Activation of resting T lymphocytes through the T cell antigen receptor complex is initiated by critical phosphorylation and dephosphorylation events that regulate the function and interaction of a number of signaling molecules. Key elements in these reactions are members of the Src, Syk and Csk families of protein tyrosine kinases (PTKs) and the phosphotyrosine phosphatases (PTPases) that regulate and/or counteract them, such as CD45. The PTKs can autophosphorylate and phosphorylate each other at multiple sites and, as the result of these (...) interactions, they are induced to phosphorylate other cellular proteins. These phosphorylation events lead to modulation of enzymatic activities and/or serve as binding sites for other signaling molecules having phosphotyrosine‐binding Src homology 2 (SH2) domains. As a result, these proteins translocate to the receptor complexes and are juxtaposed to the kinases that phosphorylate them. Some of the SH2‐domain‐containing polypeptides lack enzymatic activities and, instead, serve as adapter molecules that couple the signal to downstream effectors, such as regulators of the Ras proteins, and further into serine/threonine‐specific protein kinase cascades. Through largely unknown steps these reactions lead to the transcription of previously silent genes, activation of lymphocyte effector functions, progression through the cell cycle and cell proliferation. Anne Conway produced only one short treatise – The Principles of the Most Ancient and Modern Philosophy – but addressed key problems in the philosophy of religion which are still much discussed today. The most significant of these are the problem of religious diversity and the problem of evil. Although the sources of her ideas may be found in the Kabbalah of Isaac Luria and the work of the Cambridge Platonists and the Quaker George Keith, among others, she offers (...) her own unorthodox solutions to these problems. The idea of Christ as an indwelling mediator of the divine who assists us in our moral endeavours is designed to appeal to those of all religions and none, while her claim that God created creatures knowing that they would eventually be redeemed, and that suffering promotes moral development which leads to redemption offers a novel response to the problem of evil. Conway's work is also important for its influence on the work of Gottfried Wilhelm Leibniz, and some have argued that it prefigures, at least in outline, the field theory of modern physics. (shrink)

Due to the COVID-19 pandemic, most employees face increasing career-related stress, particularly those who work in multinational corporations, because the international travel constraints prevent them from going back to their families. Hence, it is imperative to investigate the critical impact of employees’ occupational burnout on career-related outcomes. In response, this research explores the moderating effect of OB on the relationships between career competencies and career sustainability. To achieve a more comprehensive understanding of relevant issues, we adopted a mixed-method research design (...) that includes both qualitative and quantitative analyses. Findings indicate that all the three dimensions of CCs were positively related to CS; moreover, OB negatively moderated the individual relationships of CC and BC with CS. The main contribution of this study is to enrich the existing career literature by addressing the critical moderating role of OB in the links of CC and BC individually with CS. This clause, absent (...) from earlier editions, while implicating the comic cowardice of the narrator also adumbrates a tragic sense of prudence. In the early years of the sixteenth century, not just Rabelais but the rest of his Renaissance world had come increasingly to know the heavy hand of censorship and persecution. Bookburning, of course, had lost its traditional source of terror with the invention of the printing press: no longer could a single bonfire extinguish all versions of a single manuscript. Instead, Church and State authorities had in the course of the Renaissance to invent new practices for controlling and regulating the circulation of dangerous words: bans on publication, excision of offensive passages prior to printing, indices of prohibited books, and so on. When left with no other alternative, however, one could still burn not just sample copies of the book but the offending author too, such as in the case of Giordano Bruno, whose auto-da-fé menacingly lit up the skies in the very first year of the seventeenth century. 1Today, with the advent of electronic media and the virtual extirpation of texts from the materiality of print and paper, it is even more difficult to abolish or even regulate the spread of refractory discourses. Written at a time, over half a century ago, when another wave of explicit attacks on writers shocked the intellectual world, Strauss’s essay, on the one hand, struck a hopeful chord by evoking the kinds of techniques available to writers in their efforts to evade or foil censorship, that is, by demonstrating that no censorship can fully succeed in terroristically bringing about an end to heterodox forms of expression. On the other hand, he posits at least the possibility for every text that it has an inexorable core that is esoteric. No censorship can be total, but there is also no such thing as a pristine state of letters prior to censorship. The Marquis de Sade has been labeled everything from a sadomasochistic pornographer to the fiction writer responsible for the ideas that led to the Nazi death camps. Must We Burn Sade? peels away the negative legacy that has shrouded Sade for too long. The “Burning Bush” was the name of a cultural circle in Bucharest in the 1940s, comprised of clergy and intellectuals who met periodically to discuss theology, philosophy, literature and to learn about prayer. Some of the most significant members of this group were arrested during the second repressive wave by the Romanian communist regime (1958); along with twelve elderly monks and intellectuals, four students who kept in touch with them were also arrested. Their names were George Văsâi, Șerban Mironescu, (...) Nicolae Rădulescu, and Emanoil Mihăilescu. Using memoirs, oral history interviews and documents from the Securitate archives, the paper will address the interactions between the young students and the elder members of the Burning Bush group of clergy and intellectuals. The narratives of the informative and criminal files of the Securitate regarding the content of their meetings will also be depicted, all inside a larger context of the regime’s repressive measures and relations with the Romanian Orthodox Church. Sherwood Anderson's Winesburg, Ohio, published in 1919, is an episodic collection of character sketches based mostly around the perspective of George Willard, a small-town journalist who listens to the stories of various characters, often described in grotesque terms, whose passionate inner lives contrast with their limited outwardly lived existences. The initial critical response to these stories was to regard Anderson as a sort of cheap Freudian who was making an obvious criticism of American Puritanism and conformity. One reviewer, Regis (...) Michaud, wrote that Winesburg, Ohio was "entirely in keeping with the most recent contributions of American literature to psychoanalysis";1 another reviewer, H. W. Boynton. (shrink)

