This short commentary offers a perspective on (...) this debate that is drawn from a consideration of underlying biological mechanisms.Because age is unquestionably the greatest single risk factor associated with AD and MCI, I begin by asking what we know about the intrinsic processes of aging. The central feature of current understanding of intrinsic aging is that it consists of the gradual, lifelong accumulation of a wide variety of molecular and cellular faults that begins very early in life and eventually leads to overt functional impairments resulting in age-related frailty, disability, and disease (Kirkwood 2005). This is an essentially bottom-up process. A strong consensus has emerged in recent years that there is no active program for aging and that there are no genetic factors that have evolved specifically to cause aging or age-related diseases. Unquestionably, there are genes that influence susceptibility to disease and even length of life, but these genes are genes for maintenance and repair functions, not for aging. It is the limitation in the past evolutionary pressure to invest in greater longevity than was required, within an environment where life was usually truncated by hazards such as starvation, injury, or infection, that means that our maintenance systems—good as they are—are insufficient to keep us going indefinitely. Added to this is the idea that there are probably also genes that program processes that are good for the young organism, but which may in later life have harmful effects. Processes like inflammation and apoptosis, which are seen to occur extensively in aged tissues, including brain, surely evolved to help cope with infection and cellular damage arising earlier in life. That they are called into play in aged tissues is a direct, but secondary, consequence of accumulated damage.With this general framework to understand the intrinsic biology of aging, let us now ask what we might expect a priori for the aging of an organ such [End Page 79] as the brain. Although we cannot pretend complete ignorance of how the brain actually does age, our enquiry can be based entirely on data from other organ systems, so we can develop our predictions about brain aging without prejudice.Aging begins very early during human development, faults accumulating from the first stages of embryonic development. Each time a cell divides, each of the daughter cells is likely to acquire a few new mutations. We know this from the estimates for the error rate in DNA replication and the size of the human genome. It is confirmed by direct evidence that at individual gene loci where mutations have been measured, mutation frequency increases with age (Morley 1998; Vijg 2000). In addition to replication errors, DNA is damaged on a daily basis to a very considerable degree by agents such as reactive oxygen species (free radicals), which are formed as byproducts of normal cellular metabolism. Although most of this damage is corrected by repair, the repair mechanisms themselves are not error free and additional mutations accumulate through this route. If a mutation is lethal to the cell, the cell dies and thus the mutation is eliminated. However, the majority of mutations are nonlethal, either because the mutation is recessive and the cell carries an intact gene copy on the other chromosome, because the gene is not currently required, or because the mutation produces only a delayed deleterious effect. Examples of genes that produce delayed deleterious effects include the genes responsible for familial AD. It is interesting to reflect that on statistical grounds alone, it is almost certain that each of us carries a randomly determined fraction of cells in our brains that bear amyloid precursor protein or presenilin mutations associated with AD. For example, research methods are increasingly incorporating citizen participation in the research process, and there has been an increase in collaborative research that brings academic and industry partners together. There have been numerous positive outcomes associated with both of these growing methodological and collaborative processes; nonetheless, both bring with them ethical considerations that require careful thought and attention. Using a case of intended but failed doping, the author seeks to answer the question of if an agent cheated when they intended to but failed in the case of doping due to inert, counterfeit drugs. The examination looks at the case using the concept of cheating and concludes by dividing the results of cheating into primary and secondary effects.

