IntroductionCognitive impairment is a highly prevalent non-motor feature of Parkinson’s disease. A better understanding of the underlying pathophysiology may help in identifying therapeutic targets to prevent or treat dementia. This study sought to identify metabolic alterations in the prefrontal cortex, a key region for cognitive functioning that has been implicated in cognitive dysfunction in PD.MethodsProton Magnetic Resonance Spectroscopy was used to investigate metabolic changes in the PFC of a cohort of cognitively normal individuals without PD, as well as PD participants (...) with either normal cognition, mild cognitive impairment, or dementia. Ratios to Creatine resonance were obtained for glutamate, glutamine and glutamate combined, N-acetylaspartate, myoinositol, and total choline, and correlated with cognitive scores across multiple domains administered to the PD participants only.ResultsWhen individuals retain cognitive capabilities, the presence of Parkinson’s disease does not create metabolic disturbances in the PFC. However, when cognitive symptoms are present, PFC Glu/Cre ratios decrease with significant differences between the PD-NC and PPD groups. In addition, Glu/Cre ratios and memory scores were marginally associated, but not after Bonferroni correction.ConclusionThese preliminary findings indicate that fluctuations in prefrontal glutamate may constitute a biomarker for the progression of cognitive impairments in PD. We caution for larger MRS investigations of carefully defined PD groups. (shrink)

Planctomycetes, Verrucomicrobia and Chlamydia are prokaryotic phyla, sometimes grouped together as the PVC superphylum of eubacteria. Some PVC species possess interesting attributes, in particular, internal membranes that superficially resemble eukaryotic endomembranes. Some biologists now claim that PVC bacteria are nucleus‐bearing prokaryotes and are considered evolutionary intermediates in the transition from prokaryote to eukaryote. PVC prokaryotes do not possess a nucleus and are not intermediates in the prokaryote‐to‐eukaryote transition. Here we summarise the evidence that shows why all of the PVC traits (...) that are currently cited as evidence for aspiring eukaryoticity are either analogous (the result of convergent evolution), not homologous, to eukaryotic traits; or else they are the result of horizontal gene transfers. Throughout the biological and biomedical sciences there is a growing need for, prescriptive ‘minimum information’ (MI) checklists specifying the key information to include when reporting experimental results are beginning to find favor with experimentalists, analysts, publishers and funders alike. Such checklists aim to ensure that methods, data, analyses and results are described to a level sufficient to support the unambiguous interpretation, sophisticated search, reanalysis and experimental corroboration and reuse of data sets, facilitating the extraction of maximum value from data sets (...) them. However, such ‘minimum information’ MI checklists are usually developed independently by groups working within representatives of particular biologically- or technologically-delineated domains. Consequently, an overview of the full range of checklists can be difficult to establish without intensive searching, and even tracking thetheir individual evolution of single checklists may be a non-trivial exercise. Checklists are also inevitably partially redundant when measured one against another, and where they overlap is far from straightforward. Furthermore, conflicts in scope and arbitrary decisions on wording and sub-structuring make integration difficult. This presents inhibit their use in combination. Overall, these issues present significant difficulties for the users of checklists, especially those in areas such as systems biology, who routinely combine information from multiple biological domains and technology platforms. To address all of the above, we present MIBBI (Minimum Information for Biological and Biomedical Investigations); a web-based communal resource for such checklists, designed to act as a ‘one-stop shop’ for those exploring the range of extant checklist projects, and to foster collaborative, integrative development and ultimately promote gradual integration of checklists. Can we take seriously the rhetoric of the sanctity of human life from the mouths of exponents of the death penalty? And yet there are those who consistently and (...) in good faith hold euthanasia in whatever form morally anathema. Many of those who share this view have expressed disapproval of government intervention in the Shiavo case. They cannot be accused of having a political agenda. Over the years, philosopher Peter Singer has made the case for euthanasia most forcefully. This may be a good time to consider critically his arguments, free of the immediate tumult surrounding the Shiavo case. Such consideration may even help us in our understanding of the issues involved in that case.Singer has been variously characterized as "the most controversial," "the most influential" and "the most notorious" philosopher in the world. His books cover a wide range of ethical issues, from animal liberation to euthanasia (including infanticide), and from globalization to the workings of George W. Bush's mind. The controversy, influence and notoriety come from positions he has taken, with a candor that one rarely finds in the work of a respectable philosopher, on the most sensitive issues of life and death. Whether or not one is in agreement with his views (I am not, as will soon become evident), they are argued with a provocative clarity and consistency. A radical advocate of animal [End Page 238] rights, he wants to dissolve the species distinction between human beings and non-human animals. An advocate of euthanasia and infanticide, he substitutes the commandment that honors the sanctity of human life for one that sanctifies the quality of life. In both instances, human beings are being denied a privileged place in the world simply by virtue of being human.If, as Copernicus demonstrated, the universe is not earth-centered, and if, as Darwin has shown, human beings are not the separate creation of God, there can be no justification for providing human beings with a separate ethical system, often at the expense of non-human animals. Singer calls this speciesism and analogizes it to racism. This review article engages with a rich field of scholarship on logistics that has gathered momentum over the past decade, focusing on two new publications by Laleh Khalili and Martín Arboleda. It contextualizes how and why logistics is bound up with the militarization of contemporary political and social life. I argue that the later 20th century rise of logistics can be better understood as both a response to and symptom of capitalist crisis and I situate this scholarship on war and (...) logistics in relationship to Giovanni Arrighi’s account of crisis and ‘unravelling hegemony’. I also show how logistics provides essential critical and visual resources that contribute to efforts to map global capitalism and to debates on totality and class composition in contemporary critical theory. Finally, contemporary events such as the ongoing Coronavirus crisis and the reemergence of Black Lives Matter are considered in light of this analysis with reference to the centrality of logistics to racial capitalism. This paper traces the history of artificial insemination by selected donors as a strategy for positive eugenic improvement. While medical artificial insemination has a longer history, its use as a eugenic strategy was first mooted in late nineteenth-century France. It was then developed as ‘scientific motherhood’ for war widows and those without partners by Marion Louisa Piddington in Australia following the Great War. By the 1930s AID was being more widely used clinically in Britain as a medical solution to male (...) infertility for married couples. In 1935 English postal clerk, Herbert Brewer, promoted AID as the socialization of the germ plasm in a eugenic scheme. The next year Hermann Muller, American Drosophila geneticist and eugenicist, presented his plan for human improvement by AID to Stalin. Some twenty years later, Muller, together with Robert Klark Graham, began planning a Foundation for Germinal Choice in California. This was finally opened in 1980 as the first practical experiment in eugenic AID, producing some 215 babies over the twenty years it functioned. While AID appeared to be a means of squaring a eugenic circle by separating paternity from love relationships, and so allowing eugenic improvement without inhibiting individual choice in marriage, it found very little favour with those who might use it, not least because of a couple’s desire to have their ‘own’ children has always seemed stronger than any eugenic aspirations. No state has ever contemplated using AID as a social policy. (shrink)

