This article focuses on the occasional individuals of many species that live longer than is usual for their populations – here called longevity outliers. They appear to be exceptions to the usual patterns of mortality rates that increase with age. There is no model of survivorship that accommodates all of these individuals. They are less vulnerable to the usual causes of death than most in their populations. There are hints of genetically based mechanisms in the form of life‐prolonging genes in (...) invertebrates and genetic resistance to fatal diseases in higher organisms. The reasons why longevity outliers ultimately die are not known. Based on well‐established trends, I predict that there will be many more humans reaching very old ages in the next century. (shrink)

We have synthesized a 582,970-base pair Mycoplasma genitalium genome. This synthetic genome, named M. genitalium JCVI-1.0, contains all the genes of wild-type M. genitalium G37 except MG408, which was disrupted by an antibiotic marker to block pathogenicity and to allow for selection. To identify the genome as synthetic, we inserted "watermarks" at intergenic sites known to tolerate transposon insertions. Overlapping "cassettes" of 5 to 7 kilobases (kb), assembled from chemically synthesized oligonucleotides, were joined by in vitro recombination to produce intermediate (...) assemblies of approximately 24 kb, 72 kb ("1/8 genome"), and 144 kb ("1/4 genome"), which were all cloned as bacterial artificial chromosomes in Escherichia coli. Most of these intermediate clones were sequenced, and clones of all four 1/4 genomes with the correct sequence were identified. The complete synthetic genome was assembled by transformation-associated recombination cloning in the yeast Saccharomyces cerevisiae, then isolated and sequenced. A clone with the correct sequence was identified. The methods described here will be generally useful for constructing large DNA molecules from chemically synthesized pieces and also from combinations of natural and synthetic DNA segments. 10.1126/science.1151721. 1 Evidence-based policy requires researchers to provide the answers to ecological questions that are of interest to policy makers. To find out what those questions are in the UK, representatives from 28 organizations involved in policy, together with scientists from 10 academic institutions, were asked to generate a list of questions from their organizations. 2 During a 2-day workshop the initial list of 1003 questions generated from consulting at least 654 policy makers and academics was used as a basis for (...) generating a short list of 100 questions of significant policy relevance. Short-listing was decided on the basis of the preferences of the representatives from the policy-led organizations. 3 The areas covered included most major issues of environmental concern in the UK, including agriculture, marine fisheries, climate change, ecosystem function and land management. 4 The most striking outcome was the preference for general questions rather than narrow ones. The reason is that policy is driven by broad issues rather than specific ones. In contrast, scientists are frequently best equipped to answer specific questions. This means that it may be necessary to extract the underpinning specific question before researchers can proceed. 5 Synthesis and applications. Greater communication between policy makers and scientists is required in order to ensure that applied ecologists are dealing with issues in a way that can feed into policy. What difference has the dialogue made in my life, personally or theologically?How would I respond to this topic from the point of view that I am a religious person engaged in a specific area of expertise (environment, philosophy, etc.)? Thesis The increased mobility and diversity of relationships in the contemporary world has often made religious identity a complex issue, hardening it for some people and diffusing it for others. But in either case, I wish to challenge the view that highlights beliefs as a key to defining identity and instead propose that many other factors might be more important, and that in religious life identity is a secondary issue. Being Christian I was born, baptized, and raised in the Christian tradition. My father was a United Church of Canada minister and I became a United Church of Canada minister, while my mother was a Baptist and my brother became a Baptist minister. Religious values were important in my upbringing and throughout my adult life. While I have maintained a concern for ultimate values, however, my own beliefs have undergone radical changes. I am sure that many in the Christian community would say that I am no longer a Christian, but I have never taken that view and have never taken the step to renounce my ordination as a Christian minister. Instead, I question the assumption that beliefs are the best way to define a religious life or religious identity. [End Page 9]In 1961 I had the good fortune to go to McGill University to study with Wilfred Smith, and from him I have learned not to focus on the "beliefs" of others—namely, intellectual constructs—but on the "faith" of others as much more encompassing and important. Some religious communities—like the Jain and the Confucian—do not care about one's beliefs, only about one's practice, both ritually