Pancreatitis is a complex, progressively destructive inflammatory disorder. Alcohol was long thought to be the primary causative agent, but genetic contributions have been of interest since the discovery that rare PRSS1, CFTR and SPINK1 variants were associated with pancreatitis risk. We now report two associations at genome-wide significance identified and replicated at PRSS1-PRSS2 and X-linked CLDN2 through a two-stage genome-wide study. The PRSS1 variant likely affects disease susceptibility by altering expression of the primary trypsinogen gene. The CLDN2 risk allele is (...) associated with atypical localization of claudin-2 in pancreatic acinar cells. The homozygous CLDN2 genotype confers the greatest risk, and its alleles interact with alcohol consumption to amplify risk. Significant associations have been found between specific human leukocyte antigen (HLA) alleles and organ transplant rejection, autoimmune disease development, and the response to infection. Traditional searches for disease associations have conventionally measured risk associated with the presence of individual HLA alleles. However, given the high level of HLA polymorphism, the pattern of amino acid variability, and the fact that most of the HLA variation occurs at functionally important sites, it may be that a combination of variable amino acid sites shared (...) by several alleles (shared epitopes) are better descriptors of the actual causative genetic variants. Here we describe a novel approach to genetic association analysis in which genes/proteins are broken down into smaller sequence features and then variant types defined for each feature, allowing for independent analysis of disease association with each sequence feature variant type. We have used this approach to analyze a cohort of systemic sclerosis patients and show that a sequence feature composed of specific amino acid residues in peptide binding pockets 4 and 7 of HLA-DRB1 explains much of the molecular determinant of risk for systemic sclerosis. This edited volume of original contributions deals with the economic and political thought of Michael Polanyi. Requiring little prior knowledge of Polanyi, this volume further develops a somewhat neglected side of Polanyi's work. In particular it examines the 'tacit integration', of subsidiary details into focal objects or actions as central to all knowing and action. It traces ontological counterparts in the structures of comprehensive entities and complex actions, and a multi-level universe in which lower levels have their boundary conditions, (...) the extents to which they apply, determined by those of the next higher level, whilst each possessing its own laws or operative principles. This schema of 'dual control' preserves the reality and relative autonomy of each level, and its interactions with others, against the various reductions. The essays in this volume also employ and develop important additional concepts and distinctions such as: 'corporate' and 'spontaneous' order; 'public' and 'private' liberties; 'general' and 'specific authority'; and 'moral inversion'; which, as the essays show, are necessary for understanding and maintaining a free society and the freedom of institutions within it. Among the topics treated with them are: more of the prerequisites of freedom in public liberties dedicated to principles and transcendent values; totalitarianism and society as spontaneous order; the balance of general and specific authority in society and particular institutions; reductionism, totalitarianism and consumption in consumer societies, as moral inversions; the mutual interactions of economics and politics as distinct and autonomous but interacting levels; the sociological aspects of economics; and Polanyi's own contributions to sociology. This volume explores the role of some of the most prominent twentieth-century philosophers and political thinkers as teachers. It examines what obstacles they confronted as teachers and how they overcame them in conveying truth to their students in an age dominated by ideological thinking.

Despite a large and multifaceted effort to understand the vast landscape of phenotypic data, their current form inhibits productive data analysis. The lack of a community-wide, consensus-based, human- and machine-interpretable language for describing phenotypes and their genomic and environmental contexts is perhaps the most pressing scientific bottleneck to integration across many key fields in biology, including genomics, systems biology, development, medicine, evolution, ecology, and systematics. Here we survey the current phenomics landscape, including data resources and handling, and the progress that (...) has been made to accurately capture relevant data descriptions for phenotypes. We present an example of the kind of integration across domains that computable phenotypes would enable, and we call upon the broader biology community, publishers, and relevant funding agencies to support efforts to surmount today's data barriers and facilitate analytical reproducibility. Red-2 is a computer program for red-cell antibody identification, a piece of "normal science". Abstracting from Red-2, a general problem solving mechanism is described that is especially suited for performing a form of abductive inference or best explanation finding. A problem solver embodying this mechanism synthesizes composite hypotheses by combining hypothesis parts. This is a common task of intelligence, and a component of scientific reasoning. The work addresses the question, 'How is science possible?' by showing how a simple but powerful (...) form of hypothesis synthesis is computationally feasible. (shrink)