The price of Bitcoin is once more soaring. From early October 2020 to early January 2021, the price of a single Bitcoin token went from roughly $10,000 to nearly $65,000, reinspiring the hopes of the crypto-faithful in the inevitability of a future beyond centralized banking and leaving the rest to dread the jargon of computational libertarianism. The speculative betting driving this recent price action, however, belies a more rudimentary and overlooked shift in the digital economy signaled by cryptocurrencies and Bitcoin (...) in particular. Unlike an earlier industrial logic that sought to reduce heat loss and improve efficiency to maximize surplus value, Bitcoin’s proof-of-work system shifts the basis of value production from efficiency to inefficiency. Moreover, it does so by using a cryptographic algorithm whose purpose is to destroy the meaning of its inputs. Through an exploration of Bitcoin’s proof-of-work technics and its inversion of traditional models of value extraction, the text argues that Bitcoin reveals a profound transformation in the nature of surplus represented by computational capitalism. The continuing viability of the free‐radical theory of ageing has been questioned following apparently incompatible recent results. We show by modelling positional effects of the generation and primary targets of reactive oxygen species that many of the apparently negative results are likely to be misleading. We conclude that there is instead a need to look more closely at the mechanisms by which free radicals contribute to age‐related dysfunction in living systems. There also needs to be deeper understanding of the dynamics (...) of accumulation and removal of the various kinds of molecular damage, in particular mtDNA mutations. Finally, the expectation that free‐radical damage on its own might cause ageing needs to be relinquished in favour of the recognition that the free‐radical theory is just one of the multiple mechanisms driving the ageing process. This narrative symposium examines the relationship of bioethics practice to personal experiences of illness. A call for stories was developed by Tod Chambers, the symposium editor, and editorial staff and was sent to several commonly used bioethics listservs and posted on the Narrative Inquiry in Bioethics website. The call asked authors to relate a personal story of being ill or caring for a person who is ill, and to describe how this affected how they think about bioethical questions and the (...) practice of medicine. Eighteen individuals were invited to submit full stories based on review of their proposals. Twelve stories are published in this symposium, and six supplemental stories are published online only through Project MUSE. Authors explore themes of vulnerability, suffering, communication, voluntariness, cultural barriers, and flaws in local healthcare systems through stories about their own illnesses or about caring for children, partners, parents and grandparents. Commentary articles by Arthur Frank, Bradley Lewis, and Carol Taylor follow the collection of personal narratives. (shrink)

Human life expectancy has increased dramatically through improvements in public health, housing, nutrition and general living standards. Lifespan is now limited chiefly by intrinsic senescence and its associated frailty and diseases. Understanding the biological basis of the ageing process is a major scientific challenge that will require integration of molecular, cellular, genetic and physiological approaches. A recent report by Tyner et al.1 suggests that p53 is bad for longevity. Heterozygotic mice carrying a p53 mutation that apparently enhances the stability of the wild‐type protein showed shorter lifespans and faster ageing while also developing fewer tumours. This fits with the idea that cellular ageing is the price paid for better protection against unlimited proliferation of cancer cells. But other work shows that there is a strong positive association between DNA repair‐mediated protection against cancer and ageing. Objectives: To explore how subjects in a placebo-controlled vitamin A supplementation trial among Ghanaian women aged 15–45 years perceive the trial and whether they know that not all trial capsules are the same, and to identify factors associated with this knowledge.Methods: 60 semistructured interviews and 12 focus groups were conducted to explore subjects’ perceptions of the trial. Steps were taken to address areas of low comprehension, including retraining fieldworkers. 1971 trial subjects were randomly selected for a survey measuring their knowledge (...) that not all trial capsules are the same. The subjects’ fieldworkers were also interviewed about their characteristics and trial knowledge. Factors associated with knowledge were explored using multi-level modeling.Results: Although subjects knew they were taking part in research, most thought they were receiving an active and beneficial medication. Variables associated with knowledge were education and district of residence. Radio broadcasts benefited those with some schooling. Fieldworkers’ characteristics were not associated with subjects’ knowledge.Conclusions: Research and debate on new or improved consent procedures are urgently required, particularly for subjects with little education. A problem in the text of Pindar, the interpretation of λαρετον, O. 2. 87, seems to be vanishing, swept away by a remarkable consensus of recent criticism, a consensus the more remarkable in that it accepts a false solution to a genuine difficulty. This article has two purposes, the first and more important of which is to argue that the currently prevailing answer is manifestly wrong, the second to offer evidence in support of a different approach. Simply read γαρυτων, recent (...) critics maintain, and all problems disappear. Since -ο- and -ω- were not yet distinguished in the orthography of Pindar's day, γαρυτων is as correct as the unanimous γαρύτον of the MSS, testimonia, and scholia. By this simple change, the argument proceeds, the troublesome dual of the MSS is purged and with it the ‘historicist hare’, as one critic has recently called it, which less enlightened Pindarists chased for so long. If there is no dual, there is no need to speculate as to the identity of the ‘pair’ likened to κρακς and contrasted with the ‘divine bird of Zeus’, the man who is wise φυ. We