To localize wheat ESTs on chromosomes, 882 homoeologous group 6-specific ESTs were identified by physically mapping 7965 singletons from 37 cDNA libraries on 146 chromosome, arm, and sub-arm aneuploid and deletion stocks. The 882 ESTs were physically mapped to 25 regions flanked by 23 deletion breakpoints. Of the 5154 restriction fragments detected by 882 ESTs, 2043 were localized to group 6 chromosomes and 806 were mapped on other chromosome groups. The number of loci mapped was greatest on chromosome 6B and (...) least on 6D. The 264 ESTs that detected orthologous loci on all three homoeologs using one restriction enzyme were used to construct a consensus physical map. The physical distribution of ESTs was uneven on chromosomes with a tendency toward higher densities in the distal halves of chromosome arms. About 43% of the wheat group 6 ESTs identified rice homologs upon comparisons of genome sequences. Fifty-eight percent of these ESTs were present on rice chromosome 2 and the remaining were on other rice chromosomes. Even within the group 6 bins, rice chromosomal blocks identified by 1-6 wheat ESTs were homologous to up to 11 rice chromosomes. This article deals with the research tradition of Radical Constructivism and proposes four central claims for its theoretical, methodological and epistemic orientation and status. First, Radical Constructivism should be viewed as a comprehensive empirical research tradition with an emphasis on cognition, learning, living systems and organization which, in addition, developed a new general methodology for scientific operations. Second, the main opponent of Radical Constructivism, especially in the research program of Heinz von Foerster, does not lie in philosophical or epistemological terrains (...) but in the area of the general scientific methodology and in its conventional mode of exploring the world. Third, Radical Constructivism proposed a new and alternative way for scientific explorations and world-making which produces, additionally, tangible and non-trivial effects with respect to scientific outcomes. Fourth, due to this new way of scientific world-making and due to its novel scientific methodology Radical Constructivism was only marginally interested in epistemological issues. Instead, the empirical research tradition of Radical Constructivism offered intriguing answers why the varieties of scientific realism and its allies like scientific objectivity seem so appealing and almost self-evident. -/- . Depletion of mitochondrial endo/exonuclease G‐like (EXOG) in cultured neonatal cardiomyocytes stimulates mitochondrial oxygen consumption rate (OCR) and induces hypertrophy via reactive oxygen species (ROS). Here, we show that neurohormonal stress triggers cell death in endo/exonuclease G‐like‐depleted cells, and this is marked by a decrease in mitochondrial reserve capacity. Neurohormonal stimulation with phenylephrine (PE) did not have an additive effect on the hypertrophic response induced by endo/exonuclease G‐like depletion. Interestingly, PE‐induced atrial natriuretic peptide (ANP) gene expression was completely (...) abolished in endo/exonuclease G‐like‐depleted cells, suggesting a reverse signaling function of endo/exonuclease G‐like. Endo/exonuclease G‐like depletion initially resulted in increased mitochondrial OCR, but this declined upon PE stimulation. In particular, the reserve capacity of the mitochondrial respiratory chain and maximal respiration were the first indicators of perturbations in mitochondrial respiration, and these marked the subsequent decline in mitochondrial function. Although pathological stimulation accelerated these processes, prolonged EXOG depletion also resulted in a decline in mitochondrial function. At early stages of endo/exonuclease G‐like depletion, mitochondrial ROS production was increased, but this did not affect mitochondrial DNA (mtDNA) integrity. After prolonged depletion, ROS levels returned to control values, despite hyperpolarization of the mitochondrial membrane. The mitochondrial dysfunction finally resulted in cell death, which appears to be mainly a form of necrosis. In conclusion, endo/exonuclease G‐like plays an essential role in cardiomyocyte physiology. Loss of endo/exonuclease G‐like results in diminished adaptation to pathological stress. The decline in maximal respiration and reserve capacity is the first sign of mitochondrial dysfunction that determines subsequent cell death. (shrink)

This article discusses three policy ideas that address the limitations of the traditional welfare state: universal basic income (UBI), universal basic services (UBS), and the social economy. As a lens from which to evaluate these policy alternatives, we develop a concept of active citizenship as an interactive and recursive process between people’s equal political influence and the institutional conditions in which they are placed. While the social policy discourse on active citizenship has centred on the debate between increasing individual responsibilities (...) or enhancing people’s capabilities, our conception recentres the political dimension of active citizenship as people’s capacity, not only to exercise individual self-determination, but also collective self-determination over shared conditions. We conclude that, in addition to the conditions for security and autonomy, opportunities for organised social cooperation are necessary to achieve a virtuous cycle between people’s political influence and the institutions that support it. Reading is a peculiar kind of experience. Although its practice and theory have a very long tradition, the question of aesthetic pleasure is as perplexing as ever. Why do we read? What exactly thrills us in the text? Taking the work of Roland Barthes as a central reference, the aim of this collection of essays is to investigate a variety of themes and issues associated with the question of readerly pleasure. Pleasure 'in' the text is related to the content of (...) the text and associated with various methods of representing the pleasures of 'real life', whereas pleasure 'of' the text is discovered in the literary form. The imperfect, if not erroneous, distinction between form and content, constitutes one of the main methodological techniques for identifying the two major sources of pleasure, and serves as a starting point for the inquiry. This bookdoes not merely offer a personal view of the problem in question, nor an exposition of this problem locked within the limits of a given theory, but a broader perspective consisting of the reflections of academics who critically evaluate both its theoretical and practical aspects, across disciplines such as literary theory and criticism, semiology, philosophy and psychoanalysis. In recent years, the impact of party leaders on voting behavior has attracted increasing attention, leading some scholars to identify a phenomenon of ‘presidentialization’. Many extant studies of this topic in Japan are limited to one or two electoral cycles. In order to trace long-term trends, this paper analyses longitudinal survey data to investigate the existence and magnitude of the effect party leader evaluations exert on vote choice in Japan. Empirical results show that while only dominant and forceful personalities substantially (...) influenced voters' likelihood of supporting their parties in the 1980s and 1990s, by the 2000s assessments of most major party leaders had a significant impact on their parties' electoral performance. In short, party leaders affect vote choice due not to their personalities, but instead to the position they hold. We also test the hypotheses that the association between leader appraisal and voting behavior would be particularly conspicuous among voters who lack party identification and those who are most heavily exposed to media reportage. Analysis reveals that independent voters are not more likely to vote on the basis of leader evaluations than partisans; a leader effect is found more frequently among voters with greater exposure to election coverage on television.
