Much of the current research in regenerative medicine concentrates on stem-cell therapy that exploits the regenerative capacities of stem cells when injected into different types of human tissues. Although new therapeutic paths have been opened up by induced pluripotent cells and human mesenchymal cells, the rate of success is still low and mainly due to the difficulties of managing cell proliferation and differentiation, giving rise to non-controlled stem cell differentiation that ultimately leads to cancer. Despite being still far from becoming (...) a reality, these studies highlight the role of physical and biological constraints (e.g., cues and morphogenetic fields) placed by tissue microenvironment on stem cell fate. This asks for a clarification of the coupling of stem cells and microenvironmental factors in regenerative medicine. We argue that extracellular matrix and stem cells have a causal reciprocal and asymmetric relationship in that the 3D organization and composition of the extracellular matrix establish a spatial, temporal, and mechanical control over the fate of stem cells, which enable them to interact and control (as well as be controlled by) the cellular components and soluble factors of microenvironment. Such an account clarifies the notions of stemness and stem cell regeneration consistently with that of microenvironment. (shrink)

In the current social and technological scenario, the term digital is abundantly used with an apparently transparent and unambiguous meaning. This article aims to unveil the complexity of this concept, retracing its historical and cultural origin. This genealogical overview allows to understand the reason why an instrumental conception of digital media has prevailed, considering the digital as a mere tool to convey a message, as opposed to a constitutive conception. The constitutive conception places the digital phenomenon in the broader ground (...) of media studies, and it considers digital technologies as an interface between the subject and the world. In this perspective, the media is not added to the experience of the person, but it shapes it from within on a cognitive, expressive and communicative level. The article makes use of two powerful examples to show the shortcomings of an instrumental conception of the digital, and to affirm the value of a constitutive conception for current media studies regarding digital interfaces. Since the 1970s, the origin of cancer is being explored from the point of view of the Somatic Mutation Theory (SMT), focusing on genetic mutations and clonal expansion of somatic cells. As cancer research expanded in several directions, the dominant focus on cells remained steady, but the classes of genes and the kinds of extra-genetic factors that were shown to have causal relevance in the onset of cancer multiplied. The wild heterogeneity of cancer-related mutations and phenotypes, along with the increasing (...) complication of models, led to an oscillation between the hectic search of 'the' few key factors that cause cancer and the discouragement in face of a seeming 'endless complexity'. To tame this complexity, cancer research started to avail itself of the tools that were being developed by Systems Biology. At the same time, anti-reductionist voices began claiming that cancer research was stuck in a sterile research paradigm. This alternative discourse even gave birth to an alternative theory: the Tissue Organization Field Theory (TOFT). A deeper philosophical analysis shows the serious limits of both reductionist and anti-reductionist positions and of their polarization. This book demonstrates that a radical philosophical reflection is necessary to drive cancer research out of its impasses. At the very least, this will be a reflection on the assumptions of different kinds of cancer research, on the implications of what cancer research has been discovering over 40 years and more, on a view of scientific practice that is most able to make sense of the cognitive and social conflicts that are seen in the scientific community (and in its results), and, finally, on the nature of living entities with which we entertain this fascinating epistemological dance that we call scientific research. The proposed Dynamic and Relational View of carcinogenesis is a starting point in all these directions. (shrink)

Cancer research is experiencing ‘paradigm instability’, since there are two rival theories of carcinogenesis which confront themselves, namely the somatic mutation theory and the tissue organization field theory. Despite this theoretical uncertainty, a huge quantity of data is available thanks to the improvement of genome sequencing techniques. Some authors think that the development of new statistical tools will be able to overcome the lack of a shared theoretical perspective on cancer by amalgamating as many data as possible. We think instead (...) that a deeper understanding of cancer can be achieved by means of more theoretical work, rather than by merely accumulating more data. To support our thesis, we introduce the analytic view of theory development, which rests on the concept of plausibility, and make clear in what sense plausibility and probability are distinct concepts. Then, the concept of plausibility is used to point out the ineliminable role played by the epistemic subject in the development of statistical tools and in the process of theory assessment. We then move to address a central issue in cancer research, namely the relevance of computational tools developed by bioinformaticists to detect driver mutations in the debate between the two main rival theories of carcinogenesis. Finally, we briefly extend our considerations on the role that plausibility plays in evidence amalgamation from cancer research to the more general issue of the divergences between frequentists and Bayesians in the philosophy of medicine and statistics. We argue that taking into account plausibility-based considerations can lead to clarify some epistemological shortcomings that afflict both these perspectives. (shrink)