need no longer suppose that the μαθντς are Simonides and Bacchylides – the traditional answer – or any other specific rivals. Unfortunately for this view, there is no evidence to justify taking γαρυτων as a plural, which is of course precisely what critics have been doing. It is – if anything – a third dual imperative , and every bit as much a dual as the γαρύτον of the MSS. Mr Stoneman is not alone in his ready dismissal of the ‘historicist hare’. Here is the view of Professor Lloyd-Jones: ‘… the lightest possible alteration converts the dual to a plural imperative, so that the number two vanishes’. A year before, Professor C. A. P. Ruck had chided the scholiasts for ‘reading out of Pindar's ΓΑΡΥΕΤΟΝ the dual…rather than the plural’. Bowra had declared that γαρυτων ‘would be the plural of the imperative’. The belief is widespread and persistent; those who wish a full conspectus of earlier views on the matter may consult the massive compilation made by Dr J. van Leeuwen in 1964. Not all critics and editors have endorsed the change from the traditional reading. While it has been in the successive Teubner editions since Schröder adopted it in 1900, neither Turyn nor Bowra accepted it. But among those who have argued for γαρυτων, only one has expressed any doubt that it is a plural, and that one is Theodor Bergk, who first proposed it. Ethics researchers focus on moral awareness as a precursor to ethical decision making, but they pay little attention to framing processes that precede moral awareness. This study addresses this gap in the literature to examine how a student entrepreneur starting a venture while completing an assignment frames issues and how these frames affect moral awareness (i.e., whether or not the entrepreneur considers ethical dimensions). Framing does not occur in isolation but is part of a sensemaking process involving others. Using a (...) single case study method, we capture an entrepreneur’s framing process over time as the new venture emerges and our data reveals frames that may preclude consideration of ethical dimensions, including some frames developed and reinforced through entrepreneurship education. We make a contribution to the entrepreneurship education literature by suggesting ways to incorporate ethics into entrepreneurship courses. (shrink)

Wearable robots are an emerging form of technology that allow organizations to combine the strength, precision, and performance of machines with the flexibility, intelligence, and problem-solving abilities of human wearers. Active exoskeletons are a type of wearable robot that gives wearers the ability to effortlessly lift up to 200 lbs., as well as perform other types of physically demanding tasks that would be too strenuous for most humans. Synchronization between exoskeleton suits and wearers is one of the most challenging requirements (...) to operate these technologies effectively. In this conceptual paper, we extend interpersonal adaption theory (IAT) to the exoskeleton context and explicate (a) the antecedents that are most likely to shape synchrony in human-exoskeleton interactions, (b) automatic and strategic synchrony as adaptive behaviors in human-exoskeleton interactions, and (c) outcome variables that are especially important in these processes. Lastly, we offer a discussion of key methodological challenges for measuring synchrony in human-exoskeleton interactions and offer a future research agenda for this important area. (shrink)

Mitochondrial dysfunction has long been considered a key mechanism in the ageing process but surprisingly little attention has been paid to the impact of mitochondrial number or density within cells. Recent reports suggest a positive association between mitochondrial density, energy homeostasis and longevity. However, mitochondrial number also determines the number of sites generating reactive oxygen species (ROS) and we suggest that the links between mitochondrial density and ageing are more complex, potentially acting in both directions. The idea that increased density, (...) especially when combined with mitochondrial dysfunction, might accelerate ageing is supported by a negative correlation between mitochondrial density and maximum longevity in an interspecies comparison in mammals, and by evidence for an intimate interconnection between cellular ROS levels, mitochondrial density and cellular ageing. Recent data suggest that retrograde response, which activates mitochondrial biogenesis, accompanies cellular ageing processes. We hypothesise that increased mitochondrial biogenesis, and possibly also impaired degradation and segregation of mitochondria, if occurring as adaptation to pre‐existing mitochondrial dysfunction, might aggravate ROS production and thus actively contribute to ageing. As moral injury is a still-emerging concept within the area of military mental health, prevalence estimates for moral injury and its precursor, potentially morally injurious events (PMIEs), remain unknown for many of the world’s militaries. The present study sought to estimate the prevalence of PMIEs in the Canadian Armed Forces (CAF), using data collected from CAF personnel deployed to Afghanistan, via logistic regressions controlling for relevant sociodemographic, military, and deployment characteristics. Analyses revealed that over 65% of CAF members reported exposure (...) to at least one event that would be considered a PMIE. The most commonly PMIEs individuals reported included seeing ill or injured women and children they were unable to help (48.4%), being unable to distinguish between combatants and non-combatants (43.6%), and finding themselves in a threatening situation where they were unable to respond due to the rules of engagement under which they were required to operate (35.4%). These findings provide support for both the presence of exposure to PMIEs in CAF members and the need for formal longitudinal data collection regarding PMIE exposure and moral injury development. The context derives from the initiative of the Scottish Executive to enhance the status and working conditions of teachers. We consider the extent to which continuing professional development activities arising out of the Chartered Teacher Programme encourage teachers to value research, equip them to become research-minded and support them to engage in research and enquiry in their own professional contexts.
