Collagen is the most abundant vertebrate protein and forms a stable fibrous architecture in connective tissues, such as bone, cartilage, skin and tendon. Much recent research has been directed towards an understanding of the molecular and cellular mechanisms involved in the synthesis and degradation of collagen, because a change in the normal balance, or an increased destruction of collagen, can cause loss of function of specialized tissues. This short review attempts to summarize present knowledge about the proteolytic destruction of collagen (...) and the exciting new work on the factors that are emerging as important controlling agents. An understanding of the regulatory processes is yielding some important clues regarding the levels at which they can fail in pathological conditions and we speculate about evolving strategies for preventing uncontrolled resorption. In the legal judgement reason demands that it extend itself beyond the mere subjective limits of the self in order that it might fashion a judgement that speaks for the other. This is the universal necessity of the judgement. No claim of truth or the moral law can guarantee that others will agree with this judgement: thus disputation is the risk which reason takes in order to judge at all. When we are fatigued, we feel that our performance is worse than when we are fresh. Yet, for over 100 years, researchers have been unable to identify an objective, behavioral measure that covaries with the subjective experience of fatigue. Previous work suggests that the metrics of signal detection theory —response bias and perceptual certainty —may change as a function of fatigue, but no work has yet been done to examine whether these metrics covary with fatigue. Here, we investigated cognitive fatigue (...) using SDT. We induced fatigue through repetitive performance of the n-back working memory task, while functional magnetic resonance imaging data was acquired. We also assessed cognitive fatigue at intervals throughout. This enabled us to assess not only whether criterion and d’ covary with cognitive fatigue but also whether similar patterns of brain activation underlie cognitive fatigue and SDT measures. Our results show that both criterion and d’ were correlated with changes in cognitive fatigue: as fatigue increased, subjects became more conservative in their response bias and their perceptual certainty declined. Furthermore, activation in the striatum of the basal ganglia was also related to cognitive fatigue, criterion, and d’. These results suggest that SDT measures represent an objective measure of cognitive fatigue. Additionally, the overlap and difference in the fMRI results between cognitive fatigue and SDT measures indicate that these measures are related while also separate. In sum, we show the relevance of SDT measures in the understanding of fatigue, thus providing researchers with a new set of tools with which to better understand the nature and consequences of cognitive fatigue. The dominant culture in India in the Buddha’s day, Brahmanical culture, took as axiomatic the existence of a supernatural creator deity. This deity, termed ‘Brahma’, was conceived as being ‘the all-seeing, the all-powerful, the Lord, the maker and creator, ruler, appointer and orderer, father of all that have been and will be’. Although the Buddha completely rejected such apparent metaphysical speculation as a ‘thicket of views’, he nowhere formulated a systematic repudiation of theism. In one canonical text, however, the Buddha, (...) encountering a young Brahmin espousing theistic beliefs, gives a series of analogies and similes that help to illuminate his views on the matter. In short, the Buddha saw such a belief as being dangerously re- flexive, and hence as a symptom of a debilitating conceptual and affective disorder. Thus, in the dialogue, the Buddha aims to ease this ailment of his interlocutor through a threefold strategy: (1) displaying the language usage that under girds the problem; (2) reorienting the interlocutor towards the primacy of his conceptual apparatus as the proper locus of concern; and (3) providing a practice through which the interlocutor may develop the skills necessary for conceptual and affective health. The parameters of the discussion in this sutta are wide enough to render it of relevance to contemporary debates on theism. That is, the issue at stake in the sutta may be read as being not only about a restricted local notion of deity, but about God, broadly conceived. An abiding concern of Mahāyāna Buddhists has been the accessibility of a buddha’s power in the world.1 Some Buddhists, notably philosophers and their commentators, have grappled with the very coherence of such a possibility. Viewing the question from a logical perspective, it has been necessary for such systematic thinkers to reconcile the apparent inconsistency ensuing from the two essential qualities deemed definitive of a buddha. A buddha is one who, by virtue of his awareness of the nature of reality, is (...) completely liberated from the life-impelling force of mental defilements, and is thus beyond the scope of our world; and he is one who, by virtue of his profound compassion, is naturally compelled to continue engagement with beings still delusively ensnared in the world. Logically, these two qualities are at odds. Not all Buddhists, however, have sought a solution to the dilemma of accessing a buddha’s power on the basis of logical or epistemological theory. Some Buddhists have, rather, sought an imaginative-cultic solution. Relics, statues, paintings, architectural monuments, books, remembrance, meditation, guru veneration, and visualization have, at various times and in various places, been held to be the most effective means of rendering present the otherwise inaccessible or obscured power that accompanies a buddha. In this article, I trace the idea of the mantra as a vehicle of enlightened presence as it was presented to Indian Mahāyāna Buddhists in the medieval period. (shrink)