However, the foundations of this new discipline need to be fleshed out more carefully. In this paper, we claim that a relational ontology is a necessary tool to ground both the conceptual and explanatory aspects of Systems Biology. A relational ontology holds that relations are prior—both conceptually and explanatory—to entities, and that in the biological realm entities are defined primarily by (...) the context they are embedded within—and hence by the web of relations they are part of. In the last few decades, philosophy of science has increasingly focused on multilevel models and causal mechanistic explanations to account for complex biological phenomena. On the one hand, biological and biomedical works make extensive use of mechanistic concepts; on the other hand, philosophers have analyzed an increasing range of examples taken from different domains in the life sciences to test—support or criticize—the adequacy of mechanistic accounts. The article highlights some challenges in the elaboration of mechanistic explanations with a focus on (...) cancer research and neuropsychiatry. It jointly considers fields, which are usually dealt with separately, and keeps a close eye on scientific practice. The article has a twofold aim. First, it shows that identification of the explananda is a key issue when looking at dynamic processes and their implications in medical research and clinical practice. Second, it discusses the relevance of organizational accounts of mechanisms, and questions whether thorough self-sustaining mechanistic explanations can actually be provided when addressing cancer and psychiatric diseases. While acknowledging the merits of the wide ongoing debate on mechanistic models, the article challenges the mechanistic approach to explanation by discussing, in particular, explanatory and conceptual terms in the light of stances from medical cases. Cancer research has been at the forefront of biomedical activity in recent decades, and advances in molecular biology have provided a growing amount of information on the mechanisms involved in the etiopathogenesis of tumors. Nevertheless, despite these advances, the complexity of cancer is more evident, especially as different levels of phenomena are considered to explain the heterogeneity of the neoplastic process. A synthetic analysis of advances in cancer research illustrates these changes. In attempting to overcome the limits of epistemological reductionism, (...) there is a move from a view of a genetic basis for cancer to a more comprehensive perspective aiming to integrate a large amount of information and to understand the neoplastic phenomenon at higher levels of biological complexity through a new interpretative framework. This article aims at discussing an interesting variant of scientific realism recently proposed and defended by Sandra Mitchell (forthcoming), namely an affordances-based and pragmatist variant of scientific realism. We firstly place Mitchell’s proposal in the context of the current state of the debate over scientific realism. Secondly, we summarize the salient features of Mitchell’s proposal. Thirdly, we point out some aspects of that proposal that might require some further refinement and clarification in order to make it less prone to criticisms (...) by both realists and antirealists. More precisely, in this paper we address the following issues: 1) whether Mitchell’s proposal can be classified as a genuine form of scientific realism; 2) whether the fact that in Mitchell’s proposal figure some variants of the no miracle argument is in tension with some other of its features. This volume reviews examples and notions of robustness at several levels of biological organization. It tackles many philosophical and conceptual issues and casts an outlook on the future challenges of robustness studies in the context of a practice-oriented philosophy of science. The focus of discussion is on concrete case studies. These highlight the necessity of a level-dependent description of robust biological behaviors.Experts from the neurosciences, biochemistry, ecology, biology, and the history and the philosophy of life sciences provide a multiplex perspective (...) on the topic. Contributions span from protein folding, to cell-level robustness, to organismal and developmental robustness, to sensorimotor systems, up to the robustness of ecological systems.Several chapters detail neurobiological case-studies. The brain, the poster child of plasticity in biology, offers multiple examples of robustness. Neurobiology explores the importance of temporal organization and multiscalarity in making this robustness-with-plasticity possible. The discussion also includes structures well beyond the brain, such as muscles and the complex feedback loops involved in the peculiar robustness of music perception. From biological, neurological and societal systems, arising by the interplay of self-organizing dynamics and environmental pressures, to the current sophisticated engineering that aims at artificially reproducing the adaptability and (...) resilience of living systems in front of perturbations in man-made devices, robustness seems to hold the key for orchestrating stability and change. This introduction offers a general survey of the contribution that the notion of robustness is providing to reframing major concepts within the life sciences, such as development, evolution, time and environment, and to reframing the relationship between biology and engineering, as well as between biology and physics. Se muestra cómo la indeterminación característica de la complejidad biológica contribuyó al surgimiento de las perspectivas sistémicas en la biología contemporánea y el modo en el que estas pueden admitir un determinismo biológico. Se mencionan algunas características que adquieren en este contexto las nociones de causalidad y de información biológica, para indicar por qué el estudio del indeterminismo y del determinismo biológico abre el camino hacia una comprensión más amplia de la causalidad.

