In this study, we discuss the psychological acceptability of an utterance strategy used by the Driving Support Agent. Previous literature regarding DSA suggests that the adoption of a small robot as a form will increase acceptability. However, the agent’s utterance has been reported as a problem faced by the user. Therefore, in this study, we designed the agent’s utterance using politeness strategy as described by Brown and Levinson’s famous sociolinguistics and pragmatics theory and analyzed its acceptability through a participant-based experiment. (...) In this experiment, we used positive and negative politeness strategies. In general, PPS is utilized to reflect the desire to be liked/recognized by others, whereas NPS is utilized to reflect the need for not wanting to be disturbed by others. Based on our results, PPS was rated high compared to NPS. Therefore, many participants highly evaluated PPS. However, there was a group of participants who appreciated NPS. There were also participants who evaluated the two strategies equally. The number of participants in these three groups was observed at 4:1:1. Correct chromosome segregation in mitosis relies on chromosome biorientation, in which sister kinetochores attach to microtubules from opposite spindle poles prior to segregation. To establish biorientation, aberrant kinetochore–microtubule interactions must be resolved through the error correction process. During error correction, kinetochore–microtubule interactions are exchanged (swapped) if aberrant, but the exchange must stop when biorientation is established. In this article, we discuss recent findings in budding yeast, which have revealed fundamental molecular mechanisms promoting this “swap and stop” process for error correction. (...) Where relevant, we also compare the findings in budding yeast with mechanisms in higher eukaryotes. Evidence suggests that Aurora B kinase differentially regulates kinetochore attachments to the microtubule end and its lateral side and switches relative strength of the two kinetochore–microtubule attachment modes, which drives the exchange of kinetochore–microtubule interactions to resolve aberrant interactions. However, Aurora B kinase, recruited to centromeres and inner kinetochores, cannot reach its targets at kinetochore–microtubule interface when tension causes kinetochore stretching, which stops the kinetochore–microtubule exchange once biorientation is established. The electron double-slit interference is re-examined from the point of view of temporal topos. Temporal topos (or t-topos) is an abstract algebraic (categorical) method using the theory of sheaves. A brief introduction to t-topos is given. When the structural foundation for describing particles is based on t-topos, the particle-wave duality of electron is a natural consequence. A presheaf associated with the electron represents both particle-like and wave-like properties depending upon whether an object in the site (t-site) is specified (particle-like) or (...) not (wave-like). It is shown that the localization of the electron at one of the slits is equivalent to choosing a particular object in the t-site and that the electron behaves as a wave when it passes through a double-slit because there are more than one object in the t-site. Also, the single-slit diffraction is interpreted as a result of the possibility of many different ways of factoring a morphism between two objects. (shrink)

In bacteria, PriA protein, a conserved DEXH‐type DNA helicase, plays a central role in replication restart at stalled replication forks. Its unique DNA‐binding property allows it to recognize and stabilize stalled forks and the structures derived from them. Cells must cope with fork stalls caused by various replication stresses to complete replication of the entire genome. Failure of the stalled fork stabilization process and eventual restart could lead to various forms of genomic instability. The low viability of priA null cells (...) indicates a frequent occurrence of fork stall during normal growth that needs to be properly processed. PriA specifically recognizes the 3′‐terminus of the nascent leading strand or the invading strand in a displacement (D)‐loop by the three‐prime terminus binding pocket (TT‐pocket) present in its unique DNA binding domain. Elucidation of the structural basis for recognition of arrested forks by PriA should provide useful insight into how stalled forks are recognized in eukaryotes. (shrink)

Since the realisation that the antigen‐binding regions of antibodies, the variable (V) regions, can be uncoupled from the rest of the molecule to create fragments that recognise and abrogate particular protein functions in cells, the use of antibody fragments inside cells has become an important tool in bioscience. Diverse libraries of antibody fragments plus in vivo screening can be used to isolate single chain variable fragments comprising VH and VL segments or single V‐region domains. Some of these are interfering antibody (...) fragments that compete with protein‐protein interactions, providing lead molecules for drug interactions that until now have been considered difficult or undruggable. It may be possible to deliver or express antibody fragments in target cells as macrodrugs per se. In future incarnations of intracellular antibodies, however, the structural information of the interaction interface of target and antibody fragment should facilitate development of binding site mimics as small drug‐like molecules. This is a new dawn for intracellular antibody fragments both as macrodrugs and as precursors of drugs to treat human diseases and should finally lead to the removal of the epithet of the ‘undruggable’ protein‐protein interactions. Actions shape what we see and memorize. A previous study suggested the interaction between motor and memory systems by showing that memory encoding for task-irrelevant items was enhanced when presented with motor-response cues. However, in the studies on the attentional boost effect, it has been revealed that detection of the target stimulus can lead to memory enhancement without requiring overt action. Thus, the direct link between the action and memory remains unclear. To exclude the effect of the target detection process (...) as a potential confounder, this study assessed the benefit of action for memory by separating items from the response cue in time. In our pre-registered online experiment, participants responded to visual Go cues by pressing a key or counting while ignoring No-go cues. In each trial, two task-irrelevant images were sequentially presented after the cue disappearance. After encoding the Go/No-go tasks, participants performed a surprise recognition memory test for those images. Importantly, we quantified the impact of overt execution of the action by comparing memories with and without motor response and the impact of covert motor processes by comparing memory between the motor and cognitive tasks. The results showed no memory differences between Go and No-go trials in the motor task. This means that the execution itself was not critical for memory enhancement. However, the memory performance in the motor No-go trials was higher than that in the cognitive No-go trials, only for the items presented away from the cues in time. Therefore, engaging the motor task itself could increase incidental memory for the task-irrelevant items compared to a passive viewing situation. We added empirical evidence on the online interaction between action and memory encoding. These memory advantages could be especially brought in action preparation and planning. We believe this fact may expand our present understanding of everyday memory, such as active learning. (shrink)

The simultaneous perception of multimodal sensory information is important for effective reactions to the external environment. In relation to the effect on time perception, voluntary movement and rhythmic stimuli have already been identified in previous studies to be associated with improved accuracy of temporal order judgments. Here, we examined whether the combination of voluntary movement and rhythmic stimuli improves the just noticeable difference in audio-tactile TOJ Tasks. Four different experimental conditions were studied, involving two types of movements and two types (...) of stimulus presentation. In the voluntary movement condition, after the auditory stimulus participants moved their right index finger voluntarily and naturally, while in the involuntary movement condition, their right index finger was moved by the tactile device. The stimuli were provided in a rhythmic or one-time only manner by hitting inside the first joint of the participants' right index finger using a tactile device. Furthermore, in the rhythmical tactile conditions, tactile stimuli were presented rhythmically to the right index finger 5 times consecutively. On the other hand, in the one-time tactile conditions, tactile stimuli was presented one-time only to the right index finger. Participants made an order judgment for the fifth tactile stimuli and the first and only auditory stimuli. In our TOJ tasks, auditory-tactile stimulus pairs were presented to participants with varying stimulus-onset asynchronies. For the two stimuli presented at a time that were shifted by the SOA, the participants were asked to judge which one was presented first, and they were given a two-choice answer. Using a non-parametric test, our results showed that voluntary movement and rhythmic tactile stimuli were both effective in improving the JNDs in TOJ Tasks. However, in the combination of voluntary movement and rhythmic tactile stimuli, we found that there was no significant difference in JNDs in our experiments. (shrink)
