The presence of vegetation on railway tracks threatens track safety and longevity. However, vegetation inspections in Sweden are currently being carried out manually. Manually inspecting vegetation is very slow and time consuming. Maintaining an even quality standard is also very difficult. A machine vision-based approach is therefore proposed to emulate the visual abilities of the human inspector. Work aimed at detecting vegetation on railway tracks has been split into two main phases. The first phase is aimed at detecting vegetation on (...) the tracks using appropriate image analysis techniques. The second phase is aimed at detecting the rails in the image to determine the cover of vegetation that is present between the rails as opposed to vegetation present outside the rails. Results achieved in the current work indicate that the machine vision approach has performed reasonably well in detecting the presence/absence of vegetation on railway tracks when compared with a human operator. This article shows that William E. Connolly’s work holds resources for projects of racial justice but must be revised to fully meet the challenge of racial inequality. There are two interrelated problems in Connolly’s theory: first, the drive to destabilize identity, for which he argues, rejects the need for collective identity, which is necessary in democratic politics. Furthermore, because domination renders identity unstable, the call to destabilize identity places too great a burden on already marginalized groups. The problem of destabilizing (...) identity is underwritten by a second problem: the white working class and its grievances occupy too important a position in Connolly’s analysis. This article uses insights provided by James Baldwin to amend Connolly’s contributions. The article argues that there is a need to destabilize the ‘we’ of American politics that allows white, male identity to occupy the ‘heart’ of liberal democracy. Baldwin’s insights help solve the problem of the ‘black hole’ at the center of liberal democracy which, Connolly argues, sustains fundamentalism and racial injustice. This article’s contribution is to amend Connolly’s work to meet the challenge of racial injustice, offer a novel reading of the place of political imagination in Baldwin’s work, and show how political imagination adds depth to conversations about democracy, racial justice, and pluralism. Spatially and temporally restricted expression of Hox genes requires multiple mechanisms at both the transcriptional and the post-transcriptional levels. New insight into this precise expression mechanism comes from recent findings of a novel sense–antisense miRNA combination from the bithorax complex of Drosophila melanogaster.1-4 These two miRNAs encoded from the same locus target 3′ untranslated regions of anterior hox genes, Antp, Ubx and abd-A to establish the dominance of posterior hox gene Abd-B in its expression domain. . Speaker recognition has been an active research area for many years. Methods to represent and quantify information embedded in speech signal are termed as features of the signal. The features are obtained, modeled and stored for further reference when the system is to be tested. Decision whether to accept or reject speakers are taken based on parameters of the data modeling techniques. Real world offers various degradations to the signal that hamper the signal quality. The degradations may be due (...) to ambient background noise, reverberation or multispeaker scenario. This paper presents a survey of various feature extraction, data modeling methods, metrics that are used to take the decisions and methods that can be used to preprocess the degraded data that have been used to perform the task of speaker recognition. Serial analysis of gene expression (SAGE) is a powerful technique that can be used for global analysis of gene expression. Its chief advantage over other methods is that it does not require prior knowledge of the genes of interest and provides qualitative and quantitative data of potentially every transcribed sequence in a particular cell or tissue type. This is a technique of expression profiling, which permits simultaneous, comparative and quantitative analysis of gene‐specific, 9‐ to 13‐basepair sequences. These short sequences, called (...) SAGE tags, are linked together for efficient sequencing. The sequencing data are then analyzed to identify each gene expressed in the cell and the levels at which each gene is expressed. The main benefit of SAGE includes the digital output and the identification of novel genes. In this review, we present an outline of the method, various bioinformatics methods for data analysis and general applications of this important technology. Phosphatidylinositol 4,5‐bisphosphate (PI4,5P2) is a key lipid signaling molecule that regulates a vast array of biological activities. PI4,5P2 can act directly as a messenger or can be utilized as a precursor to generate other messengers: inositol trisphosphate, diacylglycerol, or phosphatidylinositol 3,4,5‐trisphosphate. PI4,5P2 interacts with hundreds of different effector proteins. The enormous diversity of PI4,5P2 effector proteins and the spatio‐temporal control of PI4,5P2 generation allow PI4,5P2 signaling to control a broad spectrum of cellular functions. PI4,5P2 is synthesized by phosphatidylinositol phosphate kinases (...) (PIPKs). The array of PIPKs in cells enables their targeting to specific subcellular compartments through interactions with targeting factors that are often PI4,5P2 effectors. These interactions are a mechanism to define spatial and temporal PI4,5P2 synthesis and the specificity of PI4,5P2 signaling. In turn, the regulation of PI4,5P2 effectors at specific cellular compartments has implications for understanding how PI4,5P2 controls cellular processes and its role in diseases. (shrink)