The philosophical discussion of emergence is often focused on properties of ‘wholes’ that are evaluated as emergent with respect to the properties of ‘parts’. Downward causation is, consequently, evaluated as some kind of causal influence of whole properties over parts properties. Yet, several important cases in scientific practice seem to be pursuing hypotheses of parts properties emerging from wholes properties, inverting the instinctive association of emergence with wholes. Furthermore, some areas of reflection which are very important for emergence, e.g., the (...) philosophy of consciousness, do not allow mapping properties onto part-whole organizations. The conceptual puzzle is solved by constructing a framework that disentangles the mereological dimension from the superventional dimension. By liberalizing the spatio-temporal allocation of emergent properties, the proposed dual framework could better capture the way in which emergence and downward causation are addressed in scientific practice. In the process of scientific discovery, knowledge ampliation is pursued by means of non-deductive inferences. When ampliative reasoning is performed, probabilities cannot be assigned objectively. One of the reasons is that we face the problem of the unconceived alternatives: we are unable to explore the space of all the possible alternatives to a given hypothesis, because we do not know how this space is shaped. So, if we want to adequately account for the process of knowledge ampliation, we need to (...) develop an account of the process of scientific discovery which is not exclusively based on probability calculus. We argue that the analytic view of the method of science advocated by Cellucci is interestingly suited to this goal, since it rests on the concept of plausibility. In this perspective, in order to account for how probabilities are in fact assigned in uncertain contexts and knowledge ampliation is really pursued, we have to take into account plausibility-based considerations. La biodiversidad suele reconocerse en diferentes disciplinas como un valor universal. Ésta apunta a la heterogeneidad de las propiedades que caracterizan al mundo biológico. Sin embargo, a pesar de su uso común, el análisis crítico de la literatura filosófica pone en evidencia cierta dificultad a conceptualizar la biodiversidad, dada una aparente dicotomía entre los elementos normativos y descriptivos del término mismo. En este artículo se sostiene que es necesario considerar el aspecto relacional de la biodiversidad con el n de resolver (...) esta dicotomía. Esto significa que para ser un valor, cualquier diferencia en el mundo natural que sea definida en términos de biodiversidad implicará, a nivel conceptual y explicativo, la relación intrínseca entre lo que tenga en común con las entidades y lo que sea específico de éstas. De esta manera la biodiversidad será un concepto explicativo por sí mismo. Una visión relacional de la biodiversidad también hace reconocer el carácter multidimensional de dicha noción, lo que ha probado ser realmente útil en diferentes contextos, pudiéndose caracterizar propiamente en términos de “riqueza” implicada por el concepto de la biodiversidad. In this article, we argue, first, that there are very different research projects that fall under the heading of “systems biology of cancer.” While they share some general features, they differ in their aims and theoretical commitments. Second, we argue that some explanations in systems biology of cancer are concerned with properties of signaling networks and how they may play an important causal role in patterns of vulnerability to cancer. If well-designed, the results of a Randomised Clinical Trial can justify a causal claim between treatment and effect in the study population; however, additional information might be needed to carry over this result to another population. RCTs have been criticized exactly on grounds of failing to provide this sort of information Evidence, inference and enquiry. On (...) the other side, so-called In Silico Clinical Trials face the same criticisms addressed against standard modelling and simulation techniques, and cannot be equated to experiments Philosophy of molecular medicine: foundational issues in research and practice, Routledge, New York, 2017; Parker in Synthese 169:483–496, 2009; Parke in Philos Sci 81:516–536, 2014; Diez Roux in Am J Epidemiol 181:100–102, 2015 and related discussions in Frigg and Reiss in Synthese 169:593–613, 2009; Winsberg in Synthese 169:575–592, 2009; Beisbart and Norton in Int Stud Philos Sci 26:403–422, 2012). We undertake a formal analysis of both methods in order to identify their distinct contribution to causal inference