The definition of precision medicine has continued to evolve, partly in response to criticism of the original concept. However, whatever the definition or current state of the brand, it fundamentally relies on a putatively tight linkage between genotype and complex human traits. If such a linkage is truly robust, then it should be possible to predict the occurrence of complex traits, both good and bad. If such prediction is possible, it should also be feasible to intervene to prevent or preempt (...) disease-related traits. Stiff Person Syndrome is a rare autoimmune disorder associated with antibodies against glutamic acid decarboxylase, the key enzyme in γ -aminobutyric acid synthesis. In order to investigate the role of cerebral benzodiazepinereceptor binding in SPS, we performed [ 11 C]flumazenil positron emission tomography in a female patient with SPS compared to nine healthy controls. FMZ is a radioligand to the postsynaptic central benzodiazepine receptor which is co-localized with the GABA-A receptor. In the SPS patient, we found a global reduction of (...) cortical FMZ binding. In addition, distinct local clusters of reduced radiotracer binding were observed. These data provide first in vivo evidence for a reduced postsynaptic GABA-A receptor availability which may reflect the loss of GABAergic neuronal inhibition in SPS. (shrink)

The increasing digitalization in the field of psychological and educational testing opens up new opportunities to innovate assessments in many respects (e.g., new item formats, flexible test assembly, efficient data handling). In particular, computerized adaptive testing provides the opportunity to make tests more individualized and more efficient. The newly developed continuous calibration strategy (CCS) from Fink, Born, Spoden, and Frey (2018) makes it possible to construct computerized adaptive tests in application areas where separate calibration studies are not feasible. Due (...) to the goal of reporting on a common metric across test cycles, the equating is crucial for the CCS. The quality of the equating depends on the common items selected and the scale transformation method applied. Given the novelty of the CCS, the aim of the study was to evaluate different equating setups in the CCS and to derive practical recommendations. The impact of different equating setups on the precision of item parameter estimates and on the quality of the equating was examined in a Monte Carlo simulation, based on a fully crossed design with the factors common item difficulty distribution (bimodal, normal, uniform), scale transformation method (mean/mean, mean/sigma, Haebara, Stocking-Lord), and sample size per test cycle (50, 100, 300). The quality of the equating was operationalized by three criteria (proportion of feasible equatings, proportion of drifted items, and error of transformation constants). The precision of the item parameter estimates increased with increasing sample size per test cycle, but no substantial difference was found with respect to the common item difficulty distribution and the scale transformation method. With regard to the feasibility of the equatings, no differences were found for the different scale transformation methods. However, when using the moment methods (mean/mean, mean/sigma), quite extrem levels of error for the transformation constants A and B occurred. Among the characteristic curve method the performance of the Stocking-Lord method was slightly better than for the Haebara method. Thus, while no clear recommendation can be made with regard to the common item difficulty distribution, the characteristic curve methods turned out to be the most favorable scale transformation methods within the CCS. This paper clarifies Wittgenstein’s critique of Moore in _On Certainty,_ and argues that this critique is largely misunderstood, for two reasons. Firstly, Wittgenstein partly misrepresents Moore. Secondly, Wittgenstein is wrongly taken to be an internalist regarding justification for knowledge. Once we realize these two points, we can understand Wittgenstein’s critique properly as a grammatical argument in that Moore fails to see how the concepts of knowledge and certainty relate to those of justification and evidence. On this reading, we can also (...) understand that Moore and Wittgenstein were in more agreement than many people have thought, even though Moore was not able to exploit and express his philosophical insights properly. There is tension between commonly held views concerning phenomenal imagery on the one hand and our first-person epistemic access to it on the other. This tension is evident in many individual issues and experiments in philosophy and psychology (e.g. inattentional and change blindness, the speckled hen, dream coloration, visual periphery). To dissolve it, we can give up either (i) that we lack full introspective access to the phenomenal properties of our imagistic experiences, or (ii) that phenomenal imagery is fully determined, (...) or (iii) that phenomenal imagery does not exist. Which option is preferable? I explore the feasibility of option (ii) in more details by distinguishing between di erent kinds of indeterminacy. One of the most often proclaimed versions of indeterminacy is tied to Representationalism. However, I argue that in this context, Representationalism appears to be the least satisfying option, for which I give ten reasons. By abduction, we should reject Representationalism in favour of (i) or (iii)—the most parsimonious option being eliminativism, and the most conservative being the rejection of privileged access. The decision between these two, however, should be made on a case-by-case basis, depending on which is most adequate given the data. I illustrate this with the comparison between visual agnosia and Charles-Bonnet-Syndrome. (shrink)