Objective: Deep brain stimulation targeted to the ventral intermediate nucleus of the thalamus is effective for motor symptoms in essential tremor, but there is limited data on cognitive outcomes. We examined cognitive outcomes in a large cohort of ET DBS patients.Methods: In a retrospective analysis, we used repeated-measures ANOVA testing to examine whether the age of tremor onset, age at DBS surgery, hemisphere side implanted with lead, unilateral vs. bilateral implantations, and presence of surgical complications influenced the cognitive outcomes. Neuropsychological (...) outcomes of interest were verbal memory, executive functioning, working memory, language functioning, visuospatial functioning, and general cognitive function.Results: We identified 50 ET DBS patients; 29 males; the mean age of tremor onset was 35.84 years with a median age of 38 years. The mean age at DBS was 68.18 years. There were 37 unilateral 30 left, seven right, and 13 bilateral brain implantations. In the subgroup analysis, there was a significant interaction between assessment and age of tremor onset ; F = 4.47; p = 0.043 for working memory. The post hoc testing found improvements for younger onset ET. Similarly, there was a significant interaction between assessment and complications vs. no complications subgroups; F = 4.34; p = 0.043 for verbal memory with worsening scores seen for ET patients with complications. The remaining tests were not significant.Conclusion: In this large cohort of ET patients with, DBS was not accompanied by a significant decline in many cognitive domains. These outcomes were possibly related to the selection of patients with normal cognitive functioning before surgery, unilateral DBS implantations for the majority, and selection of patients with optimal response to DBS. We estimate that 208,000 deep brain stimulation (DBS) devices have been implanted to address neurological and neuropsychiatric disorders worldwide. DBS Think Tank presenters pooled data and determined that DBS expanded in its scope and has been applied to multiple brain disorders in an effort to modulate neural circuitry. The DBS Think Tank was founded in 2012 providing a space where clinicians, engineers, researchers from industry and academia discuss current and emerging DBS technologies and logistical and ethical issues facing the field. (...) The emphasis is on cutting edge research and collaboration aimed to advance the DBS field. The Eighth Annual DBS Think Tank was held virtually on September 1 and 2, 2020 (Zoom Video Communications) due to restrictions related to the COVID-19 pandemic. The meeting focused on advances in: (1) optogenetics as a tool for comprehending neurobiology of diseases and on optogenetically-inspired DBS, (2) cutting edge of emerging DBS technologies, (3) ethical issues affecting DBS research and access to care, (4) neuromodulatory approaches for depression, (5) advancing novel hardware, software and imaging methodologies, (6) use of neurophysiological signals in adaptive neurostimulation, and (7) use of more advanced technologies to improve DBS clinical outcomes. There were 178 attendees who participated in a DBS Think Tank survey, which revealed the expansion of DBS into several indications such as obesity, post-traumatic stress disorder, addiction and Alzheimer’s disease. The affinities of antibodies (Abs) for their target antigens (Ags) gradually increase in vivo following an infection or vaccination, but reach saturation at values well below those realisable in vitro. This ‘affinity ceiling’ could in many cases restrict our ability to fight infections and compromise vaccines. What determines the affinity ceiling has been an unresolved question for decades. Here, we argue that it arises from the strength of the chain of protein complexes that is pulled by B cells during the (...) process of Ag acquisition. The affinity ceiling is determined by the strength of the weakest link in the chain. We identify the weakest link and show that the resulting affinity ceiling can explain the Ab affinities realized in vivo, providing a conceptual understanding of Ab affinity maturation. We explore plausible evolutionary underpinnings of the affinity ceiling, examine supporting evidence and alternative hypotheses and discuss implications for vaccination strategies. (shrink)

Patients with advanced Alzheimer's disease experience cognitive impairment and physical disabilities in daily life. Currently, there are no treatments available to slow down the course of the disease, and limited treatments exist only to treat symptoms. However, deep brain stimulation of the nucleus basalis of Meynert has been reported to improve cognitive function in individuals with AD. Here, we report the effects of NBM-DBS on cognitive function in a subject with severe AD. An 80-year-old male with severe AD underwent surgery (...) for bilateral NBM-DBS electrode placement. After 10 weeks of stimulation, Mini-Mental State Examination assessment improved from a score of 5 to 9 points, and assessment using the Alzheimer's Disease Assessment Scale–Cognitive Subscale showed a marked reduction in total score from 43 to 33 points, suggesting cognitive benefits from NBM-DBS. The patient's postoperative course was complicated by a subdural effusion that occurred several days after surgery, with complete recovery. Interestingly, the subject also displayed abnormal thermoregulation with stimulation initiation and stimulation parameter modifications. In this article, I will elucidate the Indian government’s two primary discourses concerning yoga since 2014 as right-wing Hindu Nationalist Prime Minister Narendra Modi and his Hindu Nationalist political party, the Bharatiya Janata Party, have interacted with both international and domestic audiences. These discourses can be broadly grouped into two categories, or what I refer to as Modi and the BJP’s “double discourse”: Yoga as a global soft power solution to counter the Global North’s climate change privilege on the (...) international stage and Yoga as biopower for the advancement of India’s depressed economy on the domestic front. Relying on yoga’s polyvalent character, Modi and the BJP are able to frame yoga in these two particular ways—which together signal their commitment to neoliberal economic ideology—by drawing from historical and contemporaneous precedents which I also outline in the article. I conclude with a brief visit to the preparations for the BJP’s 2019 International Day of Yoga, where this double discourse becomes most evident in the two divergent themes announced for the event.