in the clinical setting. Britton et al.’s study :E2098–E2105, 2013) on the impact of ion current variability on cardiac electrophysiology is used for illustrative purposes. We deduce that, by predicting variability through interpolation, ISCTs aid with problems regarding extrapolation of RCTs results, and therefore in assessing their external validity. Furthermore, ISCTs can be said to encode “thick” causal knowledge —as opposed to “thin” difference-making information inferred from RCTs. Hence, ISCTs and RCTs cannot replace one another but rather, they are complementary in that the former provide information about the determinants of variability of causal effects, while the latter can, under certain conditions, establish causality in the first place. In this paper we have two main goals. The first is to challenge two key elements of reductionist thinking concerning human work inherited from the Information and Communication Technology revolution that have significantly shaped current concepts of work at both the individual and institutional level: the ‘flexible man’ model and the obsession with the objective function of economic productivity. We show how, combined with the logic of productivity as a means for continuous economic growth, these elements justify the overlapping of (...) value and price. This overlap characterizes current meritocratic paradigms. Our second goal is to show how and why an emerging integrated paradigm is a more suitable model for taking into consideration the specifically human dimensions of work. These dimensions encompass the sphere of caring and are mostly buried in the functionalist-mechanistic system. In this context, we argue that the COVID-19 pandemic can work as an exogenous shock useful to boost the rise of this new paradigm of human work. (shrink)

The possibility of curing aging is currently generating hopes and concerns among entrepreneurs, experts, and the general public. This article aims to clarify some of the key assumptions of the Strategies for Engineered Negligible Senescence agenda, one of the most prominent paradigms for rejuvenation. To do this, we present the three fundamental claims of this research program: (1) aging can be repaired; (2) rejuvenation is possible through the reversal of all molecular damage; (3) and the human organism is a sophisticated (...) machine. Secondly, we argue that this agenda fits with a machine conception of the organism (described by Daniel Nicholson); we show that, if aging is understood from this philosophical approach, there is an internal confusion in the research program between what is repair and what is rejuvenation. Finally, we state that this theoretical viewpoint connects with scientific criticism and reinforces the idea that there are limits to the aspirations to live indefinitely young. (shrink)

Riassunto: Nel presente articolo si analizzerà come, a fronte del paradigma transumanista che opera per una progressiva inclusione dell’uomo nel suo stesso processo di ‘miglioramento’ del mondo, è sempre più pressante la domanda sulla dinamica tra verità (truth) e fiducia (trust) nella relazione uomo-macchina e nella riflessione etica degli enti che orientano gli odierni processi di innovazione bio-tecnologica. Le visioni che emergono – frutto rispettivamente del paradigma transumanista e di un’ecologia umana che restituisce all’uomo il senso della sua storia e (...) del suo vivere in relazione – non sono banalmente antitetiche: poggiano su due visioni dell’uomo scientifico profondamente diverse. Si propone quindi che un modo costruttivo per coniugare verità e fiducia nella nostra società contemporanea, dipenderà sempre più dall’idea di uomo che con la scienza vogliamo servire, coniugando il progresso bio-tecnologico con l’incompletezza biologica che caratterizza l’uomo. Parole chiave: Transumanesimo; Estraneità; Dignità; Verità; Fiducia. Resumen: Analizaremos cómo, ante el paradigma transhumanista, que intenta incluir al ser humano en el proceso de “mejora” del mundo, es cada vez más apremiante la pregunta por la relación entre la verdad (truth) y la confianza (trust). Nos referimos a la relación entre una narrativa verdadera sobre la tecnología y la confianza que en dicha tecnología podemos depositar. Esta pregunta se plantea en el ámbito de la relación hombre-máquina, así como en la reflexión ética sobre la innovación biotecnológica. Emergen aquí dos visiones, que no son simplemente antitéticas: la propia del paradigma transhumanista y la propia de una ecología humana que nos devuelve el sentido histórico y social de nuestra vida. Estas dos visiones se corresponden también con dos concepciones profundamente diferentes del ser humano. Entendemos, pues, que