and morally. Wilfred Smith called it orthopraxy, rather than orthodoxy, and it provides an alternative means of marking religious identity.Why should I feel bound by the efforts of Emperor Constantine when he convened a church council in 325 CE to define Christian identity or by subsequent efforts of church institutions to do the same? Today, my Christian identity is nurtured and expressed by a constellation of moral values and cosmic trust, of faith and actions, that are connected to the early Jesus movement, especially as articulated by John Dominic Crossan. Living as we do in the modern era, we have very different beliefs about history and cosmology from these early Christians, as John Robinson discussed forty years ago in his book Honest to God. My affection for Christian church life and the early Jesus movement involves other factors than the concerns of those who formed Christian creeds. The Trinitarian doctrine makes sense as a conglomeration made from the conceptualization of several historical experiences important in the early Christian community, but the Trinitarian concept does not clarify or enhance my contemporary experience.In spite of the Trinitarian formulation, I still honor and value the powerful and enduring influence of my Christian upbringing. Auxier’s goal is to achieve an all-round (...) reappraisal of Royce, setting the record straight about major misconceptions accrued throughout the course of Royce scholarship over the past 65 years—if not longer.One of the most significant events in Royce scholarship during the 20th Century was the appearance of John E. Smith’s Royce’s Social Infinite: The Community of Interpretation.1 As Auxier notes: “For the purposes for which it was intended, Smith’s book will never be surpassed in its excellence. Smith’s book is outstanding in numerous ways … but its clarity and brevity are high among its chief virtues” (13). The problem with Smith’s book, for Auxier, is that readers have used it for a purpose it was never intended to serve—that of a crutch or “shortcut to a full understanding of Royce” (13). This tendency has had a compounding effect: “[N]ot only scholarship, but scholarly consciousness about Royce is formed by Smith’s book” (14). But the most damning consequence of Smith’s book is its claim that Royce’s thought underwent a decisive shift in its later stages: “[T]he all-important change in Royce’s conception of the Absolute consists in the shift from the idea that the Infinite thought is an all-embracing consciousness apprehending at a glance all truth … to the idea that the Infinite is actual as a well-ordered system … having a generally triadic form and involving a type of cognition called interpretation” (19).2 Once a bifurcation between the “early” and the “later” Royce is introduced, the tendency is to identify Royce’s “Peircean moment” of 1912 as the decisive point of rupture between the “two Royces”—an incorrect view stemming from the inability to take a longer view of the Peirce-Royce relationship:But our recognition of the intense, constant and long term character of their exchange also eliminates the supposition that that Royce took a sudden turn in 1912. We highlight the various domains of the flavivirus virulence factor NS1 and speculate on potential implications of the NS1 3D structure in understanding its role in flavivirus pathogenesis. Flavivirus non‐structural protein 1 (NS1) is a virulence factor with dual functions in genome replication and immune evasion. Crystal structures of NS1, combined with reconstructions from electron microscopy (EM), provide insight into the architecture of dimeric NS1 on cell membranes and the assembly of a secreted hexameric NS1‐lipid complex found in patient sera. (...) Three structural domains of NS1 likely have distinct roles in membrane association, replication complex assembly, and immune system avoidance. A conserved hydrophobic inner face is sequestered either on the membrane or in the interior of the secreted hexamer and contains regions implicated in viral replication. The exposed variable outer face is presented to cellular and secreted components of the immune system in infected patients and contains candidate regions for immune system modulation. We anticipate that knowledge of the distinct NS1 domains and assembly will lead to advances in elucidating virus‐host interactions mediated through NS1 and in dissecting the role of NS1 in viral genome replication. David Hume is the greatest figure in the empiricist tradition in philosophy and was a particular source of inspiration for the logical positivists (see logical positivism). Hume was born in 1711 and entered Edinburgh University at the age of 12. After graduating, he had a varied career in commerce, diplomacy, as a librarian, and as a writer of history. Twice he was secretary to General St Clair and on one occasion set off with him on an expedition to drive (...) the French out of Canada. Forced back by the wind, it was decided instead to make a brief incursion on the coast of Brittany. This failed, for the expedition had maps of Canada, but none of Brittany. During a three‐year stay in France he wrote his major philosophical work Treatise of Human Nature. It appeared in 1739 and, in his own words, “fell dead‐born from the Press.” His Inquiry Concerning Human Understanding, which was intended to be a more popular version of book I of his Treatise, was published in 1748 but was only a little more favorably received. His six‐volume History of England, on the other hand, was a great success. He died in 1776, never having succeeded in his ambition to be appointed to a professorship of philosophy in Scotland, due to his religious skepticism. (shrink)

