Replication forks inevitably stall at damaged DNA in every cell cycle. The ability to overcome DNA lesions is an essential feature of the replication machinery. A variety of specialized polymerases have recently been discovered, which enable cells to replicate past various forms of damage by a process termed translesion synthesis. Alternatively, homologous recombination can be used to restart DNA replication across the lesion. Genetic and biochemical studies have shed light on the impact of these two post‐replication repair pathways in bacteria (...) and yeast. In vertebrates, however, a genetic approach to study post‐replication repair has been compromised because many of the genes involved appear to be essential for embryonic development. We have taken advantage of the chicken cell line DT40 to perform a genetic analysis of translesion synthesis and homologous recombination and to characterize genetic interactions between these two pathways in vertebrates. In this article, we aim to summarize our current understanding of post‐replication repair in DT40 in the perspective of bacterial, yeast and mammalian genetics. This article reports an interesting link between the psychophysical property of intentional control of perceptual switching and the underlying neural activities. First, we revealed that the timing of perceptual switching for a dynamical dot quartet can be controlled by the observers' intention, without eye movement. However, there is a clear limitation to this control, such that each animation frame of the stimulus must be presented for a sufficiently long time length; in other words, the frequency of the stimulus alternation must (...) be sufficiently slow for the control. The typical stimulus onset asynchrony for a 50% level of success was about 275 ms for an average of 10 observers. On the basis of psychophysical property, we designed three experiments for investigating the neural process with a magnetoencephalography. They revealed that: (1) a peak component occurring about 300 ms after a reversal was stronger when the direction of perceived motion was switched intentionally than when it was not switched, and (2) neural components about 30–40 ms and 240–250 ms after the reversal of the stimulus animation were stronger when perception was altered intentionally than when it was switched unintentionally. The 300 ms component is consistent with a previous study about passive perceptual switching (Struber and Herrmann [2002]: Cogn Brain Res 14:370–382), but the intentional effect was seemed to be a different component from the well-known P300 component. In lieu of an abstract, here is a brief excerpt of the content:Religion and Science: Nishitani’s View of Nihility and Emptiness–A Pure Land Buddhist CritiqueRyusei TakedaIn general, philosophical critique of Nishida, Tanabe, and Nishitani, the so-called Kyoto school, has been mainly conducted from a Zen Buddhist perspective. One should not, however, overlook the fact that a profound regard for the philosophical aspects of Pure Land Buddhist thought, another major stream of Mahayana Buddhism, is deeply intertwined in the foundation of their (...) philosophies. But the ways in which they interpret it and the position they give it within their philosophical systems are not the same: they have taken different forms, often giving rise to original terminology, in accord with each thinker’s metaphysical speculation.As Alfred N. Whitehead critically argues in his Religion in the Making, the fundamental reason for the decay of both Buddhism and Christianity is that “each religion has unduly sheltered itself from the other. The self-sufficient pedantry of learning and the confidence of ignorant zealots have combined to shut up each religion in its own forms of thought. Instead of looking to each other for deeper meanings, they have remained self-satisfied and unfertilized.” Further, “both have suffered from the rise of the third tradition, which is science, because neither of them had retained the requisite flexibility of adaptation.” 1In this contemporary situation, the philosophies of Nishida, Tanabe, and Nishitani, in which the issue of religion and science has been brought into fundamental question and examined on the dimension of origin of their development, have extremely important implications for the critical interpretation of Pure Land Buddhist doctrines, particularly in the postmodern world. 2There are two points I want to make in this paper. First, the fundamental structure of Pure Land Buddhism, in a way an antipode to Zen, will be examined in light of Nishitani’s metaphysical analysis of the relationship between human experience and the laws of nature, particularly bringing into focus his existential realization of ‘nihility.’And second, I want to challenge Nishitani’s understanding of ‘emptiness’ from my perspective as a Pure Land Buddhist, and also to uncover some fundamental [End Page 155] problems underlying his view of ‘emptiness’ by attempting to relate it to Nagarjuna’s negation of ‘substance’ in his concept of ‘non-self being,’ or ‘nih-svabhava.’ 3What is Religion for Nishida, Tanabe, and Nishitani?Before getting into those two topics, I want to make a rough sketch indicating about where Nishida, Tanabe, and Nishitani see that religious experience is to be truly realized.The starting point of Nishida’s understanding of religion is found in his argument that “it is on the ground of fact that religion ought to be considered.” The fact of religious experience is in his view none other than self-awareness of ‘absolute nothingness,’ which is also the foundation of what we consider as objective knowledge. The self-awareness manifests itself in the midst of such fact and serves to reveal that fact. He argues that the ultimate goal of Mahayana Buddhism is to penetrate into the depth of this fact.In Tanabe’s philosophy, one’s resurrection and return to the world (genso) and self-renunciation through ‘metanoetics’ (zangedo), a philosophy of absolute Other-power, are required and witnessed to in order for one to establish a genuine religious standpoint. Neither/nor, instead of both/and, is the only relationship in which the self negates itself by means of contradiction, so that at the same time as the self is extinguished in the negation, the contradiction of that which opposes the self is also extinguished. Converted into the neither/nor relationship, both self and contradiction cease to be contradictory opposites. 