Throughout the biological and biomedical sciences there is a growing need for, prescriptive ‘minimum information’ (MI) checklists specifying the key information to include when reporting experimental results are beginning to find favor with experimentalists, analysts, publishers and funders alike. Such checklists aim to ensure that methods, data, analyses and results are described to a level sufficient to support the unambiguous interpretation, sophisticated search, reanalysis and experimental corroboration and reuse of data sets, facilitating the extraction of maximum value from data sets (...) them. However, such ‘minimum information’ MI checklists are usually developed independently by groups working within representatives of particular biologically- or technologically-delineated domains. Consequently, an overview of the full range of checklists can be difficult to establish without intensive searching, and even tracking thetheir individual evolution of single checklists may be a non-trivial exercise. Checklists are also inevitably partially redundant when measured one against another, and where they overlap is far from straightforward. Furthermore, conflicts in scope and arbitrary decisions on wording and sub-structuring make integration difficult. This presents inhibit their use in combination. Overall, these issues present significant difficulties for the users of checklists, especially those in areas such as systems biology, who routinely combine information from multiple biological domains and technology platforms. To address all of the above, we present MIBBI (Minimum Information for Biological and Biomedical Investigations); a web-based communal resource for such checklists, designed to act as a ‘one-stop shop’ for those exploring the range of extant checklist projects, and to foster collaborative, integrative development and ultimately promote gradual integration of checklists. In this book philosophers, scholars of religion, and activists address the theme of responsibility. Barbara Darling-Smith brings together an enlightening collection of essays that analyze the ethics of responsibility, its relational nature, and its global struggle.

My interest in gathering together a collection of this sort was generated by a fortuitous combination of historical studies under Professor Keith Lehrer and studies in cognitive science under Professor R. Michael Harnish at the University of Arizona. Work on the volume began there while I was an instructor in the Department of Linguistics and was greatly encouraged by participants in the Faculty Seminar on Cognitive Science chaired by Professor Lance J. In the mid-20th century, descriptive meta-ethics addressed a number of central questions, such as whether there is a necessary connection between moral judgment and motivation, whether moral reasons are absolute or relative, and whether moral judgments express attitudes or describe states of affairs. I maintain that much of this work in mid-20th century meta-ethics proceeded on an assumption that there is good reason to question. The assumption was that our ordinary discourse is uniform and determinate enough to vindicate one side (...) or the other of these meta-ethical debates. I suggest that ordinary moral discourse may be much less uniform and determinate than 20th century meta-ethics assumed. (shrink)

Taking my cue from Michael Smith, I try to extract a decent argument for non-cognitivism from the text of the Treatise. I argue that the premises are false and that the whole thing rests on a petitio principi. I then re-jig the argument so as to support that conclusion that Hume actually believed (namely that an action is virtuous if it would excite the approbation of a suitably qualified spectator). This argument too rests on false premises and a (...) begged question. Thus the Motivation Argument fails BOTH as an argument for noncognitivism AND as an argument for what Hume actually believed, that moral distinctions are not derived from reason and that moral properties are akin to secondary qualities. So far as the Motivation Argument is concerned, both cognitivists and rationalists can rest easy. Themes: 1) Hume’s Slavery of Reason thesis is only defensible if passions are not only desires but sometimes dispositions to acquire desires (DTADs). 2) A desire for our good on the whole, which Humeans need to posit to fend off apparent counterexamples to the Slavery of Reason Thesis, does not sit well with the Humean theory of how novel desires arise (an objection due originally to Reid). 3) Hume is wrong to suppose that ‘abstract or demonstrative reasoning never influences any of our actions, but only as it directs our judgment concerning causes and effects’ as the examples of Russell and Hobbes convincingly demonstrate. This ironic as both Russell and Hobbes subscribed to the Slavery of Reason Thesis. 4) I critique Michael Smith’s critique of motivational externalism. (shrink)

