This book is a guide to working with social science concepts. Concepts are the prisms through which we see the social world. They are foundational to the social science enterprise, and the quality of investigations hinges in part on how well researchers make use of them. Most social science concepts are drawn from ordinary language used in everyday ways; however, many social scientists "reconfigure" ordinary words to meet their research needs. They tinker with the meanings of words to fit their (...) theoretical aims and make them precise, useful tools of measurement and comparison. This book examines social science concepts through an interpretivist lens with the aim of providing concrete conceptual guidance for future research. Specifically, this book seeks to: 1) identify characteristic dangers that attend the making and use of reconfigured concepts; 2) lay out ways in which a select number of interpretivist approaches have been used to mitigate these dangers; 3) introduce practical tools that rework these interpretivist approaches into explicit and useable research methods; 4) show concretely how these elucidation tools can be gainfully used by social scientists working both within and outside of the interpretivist tradition. Analysis of sites of newly integrated DNA in cellular genomes is important to several fields, but methods for analyzing and visualizing these datasets are still under development. Here, we describe tools for data analysis and visualization that take as input integration site data from our INSPIIRED pipeline. Paired-end sequencing allows inference of the numbers of transduced cells as well as the distributions of integration sites in target genomes. We present interactive heatmaps that allow comparison of distributions of integration sites to (...) genomic features and that support numerous user-defined statistical tests. To summarize integration site data from human gene therapy samples, we developed a reproducible report format that catalogs sample population structure, longitudinal dynamics, and integration frequency near cancer-associated genes. We also introduce a novel summary statistic, the UC50, which provides a single number summarizing possible clonal expansion. Using these tools, we characterize ongoing longitudinal characterization of a patient from the first trial to treat severe combined immunodeficiency-X1, showing successful reconstitution for 15 years accompanied by persistence of a cell clone with an integration site near the cancer-associated gene CCND2. The new National Institutes of Health (NIH) Policy on the Use of a Single Institutional Review Board (sIRB) for Multi-Site Research was adopted primarily to simplify and speed the review of complex multisite clinical trials. However, speeding review requires overcoming a number of obstacles. Perhaps the most substantial obstacle is the time and effort needed to develop reliance agreements among the participating sites. We conducted 102 semistructured interviews with sIRB personnel, including directors, chairs, reviewers, and staff, from 20 IRBs that (...) acted as sIRBs for multisite research, including 6 commercial/independent sIRBs, and 10 university-based academic and 4 federal sIRBs. Almost without exception, the interviewees agreed that reliance agreements were complex, difficult to develop, and time-consuming. A major problem for relying sites was that different agreements specified different responsibilities for the relying sites. Attitudes differed about whether these problems will be resolved as IRB staff and managers become more experienced with sIRBs. However it is clear that the process of developing reliance agreements must be simplified. Federal assistance in standardizing at least some sections of reliance agreements might reduce the difficulties involved. (shrink)

Electronic institutional review board systems (eIRBs) have become an integral component in ensuring compliance with Human Research Protection Program (HRPP) and IRB requirements. Despite this, few of these systems are configured to administer the single IRB (sIRB) process mandated by the National Institutes of Health (NIH) for multisite research. We interviewed 103 sIRB administrators, chairs, members, and staff members about their experiences with sIRB multisite research review. We observed three main obstacles to adapting existing eIRB systems to accommodate the sIRB (...) process: (1) Existing systems are not designed for sIRBs and are not configured to administer sIRB responsibilities, (2) they are not interoperable, and (3) resources to improve existing systems are lacking. Our findings suggest that IRBs that act as an sIRB will need major changes to their electronic systems in order to accommodate sIRB processes. These difficulties threaten both the ability of IRBs to focus on ethical rather than bureaucratic problems and the efficiency of multisite trials. (shrink)

Nathan Glazer's The Limits of Social Policy presents an analysis of recent American social policy that Edmund Burke could applaud. A main cause of the failure of American social programs, Glazer argues, is that government ineluctably supplants and weakens the ?fine structure?; of society?the traditional institutions of family, religion, and community. Sometimes the government is merely clumsy and ignorant, but more often the ill effects are the result of ?the ruling doctrines of our age,?; equality and human rights, which cannot (...) bear to leave the convoluted and sometimes unlovely ways of custom and local mores to play themselves out unimpeded. But the very persuasiveness of Glazer's analysis of the past casts some doubt on his measured optimism about the future. Are moderate reforms going to be good enough? Perhaps things will work out for the middle class and affluent, the reviewer argues, but the underclass increasingly appears to be a casualty of moderate reforms. The reviewer offers some reasons for thinking about a return to Jeffersonian self?government, unpragmatic as that may seem in 1991. (shrink)

Samuel Butler, a contemporary critic of Charles Darwin, proffered an alternative, vitalistic account of evolution. At the same time, he put into question all modern naturalistic treatments of this fundamental idea which presuppose that evolution is mainly a scientific problem. On the contrary, Butler in effect insists, this extremely vague idea calls for not an `explanation' but rather a fairly comprehensive, plausible story that helps elucidate an inherently complex idea. Butler can thus be read as outlining an anthropomorphic metaphorics (...) that evokes a living Cosmos wherein it might be possible to do justice to the problem which Darwin left unresolved---the problem of heredity. In this picture of the Cosmos Butler links the fundamental notion of organization not to the allegedly universal and immutable `laws of nature,' as the moderns would have it, but rather to dynamically evolving relationships between only more or less stable habits. The variations in extant habits that emergence elicits are moreover the products of quasi-intelligent responses to new challenges from the environment. For Butler follows Lamarck in holding that all organisms possess powers capable of responding to felt needs and/or desires to make alterations in the habits (or instincts) that characterize their modes of existence. He thus in the end effectively bequeaths to his readers a challenge to extend and amplify, if possible, his outline of a promising metaphysical imaginary that can take into account some highly unorthodox conjectures.