una forma constructiva de combinar la verdad y la confianza en nuestra sociedad contemporánea dependerá cada vez más de la idea de ser humano a la que la ciencia pretenda servir. Para servir realmente al ser humano, la ciencia habrá de combinar el progreso biotecnológico con la incompletitud biológica que nos caracteriza. Organoids and organs-on-a-chip are currently the two major families of 3D advanced organotypic in vitro culture systems, aimed at reconstituting miniaturized models of physiological and pathological states of human organs. Both share the tenets of the so-called “three-dimensional thinking”, a Systems Physiology approach focused on recapitulating the dynamic interactions between cells and their microenvironment. We first review the arguments underlying the “paradigm shift” toward three-dimensional thinking in the in vitro culture community. Then, through a historically informed account of the technical (...) affordances and the epistemic commitments of these two approaches, we highlight how they embody two distinct experimental cultures. We finally argue that the current systematic effort for their integration requires not only innovative “synergistic” engineering solutions, but also conceptual integration between different perspectives on biological causality. Cancers rely on multiple, heterogeneous processes at different scales, pertaining to many biomedical fields. Therefore, understanding cancer is necessarily an interdisciplinary task that requires placing specialised experimental and clinical research into a broader conceptual, theoretical, and methodological framework. Without such a framework, oncology will collect piecemeal results, with scant dialogue between the different scientific communities studying cancer. We argue that one important way forward in service of a more successful dialogue is through greater integration of applied sciences (experimental and clinical) (...) with conceptual and theoretical approaches, informed by philosophical methods. By way of illustration, we explore six central themes: (i) the role of mutations in cancer; (ii) the clonal evolution of cancer cells; (iii) the relationship between cancer and multicellularity; (iv) the tumour microenvironment; (v) the immune system; and (vi) stem cells. In each case, we examine open questions in the scientific literature through a philosophical methodology and show the benefit of such a synergy for the scientific and medical understanding of cancer. The ageing of the population is one of the most significant social transformations that the twenty first century is showcasing and a challenge that impacts society at large. The elderly, inasmuch as everybody else, are confronted with continuous transformations that are induced by technology, although they seldom benefit from the opportunities that technology entails. The digital divide amongst various segments of the population is often age-related and due to different reasons, including biological, psychological, social and financial ones. There is an (...) ongoing reflection pertaining to the factors that hinders the full adoption of ICTs by the elderly and a question regarding what can be done to overcome their poor involvement in technology. This article, based on the results of a recent research, which has been conducted in Italy, aims at highlighting the importance of engaging the elderly in the use of technology as a key to building bridges between generations. In our everyday interaction with the environment, we often perceive objects and spaces as opportunities to feel, maintain, enhance, and change our affective states and processes. The concept of affective affordance was coined to accommodate this aspect of ordinary perception and the many ways in which we rely on the material environment to regulate our emo- tions. One natural way to think of affective affordances in emotion regulation is to interpret them as tools for regulating felt affective states. We argue (...) that this way of conceiving of affective affordances is too restrictive. By analyzing the role of expressive properties of objects and spaces in emotion regulation, we show that our environment does not need to elicit felt affective states to regulate our emotions. Thus, we revise the concept of affective affordance to accommodate these cases. This revised concept explains more with less. Hence, it should be preferred in wide-reaching philosophical projects that aim to explain human affectivity as a situated phenomenon. (shrink)