In `Underestimating Self-Control' (1997a), I argued that Jeanette Kennett and Michael Smith (1996) underestimate our capacity for synchronic self-control. They argued for a solution to a puzzle about such self-control that features non-actional exercises' of self-control. I argued in response that `a more robust, actional exercise of self-control is open to agents in scenarios of the sort in question' (1997a: 119). They disagree (Kennett and Smith 1997).In Mele 1997a, I resisted the temptation to criticize Kennett and (...) class='Hi'>Smith's attempted resolution, because I thought it better to concentrate on sketching an alternative resolution. Here, I will argue that the resolution they offer is problematic and reply to their criticism (1997) of my suggested resolution. (shrink)

Hume is widely regarded as the grandfather of emotivism and indeed of non-cognitivism in general. For the chief argument for emotivism - the Argument from Motivation - is derived from him. In my opinion Hume was not an emotivist or proto-emotivist but a moral realist in the modern ‘response-dependent’ style. But my interest in this paper is not the historical Hume but the Hume of legend since the legendary Hume is one of the most influential philosophers of the present age. (...) According to Michael Smith ‘the Moral Problem’ – the central issue in meta-ethics - is that the premises of Hume’s argument appear to be true though the non-cognitivist conclusion appears to be false. Since the argument seems to be valid, something has got to give. Smith struggles to solve the problem by holding on to something like the premises of the argument whilst trying to fend off the conclusion. In my view this is a wasted effort. Hume was not arguing for non-cognitivsm in the first place, and the arguments for non-cognitivism that can be extracted from his writings are no good. Either the premises are false or the inferences are invalid. And this is despite the fact that Hume was substantially right about reason and the passions. Thus ‘the Moral Problem’ is not a problem, and the legendary Hume does not deserve his influence. -/- An important theme in this paper is the concept of a DTAD or a dispositions to acquire desires. These play an important role in motivation but unlike desires (with which they are sometimes confused ) they are NOT propositional attitudes. (shrink)

Contemporary ethical thought owes a great deal to David Hume whose work has inspired theories as diverse as non-cognitivism, error theory, quasi-realism, and instrumentalism about practical reason. This timely volume brings together an international range of distinguished scholars to discuss and dispute issues revolving around three closely related Humean themes which have recently come under close scrutiny. First is Hume's infamous claim that 'Reason is, and ought only to be, the slave of the passions'. Moral sentimentalism has seen a tremendous rise in popularity in recent years within contemporary meta-ethical theory, since it promises to delineate the normative domain in a naturalistically unobjectionable manner. After showing that both Michael Slote and Jesse Prinz’s sentimentalist positions fall short of fulfilling this promise, this essay argues that contemporary sentimentalists are advised to take their clues from Adam Smith rather than David Hume. While Hume was absolutely right in emphasizing the importance of empathy in the moral (...) context, his official description of the mechanisms of empathy as articulated in the Treatise falls fundamentally short for this purpose. Adam Smith’s conception of empathy, a conception that in fact is closer to some of Hume’ remarks in the Enquiry rather than the Treatise, as essentially involving perspective taking and his appeal to the impartial spectator perspective proves to be more fertile. Only in this manner do sentimentalists have any hope of accounting for the intersubjective normative and obligatory dimension of moral judgments. (shrink)