We would like to thank the commentators for their generous comments, valuable insights and helpful suggestions. We begin this response by discussing the selfishness axiom and the importance of the preferences, beliefs, and constraints framework as a way of modeling some of the proximate influences on human behavior. Next, we broaden the discussion to ultimate-level (that is evolutionary) explanations, where we review and clarify gene-culture coevolutionary theory, and then tackle the possibility that evolutionary approaches that exclude culture might be sufficient (...) to explain the data. Applied ontologies have been used more and more frequently to enhance systems engineering. In this paper, we argue that adopting principles of ontological realism can increase the benefits that ontologies have already been shown to provide to the systems engineering process. Moreover, adopting Basic Formal Ontology (BFO), an ISO standard for top-level ontologies from which more domain specific ontologies are constructed, can lead to benefits in four distinct areas of systems engineering: (1) interoperability, (2) standardization, (3) testing, and (4) data (...) exploitation. Reaping these benefits in a model-based systems engineering (MBSE) context requires utilizing an ontology’s vocabulary when modeling systems and entities within those systems. If the chosen ontology abides by the principles of ontological realism, a semantic standard capable of uniting distinct domains, using BFO as a hub, can be leveraged to promote greater interoperability among systems. As interoperability and standardization increase, so does the ability to collect data during the testing and implementation of systems. These data can then be reasoned over by computational reasoners using the logical axioms within the ontology. This, in turn, generates new data that would have been impossible or too inefficient to generate without the aid of computational reasoners. (shrink)

It remains unclear whether such genetic variants are associated with disease aggressiveness. The NCI-SPORE Genetics Working Group retrospectively collected clinicopathologic information and genotype data for 36 SNPs which at the time had been validated to be associated with PC risk from 25,674 cases with PC. Cases were grouped according to race, Gleason score and aggressiveness. Statistical analyses were used to compare the frequency (...) of the SNPs between different disease cohorts. After adjusting for multiple testing, only PC-risk SNP rs2735839 was significantly and inversely associated with aggressive and high-grade disease in European men. Similar associations with aggressive and high-grade disease were documented in African-American subjects. The G allele of rs2735839 was associated with disease aggressiveness even at low PSA levels in both European and African-American men. Our results provide further support that a PC-risk SNP rs2735839 near the KLK3 gene on chromosome 19q13 may be associated with aggressive and high-grade PC. Future prospectively designed, case-case GWAS are needed to identify additional SNPs associated with PC aggressiveness. (shrink)

In this paper we seek to draw attention to some striking and heretofore unnoticed textual connections between Adam Smith's Theory of Moral Sentiments and David Hume's A Treatise of Human Nature. We find significant textual parallels between the parable of the poor man's son of TMS 4.1 and the famous conclusion to Book 1 of Hume's Treatise. These passages are often regarded as especially intense and moving parts of their respective works. We explore the nature and substance of (...) these connections and comment on their larger significance. The nature of the connections suggests that Smith consciously engaged Hume in his work through philosophical conversation. We suggest that these related passages show both Hume and Smith exploring and developing a particular dialectic between contemplation and action in human life. Both move to invert the classical relationship between contemplation and action through what we call the elevated imagination. (shrink)

He was a contemporary of Adam Smith and read Smith's The Wealth of Nations shortly before his death. Some commentators have suggested that Hume exercised some influence over Smith's views on economics; others are not so sure. Hume's commentators over (...) the last 200 years have emphasized his theory of knowledge, doctrine of causality and belief, theory of morals and historical writing. His views on economics have been relatively neglected. The purpose of this essay is to summarize and clarify Hume's views on economics. Commerce and Trade Hume's essay "Of Commerce" is an extension of his view of property rights. Just as government should not impair property rights, it should not impair commerce, which is the freedom to contract between consenting parties. But Hume was not a 'pure' advocate of laissez-faire. He supported the infant industry argument: A tax on