In this article, we offer a systematic description of intermedia relations across music, literature, the visual arts, and film. Beginning with some general reflections on materiality and medium in these diverse fields of art, we then offer various examples consisting of two media (e.g. music film, song, images in writing). We pursue the question if, and how, one of the two media may take priority over the other. In our conclusion, we deal with „intermedia relations in monomediality“. This section focuses (...) on artistic representations made in one medium, but based on reflections on another medium. For instance, this is the case when a novelist writes about a picture without having this picture reprinted in the text. (shrink)

Is a phenomenal pain a conscious primitive or composed of more primitive phenomenal states? Are pain experiences necessarily or only contingently unpleasant? Here, I sketch how to answer such questions concerning intra-phenomenal metaphysics using the example of pain and unpleasantness. Arguments for a symmetrical metaphysical independence of phenomenal pain and unpleasant affect are presented, rejecting a composite view like the IASP definition and dimensional views. The motivating intuition of these views is explained by common binding mechanisms in consciousness and characterized (...) as fallacious if generalized. There are, however, underlying commonalities between pain perception and unpleasant affect, e.g. formal content or evolutionary ancestry. (shrink)

This paper presents a new argument against the view that structural (or attitude-based) irrationality consists in failing to respond correctly to normative reasons. According to this view, a pattern of attitudes is structurally irrational if and only if that pattern guarantees that one has at least one attitude one ought not to have. I argue that this ought-violation view comes with three indispensable theoretical commitments. These commitments concern various relationships between normative permissions and oughts that govern beliefs and intentions, and (...) they may appear plausible when viewed in isolation. However, once they are collectively conjoined, they imply oughts that are incompatible with the ought-violation view of structural irrationality. In particular, on the ought-violation view, these oughts entail that you are necessarily structurally irrational whenever [you intend to p] and [you believe that it is not the case that you ought to p]. However, since you can rationally intend things that you believe to be normatively optional (e.g. merely permitted or supererogatory), the ought-violation view faces the following dilemma: it must either deny one of its essential commitments or accept as irrational a set of attitudes that are clearly not. Either way suffices to show that the ought-violation view cannot serve as a correct analysis of structural irrationality. (shrink)