George Molnar came to see that the solution to a number of the problems of contemporary philosophy lay in the development of an alternative to Hume's metaphysics. This alternative would have real causal powers at its centre. Molnar set about developing a thorough account of powers that might persuade those who remained, perhaps unknowingly, in the grip of Humean assumptions. He succeeded in producing something both highly focused and at the same time wide-ranging. He showed both that the notion (...) of a power was central and that it could serve to dispel a number of long-standing philosophical problems.Molnar's account of powers is as realist as any that has so far appeared. He shows that dispositions are as real as any other properties. Specifically, they do not depend for their existence on their manifestations. Nevertheless, they are directed towards such manifestations. Molnar thus appropriates the notion of intentionality, from Brentano, and argues that it is the essential characteristic of powers. He offers a persuasive case for there being some basic and ungrounded powers, thus ruling out the reducibility of the dispositional to the non-dispositional. However, he does allow that there are non-power properties as well as power properties. In this respect, his final position is dualistic.This is contemporary metaphysics of the highest quality. It is a work that was almost complete when its author died. It has been edited for publication by another specialist in the subject, Stephen Mumford, who has also provided an introduction that will allow non-specialists to become acquainted with the issues. The American Medical Association enacted its Code of Ethics in 1847, the first such national codification. In this volume, a distinguished group of experts from the fields of medicine, bioethics, and history of medicine reflect on the development of medical ethics in the United States, using historical analyses as a springboard for discussions of the problems of the present, including what the editors call "a sense of moral crisis precipitated by the shift from a system of fee-for-service medicine to a (...) system of fee-for-system medicine, better known as 'managed care.'" Historians go to great lengths to avoid confronting discontinuity, searching for explanations as to why such events as the fall of the Berlin Wall, George W. Bush's invasion of Iraq, and the introduction of the euro logically develop from what came before. _Moved by the Past_ radically breaks with this tradition of predating the past, incites us to fully acknowledge the discontinuous nature of discontinuities, and proposes to use the fact that history is propelled by unforeseeable leaps and bounds (...) as a starting point for a truly evolutionary conception of history. Integrating research from a variety of disciplines, Eelco Runia identifies two modes of being "moved by the past": regressive and revolutionary. In the regressive mode, the past may either overwhelm us--as in nostalgia--or provoke us to act out what we believe to be solidly dead. When we are moved by the past in a revolutionary sense, we may be said to embody history: we burn our bridges behind us and create accomplished facts we have no choice but to live up to. In the final thesis of _Moved by the Past_, humans energize their own evolution by habitually creating situations ("catastrophes" or sublime historical events) that put a premium on mutations. This book therefore illuminates how every now and then we chase ourselves away from what we were and force ourselves to become what we are. We set out by noting the preference for circular flows in ecological thought, and the related abhorrence of inefficiency and waste that Western ecology shares with mainstream economic thinking. This has often been manifest in a shared disdain both for uncontained, free-burning fire and for ‘unmanaged’ sexual desire. The paper constructs a ‘pyrosexual’ counter-narrative that explores the mutually constitutive and generative implication of sex and fire. Bringing together the solar ecology of Georges Bataille, feminist and queer thinking about sexuality and (...) reproduction, and a range of ways of theorising biological life and fire, we explore how fire mediates between organismic desire and the energetic dynamics of the earth and solar system. The first section takes a genealogical approach to fire and sex that traces their entanglement from the initial ‘assembling’ of fire through to the emergence of a fire-handling creature. The second section looks at how fire has been contained and intensified by human actors, and the role that heat-driven transformations of inorganic matter have played in the incitement and channeling of desire in urban spaces. The third section addresses the development of industrial ‘heat engines’ and the implications for desire and reproduction of tapping vast reservoirs of subterranean solar energy. We round off by beginning to consider what alternative possibilities might lie in the renegotiation of sex and fire on a planet undergoing rapid change. (shrink)

In works of literary fiction, it is a part of the fiction that the words of the text are being recounted by some work-internal 'voice': the literary narrator. One can ask similarly whether the story in movies is told in sights and sounds by a work-internal subjectivity that orchestrates them: a cinematic narrator. George M. Wilson argues that movies do involve a fictional recounting (an audio-visual narration ) in terms of the movie's sound and image track. Viewers are usually (...) prompted to imagine seeing the items and events in the movie's fictional world and to imagine hearing the associated fictional sounds. However, it is much less clear that the cinematic narration must be imagined as the product of some kind of 'narrator' - of a work-internal agent of the narration. Wilson goes on to examine the further question whether viewers imagine seeing the fictional world face-to-face or whether they imagine seeing it through some kind of work-internal mediation . It is a key contention of this book that only the second of these alternatives allows one to give a coherent account of what we do and do not imagine about what we are seeing on the screen. Having provided a partial account of the foundations of film narration, the final chapters explore the ways in which certain complex strategies of cinematic narration are executed in three exemplary films: David Fincher's Fight Club , von Sternberg's The Scarlet Empress , and the Coen brothers' The Man Who Wasn't There.