German linen encourages home manufacturers, and thereby multiplies our people and industry. A tax on brandy encreases the sale of rum, and supports our southern colonies. The bulk of workers are employed either in argiculture or manufacturing. In the more primitive societies, almost everyone is employed in agriculture (husbandry), and as agricultural techniques are improved, the land becomes able to support more nonagricultural workers, including artisans and providers of luxuries as well as manufacturers. Some of these 185 surplus (i.e., nonagricultural or manufacturing) workers are sometimes appropriated by the sovereign for use in the army or navy (OC 256), which prevents them from producing luxury goods and services, and leads to a different kind of state than would exist if their labor were not so appropriated. Increases in industry, the arts and trade generally increase both the power of the sovereign and the happiness of the people (OC 260). Trade is something that is mutually beneficial -- both parties gain. Hume spends a great deal of time refuting the idea that one nation's gain is another's trade loss. Thus, trade is a win-win rather than a win-lose situation, as some modern-day mercantilists (such as Thurow fThe Zero Sum Gamel and Batra G The Great Crash of 1989l)would have us believe. Hume views everything in the world as purchased by labor, and sees labor as being caused by people's passions (OC 261). In modern language, one could say that wealth is produced either from physical or mental labor, and that it is individual incentive that causes the human action to begin in the first place. Of course, this view overlooks the fact that labor does not exist in a vacuum, but is mixed with land and perhaps capital (which can be seen as the result of accumulated labor). Exchange occurs when producers of a commodity produce more than they can consume. At that point, they trade their surplus for other goods and services (OC 261). In peacetime, the farmers' surplus goes toward the acquisition of manufactures or the cultivation of the liberal arts. In war, the surplus can go toward the support of an army or navy. The state becomes more powerful as it can produce increasingly beyond subsistence, what Hume refers to as employment beyond mere necessities (OC 186 262). But rather than encouraging the increase in state power by force, such increase is best accomplished by allowing individuals to produce as much as they want, which they will do automatically in the absence of coercion, and then appropriate part of the surplus: It is a violent method, and in most cases impracticable, to oblige the labourer to toil, in order to raise from the land more than what subsists himself and family. Furnish him with manufactures and commodities, and he will do it himself. Afterwards you will find it easy to seize some part of his superfluous labour, and employ it in the public service, without giving him his wonted return. This is an in-depth study of J.G. Fichte’s philosophy of mathematics and theory of geometry. It investigates both the external formal and internal cognitive parallels between the axioms, intuitions and constructions of geometry and the scientific methodology of the Fichtean system of philosophy. In contrast to “ordinary” Euclidean geometry, in his Erlanger Logik of 1805 Fichte posits a model of an “ursprüngliche” or original geometry – that is to say, a synthetic and constructivistic conception grounded in ideal archetypal elements that (...) are grasped through geometrical or intelligible intuition. -/- Accordingly, this study classifies Fichte’s philosophy of mathematics as a whole as a species of mathematical Platonism or neo-Platonism, and concludes that the Wissenschaftslehre itself may be read as an attempt at a new philosophical mathesis, or “mathesis of the mind.” -/- “This work testifies to the author's exact and extensive knowledge of the Fichtean texts, as well as of the philosophical, scientific and historical contexts. Most academic philosophers and intellectual historians are familiar with the major historical figures and intellectual movements coming out of Scotland in the 18 th Century. These scholars are also familiar with the works of Immanuel Kant and his influence on Western thought. But with the exception of discussion examining David Hume’s influence on Kant’s epistemology, metaphysics, and moral theory, little attention has been paid to the influence of the Scottish Enlightenment thinkers on Kant’s philosophy. _Kant and The Scottish Enlightenment_ (...) aims to fill this perceived gap in the literature and provides a starting point for future discussions looking at the influence of Hume, Thomas Reid, Adam Smith, Francis Hutcheson, and other Scottish Enlightenment thinkers on Kant’s philosophy. The chapters are laid out according to a natural progression of ideas beginning with the influence of Scottish Enlightenment thinkers on Kant’s moral thought, including the role of feelings, reason, and religion. From there, the discussion moves to an examination of the relationship between truth, freedom, and responsibility in the texts coming out of the Scottish Enlightenment and its connection to Kant’s metaphysics and aesthetics. The volume concludes with a discussion of the influence of Scottish Enlightenment thinkers on Kant’s anthropology.