In contrast, Mandeville was not so neglected in his own age, a point well-chronicled in F. B. Kaye's introduction to The Fable of the Bees, and substantiated, tangibly, by this collection of writings excellently assembled and edited by J. Martin Stafford. In the eighteenth century and, more particularly, (...) in the decade between the publication of the 1723 edition of the Fable and Mandeville's death, numerous sermons, essays, letters, and books were published with the single intent of refuting what one eighteenth-century critic considered "so monstrous an Opinion", namely, that private vice might render a benefit to the public. What one discovers on reading these early critics is that it is not so much Mandeville's opinion that is monstrous as it is the incessant misinterpretations that are so often used against him. Otto Poggeler’s Der Denkweg Martin Heidegger was first published in 1963, one year after the English translation of Heidegger’s Sein und Zeit. Widely acclaimed as the most comprehensive and accurate study of the development of Heidegger’s thought, a second unrevised edition was make available in Germany in 1983. Now thanks to the translators, Daniel Magurshak and Sigmund Barber, this important work is available in English for the frist time. Included is a Preface to what Poggeler calls the “American Edition” (...) and an “Afterword to the Second Edition” in which Poggeler discusses recent developments in Heidegger studies including Heidegger’s political entanglements. For more than thirty years, the question of how sentences about God manage to refer has been in the background and often in the foreground of discussions of religious language and metaphysics. In some cases philosophers of religion and theologians have spoken vaguely about or given up all together claims to depict reality in religious discourse. Janet Martin Soskice challenges these views on the grounds that they are rooted in a bankrupt form of empiricism and that they fail to (...) be consistent with the aim of traditional Christian faith to refer to God as a reality. In 1856, the State of Ohio began an enumeration of its population to count and identify people with disabilities. This paper examines the ethical role of the accounting profession in this project, which supported the transatlantic eugenics movement and its genocidal attempts to eliminate disabled persons from the population. We use a theoretical approach based on Levinas who argued that the self is generated through engagement with the Other, and that this engagement presupposes a responsibility to and for the Other. We show that successive waves of legislation relied on State and County auditors along with Township clerks and assessors to conduct the mechanics of the enumeration of the population, which focused on the identification, categorization, and counting of the disabled people of the State. We argue that the accounting-based technologies of enumeration and reporting objectify the enumerated persons and deny the auditor’s pre-existing ethical obligation to this new Other. We show how the financial expertise and structures of the State were engaged in the execution of this mandate, which remained in place for over a century and supported a program of institutionalization. We consider the ramifications of this for our understanding of the ethical role of public sector accounting in the United States over this period, which has been under-explored. (shrink)

The hypothesis that anatomically modern homo sapiens could have undergone changes akin to those observed in domesticated animals has been contemplated in the biological sciences for at least 150 years. The idea had already plagued philosophers such as Rousseau, who considered the civilisation of man as going against human nature, and eventually.