The relationship between philosophy and science has always been complementary. Today, while science moves increasingly fast and philosophy shows some problems in catching up with it, it is not always possible to ignore such relationships, especially in some disciplines such as philosophy of mind, cognitive science, and neuroscience. However, the methodological procedures used to analyze these data are based on principles and assumptions that require a profound dialogue between philosophy and science. Following these ideas, this work aims to raise the (...) problems that a classical connectionist theory can cause and problematize them in a cognitive framework, considering both philosophy and cognitive sciences but also the disciplines that are near to them, such as AI, computer sciences, and linguistics. For this reason, we embarked on an analysis of both the computational and theoretical problems that connectionism currently has. The second aim of this work is to advocate for collaboration between neuroscience and philosophy of mind because the promotion of deeper multidisciplinarity seems necessary in order to solve connectionism’s problems. In fact, we believe that the problems that we detected can be solved by a thorough investigation at both a theoretical and an empirical level, and they do not represent an impasse but rather a starting point from which connectionism should learn and be updated while keeping its original and profoundly convincing core. Section “The Concept of Enactive Memory” introduces the concept of enactive memory, clarifies its central theoretical com- mitments and goals, and describes how enactivists have explained episodic remember- ing. Section “The Philosophical Context of Enactive Memory” provides a brief contextu- alization of the enactive approach to memory with respect to approaches and theories with which, for one reason or another, it shares some theoretical insights, like the embodied, the extended, and the distributed approach, and the simulation theory. Section “Forgetting as a Challenge for Enactive Memory” explains why forgetting poses an important explanatory challenge to the enactive approach to memory and describes one philosophical proposal that has been developed to address this challenge. Section “Summary” provides a summary of the chapter, mentions some ongoing projects on enactive memory, and identifies some topics that advocates of the enactive approach to memory may want to investigate in the future. This book presents a novel interpretation of Aristotle's account of how shame instils virtue, and defends its philosophical import. Shame is shown to provide motivational continuity between the actions of the learners and the virtuous dispositions that they will eventually acquire.

This book supports a version of the trope-bundle view of individual substances matching also with a coherent account of change, individuation and individual essences. In particular, it is argued that qualitative individuation and qualitative individual essences can be tackled within the frames of a trope account. The adoption of a trope BT together with the individuation of tropes via the bearer substance might create the feeling of circularity since tropes and substances seem mutually to individuate each other. The novel solution (...) to the problem developed here consists in showing that the individuation of concrete individual substances is independent, in crucial respects, from the fact that they are construed as bundles of tropes. Apart from metaphysician colleagues, the book is recommended for advanced students in analytic metaphysics. (shrink)

¿Qué tenemos en común y qué nos separa de los animales? ¿Somos un animal más? Las humanidades clásicas, basadas en el excepcionalismo humano, han dado paso a las posthumanidades, que incluyen los estudios animales, así como otras perspectivas que desafían la noción tradicional del sujeto humano, como los estudios de género, poscoloniales y decoloniales, la ecocrítica o la teoría queer. Pensarnos en nuestra relación con los animales tiene consecuencias filosóficas en ámbitos como la ciencia, los afectos, el trabajo y el (...) desarrollo de políticas concretas de salud y de alimentación; todas ellas, zonas de contacto entre humanos y animales. Sobre estos encuentros, aunque nunca reparemos en ello, edificamos nuestro mundo. Este ensayo imprescindible aglutina parte de los conocimientos desarrollados hasta ahora por quienes se han acercado a la relación humano-animal, y los vincula con la historia, los movimientos políticos y sociales, las nuevas concepciones del género, de la ciencia y del conocimiento. Los argumentos, contradicciones, contextos e incluso tabúes que acoge este debate interdisciplinar ayudan a imaginar y a matizar unos presupuestos éticos posibles sobre los que replantear nuestra relación con los animales y, en último término, con nosotros mismos.
