Louis Pasteur plays a role in the creation myth of biotechnology which resembles the heroic position of his great antagonist Liebig in the story of agricultural chemistry. His intellectual development, expressed in a great book, supposedly underlay a revolution in practice. Similarly, biotechnology is conventionally traced back to Pasteur, through whose influence, it has been assumed, ancient crafts were transformed into an applicable science of microbiology. The emphasis on Pasteur's work in the history of biotechnology has served to bolster the (...) image of progress in the technology following from periodic scientific breakthroughs. Elsewhere I have argued that biotechnology can be better seen as a boundary object, to use Star and Griesemer's terminology, between biology and engineering. As such it has been significant throughout this century, and the word has been used since 1917. (shrink)

The Dutch microbiologist/biochemist Albert Jan Kluyver was an early proponent of the idea of biochemical unity, and how that concept might be demonstrated through the careful study of microbial life. The fundamental relatedness of living systems is an obvious correlate of the theory of evolution, and modern attempts to construct phylogenetic schemes support this relatedness through comparison of genomes. The approach of Kluyver and his scientific descendants predated the tools of modern molecular biology by decades. Kluyver himself is poorly recognized (...) today, yet his influence at the time was profound. Through lens of today however, it has been argued that the focus by Kluyver and others to create taxonomic and phylogenetic schemes using morphology and biochemistry distorted and hindered progress of the discipline of microbiology, because of a perception that the older approaches focused too much on a reductionist worldview. This essay argues that in contrast the careful characterization of fundamental microbial metabolism and physiology by Kluyver made many of the advances of the latter part of the twentieth century possible, by offering a framework which in many respects anticipated our current view of phylogeny, and by directly and indirectly training a generation of scientists who became leaders in the explosive growth of biotechnology. The origins of oxidative phosphorylation, initially known as aerobic phosphorylation, grew out of three research areas of muscle metabolism, creatine phosphorylation, aerobic metabolism of lactic acid in muscle, and studies on the nature and role of adenosine triphosphate . Much of this work centred round the laboratory of Otto Meyerhof, and most of those contributing to the study of aerobic phosphorylation were influenced by that laboratory: particularly Lipmann and also Ochoa. The work of Engelhardt on ATP levels in blood also (...) appears to have been influenced by the studies of Meyerhof’s laboratory. However, with the work of Kalckar, influenced by Lipmann, biochemists began to realise the potential importance of the process. This was confirmed and extended by Belitzer and Ochoa and the theoretical contribution of Lipmann. Thus it is not easy to identify a single point that marked the initiation of studies in this field.The early work was based on the use of tissue homogenates and cells but ultimately this approach limited research possibilities. The development of techniques based on mitochondria opened up new possibilities and brought this first phase of the study of oxidative phorphorylation to a close. (shrink)

In his mature writings, Kuhn describes the process of specialisation as driven by a form of incommensurability, defined as a conceptual/linguistic barrier which promotes and guarantees the insularity of specialties. In this paper, we reject the idea that the incommensurability among scientific specialties is a linguistic barrier. We argue that the problem with Kuhn’s characterisation of the incommensurability among specialties is that he presupposes a rather abstract theory of semantic incommensurability, which he then tries to apply to his description of (...) the process of specialisation. By contrast, this paper follows a different strategy: after criticising Kuhn’s view, it takes a further look at how new scientific specialties emerge. As a result, a different way of understanding incommensurability among specialties will be proposed. (shrink)

In this article, I examine various aspects of the application of Heidegger's motif of interpretative articulation (the core phenomenological motif of existential analytic) to the constitutional analysis of meaningful objects in scientific research that are contextually ready-to-hand. It is my contention that not only the concepts of the ‘fore-structure of understanding’ and the ‘as-structure of interpretation’, but also the extended concepts of the ‘hermeneutic fore-structure of meaning constitution’ and ‘characteristic hermeneutic situation’ are the keys to understanding the interpretative nature of (...) scientific research. The paper applies the constitutional analysis of hermeneutic phenomenology to several phenomena of scientific research—constitution of meaningful objects, situational fulfilment of a domain's general project, production of a domain's thematically given objects, implementation of readable technologies to what is contextually ready-to-hand, reading hypothetical theoretical objects, and exegetical textualizing within interrelated practices. The study is supplied with exemplary illustrations from enzymology and biochemistry. The correlation between a domain's interpretative articulation and the appropriation of research possibilities within the research everydayness is addressed. Special attention is paid to the belief in and reading of intracellular enzymes as hypothetical theoretical objects. (shrink)