For many decades, race and racicsm have been common areas of study in departments of sociology, history, politcal science, English, and athropology. Much more recently, as the historical concept of race and racial categories have faced signifcant scientific and politcal challenges, philosophers have become more interested in these areas. This changing understanding of the ontology of race has invited inquiry from researchers in moral philosophy, metaphysics, epistemology, philosophy of science, philosophy of language, and aesthetics. The Routlege Companion to Philosophy of (...) Race offers in one comprehensive volume newly written articles on race from the world’s leading Analytic and Continental philosophers. Two major reports in the UK and USA have recently sanctioned as ethically acceptable genome editing of future generations for the treatment of serious rare inherited conditions. This marks an important turning point in the application of recombinant DNA techniques to humans. The central question this paper addresses is how did it became possible for human genetic engineering (HGE) of future generations to move from an illegitimate idea associated with eugenics in the 1980s to a concrete proposal sanctioned by scientists (...) and bioethicists in 2020? The paper uses the concept of a regime of normativity to understand the co-evolution and mutual shaping of technology, imaginaries, norms and governance processes in debates about HGE in the USA and UK. It will be argued that interlinked discursive, institutional, political and technological changes have made proposals for the use of genome editing in the genetic engineering of future generations both “thinkable” and legitimate. This paper approaches the question of awareness outside of attention through a broader psychological examination of human consciousness. Questions regarding the boundaries of conscious awareness, as well as the possibility of 'subconscious' or 'unconscious' mental processes, were widely discussed 100 years and more ago when they played a central role in the thinking of turn-of-thecentury theorists such as William James, F.W.H. Myers, Jean-Martin Charcot, and Pierre Janet, all of whom were interested in dissociative phenomena suggestive of consciousness, or awareness, (...) beyond the margins of attention. Such phenomena included hypnosis, hysteria, trance states, and motor automatisms, and for many scholars also sleep related conditions such as dreaming and hypnogogic states. (shrink)

ABSTRACTTime-consciousness has long been a focus of research in phenomenology and phenomenological psychology. We advance and extend this tradition of research by focusing on the character of temporal experience under conditions of mania. Symptom scales and diagnostic criteria for mania are peppered with temporally inflected language: increased rate of speech, racing thoughts, flight-of-ideas, hyperactivity. But what is the underlying structure of temporal experience in manic episodes? We tackle this question using a strategically hybrid approach. We recover and reconstruct three hypotheses (...) regarding manic temporality that were advanced and modeled by two pioneers of clinical phenomenology: Eugène Minkowski and Ludwig Binswanger. We then test, critique, and refine these hypotheses using heterophenomenological methods in an interview-based study of persons with a history of bipolar and a current diagnosis of acute mania. Psychiatric disorders present distinct clinical challenges which are partly attributable to their multifactorial aetiology and the absence of laboratory tests that can be used to confirm diagnosis or predict risk. Psychiatric disorders are highly heritable, but also polygenic, with genetic risk conferred by interactions between thousands of variants of small effect that can be summarized in a polygenic risk score. We discuss four areas in which the use of polygenic risk scores in psychiatric research and clinical contexts could have ethical (...) implications. First, there is concern that clinical use of polygenic risk scores may exacerbate existing health inequities. Second, research findings regarding polygenic risk could be misinterpreted in stigmatising or discriminatory ways. Third, there are concerns associated with testing minors as well as eugenics concerns elicited by prenatal polygenic risk testing. Fourth, potential challenges that could arise with the feedback and interpretation of high polygenic risk for a psychiatric disorder would require consideration. While there would be extensive overlap with the challenges of feeding back genetic findings in general, the potential clinical use of polygenic risk scoring warrants discussion in its own right, given the recency of this possibility. To this end, we discuss how lay interpretations of risk and genetic information could intersect. Consideration of these factors would be necessary for ensuring effective and constructive communication and interpretation of polygenic risk information which, in turn, could have implications for the uptake of any therapeutic recommendations. Recent advances in polygenic risk scoring have major implications for its clinical potential, however, care should be taken to ensure that communication of polygenic risk does not feed into problematic assumptions regarding mental disorders or support reductive interpretations.