Michael Smith has resisted Harry Frankfurt's claim that moral responsibility does not require the ability to have done otherwise. He does this by claiming that, in Frankfurt cases, the ability to do otherwise is indeed present, but is a disposition that has been `finked' or masked by other factors. We suggest that, while Smith's account appears to work for some classic Frankfurt cases, it does not work for all. Due to the terrorist attacks of 9/11 and the anthrax letters of a few weeks later, the concept of dual use has spread widely in the life sciences during the past decade. This article is aimed at a clarification of the dual use concept and its scope of application for the life sciences. Such a clarification would greatly facilitate the work of policymakers seeking to ensure security while avoiding undesirable interventions of government in the conduct of science. The article starts (...) with an overview of the main developments in life sciences in relation to dual use. This is illustrated by discussions on synthetic biology and dual use. The findings lead to a reconsideration of the dual use concept. An area in need of further attention is to what extent threats and intentions should have impact on the definition of dual use. Possible threats are analyzed against the background of the phenomenon of securitization of health care and life sciences: considering these sectors of society in security terms. Some caveats that should be taken into account in a dual use policy are described. An acceptable, adequate and applicable definition of the dual use concept could help researchers, universities, companies and policy makers. Such a definition should build upon, but go beyond, the view developed in the influential Fink-report, which concentrates on the so-called ‘experiments of concern’, e.g. experiments that enhance the virulence of pathogens (National Research Council of the National Academies 2004) It will be argued that—in addition to these more technical aspects—a definition of dual use should include the aspect of threats and intentions. Concerns about “classroom advocacy” and “politicized teaching” have recycled into near-critical mass, even in the mass media. The book is well-arranged, too, with a (...) solid sample of deconstructionist, New Historicist, Marxist, feminist, and cultural studies, and more mix-and-match theorists describing their classroom aims and means in twenty-one essays grouped into four sections. Sadoff and Cain offer elegantly evasive introductions—not only to these essays per se, but to the issues these essays purport to address. In “Surveying Theory,” David B. Downing, John Kucich, Susan S. Lanser, Lynette Felber, Donald G. Marshall, and Gary Waller lay broadly “anti-foundational” groundwork. In “Focusing,” Diana Fuss, Sandra A. Grayson, Beverly Lyon Clark, Evan Carton, Laurie A. Finke, Jonathan Arac, and Cary Nelson deconstruct things in more detail. In “Theory and Culture,” Huston Diehl, Cannon Schmitt and Donald Ulin, Lindon Barrett, Simon Gikandi, D. N. Rodowick, and Jan Gorak again widen the scope of “theorized teaching,” aiming regulation anticanonical shrapnel at every bastion of coded convention. And while readers may pause over [End Page 336] its backcover promise of “lively and accessible prose,” on the whole, the book is unusually readable for an MLA publication about theory.Since Teaching Contemporary Theory to Undergraduates is a how-to directed at true-believers, it might seem like critical blindsiding to address its fundamental assumptions rather than its tactical suggestions, to focus on its “theory” rather than on its pedagogy. Yet such a focus is not only fair but necessary. The pedagogy is in this book a pawn of the theory. And the theory is, at best, a lesson in how not to teach students how to learn. For despite all its noisy celebrations of methodological, cultural, ontological, and occasionally even human “difference,” the anthology is really a chorus of self-defeating dogma. 1 Like the virtue of variety, even the relative absence of jargon here works as much against as for the book, revealing self-contradictions so perfectly unwitting that they are twice as funny. It is a good thing, then, that the authors eventually contradict these claims, too—even if they know not that they do.The canon? In accordance with postmodern tradition, “the canon” can be calibrated to be whatever best attacks whatever other tradition a particular theorist is aiming at. It is a very modular “monolith.” And theorists never try to “overdetermine” definitions, anyway. The classroom? As an “overdetermined” site of class-struggle, “the classroom (and the academy at large) is... a social institution designed to reproduce the interests, values, and politics of the dominant social classes” (p. 104). Convention? Anything “mainstream” is so polluted with “the notion of mastery, of total and complete knowledge” that even assigning “mainstream” theorists in a theory class “reproduces that hegemony, no matter how critically we read them” (pp. 105, 61). The runoff of mainstream theorists quoted here—conventional MLA Style—makes the same point. Culture? As always already—and only—“a struggle between power and its disenfranchised shadow,” culture “systematically excludes and marginalizes disenfranchised social members” (pp. 263–64, 20–21).Education? As Paulo Freire has revealed, 2 on the one, bad, goatish, wrong Right-wing hand of God, there is the repressive “banking” model of education; on the other, good-shepherdish, right Left-wing side, there is the liberating “problem-posing” model. You are either a droning clerk bent on subduing students into serfdom, or you are a resistance fighter lighting fuses of freedom. “Instrumental activity” does “pedagogical violence,” making the teacher a tyrant and the student a victim, a “submissive imitator” in a “system” that’s “abidingly committed [End Page 337] to the vision of a white-supremacist oligarchy.” Even asking students what level of “guidance” they feel they need is imposing “rhetorical control” (pp. 32, 23, 222, 135).Facts? James Connelly and Giuseppina D'Oro present a new edition of R. G. Collingwood's classic work of 1933, supplementing the original text with important related writings from Collingwood's manuscripts which appear here for the first time. The editors also contribute a substantial new introduction. The volume will be welcomed by all historians of twentieth-century philosophy.

John Buridan was a fourteenth-century philosopher who enjoyed an enormous reputation for about two hundred years, was then totally neglected, and is now being 'rediscovered' through his relevance to contemporary work in philosophical logic. The final chapter of Buridan's Sophismata deals with problems about self-reference, and in particular with the semantic paradoxes. He offers his own distinctive solution to the well-known 'Liar Paradox' and introduces a number of other paradoxes that will be unfamiliar to most logicians. Buridan also moves on (...) from these problems to more general questions about the nature of propositions, the criteria of their truth and falsity and the concepts of validity and knowledge. This edition of that chapter is intended to make Buridan's ideas and arguments accessible to a wider range of readers. In this paper, I argue that the claim that changing the past entails a contradiction ultimately rests upon an empirical assumption, and so the conclusion that changing the past is logically impossible is to be resisted. I then present and discuss a model of time which drops the empirical assumption and coherently models changing the past. Finally, I defend the model, and changing the past, (...) against objections.