4 This is Tanabe’s criticism directed at Nishida’s notion of the self-identity of absolute contradiction. Nothingness can never be intuited as such self-identity. The self-consciousness of nothingness is only in resurrection and return to the world, which is witnessed to through practice based upon faith and performed in obedience to Other-power. 5For Nishitani the question of the essence of religion must be answered through tracing the process of the genuine pursuit of true reality. The aim of this paper is to propose an antithesis to the overreliance on scientific facts and objectivity to counter mis- and disinformation in media literacy education. Through conceptual examination of meaning, care, and facts, I demonstrate the ontological priority of meaning and values in the life-world. I then discuss conspiracy theories as a textual genre in which the crisis of meaning manifests as a prominent factor. Given the centrality of meaning, I claim that literacy education needs to go beyond (...) the matters of fact and address matters of concern in this supposedly post-truth world. (shrink)

Mental imagery of movement is a potentially valuable rehabilitation task, but its therapeutic efficacy may depend on the specific cognitive strategy employed. Individuals use two main strategies to perform the hand mental rotation task, which involves determining whether a visual image depicts a left or right hand. One is the motor imagery strategy, which involves mentally simulating one’s own hand movements. In this case, task performance as measured by response time is subject to a medial–lateral effect wherein the RT is (...) reduced when the fingertips are directed medially, presumably as the actual motion would be easier. The other strategy is to employ visual imagery, which involves mentally rotating the picture and is not subject to this medial–lateral effect. The rehabilitative benefits of the HMRT are thought to depend on the MI strategy, so it is essential to examine the effects of individual factors such as age, image perspective, and innate ability on the strategy adopted. When presented with pictures of the palm, all subjects in the current study used the MI strategy, regardless of age and ability. In contrast, when subjects were presented with pictures of the back of the hand, the VI strategy predominated among the young age group regardless of performance, while the strategy used by middle-age and elderly groups depended on performance ability. In the middle-age and elderly groups, the VI approach predominated in those with high performance skill, whereas the MI strategy predominated among those with low performance skill. Thus, higher-skill middle-aged and elderly individuals may not necessarily form a motion image during the HMRT, potentially limiting rehabilitation efficacy. (shrink)

The sample size was 297, 370, and 74 respectively. Whilethere is better awareness of GMOs in 2000 compared to 1991; thetrend shows an increase in the perceived risks of GMOs followedby growing resistance in Japan. While a majority of personsbelieved genetic engineering would make life better (...) over the nexttwenty years , the proportion of respondents who thoughtgenetic engineering would make life worse over the next twentyyears doubled from 1997 to 2000 .Respondents were asked whether they had heard aboutapplications in several areas and the order of familiarity was: pest-resistant crops, human genes in bacteria, mouseto develop cancer, food and drinks, pigs with human hearts andpre-implantation diagnosis. A divide of opinion can be seen whenthe results on benefit, risk and moral acceptability of applicationsof biotechnology by the public are compared to the forum andscientist samples.A significant change in the acceptance of the public occurred in2000 where only 22% agreed on the moral acceptability of GMfood compared to 41% in 1997. In 2000 fewer people said they arewilling to buy genetically modified fruits that taste bettercompared to 1997 . The results show less public support foruse of gene therapy than 1993 and twice as many scientistsrejected gene therapy than they did in 1991.When asked whom is best placed to regulate modernbiotechnology, the respondents were overwhelmingly in favor ofinternational regulatory bodies, such as the United Nations andthe World Health Organization , rather than national bodies.The comparison between scientists and public is interesting,however the more enthusastic sample were participants from theNovartis Life Science Forum with its mixed occupations. Neuronal activities have recently been reported to exhibit power-law scaling behavior. However, it has not been demonstrated that the power-law component can play an important role in human perceptual functions. Here, we demonstrate that the power spectrum of magnetoencephalograph recordings of brain activity varies in coordination with perception of subthreshold visual stimuli. We observed that perceptual performance could be better explained by modulation of the power-law component than by modulation of the peak power in particular narrow frequency ranges. The results (...) suggest that the brain operates in a state of self-organized criticality, modulating the power spectral exponent of its activity to optimize its internal state for response to external stimuli.