This dissertation argues for a metaethical theory I call "Expressive-Assertivism." Expressive-Assertivism is a distinctive, substantial refinement of dual-use metaethical theories traditionally associated with R. M. Hare, C. L. Stevenson, and, more recently, with David Copp. If true, Expressive-Assertivism clarifies, resolves, or dissolves---without, in turn, raising additional difficulties---a number of philosophical problems, including what Michael Smith calls "The Moral Problem," which many consider to be the central organizing problem in contemporary metaethics. The following are the three most important features (...) of Expressive-Assertivism. ;1. The central tenet. The proper, literal use of an ethical sentence, such as 'Donating to charity is right', is the performance of a direct expressive illocutionary act and a direct assertive illocutionary act. Recognizing that ethical sentences can be used to both directly express a speaker's attitude and directly assert something about the world captures what is intuitively compelling about "expressivist" theories---namely, that there is some strong connection between a speaker's sincere moral utterance and his or her motivations to act---and what is intuitively compelling about "descriptivist" theories---namely, that moral utterances appear to have descriptive or cognitive content and, therefore, are truth-evaluable. The Central Tenet is the feature of Expressive-Assertivism that dissolves The Moral Problem. ;2. The generality principle. The proper, literal use of an ethical sentence directly expresses an attitude, not toward the subject of the sentence, but toward things of a more general kind, namely, things that have the property picked out by the ethical predicate . ;3. The extensionality principle. A speaker directly expresses an attitude toward things that have the property picked out by an ethical predicate whenever that predicate appears in any extensional context. Together, the Extensionality and Generality Principles shield Expressive-Assertivism from many objections to traditional expressivist theories, including "The Embedding Objection," which many consider to be the most difficult problem faced by expressivists. History, Philosophy and Science Teaching argues that science teaching and science teacher education can be improved if teachers know something of the history and philosophy of science and if these topics are included in the science curriculum. The history and philosophy of science have important roles in many of the theoretical issues that science educators need to address: the goals of science education; what constitutes an appropriate science curriculum for all students; how science should be taught in traditional cultures; what (...) integrated science is; how scientific literacy can be promoted; and the conflict which can occur between science curriculum and deep-seated religious or cultural values and knowledge. In part, answers to these questions hinge on views about the nature of science, views that are best informed by historical and philosophical study. Outlining the history of liberal, or contextual, approaches to the teaching of science, Michael Matthews elaborates contemporary curriculum developments that explicitly address questions about the nature and the history of science. We all have moral beliefs. But what if one beleif conflicts with another? DePaul argues that we have to make our beliefs cohere, but that the current coherence methods are seriously flawed. It is not just the arguments that need to be considered in moral enquiry. DePaul asserts that the ability to make sensitive moral judgements is vital to any philosophical inquiry into morality. The inquirer must consider how her life experiences and experiences with literature, film and theatre have influenced (...) her capacity for making moral judgments and attempt to ensure that this capacity is neither naive nor corrupted. This article offers three contrasting cases of the use of neutrality and drift in molecular evolution. In the first, neutrality is assumed as a simplest case for modeling. In the second and third, concepts of drift and neutrality are developed within the context of population genetics testing and the development and application of the molecular clock.

The National Institute of Mental Health (Bethesda, MD) reports that approximately 5.2 million Americans experience post-traumatic stress disorder (PTSD) each year. PTSD can be severely debilitating and diminish quality of life for patients and those who care for them. Studies have indicated that propranolol, a beta-blocker, reduces consolidation of emotional memory. When administered immediately after a psychic trauma, it is efficacious as a prophylactic for PTSD. Use of such memory-altering drugs raises important ethical concerns, including some futuristic dystopias put forth (...) by the President's Council on Bioethics. We think that adequate informed consent should facilitate ethical research using propranolol and, if it proves efficacious, routine treatment. Clinical evidence from studies should certainly continue to evaluate realistic concerns about possible ill effects of diminishing memory. If memory-attenuating drugs prove effective, we believe that the most immediate social concern is the over-medicalization of bad memories, and its subsequent exploitation by the pharmaceutical industry.