Buddhist practitioner contained in the collection of works by Advayavajra (ca. 1007–10851) known as the Advayavajrasaçgraha. The prescriptions for this training, called the ådikarma (literally: preliminary practice), is contained in the first text of the collection, the Kud®≈† inirghåtana2 (The Refutation of False Views). The article is divided into two parts. The first provides some context for the ritual prescriptions, the translation of which constitutes the second part. My contention is that Advayavajra, in prescribing the ådikarma in the rhetorical manner (...) that he does, is aiming to accomplish several aims. These can be grouped under two broad concerns. The first is institutional in nature, the second, ritual. First, Advayavajra seems to be creating a bridge between the antinomian, extra-monastic forms of Buddhist practice that were gaining influence in his day, and the established institutional structures that had, for centuries already, represented the norm for Buddhist learning and practice. This reconciliatory project required him to argue for the legitimization of certain ritual and doctrinal innovations that were, in fact, divergent from established practices and views. Second, in his prescriptions for the ådikarma itself, Advayavajra aims to establish a clear relationship between preliminary training and expert accomplishment. He connects this concern with the first by founding unconventional expertise on conventional training. The strategy employed by Advayavajra in this regard is to prescribe the ådikarma not merely as “preliminary,” as is generally the case, but as “primary,” in the sense of a continuously constituted foundation. SCL (TAL1/TCL5) is a member of the helix‐loop‐helix family of transcription factors. Originally identified because of its involvement in a tumour‐specific chromosomal translocation, overexpression of the SCL gene is the most common molecular abnormality found in human T cell leukaemia. Transgenic models have now formally demonstrated that overexpression of SCL within the T cell lineage is capable of causing malignant transformation. Gene targeting experiments have revealed that the SCL gene is crucial for the development of primitive haematopoiesis in the mouse (...) and is also required for the generation of all adult haematopoietic lineages. Biochemical studies have indicated some of the proteins which interact with SCL and this has refined the hypotheses concerning the mechanisms by which SCL plays a role in leukaemogenesis and haematopoietic development. Aims: The aims of the study were to explore expert opinion on the distinction between “research” and “audit”, and to determine the need for review by a National Health Service (NHS) Research Ethics Committee (REC). Background: Under current guidelines only “research” projects within the NHS require REC approval. Concerns have been expressed over difficulties in distinguishing between research and other types of project, and no existing guidelines appear to have been validated. The implications of this confusion include unnecessary REC applications, (...) and crucially, the potential for ethically unsound projects to escape review. Methods: A three-stage Delphi method was chosen to explore expert opinion and develop consensus. Stage 1 comprised ten semi-structured interviews gathering opinion on distinguishing between types of project and how to determine need for ethical review. Stages 2 and 3 were questionnaires, asking 24 “experts” to rate levels of ethical concern and types of project for a series of questions. Anonymised responses from stage 2 were fed back in stage 3. The final responses were analysed for consensus. Results: Of 46 questions, consensus was achieved for 14 (30.4%) for level of ethical concern and for 15 (32.6%) for type of project. Conclusions: Several ideas proved discriminatory for classifying the type of project and assessing level of ethical concern, and they can be used to develop an algorithm to determine need for ethical review. There was little relationship between assessment of the level of ethical concern and classification of the project. There was inconsistency in defining and classifying studies as something other than “research” or “audit”. (shrink)

The experience of embodiment may be studied using the rubber hand illusion. Little is known about the cognitive mechanism that elicits the feeling of embodiment. In previous models of the rubber hand illusion, bodily signals are processed sequentially. Such models cannot explain some more recent findings. Carruthers (2013) proposed a multidimensional model of embodiment, in which the processing of embodiment is understood in terms of conceptual hand space. Visual features of hands are represented along several dimensions. The rubber hand illusion (...) is then explained as the erroneous matching of the online representation of the artificial hand to the stored prototype in a space defined by those dimensions. We conducted the first experimental tests to investigate the multidimensional conceptual space account. First, participants performed a series of odd-one-out judgments for triads of hand images (including their own hand) and we then employed multidimensional scaling analyses. We found that a multidimensional model of perceived hand similarity could be fitted to our data. Second, we tested if a multisensory bodily signal manipulation (the rubber hand illusion) influences the position of the viewed artificial hand in hand space. We employed synchronous and asynchronous stroking and found that in the synchronous condition, which elicits the rubber hand illusion, compared to the asynchronous condition, which does not, the artificial hand was closer to the center of hand space; that is, to a prototype hand. We discuss these findings in the context of the Carruthers (2013) conceptual space model as well as other rubber hand illusion models. (shrink)
