Fundamental problems in developmental biology, such as the generation of asymmetry, differential transcription, and the execution of positional information, are all exhibited by a prokaryotic system, the Caulobacter cell cycle. Progeny cells resulting from each cell division have different developmental programs with respect to patterns of transcription and the initiation of DNA replication. The binary fission that yields dissimilar progeny results from asymmetry generated in the parent cell. An aspect of this fundamental developmental event is the temporally‐controlled biogenesis of a (...) polar flagellum. Several flagellar promoters require the histone‐like protein, integration host factor (IHF) for transcription activation. IHF introduces bends in DNA resulting in the reorganization of the higher‐order structure of the promoter region. These promoters appear to be transcribed from the chromosome in the swarmer pole of the predivisional cell. The newly replicated chromosomes in the predivisional cell differ in sedimentation coefficient and in the ability to initiate replication in the progeny cell. We hypothesize that the differential higher order structure of the two chromosomes contributes to both replication potential and transcription specificity. (shrink)

An early step in sporulation of the bacterium Bacillus subtilis, is the formation of two compartments in the developing sporangium: the mother cell and the forespore. These compartments differ in their programs of gene expression and developmental fate. The establishment of cell type within this simple developmental program, is accomplished by the compartmentalization of sigma subunits of RNA polymerase. The localization of these sigma factors results in compartment‐specific gene expression. Recent experiments have elucidated some of the early steps in the (...) establishment of 3V cell type. After septum formation, the activity of the sigma factor, σF, is confined to the forespore compartment. This, in turn, results in the localized expression of another developmental sigma factor, σG. The forespore localization of these two sigma factors, establishes the forespore line of gene expression. σF and σG also regulate mother cell events. σF activity in the forespore regulates the proteolytic processing of σE within the mother cell compartment. The localization σE activity leads to mother cell expression of another sigma factor, pro‐σK The proteolytic processing of pro‐σK to mature σK is controlled by the forespore sigma factor, σG Mature σK then directs the transcription of mother cell specific genes. Therefore, the initial localization of σF activity to the forespore compartment, orchestrates the establishment of cell type in both forespore and mother cell compartments. (shrink)

What is new about the present study is that the author approaches the religious metamorphosis from the perspective of the evolution of consciousness itself. The result of his exploration made the author realize that the development of European consciousness was not just an accidental historical process.

Critics of South Park make claims that are strikingly similar to those that have been leveled against Western philosophy since its beginnings. Philosophy, it's been charged, also mocks religious beliefs, leads younger folks to question accepted authority and values, and corrupts our children and culture. These condemnations formed the basis for Socrates' trial and execution in Athens. This chapter explores the heretical possibility that people perceive South Park as dangerous precisely because it is a form of philosophy. The “danger” South (...) Park poses has to do with its depiction of dialogue and free thought. In the end, one can learned something: like Socrates, the South Park harms no one. Ethicists regularly turn to virtue in order to negotiate features of climate change that seem to overwhelm moral agency. Appeals to virtue in climate ethics differ by how they connect individual flourishing with collective responsibilities and by how they interpret Anthropocene relations. Differences between accounts of climate virtue help critique proposals to reframe global ecological problems in terms of resilience and planetary stewardship, the intelligibility of which depends on connecting what would be good for the species with what would be (...) good for an individual life. A pragmatic way of establishing that connection may need a strong role for respect of nature. (shrink)

The fields of environmental ethics and of religion and ecology have been shaped by Lynn White Jr.'s thesis that the roots of ecological crisis lie in religious cosmology. Independent critical movements in both fields, however, now question this methodological legacy and argue for alternative ways of inquiry. For religious ethics, the twin controversies cast doubt on prevailing ways of connecting environmental problems to religious deliberations because the criticisms raise questions about what counts as an environmental problem, how religious traditions change, (...) and whether ethicists should approach problems and traditions with reformist commitments. This article examines the critiques of White's legacy and presents a pluralist alternative that focuses religious ethics on the contextual strategies produced by moral communities as they confront environmental problems.
