, en dator? Vad menas med en maskin (dator)? Hur kan vi veta (avgöra) om ett ting har intelligens, tankar, känslor eller upplevelser? I denna uppsats avser vi att närmare belysa sambandet mellan filosofi och AI samt kritiskt granska några av AI-forskningens förutsättningar inom filosofi och logik. (shrink)

Mein Aufsatz rekonstruiert ein Formexperiment der experimentellen deutschsprachigen Prosa der 1960er Jahre, das sich in Texten aus der Frühphase der edition suhrkamp bei Peter Weiss, Jürgen Becker, Ror Wolf und Peter Handke entwickelte. Anhand der von Hans Magnus Enzensberger geprägten Formel vom »Prinzip der Felder« werden die intensiven Bezüge und Strukturähnlichkeiten, aber auch die Variationsbreite dieser experimentellen Prosa diskutiert. Ein abschließender Blick auf zeitnahe Theorien zum Zusammenspiel von Feldern (Aron Gurwitsch) und Rändern (William James) des Bewusstseins soll zudem die von (...) Jürgen Becker entwickelte These bestärken, nach welcher die »Vielstimmigkeit des Bewusstseins« erstmals in dieser experimentellen Prosa der 1960er Jahre zur Darstellung kam. (shrink)
